1Oral immunization of animals with transgenic cherry tomatillo expressing HBsAg显示文摘AIM: To investigate the expression of recombinant HBsAg (rHBsAg) in transgenic cherry tomatillo in order to explore the feasibility of producing HBV oral vaccine with cherry tomatillo by animal immune tests.METHODS: The recombinant plant expression vector containing HBsAg gene was constructed. Mediated with Agrobacterium tumefaciens, HBsAg gene was transferred into cotyledons of cherry tomatillo. Transformed cherry tomatillos were obtained through hygromycin delay-selection. Integrated DNA in transgenic cherry tomatillo was confirmed by hygromycin resistance selection, Gus detection, polymerase chain reaction (PCR) and dot blotting analysis. Antigenicity of rHBsAg was examined by ELISA and the immunogenicity of rHBsAg derived from transgenic cherry tomatillo tissues was confirmed by oral feed of transformed tissues to BALB/c mice primed with commercial HBV vaccines. Specific antibody titers in mice's serum were examined by ELISA every week.RESULTS: By far, 10 positive lines of transgenic cherry tomatillos containing HBsAg gene were obtained. Among different organs of the same transgenic cherry tomatillo,level of rHBsAg expressed in leaves was the highest with the yield up to 300ng/g fresh weight. And the rHBsAg expression level in fruits was about 10 ng/g fresh weight.In animal immune tests, oral delivery with transgenic tissues to mice primed with commercial vaccine instead of naive mice resulted in significant immune response.CONCLUSION: The result of this animal immune test indicated the rHBsAg derived from transgenic cherry tomatillo possessed normal immunogenicity. 2Liposomes and lipid disks traverse the BBB and BBTB as intact forms as revealed by two-step F?rster resonance energy transfer imaging显示文摘The blood–brain barrier(BBB) and the blood–brain tumor barrier(BBTB) prevent drug and nano-drug delivery systems from entering the brain. However, ligand-mediated nano-drug delivery systems have significantly enhanced the therapeutic treatment of glioma. In this study we investigated the mechanism especially the integrity of liposomes and lipid disks while traversing the BBB and BBTB both in vitro and in vivo. Fluorophores(DiO, DiI and DiD) were loaded into liposomes and lipid disks to form F?rster resonance energy transfer(FRET) nano-drug delivery systems. Using brain capillary endothelial cells as a BBB model, we show that liposomes and disks are present in the cytoplasm as their intact forms and traverse the BBB with a ratio of 0.68‰ and 1.67‰, respectively. Using human umbilical vein endothelial cells as BBTB model, liposomes and disks remained intact and traversed the BBTB with a ratio of 2.31‰and 8.32‰ at 3 h. Ex vivo imaging and immunohistochemical results revealed that liposomes and disks could traverse the BBB and BBTB in vivo as intact forms. 3A rapid and efficient method to express target genes in mammalian cells by baculovirus显示文摘AIM: To investigate the modification of baculovirus vector and the feasibility of delivering exogenous genes into mammalian cells with the culture supernatant of Spodoptera frugiperta (Sf9) cells infected by recombinant baculoviruses. METHODS: Two recombinant baculoviruses (BacV-CMV-EGFPA, BacV-CMV-EGFPB) containing CMV-EGFP expression cassette were constructed. HepG2 cells were directly incubated with the culture supernatant of Sf9 cells infected by recombinant baculoviruses, and reporter gene transfer and expression efficiencies were analyzed by flow cytometry (FCM). The optimal transduction conditions were investigated by FCM assay in HepG2 cells. Gene-transfer and expression efficiencies in HepG2 or CV1 cells by baculovirus vectors were compared with lipofectAMINE, recombinant retrovirus and vaccinia virus expression systems. Twenty different mammalian cell lines were used to investigate the feasibility of delivering exogenous genes into different mammalian cells with the culture supernatant of infected Sf9 cells. RESULTS: CMV promoter could directly express reporter genes in Sf9 cells with a relatively low efficiency. Target cells incubated with the 1:1 diluted culture supernatant (moi=50) for 12 h at 37 ℃ could achieve the highest transduction and expression efficiencies with least impairment to cell viability. Under similar conditions the baculovirus vector could achieve the highest gene-transfer and expression efficiency than lipofectAMINE, recombinant retrovirus and vaccinia virus expression systems. Most mammalian cell lines could be transduced with recombinant baculovirus. In primate adherent culture cells the recombinant baculovirus could arrive the highest infection and expression efficiencies, but it was not very satisfactory in the cell lines from mice and suspended culture cells. 4Hydrophobicity of reactive site loop of SCCA1 affects its binding to hepatitis B virus显示文摘AIM: To investigate the role of SCCA2 and other SCCA1 molecules in the process of hepatitis B virus (HBV) binding to mammalian cells.METHODS: SCCA1 and SCCA2 were isolated from HepG2. Binding protein (BP) genes were obtained through PCR. Recombinant baculoviruses expressing SCCA1, SCCA2, BP, and different mutants were constructed and utilized to infect mammalian cells to investigate the binding ability of infected cells to HBV.RESULTS: A SCCA1 gene (A1) was isolated from HepG2, but it appeared to lack the binding ability of infected cells to HBV. Two mutants, A1-BP and BP-A1, were constructed by interchanging the carboxyl terminal of A1 and BP. Cells expressing A1-BP showed an increased virus bindingcapacity, but not BP-A1. Comparison of A1 sequence with the sequence of BP indicated the presence of only three amino acid changes in the carboxyl terminal, two of them were found in the reactive site loop (RSL) of SCCA1. Primary structure assay revealed that the hydrophobicity of BP and AJ515706 in this domain was strong, but A1 was relatively weak. Changing the aa349 of A1 from low hydrophobic glutamic acid to high hydrophobic valine enhanced HBV binding. In contrast, HBV binding was reduced by changing the aa349 of BP from valine to glutamic acid. 5Joint elastic-electrical properties of sediments in the Yellow Sea显示文摘We measured in the laboratory compressional wave velocity and electrical resistivity on 434 sediment samples collected from the Yellow Sea to study the joint elastic-electrical properties of marine sediments. Porosity was found to reduce both elastic velocity and electrical resistivity of the marine sediments in a non-linear fashion; velocity showed an approximate linear increase with increasing logarithm of resistivity. Various effective medium models either implemented or developed were compared with the new dataset. The model results showed that the combined self-consistent approximation and differential effective medium model using critical porosity of 0.6 and 0.5 for velocity and resistivity respectively gave a reasonable description of the joint elastic-electrical behaviors of the marine sediments. 6Expression and immunoactivity of chimeric particulate antigens of receptor binding site-core antigen of hepatitis B virus显示文摘AIM: To improve the immunogenicity of receptor binding site of hepatitis B virus (HBV) on preS1 antigen using HBV core antigen as an immuno-carrier.METHODS: One to 6 tandem copies of HBV preS1 (21-47)fragment were inserted into HBcAg at the sites of aa 78 and 82, and expressed in E. coli. ELISA, Western blot and animal immunization were used to analyze the antigenicity and immmunogenicity of purified particulate antigens. The ability to capture HBV by antibodies elicited by chimeric partides was detected with immuno-capture PCR.RESULTS: Recombinant antigens CⅠ, CⅡ, CⅢ carrying 1-3 copies of HBV preS1 (21-47) individually could form viruslike particles (VLPs), similar to HBcAg in morphology. But recombinant antigens carrying 4-6 copies of HBV preS1 (21-47) were poorly expressed in E.coli. Chimeric antigens were lacking of immunoreactivity with anti-HBc monoclonal antibodies (McAbs), but still reserved good immunoreactivity with anti-HBe McAbs. CⅠ, CⅡ, CⅢ could strongly react with anti-preS1 McAb, suggesting that preS1 (21-47) fragment was well exposed on the surface of chimeric VLPs. Three chimeric VLP antigens (CⅠ, CⅡ and CⅢ) could stimulate mice to produce high-level antibody responses, and their immunogenicity was stronger than non-particulate antigen 21-47*6, containing 6 copies of preS1 (21-47). Mouse antibodies to CⅠ, CⅡ and CⅢ were able to capture HBV virions in immuno-capture PCR assay in vitro.CONCLUSION: Chimeric particulate antigens of receptor binding site-core antigen of HBV can elicit strong antibody responses to preS1. 9Research on Dielectric Focusing Lens Antenna for HPM Atmospheric Breakdown Experiments显示文摘To distinguish the dielectric surface breakdown from the atmospheric breakdown, one method for atmospheric breakdown experiment by loading dielectric focusing lens is proposed. The focusing characteristics of dielectric focusing lens are investigated for the microwave frequency on the order of 1.3GHz to 9.3GHz, the lens focal length on the order of 0.3m to 0.9m and the dielectric constant on the order of 0.3 to 0.9. Simulations show that higher frequency, shorter focal length, and fitter dielectric constant result in better focus effect that the rising and falling edges of electric field strength change faster near the focus which is closer to the theoretical value. A dielectric focusing lens for the S-band is optimally designed with the polytetrafluoroethylene of 0.32 m thickness, 0.6m diameter,and 0.4m focal length. The focus indicator of dielectric lens is measured and the atmospheric breakdown experiment is carried out. The experimental values of atmospheric breakdown are consistent with the theoretical values. The electric field strength with the lens at the focus is 5–6times greater than that without focusing lens. 11Elastic wave propagation and scattering in prestressed porous rocks显示文摘Poro-acoustoelastic theory has made a great progress in both theoretical and experimental aspects,but with no publications on the joint research from theoretical analyses,experimental measurements,and numerical validations.Several key issues challenge the joint research with comparisons of experimental and numerical results,such as digital imaging of heterogeneous poroelastic properties,estimation of acoustoelastic constants,numerical dispersion at high frequencies and strong heterogeneities,elastic nonlinearity due to compliant pores,and contamination by boundary reflections.Conventional poroacoustoelastic theory,valid for the linear elastic deformation of rock grains and stiff pores,is modified by incorporating a dualporosity model to account for elastic nonlinearity due to compliant pores subject to high-magnitude loading stresses.A modified finite-element method is employed to simulate the subtle effect of microstructures on wave propagation in prestressed digital cores.We measure the heterogeneity of samples by extracting the autocorrelation length of digital cores for a rough estimation of scattering intensity.We conductexperimental measurements with a fluid-saturated sandstone sample under a constant confining pressure of 65 MPa and increasing pore pressures from 5 to 60 MPa.Numerical simulations for ultrasound propagation in the prestressed fluid-saturated digital core of the sample are followed based on the proposed poro-acoustoelastic model with compliant pores.The results demonstrate a general agreement between experimental and numerical waveforms for different stresses,validating the performance of the presented modeling scheme.The excellent agreement between experimental and numerical coda quality factors demonstrates the applicability for the numerical investigation of the stress-associated scattering attenuation in prestressed porous rocks.Li-Yun FU Bo-Ye FU Weijia SUN Tongcheng HAN Jianlin LIU 2020Science China Earth Sciences2020,63,9:1

15Molecular simulation studies of hydrocarbon and carbon dioxide adsorption on coal显示文摘Sorption isotherms of hydrocarbon and carbon dioxide(CO_2) provide crucial information for designing processes to sequester CO_2 and recover natural gas from unmineable coal beds.Methane(CH_4),ethane(C_2H_6),and CO_2 adsorption isotherms on dry coal and the temperature effect on their maximum sorption capacity have been studied by performing combined Monte Carlo(MC) and molecular dynamics(MD) simulations at temperatures of308 and 370 K(35 and 97 ℃) and at pressures up to10 MPa.Simulation results demonstrate that absolute sorption(expressed as a mass basis) divided by bulk gas density has negligible temperature effect on CH_4,C2H6,and CO_2 sorption on dry coal when pressure is over 6 MPa.CO_2 is more closely packed due to stronger interaction with coal and the stronger interaction between CO_2 molecules compared,respectively,with the interactions between hydrocarbons and coal and between hydrocarbons.The results of this work suggest that the 'a' constant(proportional to T^2_c/Pc) in the Peng-Robinson equation of state is an important factor affecting the sorption behavior of hydrocarbons.CO_2 injection pressures of lower than8 MPa may be desirable for CH_4 recovery and CO_2sequestration.This study provides a quantitative understanding of the effects of temperature on coal sorption capacity for CH_4,C_2H_6,and CO_2 from a microscopic perspective.Junfang Zhang Keyu Liu M.B.Clennell D.N.Dewhurst Zhejun Pan M.Pervukhina Tongcheng Han 2015Petroleum Science2015,12,4:1
16Distribution of gas hydrate in fractured reservoirs:Insights from anisotropic seismic measurements显示文摘Seismic properties of hydrate-bearing reservoirs that are affected significantly by the hydrate distribution are key for quantitative assessment of the reservoir.The knowledge of hydrate distribution in fractured reservoirs remains poorly understood.To obtain such knowledge,we measured and analyzed five anisotropic velocities needed to fully characterize the seismic anisotropy in an artificial sandstone with aligned fractures during hydrate formation associated with varying distribution.We showed that while the formation of hydrate improved the velocities,the improvement was more significant for hydrate saturation above 10%.We also showed that the increasing trends varied among the anisotropic velocities when hydrate saturation was above 10%.Specifically,the compressional wave velocity travelling vertical to the bedding plane and the shear wave velocity with polarization perpendicular to the bedding plane increased more rapidly than the other compressional and shear wave velocities,respectively.Interpretation of the anisotropic seismic results suggested that the hydrate tends to bind to the grains in the fractures at low hydrate saturation,and becomes to bridge the fracture surfaces when the hydrate saturation exceeds 10%.The results have provided new insights into the hydrate distribution and its resulting anisotropic seismic properties in fractured reservoirs.This will pave the way for the successful assessment of hydrate in fractured reservoirs.Shengbiao LIU Tongcheng HAN Li-Yun FU 2021Science China Earth Sciences2021,64,5:0
17Destroying pathogen-tumor symbionts synergizing with catalytic therapy of colorectal cancer by biomimetic protein-supported single-atom nanozyme显示文摘The crucial role of intratumoral bacteria in the progression of cancer has been gradually recognized with the development of sequencing technology.Several intratumoral bacteria which have been identified as pathogens of cancer that induce progression,metastasis,and poor outcome of cancer,while tumor vascular networks and immunosuppressive microenvironment provide shelters for pathogens localization.Thus,the mutually-beneficial interplay between pathogens and tumors,named“pathogentumor symbionts”,is probably a potential therapeutic site for tumor treatment.Herein,we proposed a destroying pathogen-tumor symbionts strategy that kills intratumoral pathogens,F.nucleatum,to break the symbiont and synergize to kill colorectal cancer(CRC)cells.This strategy was achieved by a groundbreaking protein-supported copper single-atom nanozyme(BSA-Cu SAN)which was inspired by the structures of native enzymes that are based on protein,with metal elements as the active center.BSA-Cu SAN can exert catalytic therapy by generating reactive oxygen species(ROS)and depleting GSH.The in vitro and in vivo experiments demonstrate that BSA-Cu SAN passively targets tumor sites and efficiently scavenges F.nucleatum in situ to destroy pathogentumor symbionts.As a result,ROS resistance of CRC through elevated autophagy mediated by F.nucleatum was relieved,contributing to apoptosis of cancer cells induced by intracellular redox imbalance generated by BSA-Cu SAN.Particularly,BSA-Cu SAN experiences renal clearance,avoiding long-term systemic toxicity.This work provides a feasible paradigm for destroying pathogen-tumor symbionts to block intratumoral pathogens interplay with CRC for antitumor therapy and an optimized trail for the SAN catalytic therapy by the clearable protein-supported SAN.Xinyue Wang Qian Chen Yefei Zhu Kairuo Wang Yongliang Chang Xiawei Wu Weichao Bao Tongcheng Cao Hangrong Chen Yang Zhang Huanlong Qin 2023Signal Transduction and Targeted Therapy2023,8,8:0
