| 1 | Highly sensitive potentiometric immunosensor for hepatitis B surface antigen diagnosis显示文摘A highly sensitive potentiometric immunosensor for the diagnoses of epidemic dis- eases has been developed by means of self-assembly to immobilize hepatitis B surface antibody (HBsAb) for the detection of hepatitis B surface antigen (HBsAg) as a model. At first, the Nafion containing ?SO3 groups was immobilized on a platinum electrode surface to absorb the ?NH3 ? + groups of antibody molecules via the opposite-charged adsorption technique, in the meantime, hepatitis B surface antibodies were adsorbed onto the surface of Au nanoparticles, then hepatitis B surface antibodies and Au nanopartilces were entrapped into polyvinyl butyral on the surface of Nafion film. The modified procedure was further characterized by electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). The influence and factors influencing the per- formance of resulting immunosensor were studied in detail. The resulting immunosensor exhib- ited sigmoid curve with log HBsAg concentrations, high sensitivity, wide linear range from 26 to 1280 ng·mL?1 with a detection limit of 3.1 ng·mL?1, rapid potentiometric response (<3 min) and long-term stability (>4 months). | 2 | Metal sulfide quantum dots-aggregated PAMAM dendrimer for cadmium ion-selective electrode-based immunoassay of alpha-fetoprotein显示文摘An ion-selective electrode(ISE)-based immunoassay has been innovatively designed for the sensitive detection of liver cancer biomarker(alpha-fetoprotein,AFP),using metal sulfide quantum dot(QD)-based nano labels.Cd S QDs-aggregated PAMAM dendrimer(QD-DE)was first synthesized and functionalized with polyclonal rabbit anti-human AFP antibodies.Thereafter,a sandwich immunoreaction was implemented on monoclonal mouse anti-human AFP antibody-coated microplate by using antibody-functionalized QD-DE as the secondary antibody.Accompanying the immunocomplex,subsequent potentiometric detection of cadmium ion dissolved from the QD-DE under acidic condition was conducted on a portable cadmium ion-selective electrode(Cd-ISE).Results revealed that the electrode potential of the Cd-ISE increased with the increment of AFP concentration from 0.1 to 100 ng m L^(-1)at a detection limit(LOD)of 68 pg m L^(-1).The relative standard deviations(RSD)were below9.09%and 10.54%for the intra-and inter-assay,respectively.Additionally,six human serum specimens were determined on CdISE-based immunosensor by using commercial human AFP ELISA kit as the reference,and gave good relationship between two methods.Importantly,Cd-ISE-based immunoassay offers the promise for simple and cost-effective screening of disease-related biomarkers. | Qunfang Li Jing Jin Fangming Lou Dianping Tang | 2018 | Science China Chemistry2018,61,6: | 2 |

| 5 | Enzyme-conjugated hybridization chain reaction for magneto-controlled immunoassay of squamous cell carcinoma antigen with pH meter显示文摘A nanoparticle-based potentiometric immunoassay was designed for the sensitive detection of squamous cell carcinoma antigen(SCCA;cervical carcinoma marker) on a portable pH meter coupling enzyme-labeled hybridization chain reaction(HCR) with two alternating hairpin DNA probes for the signal amplification. Initially, a sandwich-type immunoreaction was carried out between anti-SCCA capture antibody-conjugated magnetic bead and detection antibody/initiator strand-coated gold nanoparticle(AuNP). Then, the HCR reaction was readily executed between two glucose oxidase(GOx)-labeled hairpins through the initiator strand to form numerous GOx concatamers on the AuNP via the long nicked double-helix. The concatenated GOx oxidized glucose into gluconic acid and hydrogen peroxide, thus resulting in the pH change of the detection solution on a handheld pH meter. Several labeling protocols including GOx-antibody, GOx-AuNP-antibody and GOx-HCR-AuNP-antibody were investigated for detection of target SCCA, and improved analytical features were obtained with the immune-HCR assay. Under optimum conditions, the immune-HCR assay exhibited good pH responses for the determination of SCCA at a concentration as low as 5.7 pg/mL.
