| 1 | Connexin 43 hemichannels protect bone loss during estrogen deficiency显示文摘Estrogen deficiency in postmenopausal women is a major cause of bone loss,resulting in osteopenia,osteoporosis,and a high risk for bone fracture.Connexin 43 (Cx43) hemichannels (HCs) in osteocytes play an important role in osteocyte viability,bone formation,and remodeling.We showed here that estrogen deficiency reduced Cx43 expression and HC function.To determine if functional HCs protect osteocytes and bone loss during estrogen deficiency,we adopted an ovariectomy model in wild-type (WT) and two transgenic Cx43 mice:R76W (dominant-negative mutant inhibiting only gap junction channels) and Cx43 Δ130–136 (dominant-negative mutant compromising both gap junction channels and HCs).The bone mineral density (BMD),bone structure,and histomorphometric changes of cortical and trabecular bones after ovariectomy were investigated.Our results showed that the Δ130–136 transgenic cohort had greatly decreased vertebral trabecular bone mass compared to WT and R76W mice,associated with a significant increase in the number of apoptotic osteocyte and empty lacunae.Moreover,osteoclast surfaces in trabecular and cortical bones were increased after ovariectomy in the R76W and WT mice,respectively,but not in Δ130–136 mice.These data demonstrate that impairment of Cx43 HCs in osteocytes accelerates vertebral trabecular bone loss and increase in osteocyte apoptosis,and further suggest that Cx43 HCs in osteocytes protect trabecular bone against catabolic effects due to estrogen deficiency. | Liang Ma Rui Hua Yi Tian Hongyun Cheng Roberto Jose Fajardo Joseph J. | 2 | Clostridium perfringens bacteremia caused by choledocholithiasis in the absence of gallbladder stones显示文摘A 67-years-old male presented with periumbilical abdominal pain, fever and jaundice. His anaerobic blood culture was positive for clostridium perfringens. Computed tomogram scan of the abdomen and abdominal ultrasound showed normal gallbladder and common bile duct (CBD). Subsequently magnetic resonance cholangiopancreaticogram showed choledocholithiasis. Endoscopic retrograde cholangiopancreaticogram-with sphincterotomy and CBD stone extraction was performed. | 3 | Graphene for energy harvesting/storage devices and printed electronics显示文摘Graphene-based materials are intriguing from the perspective of fundamental science and technology because they are non-toxic, chemically and thermally tolerant, and mechanically robust. Graphene exhibits superior electrical conductivity, high surface area and a broad electrochemical window that may be particularly advantageous for their applications in energy storage devices. In addition, graphene can be prepared in the form of a colloidal suspension with adjustable solubility and thus is suitable for printing applications and offers both transparency and good conductivity at the same time. In this review, applications of graphene in solar cells, batteries, supercapacitors and fuel cells are summarized with the latest developments. | 4 | Structure-based Comparative Analysis and Prediction of N-linked Glycosylation Sites in Evolutionarily Distant Eukaryotes显示文摘The asparagine-X-serine/threonine (NXS/T) motif, where X is any amino acid except proline, is the consensus motif for N-linked glycosylation. Significant numbers of high-resolution crystal structures of glycosylated proteins allow us to carry out structural analysis of the N-linked glycosylation sites (NGS). Our analysis shows that there is enough structural information from diverse glycoproteins to allow the development of rules which can be used to predict NGS. A Python-based tool was developed to investigate asparagines implicated in N-glycosylation in five species: Homo sapiens, Mus musculus, Drosophila melanogaster, Arabidopsis thaliana and Saccharomyces cerevisiae. Our analysis shows that 78% of all asparagines of NXS/T motif involved in N-glycosylation are localized in the loop/turn conformation in the human proteome. Similar distribution was revealed for all the other species examined. Comparative analysis of the occurrence of NXS/T motifs not known to be glycosylated and their reverse sequence (S/TXN) shows a similar distribution across the secondary structural elements, indicating that the NXS/T motif in itself is not biologically relevant. Based on our analysis, we have defined rules to determine NGS. Using machine learning methods based on these rules we can predict with 93% accuracy if a particular site will be glycosylated. If structural information is not available the tool uses structural prediction results resulting in 74% accuracy. The tool was used to identify glycosylation sites in 108 human proteins with structures and 2247 proteins without structures that have acquired NXS/T site/s due to non-synonymous variation. | 5 | DK Crush技术治疗真性冠状动脉分叉病变:与经典Crush技术的对比显示文摘目的检验作者改良的DK Crush技术能否确保最终对吻扩张的成功。方法本研究属于连续、非随机和开放式研究,共入选了88例真性分叉病变(分支血管直径>2.0mm)的患者。其中2004年10月至2005年1月间入选的44例患者接受经典Crush技术治疗,2005年1月至2005年6月间入选的44例患者接受DK Crush技术治疗。比较两组患者术前、术中和术后30d内的诸项参数。结果DK Crush组与经典Crush组比较,患者分支血管病变长度长(13.5±3.4mm比7.8±3.1mm,P<0.05)、经皮冠状动脉介入术需时短(44±12min比68±17min,P<0.05)、最终对吻扩张成功率高(100%比70%,P<0.01)、使用球囊数量少(1.6±0.4个比2.7±0.7个,P<0.05)及造影剂使用量小(102±38mL比176±46mL,P<0.05)。DKCrush组与经典Crush组患者的主干与分支血管之间的夹角[(57±18)°比(47±15)°],主干血管病变长度(24.3±8.6mm比21.1±7.3mm)差异均无统计学意义(P均>0.05)。经典Crush组有两例(4.3%)最终对吻失败的患者出现亚急性血栓栓塞。DK Crush组术后即刻分支血管开口部位最小血管直径显著大于经典Crush组(3.01±0.13mm比2.74±0.12mm,P<0.01),而残余狭窄却显著小于后者(7.3%±8.6%比17.4%±11.2%,P<0.05)。经典Crush组有5例患者术后即刻分支血管开口残余狭窄>30%(对吻失败)。结论经典Crush技术存在的技术缺陷是导致预后不良的主要原因。改良的DK Crush技术能显著提高对吻扩张的成功率,进一步的随机研究可以明确后者的有效性。 | 陈绍良 方唯一 魏盟 何奔 张瑞岩 陈韵岱 张俊杰 叶飞 朱中生 林松 单守杰 杨松 钱均 Damras Michael Lee Tan Huaycheem Tejas Patel | 2007 | 中国介入心脏病学杂志2007,15,2: | 7 |

| 7 | Cervical inlet patch-optical coherence tomography imaging and clinical significance显示文摘AIM:To demonstrate the feasibility of optical coherence tomography(OCT)imaging in differentiating cervical inlet patch(CIP)from normal esophagus,Barrett'sesophagus(BE),normal stomach and duodenum. METHODS:This study was conducted at the Veterans Affairs Boston Healthcare System(VABHS).Patients undergoing standard esophagogastroduodenoscopy at VABHS,including one patient with CIP,one representative patient with BE and three representative normal subjects were included.White light video endoscopy was performed and endoscopic 3D-OCT images were obtained in each patient using a prototype OCT system.The OCT imaging probe passes through the working channel of the endoscope to enable simultaneous video endoscopy and 3D-OCT examination of the human gastrointestinal(GI)tract.Standard hematoxylin and eosin(H and E)histology was performed on biopsy or endoscopic mucosal resection specimens in order to compare and validate the 3D-OCT data. RESULTS:CIP was observed from a 68-year old male with gastroesophageal reflux disease.The CIP region appeared as a pink circular lesion in the upper esophagus under white light endoscopy.OCT imaging over the CIP region showed columnar epithelium structure,which clearly contrasted the squamous epithelium structure from adjacent normal esophagus.3D-OCT images obtained from other representative patients demonstrated distinctive patterns of the normal esophagus,BE,normal stomach,and normal duodenum bulb.Microstructures,such as squamous epithelium,lamina propria, muscularis mucosa,muscularis propria,esophageal glands,Barrett's glands,gastric mucosa,gastric glands, and intestinal mucosal villi were clearly observed with OCT and matched with H and E histology.These results demonstrated the feasibility of using OCT to evaluate GI tissue morphology in situ and in real-time. | 10 | Effect of hydroalcoholic extract of Hibiscus rosa sinensis Linn.leaves in experimental colitis in rats显示文摘Objective:To elucidate the ameliorative effect of liydrualcuholic extract of leaves of Hibiscus rosa sinensis(HRS) in acetic acid induced experimental colitis in male wistar rats.Methods: The animals were administered with 2 mL acetic acid(4%) via intra rectal.The animals were divided into various treatment groups(n=6).Prednisolone was used as standard drug and HRS was administered at a dose of 50,100 and 200 mg/kg p.o.The control group of animals received 1 mL of vehicle(distilled water).Ulcer area,ulcer index,spleen weight,colon weight to length ratio, macroscopic score,haemalological parameters,colonic superoxide dismutase(SOD),glutathione(GSH),myeloperoxidase(MPO),malondialdchyde(MDA),tumor necrosis factor-α(TNK- α), nitric oxide(NO) and histological changes were recorded after the treatment regimen of 11 days. | 11 | Gut microbiota and obesity:Impact of antibiotics and prebiotics and potential for musculoskeletal health显示文摘Obesity is a complex disease with multiple contributing factors.One of the most intensely studied factors during the past decade has been the gut microbiota,which is the community of all microbes in the intestinal tract.The gut microbiota,via energy extraction,inflammation,and other actions,is now recognized as an important player in the pathogenesis of obesity.Dysbiosis,or an imbalance in the microbial community,can initiate a cascade of metabolic disturbances in the host.Early life is a particularly important period for the development of the gut microbiota,and perturbations such as with antibiotic exposure can have long-lasting consequences for host health.In early life and throughout the life span,diet is one of the most important factors that shape the gut microbiota.Although diets high in fat and sugar have been shown to contribute to dysbiosis and disease,dietary fiber is recognized as an important fermentative fuel for the gut microbiota and results in the production of short-chain fatty acids that can act as signaling molecules in the host.One particular type of fiber,prebiotic fiber,contributes to changes in the gut microbiota,the most notable of which is an increase in the abundance of Bifidobacterium.This review highlights our current understanding of the role of gut microbiota in obesity development and the ways in which manipulating the microbiota through dietary means,specifically prebiotics,could contribute to improved health in the host,including musculo skeletal health. | Teja Klancic Raylene A.Reimer | 2020 | Journal of Sport and Health Science2020,9,2: | 2 |
