2Spatial organization of bacterial flora in normal and inflamed intestine: A fluorescence in situ hybridization study in mice显示文摘AIM: To studythe role of intestinal flora in inflammatory bowel disease (IBD).METHODS: The spatial organization of intestinal flora was investigated in normal mice and in two models of murine colitis using fluorescence in situ hybridization.RESULTS: The murine small intestine was nearly bacteriafree. The normal colonic flora was organized in three distinct compartments (crypt, interlaced, and fecal), each with different bacterial compositions. Crypt bacteria were present in the cecum and proximal colon. The fecal compartment was composed of homogeneously mixed bacterial groups that directly contacted the colonic wall in the cecum but were separated from the proximal colonic wall by a dense interlaced layer. Beginning in the middle colon, a mucus gap of growing thickness physically separated all intestinal bacteria from contact with the epithelium. Colonic inflammation was accompanied with a depletion of bacteria within the fecal compartment, a reduced surface area in which feces had direct contact with the colonic wall, increased thickness and spread of the mucus gap, and massive increases of bacterial concentrations in the crypt and interlaced compartments. Adhesive and infiltrative bacteria were observed in inflamed colon only, with dominant Bacteroides species.CONCLUSION: The proximal and distal colons are functionally different organs with respect to the intestinal flora, representing a bioreactor and a Segregation device.The highly organized structure of the colonic flora, its specific arrangement in different colonic segments, and its specialized response to inflammatory stimuli indicate that the intestinal flora is an innate part of host immunity that is under complex control.Alexander Swidsinski Vera Loening-Baucke Herbert Lochs Laura P.Hale 2005World Journal of Gastroenterology2005,11,8:27

4Prognostic angiogenic markers(endoglin, VEGF, CD31) and tumor cell proliferation(Ki67) for gastrointestinal stromal tumors显示文摘AIM: To evaluate the correlation between the immunoexpression of angiogenic markers [CD31, CD105 and vascular endothelial growth factor(VEGF)], proliferative index(Ki67), and prognosis of patients with gastrointestinal stromal tumors(GIST).METHODS: This is a retrospective study of 54 GIST cases. Medical records were searched to obtain the GIST patients' demographic and clinical data, and paraffin-embedded blocks of tumor samples were retrieved from the hospital archives to conduct a new immunohistochemical evaluation. The tumor samples of GIST patients were subject to immunohistochemical evaluation for endoglin(CD105), CD31, VEGF, and Ki67 expression. The CD105 and CD31 intratumoral microvascular density(IMVD) was measured using automated analysis. We determined the correlation between the immunoexpression of CD105, CD31, VEGF,Ki67 and prognosis. In addition, we conducted a cutoff analysis using the receiver-operating characteristic curve. VEGF positivity was classified as either null/weak or strong. Ki67 was evaluated using a cutoff of 5%positive cells. The prognosis was classified as good(patient alive without recurrence) or poor(patient with recurrence/death).RESULTS: The distribution of tumor sites among the54 analyzed samples was as follows: 27(50%) in the stomach, 20(37.1%) in the small intestine, 6(11.1%)in the colon, and 1(1.8%) in the esophagus. The size of the tumors ranged from 2 to 33 cm(median: 8cm); in 12 cases(22.2%), the tumor was below 5 cm at the largest diameter, but in 42 cases(77.7%), the tumor was larger than 5 cm. The means of CD105 and CD31 were significantly higher in the group with poor prognosis(P < 0.001). The cut-off values of CD105(>1.2%) and CD31(> 2.5%) in the receiver-operating characteristic curve were related to a poorer prognosis.Cases with a better prognosis showed significantly null/weak staining for VEGF(P < 0.001). Ki-67 expression of≥ 5% was strongly correlated with a worse prognosis(P< 0.001). 8Use of butyrate or glutamine in enema solution reduces inflammation and fibrosis in experimental diversion colitis显示文摘AIM:To investigate whether butyrate or glutamine enemas could diminish inflammation in experimental diversion colitis.METHODS:Wistar specific pathogen-free rats were submitted to a Hartmann's end colostomy and treated with enemas containing glutamine,butyrate,or saline.Enemas were administered twice a week in the excluded segment of the colon from 4 to 12 wk after the surgical procedure.Follow-up colonoscopy was performed every 4 wk for 12 wk.The effect of treatment was evaluated using video-endoscopic and histologic scores and measuring interleukin-1β,tumor necrosis factor-alpha,and transforming growth factor beta production in organ cultures by enzyme linked immunosorbent assay.RESULTS:Colonoscopies of the diverted segment showed mucosa with hyperemia,increased number of vessels,bleeding and mucus discharge.Treatment with either glutamine or butyrate induced significant reductions in both colonoscopic(P < 0.02) and histological scores(P < 0.01) and restored the densities of collagen fibers in tissue(P = 0.015;P = 0.001),the number of goblet cells(P = 0.021;P = 0.029),and the rate of apoptosis within the epithelium(P = 0.043;P = 0.011) to normal values.The high levels of cytokines in colon explants from rats with diversion colitis significantly decreased to normal values after treatment with butyrate or glutamine.CONCLUSION:The improvement of experimental diversion colitis following glutamine or butyrate enemas highlights the importance of specific luminal nutrients in the homeostasis of the colonic mucosa and supports their utilization for the treatment of human diversion colitis.Rodrigo Goulart Pacheco Christiano Costa Esposito Lucas CM Müller Morgana TL Castelo-Branco Leonardo Pereira Quintella Vera Lucia A Chagas Heitor Siffert P de Souza Alberto Schanaider 2012World Journal of Gastroenterology2012,18,32:9

12Collagen/hyaluronan based hydrogels releasing sulfated hyaluronan improve dermal wound healing in diabetic mice via reducing inflammatory macrophage activity显示文摘Sustained inflammation associated with dysregulated macrophage activation prevents tissue formation and healing of chronic wounds.Control of inflammation and immune cell functions thus represents a promising approach in the development of advanced therapeutic strategies.Here we describe immunomodulatory hyaluronan/collagen(HA-AC/coll)-based hydrogels containing high-sulfated hyaluronan(sHA)as immunoregulatory component for the modulation of inflammatory macrophage activities in disturbed wound healing.Solute sHA downregulates inflammatory activities of bone marrow-derived and tissue-resident macrophages in vitro.This further affects macrophage-mediated pro-inflammatory activation of skin cells as shown in skin ex-vivo cultures.In a mouse model of acute skin inflammation,intradermal injection of sHA downregulates the inflammatory processes in the skin.This is associated with the promotion of an anti-inflammatory gene signature in skin macrophages indicating a shift of their activation profile.For in vivo translation,we designed HA-AC/coll hydrogels allowing delivery of sHA into wounds over a period of at least one week.Their immunoregulatory capacity was analyzed in a translational experimental approach in skin wounds of diabetic db/db mice,an established model for disturbed wound healing.The sHA-releasing hydrogels improved defective tissue repair with reduced inflammation,augmented pro-regenerative macrophage activation,increased vascularization,and accelerated new tissue formation and wound closure.Sophia Hauck Paula Zager Norbert Halfter Elke Wandel Marta Torregrossa Ainur Kakpenova Sandra Rother Michelle Ordieres Susann Räthel Albrecht Berg Stephanie Möller Matthias Schnabelrauch Jan C.Simon Vera Hintze Sandra Franz 2021Bioactive Materials2021,6,12:7
13Engineering the Smart Factory显示文摘The fourth industrial revolution promises to create what has been called the smart factory. The vision is that within such modular structured smart factories, cyber-physical systems monitor physical processes, create a virtual copy of the physical world and make decentralised decisions. This paper provides a view of this initiative from an automation systems perspective. In this context it considers how future automation systems might be effectively configured and supported through their lifecycles and how integration,application modelling, visualisation and reuse of such systems might be best achieved. 14Institutional experience of PTH evaluation on fine-needle washing after aspiration biopsy to locate hyperfunctioning parathyroid tissue显示文摘Assaying parathyroid hormone(PTH) in the washing liquid after fine-needle aspiration biopsy(FNAB) seems to be a valid approach to locate parathyroid tissue.PTH-FNAB was evaluated in 47 patients with a clinical picture of primary hyper-parathyroidism(PHP) and ultrasonography(US) suggestive of parathyroid lesion.The patients were subdivided into two groups on the basis of the absence or presence of US thyroid alterations.The result of PTH-FNAB was compared with those of cytology,scintigraphy and,in 24 patients,surgical outcome.PTH-FNAB samples with a value higher than that recorded in the serum and higher than our institutional cut-off were deemed to be probable samples of parathyroid tissue.Cytology proved diagnostic for benign thyroid lesions,non-diagnostic for thyroid lesions,hyperplastic parathyroid tissue,undetermined or malignant thyroid lesions and other lesions in 45%,30%,17%,4%,and 4% of cases,respectively.In 47% of cases,PTH-FNAB indicated that the sample had been taken in parathyroid tissue.In patients without US alterations,the diagnostic accuracy of PTH-FNAB was greater than that of scintigraphy.After surgery,comparison between the results of PTH-FNAB and scintigraphy,in terms of positive predictive value(PPV),revealed the superiority of PTH-FNAB;PPV was 94% for FNAB and 71% for scintigraphy,while sensitivity was 83% and 69%,respectively.PTH-FNAB evaluation after FNAB appears to be more diagnostic than cytology and scintigraphy.Of all the procedures used,PTH-FNAB appears to be the method of choice when the target is US suggestive and reachable.PTH-FNAB appears to be a useful method of guiding surgical intervention.Massimo GIUSTI Mara DOLCINO Lara VERA Carla GHIARA Francesca MASSARO Laura FAZZUOLI Diego FERONE Michele MUSSAP Francesco MINUTO 2009Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2009,10,5:6
15Quantitative genetic analysis of chlorophyll a fluorescence parameters in maize in the field environments显示文摘Chlorophyll fluorescence transient from initial to maximum fluorescence('P' step) throughout two intermediate steps('J' and 'I')(JIP‐test) is considered a reliable early quantitative indicator of stress in plants. The JIP‐test is particularly useful for crop plants when applied in variable field environments. The aim of the present study was to conduct a quantitative trait loci(QTL) analysis for nine JIP‐test parameters in maize during flowering in four field environments differing in weather conditions. QTL analysis and identification of putative candidate genes might help to explain the genetic relationship between photosynthesis and different field scenarios in maize plants. The JIP‐test parameters were analyzed in the intermated B73 Mo17(IBM) maize population of 205 recombinant inbred lines. A set of 2,178 molecular markers across the whole maize genome was used for QTL analysis revealing 10 significant QTLs for seven JIP‐test parameters, of which five were co‐localized when combinedover the four environments indicating polygenic inheritance and pleiotropy. Our results demonstrate that QTL analysis of chlorophyll fluorescence parameters was capable of detecting one pleiotropic locus on chromosome 7, coinciding with the gene gst23 that may be associated with efficient photosynthesis under different field scenarios.Domagoj ?imi? 17性别和年龄对中国人冠状动脉旁路移植术后住院病死率的影响显示文摘目的确定中国人冠状动脉旁路移植术后住院病死率是否存在性别差异以及该差异是否与年龄相关.方法回顾性分析1997年1月1日至2001年12月31日阜外心血管病医院外科全部接受冠状动脉旁路移植术的患者共2682例(男性2316例,女性366例).按照性别和年龄分组调查它们之间的围手术期并发症及手术后住院病死率的差异.结果女性术前合并主要疾病的比例比男性高,术后并发症多,但她们术前的左心室射血分数较男性好,且冠状动脉病变数量少.然而,女性患者的术后住院病死率是男性的3倍(女性3.01%,男性1.12%,P=0.001),病死率的性别差异在低龄组更有统计学意义(女性2.6%,男性0.5%,P=0.001,ORs:4.844,95%可信区间:1.549~15.142),在老年患者中则无统计学意义(女性3.7%,男性2.4%,P=0.383).结论中国女性冠状动脉旁路移植术的住院病死率高于男性,特别是低龄女性.低龄组女性是住院病死率的独立危险因素.死亡率的性别差异随年龄增加而减少.需要进一步的研究来阐明病死率的性别差异在低龄组更显著的原因.宫路佳 Elke Lehmkuhl 胡盛寿 Friederike Kendel Stein Kandle Yong Du 章晏 杜茗 吴清玉 许建屏 孙立忠 朱晓东 Vera Regitz-Zagrosek 2006中华心血管病杂志2006,34,5:5
18Monoclonal immunoscintigraphy for detection of metastasis and recurrence of colorectal cancer显示文摘AIM:To assess the clinical role of monoclonal immunoscintigraphy for the detection of metastasis and recurrence of colorectal cancer.METHODS:Monoclonal immunoscintigraphy was performed in patients operated on for colorectal adenocar-cinoma suspected of local recurrence and metastatic disease.The results were compared with conventional diagnostics.RESULTS:Immunoscintigraphic investigation was done in 53 patients.Tumor recurrence occurred in 38 patients,and was confirmed by other diagnostic modalities in 35.In 15 patients,immunoscintigraphic findings were negative,and confirmed in 14 with other diagnostic methods.Comparative analysis confirmed good correlation of immunoscintigraphic findings and the results of conventional diagnostics and the level of tumor marker carcinoembryonic antigen.Statistical analysis of parameters of radiopharmaceutical groups imacis,indimacis and oncoscint presented homogenous characteristics all of three radiopharmaceuticals.The analysis of immunoscintigraphic target focus was clearly improved using tomography.CONCLUSION:Immunoscintigraphy is highly specific and has a good predictive value in local recurrence of colorectal cancer.Vera Artiko Ana Koljevic Markovi Dragana obi-aranovi Milorad Petrovi Andrija Anti Mirjana Stojkovi Marinko Zuvela Djordjije aranovi Milica Stojkovi Neboja Radovanovi Danijel Galun Aleksandar Milovanovi Jovica Milovanovi Anica Bobi-Radovanovi Zoran Krivokapic Vladimir Obradovi 2011World Journal of Gastroenterology2011,17,19:4
19Cerium dioxide as a new reactive sorbent for fast degradation of parathion methyl and some other organophosphates显示文摘Cerium dioxide was used for the first time as reactive sorbent for the degradation of the organophosphate pesticides parathion methyl,chlorpyrifos,dichlofenthion,fenchlorphos,and prothiofos,as well as of some chemical warfare agents—nerve gases soman and O-ethyl S-[2-(diisopropylamino) ethyl] methylphosphonothioate(VX).CeO2 specimens were prepared by calcination of basic cerous carbonate obtained by precipitation from an aqueous solution.The CeO2 samples containing certain amounts(1 wt.%–5 wt.%) of the neighboring lanthanides(La,Pr,Nd) were prepared in a similar way from pure lanthanide salts.It was shown that ceria accelerated markedly the decomposition of parathion methyl causing the cleavage of the P-O-aryl bond in the pesticide molecule.A similar reaction mechanism was proposed for the degradation of other organophosphate pesticides and nerve agents.The degradation times(reaction half-times) were in an order of minutes in the presence of CeO2,compared to hours or days under common environmental conditions.The reaction in suitable organic solvents allowed conversions of about 90% for parathion methyl loading of 20 mg pesticide/g CeO2 within 2 h with a reactant half-life in the order of 0.1 min.The key parameter governing the degradation efficiency of CeO2 was the temperature during calcination.At optimum calcination temperature(about 773.15 K),the produced ceria retained a sufficiently high surface area,and attained an optimum degree of crystallinity(related to a number of crystal defects,and thus potential reactive sites).The presence of other lanthanides somewhat decreased the reaction rate,but this effect was not detrimental and permitted the possible use of chemically impure ceria as a reactive sorbent.A fast organophosphate degradation was demonstrated not only in non-polar solvents(such as heptane),but also in polar aprotic solvents(acetonitrile,acetone) that are miscible with water.This opens new possibilities for designing more versatile decontamination strategies.The cleavage of phosphate ester bonds is of a great importance not only for the degradation of dangerous chemicals(chemical weapons,pesticides),but also for interactions of ceria(especially the nano-sized one) in biologically relevant systems.Pavel Janos Pavel Kuran Martin Kormunda Vaclav Stengl Tomas Matys Grygar Marek Dosek Martin Stastny Jakub Ederer Vera Pilarova Lubos Vrtoch 2014Journal of Rare Earths2014,32,4:4
20Characteristics of the active-layer under the China-Russia Crude Oil pipeline显示文摘Active layer is a key component for permafrost environment studies as many subsurface biological,biogeochemical,ecological,and pedogenic activities prevail in this layer.This study focuses on active layer temperature monitoring in an area with sporadic permafrost at two adjacent sites along China-Russia Crude Oil Pipeline(CRCOP),NorthEast China.Site T1 is located in disturbed ground on the right-of-way(on-ROW)2 m away from the center of the oil pipeline.T2 is located in a natural and undisturbed site,around 16.6 m off-ROW.Our objective was to study seasonal variability of the active layer depth and thermal regime from October 2017 to September 2018.The monitoring sites consist of soil temperature probes arranged in a vertical array at different depths at both sites.The following parameters were computed:number of isothermal days(ID),freezing days(FD),thawing days(TD),freezing degree days(FDD),thawing degree days(TDD),number of freeze-thaw days(FTD).The mean air temperature in the monitoring period reached-3.2℃.The temperature profile indicates that the maximum active layer thickness observed during the study period was 10 m at T1 and 2 m at T2.The majority of the soil temperatures were above or close to 0℃,resulting in great values of TDD,especially in the first 4 m depth.TDD for T1 were predominant and ranged between 600-1160℃·days(0-4 m depth)reflecting the influence of oil temperature from the pipeline.In T2 borehole FDD were predominant for all the soil layer depths resulting in less permafrost degradation.This comparison emphasizes the significant influence of vegetation removal and the dispersed heat from the pipeline on the active layer thickness.SERBAN Mihaela LI Guo-yu SERBAN Raul-David WANG Fei FEDOROV Alexander VERA Samsonova CAO Ya-peng CHEN Peng-chao WANG Wei 2021Journal of Mountain Science2021,18,2:4
