4Clarifying sub-genomic positions of QTLs for flowering habit and fruit quality in U.S. strawberry (Fragaria×ananassa) breeding populations using pedigree-based QTL analysis显示文摘The cultivated strawberry(Fragaria×ananassa)is consumed worldwide for its flavor and nutritional benefits.Genetic analysis of commercially important traits in strawberry are important for the development of breeding methods and tools for this species.Although several quantitative trait loci(QTL)have been previously detected for fruit quality and flowering traits using low-density genetic maps,clarity on the sub-genomic locations of these QTLs was missing.Recent discoveries in allo-octoploid strawberry genomics led to the development of the IStraw90 single-nucleotide polymorphism(SNP)array,enabling high-density genetic maps and finer resolution QTL analysis.In this study,breeder-specified traits were evaluated in the Eastern(Michigan)and Western(Oregon)United States for a common set of breeding populations during 2 years.Several QTLs were validated for soluble solids content(SSC),fruit weight(FWT),pH and titratable acidity(TA)using a pedigree-based QTL analysis approach.For fruit quality,a QTL for SSC on linkage group(LG)6A,a QTL for FWT on LG 2BII,a QTL for pH on LG 4CII and two QTLs for TA on LGs 2A and 5B were detected.In addition,a large-effect QTL for flowering was detected at the distal end of LG 4A,coinciding with the FaPFRU locus.Marker haplotype analysis in the FaPFRU region indicated that the homozygous recessive genotype was highly predictive of seasonal flowering.SNP probes in the FaPFRU region may help facilitate marker-assisted selection for this trait.Sujeet Verma Jason D Zurn Natalia Salinas Megan M Mathey Beatrice Denoyes James F Hancock Chad E Finn Nahla V Bassil Vance M Whitaker 2017Horticulture Research2017,4,1:10
5Development of database of real-world diesel vehicle emission factors for China显示文摘A database of real-world diesel vehicle emission factors, based on type and technology, has been developed following tests on more than 300 diesel vehicles in China using a portable emission measurement system. The database provides better understanding of diesel vehicle emissions under actual driving conditions. We found that although new regulations have reduced real-world emission levels of diesel trucks and buses significantly for most pollutants in China, NOx emissions have been inadequately controlled by the current standards, especially for diesel buses, because of bad driving conditions in the real world. We also compared the emission factors in the database with those calculated by emission factor models and used in inventory studies. The emission factors derived from COPERT(Computer Programmer to calculate Emissions from Road Transport) and MOBILE may both underestimate real emission factors, whereas the updated COPERT and PART5(Highway Vehicle Particulate Emission Modeling Software) models may overestimate emission factors in China. Real-world measurement results and emission factors used in recent emission inventory studies are inconsistent,which has led to inaccurate estimates of emissions from diesel trucks and buses over recent years. This suggests that emission factors derived from European or US-based models will not truly represent real-world emissions in China. 6An experimental validation of genomic selection in octoploid strawberry显示文摘The primary goal of genomic selection is to increase genetic gains for complex traits by predicting performance of individuals for which phenotypic data are not available.The objective of this study was to experimentally evaluate the potential of genomic selection in strawberry breeding and to define a strategy for its implementation.Four clonally replicated field trials,two in each of 2 years comprised of a total of 1628 individuals,were established in 2013–2014 and 2014–2015.Five complex yield and fruit quality traits with moderate to low heritability were assessed in each trial.High-density genotyping was performed with the Affymetrix Axiom IStraw90 single-nucleotide polymorphism array,and 17479 polymorphic markers were chosen for analysis.Several methods were compared,including Genomic BLUP,Bayes B,Bayes C,Bayesian LASSO Regression,Bayesian Ridge Regression and Reproducing Kernel Hilbert Spaces.Cross-validation within training populations resulted in higher values than for true validations across trials.For true validations,Bayes B gave the highest predictive abilities on average and also the highest selection efficiencies,particularly for yield traits that were the lowest heritability traits.Selection efficiencies using Bayes B for parent selection ranged from 74%for average fruit weight to 34%for early marketable yield.A breeding strategy is proposed in which advanced selection trials are utilized as training populations and in which genomic selection can reduce the breeding cycle from 3 to 2 years for a subset of untested parents based on their predicted genomic breeding values.Salvador A Gezan Luis F Osorio Sujeet Verma Vance M Whitaker 2017Horticulture Research2017,4,1:6

8Frequency-specific abnormalities in regional homogeneity among children with attention deficit hyperactivity disorder: a resting-state f MRI study显示文摘Although many functional magnetic resonance imaging(f MRI) studies have investigated the neurophysiology of attention deficit hyperactivity disorder(ADHD),the existing studies have not yielded consistent findings.This may be related to the different properties of different frequency bands. To investigate the frequency-specific regional homogeneity(Re Ho) of spontaneous neural activities in ADHD, the current study used resting-state f MRI to explore the Re Ho properties of five frequency bands, slow-5(0.01–0.027 Hz), slow-4(0.027–0.073 Hz),slow-3(0.073–0.198 Hz), slow-2(0.198–0.25 Hz) and the extra-low frequency(0–0.01 Hz), in 30 drug-naive boys with ADHD and 30 healthy controls. Compared with controls, the ADHD group showed decreased Re Ho in the default mode network(DMN) including the medial prefrontal cortex and precuneus, middle frontal gyrus and angular gyrus. ADHD patients also showed increased Re Ho in the posterior cerebellum. Significant interactions between frequency band and group were observed predominantly in the dorsolateral prefrontal and parietal cortices, orbital frontal cortex, supplementary motor area,inferior occipital gyrus, thalamus and anterior cerebellum.In particular, we found that the between-group difference in the extra-low frequency band(0–0.01 Hz) seemed to be greater than that in the other frequency bands for most brain regions. 13Atypical clinical and pathological findings in a patient with isolated cortical vein thrombosis显示文摘Isolated cortical vein thrombosis often produces a focal lesion. Because of the rapid development of collateral circulation, increased intracranial pressure has never been reported in a patient with isolated cortical vein thrombosis. The diagnosis of isolated cortical vein thrombosis is based mainly on MRI, catheter digital subtraction angiography, and histological findings, but may be challenging. We report a patient who presented with intermittent seizures and left-sided limb weakness. Her symptoms gradually progressed, and she eventually developed signs of increased intracranial pressure. Imaging studies showed a space-occupying lesion in the right frontal lobe of the brain. As we could not diagnose isolated cortical vein thrombosis based on the preoperative findings, surgical excision of the lesion was performed under general anesthesia. Histological examination showed destruction of the brain parenchyma with infiltration of macrophages, proliferation of reactive astrocytes and small vessels, and foci of hemorrhage. Further examination found that a number of small vessels in both the subarachnoid space and brain parenchyma were filled with thrombus, some of which was organized. Elastic fiber staining showed that the obstructed vessels were veins. 14Sodium glucose co-transporter 2 inhibition reduces succinate levels in diabetic mice显示文摘BACKGROUND Type 1 diabetes(T1D) is associated with major chronic microvascular complications which contribute significantly to diabetes associated morbidity.The protein primarily responsible for glucose reabsorption in the kidney is sodium glucose co-transporter 2(SGLT2). Presently, SGLT2 inhibitors are widely used in diabetic patients to improve blood glucose levels and prevent cardiovascular and renal complications. Given the broad therapeutic application of SGLT2 inhibitors, we hypothesised that SGLT2 inhibition may exert its protective effects via alterations of the gut microbiome and tested this in a type 1 diabetic mouse model of diabetic retinopathy.AIM To determine whether the treatment with two independent SGLT2 inhibitors affects gut health in a type 1 diabetic mouse model.METHODS The SGLT2 inhibitors empagliflozin or dapagliflozin(25 mg/kg/d) or vehicle dimethylsulfoxide(DMSO) were administered to C57 BL/6 J, Akita, Kimba and Akimba mice at 10 wk of age for 8 wk via their drinking water. Serum samples were collected and the concentration of succinate and the short chain fatty acid(SCFA) butyric acid was measured using gas chromatography-mass spectrometry. Enzyme-linked immunosorbent assay(ELISA) was performed to determine the concentration of insulin and leptin. Furthermore, the norepinephrine content in kidney tissue was determined using ELISA. Pancreatic tissue was collected and stained with haematoxylin and eosin and analysed using brightfield microscopy.RESULTS Due to the presence of the Akita allele, both Akita and Akimba mice showed a reduction in insulin production compared to C57 BL/6 J and Kimba mice.Furthermore, Akita mice also showed the presence of apoptotic bodies within the pancreatic islets. The acinar cells of Akita and Akimba mice showed swelling which is indicative of acute injury or pancreatitis. After 8 wk of SGLT2 inhibition with dapagliflozin, the intermediate metabolite of gut metabolism known as succinate was significantly reduced in Akimba mice when compared to DMSO treated mice. In addition, empagliflozin resulted in suppression of succinate levels in Akimba mice. The beneficial SCFA known as butyric acid was significantly increased in Akita mice after treatment with dapagliflozin when compared to vehicle treated mice. The norepinephrine content in the kidney was significantly reduced with both dapagliflozin and empagliflozin therapy in Akita mice and was significantly reduced in Akimba mice treated with empagliflozin.In non-diabetic C57 BL/6 J and Kimba mice, serum leptin levels were significantly reduced after dapagliflozin therapy.CONCLUSION The inhibition of SGLT2 reduces the intermediate metabolite succinate, increases SCFA butyric acid levels and reduces norepinephrine content in mouse models of T1 D. 17Strawberry sweetness and consumer preference are enhanced by specific volatile compounds显示文摘Breeding crops for improved flavor is challenging due to the high cost of sensory evaluation and the difficulty of connecting sensory experience to chemical composition.The main goal of this study was to identify the chemical drivers of sweetness and consumer liking for fresh strawberries(Fragaria×ananassa).Fruit of 148 strawberry samples from cultivars and breeding selections were grown and harvested over seven years and were subjected to both sensory and chemical analyses.Each panel consisted of at least 100 consumers,resulting in more than 15,000 sensory data points per descriptor.Three sugars,two acids and 113 volatile compounds were quantified.Consumer liking was highly associated with sweetness intensity,texture liking,and flavor intensity,but not sourness intensity.Partial least square analyses revealed 20 volatile compounds that increased sweetness perception independently of sugars;18 volatiles that increased liking independently of sugars;and 15 volatile compounds that had positive effects on both.Machine learning-based predictive models including sugars,acids,and volatiles explained at least 25%more variation in sweetness and liking than models accounting for sugars and acids only.Volatile compounds such asγ-dodecalactone;5-hepten-2-one,6-methyl;and multiple medium-chain fatty acid esters may serve as targets for breeding or quality control attributes for strawberry products.A genetic association study identified two loci controlling ester production,both on linkage group 6 A.Co-segregating makers in these regions can be used for increasing multiple esters simultaneously.This study demonstrates a paradigm for improvement of fruit sweetness and flavor in which consumers drive the identification of the most important chemical targets,which in turn drives the discovery of genetic targets for marker-assisted breeding.Zhen Fan Tomas Hasing Timothy S.Johnson Drake M.Garner Michael L.Schwieterman Christopher R.Barbey Thomas A.Colquhoun Charles A.Sims Marcio F.R.Resende Vance M.Whitaker 2021Horticulture Research2021,8,1:2
