3Laser speckle imaging and wavelet analysis of cerebral blood flow associated with the opening of the blood–brain barrier by sound显示文摘The cerebral blood flow(CBF) alterations related to sound-induced opening of the blood–brain barrier(BBB) in adult mice are studied using laser speckle contrast imaging(LSCI) and wavelet analysis of vascular physiology.The results clearly show that the opening of the BBB is accompanied by the changes of venous but not microvessel circulation in the brain. The elevation of the BBB permeability is associated with the decrease of venous CBF and the increase of its complexity. 4Meso-substituted cationic 3-and 4-N-Pyridylporphyrins and their Zn(II)derivatives for antibacterial photodynamic therapy显示文摘Photodynamic inactivation of microorganisms known as antibacterial photodynamic therapy(APDT)is one of the most promising and innovative approaches for the destruction of pathogenic microorganisms.Among the photosensitizers(PSs),compounds based on cationic porphyrins/metalloporphyrins are most successfully used to inactivate microorganisms.Series of meso-substituted cationic pyridylporphyrins and metalloporphyrins with various peripheral groups in the third and fourth positions of the pyrrole ring have been synthesized in Armenia.The aim of this work was to determine and test the most e®ective cationic porphyrins and metalloporphyrins with high photoactivity against Gram negative and Gram positive microorganisms.It was shown that the synthesized cationic pyridylporphyrins/metalloporphyrins exhibit a high degree of phototoxicity towards both types of bacteria,including the methicillinresistant S.aureus strain.Zinc complexes of porphyrins are more phototoxic than metal-free porphyrin analogs.The e®ectiveness of these Zn–metalloporphyrins on bacteria is consistent with the level of singlet oxygen generation.It was found that the high antibacterial activity of the studied cationic porphyrins/metalloporphyrins depends on four factors:the presence in the porphyrin macrocycle of a positive charge(+4),a central metal atom(Zn2þÞand hydrophobic peripheral functional groups as well as high values of quantum yields of singlet oxygen.The results indicate that meso-substituted cationic pyridylporphyrins/metalloporphyrins cannd wider application in photoinactivation of bacteria than anionic or neutral PSs usually used in APDT.Aram G.Gyulkhandanyan Marina H.Paronyan Anna G.Gyulkhandanyan Karapet R.Ghazaryan Marina V.Parkhats Boris M.Dzhagarov Maria V.Korchenova Ekaterina N.Lazareva Elena S.Tuchina Grigor V.Gyulkhandanyan Valery V.Tuchin 2022Journal of Innovative Optical Health Sciences2022,15,1:1

7COMPARATIVE TREATMENT OF ACNE VULGARIS USING PALOMAR LUX APPLIQU´E TECHNIQUE AND DIRECT INTRALESIONAL INJECTION显示文摘Intralesional injection of triamcinolone(TMC)preparations is an effective therapy for cystic acne lesions.However,invasive delivery techniques limit the use of this modality to a relatively narrow class of cases.Skin permeability can be enhanced through creating a lattice of microzones(islets)of light-induced limited thermal damage in the upper layers of epidermis.In this paper,we directly compared safety and efficacy of delivering TMC acetonide with this novel technique versus conventional intralesional injection for treatment of inflammatory acne lesions.A combination of an intense pulsed light system and a specially designed appliqu´e with a pattern of absorbing centers has been used to create the lattice of islets of damage(LID).Quantitative analysis has included estimation of the following parameters:redness,diameter,and height of acne lesions.Clinical photography has been used to document dynamics of lesion development at successive visits(two hours,24 hours and one week post-treatment).Seven subjects have participated in the study.No difference in lesion dynamics between the treatment and control groups was observed at two-hours follow-up.At 24-hours/one-week follow-ups,TMC-injected and TMC-LID-delivered groups have demonstrated 82%/93%and 80%/89%improvement in height of lesions in comparison to control(60%/68%).Delivery of TMC with the newly proposed LID technique is at least as effective as intralesional injection for treating inflammatory acne lesions.Enhancement of skin permeability using LID approach is a promising technique for accelerating delivery of various compounds to their target areas in the skin.ELINA A.GENINA ALEXEY N.BASHKATOV VALERY V.TUCHIN GEORGY V.SIMONENKO VERA N.SHERSTNEVA ILYA V.YAROSLAVSKY GREGORY B.ALTSHULER 2009Journal of Innovative Optical Health Sciences2009,2,3:0

9Photodynamic therapy of brain tumors and novel optical coherence tomography strategies for in vivo monitoring of cerebral°uid dynamics显示文摘Photodynamic therapy(PDT)is a promising tool for least-invasive alternative methods for the treatment of brain tumors.The newly discovered PDT-induced opening of the blood–brain barrier(BBB)permeability open novel strategies for drug-brain delivery during post-surgical treatment of glioblastoma GBM.Here we discuss mechanisms of PDT-mediated opening of the BBB and age differences in PDT-related increase in BBB permeability,including with formation of brain edema.The meningeal lymphatic system plays a crucial role in the mechanism of brain drainage and clearance from metabolites and toxic molecules.We discuss that noninvasive photonic stimulation of°uid clearance via meningeal lymphatic vessels,and application of optical coherence tomography(OCT)for bed-side monitoring of meningeal lymphatic drainage has the promising perspective to be widely applied in both experimental and clinical studies of PDT and improving guidelines of PDT of brain tumors.A.Abdurashitov V.Tuchin O.Semyachkina-Glushkovskaya 2020Journal of Innovative Optical Health Sciences2020,,2:0
10ASSESSMENT OF TISSUE OPTICAL CLEARING AS A FUNCTION OF GLUCOSE CONCENTRATION USING OPTICAL COHERENCE TOMOGRAPHY显示文摘One of the major challenges in imaging biological tissues using optical techniques,such as optical coherence tomography(OCT),is the lack of light penetration due to highly turbid structures within the tissue.Optical clearing techniques enable the biological samples to be more optically homogeneous,allowing for deeper penetration of light into the tissue.This study investigates the effect of optical clearing utilizing various concentrations of glucose solution(10%,30%,and 50%)on porcine skin.A gold-plated mirror was imaged beneath the tissue and percentage clearing was determined by monitoring the change in reflected light intensity from the mirror over time.The ratio of percentage clearing per tissue thickness for 10%,30% and 50% glucose was determined to be 4.7±1.6%mm^(−1)(n=6),10.6±2.0%mm^(−1)(n=7)and 21.8±2.2%mm^(−1)(n=5),respectively.It was concluded that while higher glucose concentration has the highest optical clearing effect,a suitable concentration should be chosen for the purpose of clearing,considering the osmotic stress on the tissue sample.NARENDRAN SUDHEENDRAN MOHAMED MOHAMED MOHAMAD G.GHOSN VALERY V.TUCHIN KIRILL V.LARIN 2010Journal of Innovative Optical Health Sciences2010,3,3:0

13Integrated binary hologram to monitor cargo release from a drug-eluting film显示文摘Nowadays,holography translates from a pure technical tool for recording the phase and amplitude of the light wave to a widely applicable research-based method.Holographic devices are used for security enhancement,entertainment,3D display technologies and augmented reality.Binary computer-generated holograms(amplitude or phase-based)are of specific interest.They are easy to compute and their manufacturing methods are fast and robust.In this work,a method of manufacturing amplitude-only binary holograms out of biopolymers films is proposed.Opaque cargo bits made out of different bioactive substances(antibiotics,dyes,etc.)absorb or scatter specific parts of the incoming light wave.Cargo release was conducted by submerging the produced holograms into the aqueous solution at specific pH and temperature levels.Drug elution from the film was traced out by the numerical analysis of the far-field diffraction pattern,which otherwise could only be measured post-factum using sophisticated spectroscopic or chromatography devices.This approach broadens the application of general holography over to the field of biomedical research relevant to quantitative monitoring of the drug elution.Arkady S.Abdurashitov Pavel IProshin Valery V.Tuchin Gleb B.Sukhorukov 2022Light(Advanced Manufacturing)2022,3,3:0

19PHOTOTHERAPY OF GINGIVITIS:PILOT CLINICAL STUDY显示文摘The goal of this work was to evaluate the safety and efficacy of the Red Light Emitted Toothbrush(R-LETB)emitting at wavelength of 663 nm with power density of 3.3mW/cm2 in combination with 0.1%-methylene blue(MB)solution for the reduction of plaque and treatment of gingivitis.A microbiological in vitro study and a pilot clinical study were conducted.The microbiological study has shown total suppression of pathogenicflora after a 3-min exposure to the dye solution followed by a 20-sec treatment with the R-LETB.For the clinical study,37 subjects of both sexes with gingivitis were enrolled and randomly assigned to one of two groups.Subjects in thefirst(treatment)group were instructed to rinse their mouth with MB solution provided for 1 min and then brush the teeth with the R-LETB and standardized toothpaste.The second(control)group used only the toothpaste and a regular toothbrush.Subjects in both groups followed their respective procedures 2 times a day(morning and evening)for 30 days.Indices of plaque,gingival bleeding,and inflammation were evaluated at 14-day and 30-day timepoints.In the both groups,all indices improved in comparison with baseline.However,the treatment group demonstrated more pronounced improvement of the studied indices that was attributed to additional anti-microbial action of red light and MB on gum tissue.Thus,the use of R-LETB with MB appears to have a multifactor therapeutic action on oral pathological microflora:mechanical removal of the bacteria and suppressing action on microorganisms due to photodynamic reaction.ELINA A.GENINA VLADIMIR A.TITORENKO VALERY V.TUCHIN GEORGY V.SIMONENKO ALEXEY N.BASHKATOV GENNADY M.SHUB ALEXANDER V.LEPILIN ILYA V.YAROSLAVSKY GREGORY B.ALTSHULER 2011Journal of Innovative Optical Health Sciences2011,4,4:0
