1Identification of CD13,CD107a,and CD164 as novel basophil-activation markers and dissection of two response patterns in time kinetics of IgE-de-pendent upregulation显示文摘Using two-colour flow cytometry >200 antibodies submitted to the 8th International Workshop of Human Leukocyte Differentiation Antigens (HLDA8) have been analyzed for their reactivity with resting and activated CD203c+ basophils. Four antibodies either non-reactive or weakly reactive with resting basophils exhibited an increased reactivity with basophils activated by anti-IgE-mediated cross-linking of the high affinity IgE receptor (FcεRI). These include antibod- ies against CD164 (WS-80160, clone N6B6 and WS-80162, clone 67D2), as well as two reagents with previously unknown specificities that were identified as CD13 (WS-80274, clone A8) and CD107a (WS-80280, clone E63-880). The activation patterns followed either the “CD203c-like” or “CD63-like” activation profile. The CD203c profile is characterized by a rapid and significant upregulation (of CD13, CD164, and CD203c), reaching maximum levels after 5- 15 min of stimulation. The phosphoinositide-3-kinase (PI3K)-specific inhibitor wortmannin inhibited the upregulation of these markers whereas 12-O-tetradecanoyl-phorbol-13-acetate (TPA) induced a rapid and FcεRI-independent upregulation within 1-2 min. In the CD63 profile, maximum upregulation (of CD63 and CD107a) was detected only after 20-40 min, and upregulation by TPA reached maximum levels after 60 min. In summary, our data identify CD13, CD107a, and CD164 as novel basophil-activation antigens. 4A GOCE only gravity model GOSG01S and the validation of GOCE related satellite gravity models显示文摘We compile the GOCE-only satellite model GOSG01 S complete to spherical harmonic degree of 220 using Satellite Gravity Gradiometry(SGG) data and the Satellite-to-Satellite Tracking(SST) observations along the GOCE orbit based on applying a least-squares analysis.The diagonal components(V_(xx),V_(yy),V_(zz)) of the gravitational gradient tensor are used to form the system of observation equations with the band-pass ARMA filter.The point-wise acceleration observations(a_x,a_y,a_z) along the orbit are used to form the system of observation equations up to the maximum spherical harmonic degree/order 130.The analysis of spectral accuracy characteristics of the newly derived gravitational model GOSG01 S and the existing models GOTIM04 S,GODIR04S,GOSPW04 S and JYY_GOCE02S based on their comparison with the ultrahigh degree model EIGEN-6C2 reveals a significant consistency at the spectral window approximately between 80 and 190 due to the same period SGG data used to compile these models.The GOCE related satellite gravity models GOSG01 S,GOTIM05S,GODIR05 S,GOTIM04S,GODIR04 S,GOSPW04S,JYY_-GOCE02 S,EIGEN-6C2 and EGM2008 are also validated by using GPS-leveling data in China and USA.According to the truncation at degree 200,the statistic results show that all GGMs have very similar differences at GPS-leveling points in USA,and all GOCE related gravity models have better performance than EGM2008 in China.This suggests that all these models provide much more information on the gravity field than EGM2008 in areas with low terrestrial gravity coverage.And STDs of height anomaly differences in China for the selected truncation degrees show that GOCE has improved the accuracy of the global models beyond degree 90 and the accuracies of the models improve from 24 cm to 16 cm.STDs of geoid height differences in USA show that GOSGOIS model has best consistency comparing with GPSleveling data for the frequency band of the degree between 20 and 160.Xinyu Xu Yongqi Zhao Tilo Reubelt Robert Tenzer 2017Geodesy and Geodynamics2017,8,4:5
5Novel fiber-optical interferometer with miniaturized probe for in-hole measurements显示文摘Today, micro-system technology and the development of new MEMS (Micro-Electro-Mechanical Systems) are emerging rapidly. In order for this development to become a success in the long run, measurement systems have to ensure product quality. Most often, MEMS have to be tested by means of functionality or destructive tests. One reason for this is that there are no suitable systems or sensing probes available which can be used for the measurement of quasi inaccessible features like small holes or cavities. We present a measurement system that could be used for these kinds of measurements. 8Comparative analysis of DNA extraction protocols for ancient soft tissue museum samples显示文摘DNA studies of endangered or extinct species often rely on ancient or degraded remains.The majority of ancient DNA(aDNA)extraction protocols focus on skeletal elements,with skin and hair samples rarely explored.Similar to that found in bones and teeth,DNA extracted from historical or ancient skin and fur samples is also extremely fragmented with low endogenous content due to natural degradation processes.Thus,the development of effective DNA extraction methods is required for these materials.Here,we compared the performance of two DNA extraction protocols(commercial and custom laboratory aDNA methods)on hair and skin samples from decades-old museum specimens to Iron Age archaeological material.We found that apart from the impact sample-specific taphonomic and handling history has on the quantity and quality of DNA preservation,skin yielded more endogenous DNA than hair of the samples and protocols tested.While both methods recovered DNA from ancient soft tissue,the laboratory method performed better overall in terms of DNA yield and quality,which was primarily due to the poorer performance of the commercial binding buffer in recovering aDNA.Ming Zhang Peng Cao Qing-Yan Dai Yong-Qiang Wang Xiao-Tian Feng Hong-Ru Wang Hong Wu Albert Min-Shan Ko Xiao-Wei Mao Yi-Chen Liu Li Yu Christian Roos Tilo Nadler Wen Xiao E.Andrew Bennett Qiao-Mei Fu 2021Zoological Research2021,42,3:1

15Interferometric measurement of injection nozzles using ultra-small fiber-optical probes显示文摘The measurement of boreholes with diameters smaller than 500 μm is a demanding task that cannot be performed using state-of-the-art production metrology. In this letter, a miniaturized fiber probe with a diameter of 80 μm is presented. A probe is used for low-coherence interferometry to conduct highly precise measurements of form deviations of small boreholes. Measurements conducted in nozzles are also presented.
