| 1 | Long-term thermal regimes of the Qinghai-Tibet Railway embankments in plateau permafrost regions显示文摘Ten years of ground temperature data(2003–2013) indicate that the long-term thermal regimes within embankments of the Qinghai-Tibet Railway(QTR) vary significantly with different embankment structures. Obvious asymmetries exist in the ground temperature fields within the traditional embankment(TE) and the crushed-rock basement embankment(CRBE). Measurements indicate that the TE and CRBE are not conducive to maintaining thermal stability. In contrast, the ground temperature fields of both the crushed-rock sloped embankment(CRSE) and the U-shaped crushed-rock embankment(UCRE) were symmetrical. However, the UCRE gave better thermal stability than the CRSE because slow warming of deep permafrost was observed under the CRSE. Therefore, the UCRE has the best long-term effect of decreasing ground temperature and improving the symmetry of the temperature field. More generally, it is concluded that construction using the cooling-roadbed principle meets the design requirements for long-term stability of the railway and for train transport speeds of 100 km h?1. | 3 | Inactivation of TFEB and NF-kB by marchantin M alleviates the chemotherapy-driven pro-tumorigenic senescent secretion显示文摘It is critical to regulate the senescence-associated secretory phenotype(SASP) due to its effect on promoting malignant phenotypes and limiting the efficiency of cancer therapy. In this study, we demonstrated that marchantin M(Mar-M, a naturally occurring bisbibenzyl) suppressed proinflammatory SASP components which were elevated in chemotherapy-resistant cells. Mar-M treatment attenuated the pro-tumorigenic effects of SASP and enhanced survival in drug-resistant mouse models.No toxicity was detected on normal fibroblast cells or in animals following this treatment. Inactivation of transcription factor EB(TFEB) and nuclear factor-k B(NF-k B) by Mar-M significantly accounted for its suppression on the components of SASP. Furthermore, inhibition of SASP by Mar-M contributed to a synergistic effect during co-treatment with doxorubicin to lower toxicity and enhance antitumor efficacy. Thus, chemotherapy-driven pro-inflammatory activity, seen to contribute to drug-resistance, is an important target for Mar-M. | 8 | An Inhibitory Effect of MAD1 on Bladder Tumor Cellular Proliferation in Vivo显示文摘OBJECTIVE To observe the inhibitory effect on bladder tumor prolivera-tion after transfection with the expression plasmid pcDNA3.1(+)/Mad1. METHODS Bladder tumors were induced in SD rats by intravesical instillation of MNU . The tumor-bearing rats were randomly divided into 3 groups: group A, transfected with pcDNA3.1 (+)/Mad1, group B, trans-fected with an empty vector and group C, transfected with saline. Rat body weight (RBW), bladder absolute weight (BAW) and bladder relative weight (BRW) were measured and expression levels of Mad1 and TERT were assayed. Flow cytometer analysis was used to observe the effect of Mad1 on the bladder tumors. RESULTS Comparions of RBW among the 3 groups showed there were no differences (P>0.05). But the BAW and BRW for group A were significantly decreased (P<0.01, P<0.05, respectively) comparded to groups B and C. In group A, the Mad1 mRNA expression level was markedly improved, while the TERT mRNA expression level was decreased. Flow cy-tometry showed an increase in G0/G1-phase cells and a decrease of S-phase cells after transfection with Mad1. | 9 | Tea domain transcription factor TEAD4 mitigatesTGF-β signaling and hepatocellular carcinomaprogression independently of YAP显示文摘Tea domain transcription factor 4 (TEAD4) plays a pivotal role in tissue development and homeostasis by interacting with Yesassociated protein (YAP) in response to Hippo signaling inactivation. TEAD4 and YAP can also cooperate with transforminggrowth factor-β (TGF-β)-activated Smad proteins to regulate gene transcription. Yet, it remains unclear whether TEAD4 playsa YAP-independent role in TGF-β signaling. Here, we unveil a novel tumor suppressive function of TEAD4 in liver cancer viamitigating TGF-β signaling. Ectopic TEAD4 inhibited TGF-β-induced signal transduction, Smad transcriptional activity, and targetgene transcription, consequently suppressing hepatocellular carcinoma cell proliferation and migration in vitro and xenografttumorgrowth in mice. Consistently, depletion of endogenous TEAD4 by siRNAs enhanced TGF-β signaling in cancer cells. Mechanistically,TEAD4 associates with receptor-regulated Smads (Smad2/3) and Smad4 in the nucleus, thereby impairing the binding of Smad2/3to the histone acetyltransferase p300.
