1二氢嘧啶脱氢酶、胸苷酸合成酶、c-myc和p53基因在小鼠肝组织中表达的昼夜节律性研究(英文)显示文摘背景与目的:5-Fluorouracil(5-FU)主要通过抑制胸苷酸合成酶发挥抗癌作用,而二氢嘧啶脱氢酶是5-FU体内代谢的一个起始酶和限速酶。在本研究中,我们对二氢嘧啶脱氢酶、胸苷酸合成酶、c-myc和p53基因在小鼠肝脏中表达的昼夜节律进行研究。方法:本实验共采用了24只雄性B6D2F1小鼠。所有实验动物在实验中进行了4周光照同步化(12h光照:12h黑暗)。体温和休息-活动节律通过腹腔埋置的微型感受器进行动态监测。动物分别在3,7,11,15,19,23HALO(灯亮后小时数)被处死并取出肝脏冻存。肝脏标本中抽提的总RNA用LightCycler-SYBRGreenITM扩增系统进行实时定量RT-PCR。结果:小鼠的体温和休息-活动均呈现明显和相似的昼夜节律,其峰值均位于黑暗期(相当于小鼠活动相),谷值则位于光照期(相当于小鼠休息相)。二氢嘧啶脱氢酶在mRNA水平呈现出明显的昼夜节律性,其峰值时间位于16HALO左右即位于小鼠活动相(P=0.0012)。胸苷酸合成酶在mRNA水平具有昼夜节律表达的趋势,其峰值时间出现在光照期(P=0.079)。C-myc和p53在mRNA水平的节律性表达在本实验中无统计学意义。结论:二氢嘧啶脱氢酶和胸苷酸合成酶在mRNA水平呈现出昼夜节律性,二者峰值时间相差约12h,这一结果提示可以利用二者在转录水平的时间差异用5-FU进行时间化疗。吴明玮 冼励坚 李晓梅 lnnominato Pasquale lvi Francis 2004癌症2004,23,3:22
2移植于裸鼠的人鼻咽癌细胞DNA合成生物节律的初步研究显示文摘目的:探索鼻咽癌细胞DNA合成的生物节律为临床制订鼻咽癌时辰化疗方案提供必要的参数。方法:BALB/C裸小鼠15只,置于程控的独立光照系统中(12h光照,12h黑暗)中同步化至少3周。然后双侧腋下接种人鼻咽低分化鳞癌细胞裸小鼠移植瘤CNE2,成瘤后按灯亮后3、9、15、21h(即3、9、15、21HALO)的时间点取瘤块,制成单个细胞悬液固定,染色后流式细胞仪测DNA含量。用SPSS软件系统ANOVA法检验各期细胞在4个时间点差异的显著性,用Cosinor法考察G1、S、G2/M期细胞在24h的分布是否符合余弦函数。结果:G1、S、G2/M期细胞在3、9、15、21HALO的分布随时间变化有显著性差异,G1、G2/M期细胞变化符合余弦节律。结论:移植于裸鼠的人鼻咽癌细胞的DNA合成随昼夜交替呈节律性变化。孙健 冼励坚 曹弃元 叶燕丽 LIU Xu-hui LI Xiao-mei Francis Lé vi 2001癌症2001,20,2:21

4Evaluating Conservation Effectiveness of Nature Reserves Established for Surrogate Species:Case of a Giant Panda Nature Reserve in Qinling Mountains,China显示文摘Many nature reserves are established to protect the habitat needs of particular endangered species of interest but their effectiveness for protecting other species is questionable.In this study,this effectiveness was evaluated in a nature reserve network located in the Qinling Mountains,Shaanxi Province,China.The network of reserves was established mainly for the conservation of the giant panda,a species considered as a surrogate for the conservation of many other endangered species in the region.The habitat suitability of nine protected species,including the giant panda,was modeled by using Maximum Entropy(MAXENT)and their spatial congruence was analyzed.Habitat suitability of these species was also overlapped with nature reserve boundaries and their management zones(i.e.,core,buffer and experimental zones).Results show that in general the habitat of the giant panda constitutes a reasonable surrogate of the habitat of other protected species,and giant panda reserves protect a relatively high proportion of the habitat of other protected species.Therefore,giant panda habitat conservation also allows the conservation of the habitat of other protected species in the region.However,a large area of suitable habitat was excluded from the nature reserve network.In addition,four species exhibited a low proportion of highly suitable habitat inside the core zones of nature reserves.It suggests that a high proportion of suitable habitat of protected species not targeted for conservation is located in the experimental and buffer zones,thus,is being affected by human activities.To increase their conservation effectiveness,nature reserves and their management zones need to be re-examined in order to include suitable habitat of more endangered species.The procedures described in this study can be easily implemented for the conservation of many endangered species not only in China but in many other parts of the world.XU Weihua Andrés VIA QI Zengxiang OUYANG Zhiyun LIU Jianguo LIU Wei WAN Hui 2014Chinese Geographical Science2014,24,1:12
5Serum leptin levels and insulin resistance are associated with gallstone disease in overweight subjects显示文摘AIM: To establish an association between the serum leptin levels and the development of gallstone disease (GD).METHODS: We carried out a non-matched case-controlled study in a university hospital in Mexico City. Two hundred and eighty-seven subjects were included: 97 cases with gallstones and 190 controls. Body mass index (BMI), fasting plasma leptin, insulin, serum lipid, and lipoprotein levels were measured. Insulin resistance was calculated by homeostasis model assessment (HOMA-IR). Unconditional logistic regression analysis (univariate and multivariate)stratified by BMI was used to calculate the risk of GD.RESULTS: The multivariate conditional regression analysis revealed a model for those patients with BMI <30. The selected variables in the model were HOMA-IR index with OR = 1.31, P= 0.02 and leptin higher than median with OR = 2.11, P= 0.05. 9Effective plant discrimination based on the combination of local binary pattern operators and multiclass support vector machine methods显示文摘Accurate crop and weed discrimination plays a critical role in addressing the challenges of weed management in agriculture.The use of herbicides is currently the most common approach to weed control.However,herbicide resistant plants have long been recognised as a major concern due to the excessive use of herbicides.Effective weed detection techniques can reduce the cost of weed management and improve crop quality and yield.A computationally efficient and robust plant classification algorithm is developed and applied to the classification of three crops:Brassica napus(canola),Zea mays(maize/corn),and radish.The developed algorithm is based on the combination of Local Binary Pattern(LBP)operators,for the extraction of crop leaf textural features and Support vector machine(SVM)method,for multiclass plant classification.This paper presents the first investigation of the accuracy of the combined LBP algorithms,trained using a large dataset of canola,radish and barley leaf images captured by a testing facility under simulated field conditions.The dataset has four subclasses,background,canola,corn,and radish,with 24,000 images used for training and 6000 images,for validation.The dataset is referred herein as 'bccr-segset'and published online.In each subclass,plant images are collected at four crop growth stages.Experimentally,the algorithm demonstrates plant classification accuracy as high as 91.85%,for the four classes.Vi Nguyen Thanh Le Beniamin Apopei Kamal Alameh 2019Information Processing in Agriculture2019,6,1:4
10Functional relationship between ammonia-oxidizing bacteria and ammonia-oxidizing archaea populations in the secondary treatment system of a full-scale municipal wastewater treatment plant显示文摘The abundance of ammonia-oxidizing bacteria and archaea and their amo A genes from the aerobic activated sludge tanks,recycled sludge and anaerobic digesters of a full-scale wastewater treatment plant(WWTP)was determined.Polymerase chain reaction and denaturing gradient gel electrophoresis were used to generate diversity profiles,which showed that each population had a consistent profile although the abundance of individual members varied.In the aerobic tanks,the ammonia-oxidizing bacterial(AOB)population was more than 350 times more abundant than the ammonia-oxidizing archaeal(AOA)population,however in the digesters,the AOA population was more than 10 times more abundant.Measuring the activity of the amo A gene expression of the two populations using RT-PCR also showed that the AOA amo A gene was more active in the digesters than in the activated sludge tanks.Using batch reactors and dd PCR,amo A activity could be measured and it was found that when the AOB amo A activity was inhibited in the anoxic reactors,the expression of the AOA amo A gene increased fourfold.This suggests that these two populations may have a cooperative relationship for the oxidation of ammonia.Golam MIslam Peter Vi Kimberley Ann Gilbride 2019Journal of Environmental Sciences2019,31,12:4

12Endoscopic ultrasound elastography strain histograms in the evaluation of patients with pancreatic masses显示文摘AIM: To investigate the accuracy of the strain histogram endoscopic ultrasound(EUS)-based method for the diagnostic differentiation of patients with pancreatic masses. METHODS: In a prospective single center study, 149 patients were analyzed, 105 with pancreatic masses and 44 controls. Elastography images were recorded using commercially available ultrasound equipment in combination with EUS linear probes. Strain histograms(SHs) were calculated by machine integrated software in regions of interest and mean values of the strain histograms were expressed as Mode 1(over the mass) and Mode 2(over an adjacent part of pancreatic tissue, representing the reference area). The ratio between Mode 2 and Mode 1 was calculated later, representing a new variable, the strain histogram ratio. After the final diagnosis was established, two groups of patients were formed: a pancreatic cancer group with positive cytology achieved by fine needle aspiration puncture or histology after surgery(58 patients), and a massforming pancreatitis group with negative cytology and follow-up after 3 and 6 mo(47 patients). All statistical analyses were conducted in SPSS 14.0(SPSS Inc., Chicago, IL, United States).RESULTS: Results were obtained with software for strain histograms with reversed hue scale(0 represents the hardest tissue structure and 255 the softest). Based on the receiver operating characteristics(ROC) curve coordinates, the cut-off point for Mode 1 was set at the value of 86. Values under the cut-off point indicated the presence of pancreatic malignancy. Mode 1 reached 100% sensitivity and 45% specificity with overall accuracy of 66%(95%CI: 61%-66%) in detection of pancreatic malignant tumors among the patients with pancreatic masses. The positive and negative predictive values were 54% and 100%, respectively. The cut-off for the new calculated variable, the SH ratio, was set at the value 1.153 based on the ROC curve coordinates. Values equal or above the cut-off value were indicative of pancreatic malignancy. The SH ratio reached 98% sensitivity, 50% specificity and an overall accuracy of 69%(95%CI: 63%-70%). The positive and negative predictive values were 92% and 100%, respectively.CONCLUSION: SH showed high sensitivity in pancreatic malignant tumor detection but disappointingly low specificity. 16Effect of low level laser therapy on dental pulp during orthodontic movement显示文摘AIM: To validate the protocol described here to be used in future clinical trials related to the effect of laser therapy on dental pulp. METHODS: Histologically treated samples from eight human healthy premolar teeth obtained from the middle root level were distributed in four groups: group 1(G1) absolute control; group 2(G2) only laser irradiation; group 3(G3) exposed only to orthodontics; and group 4(G4) treated with orthodontics and laser. Laser treatment was performed at 830 nm wavelength, 100 mW(energy 80 J/cm2, 2.2 J), for 22 s in the vestibular surface and 22 s in the palatal surface, 1 mm away from the dental root mucosa. Three staining methods were performed: hematoxylin-eosin(HE), Masson's Trichrome method and Gomori's method.RESULTS: The pulp histology parameters were evaluated and the results classified in to 3 parts: an inflammatory response, soft tissue response(dental pulp) and hard tissue response(dentin and predentin). There was no inflammation(chronic or acute) in any of the evaluated groups. The zones of pulp necrosis were found in one premolar of G3 and in one of G4; in groups G2 and G4 there was higher angiogenesis than in the other two groups. G4 group presented the highest level of vascularization. A reduced nerve density was observed in G3. A G2 specimen showed increased nerve density. A higher rate of calcification was observed in G1 compared to G2. Denticles, either real or false, were observed in G1, G2 and G3. Sclerosis of dentin and focal dentin loss was observed among all the groups. Secondary dentin was present in one sample in G1 and G2. A necrosis zone was found in one sample of G3 and G4. No differences between groups were observed in the odontoblast irregularity layer but the layer was wider in the group treated with laser only. A notable difference was detected in reduction of the cell-free layer between the groups G1 and G4. The findings in pulp tissue favor its adaptative response against dental movement induced by orthodontics. No definitive conclusions may be derived as this is a pilot study.
