1禽流感病毒非结构蛋白1单克隆抗体的制备及其特异性分析显示文摘为研制禽流感病毒(H5N1)非结构蛋白1(NS1)的特异性单克隆抗体(mAb),并鉴定其特异性,本研究在分别表达了具有良好抗原性的A/Vietnam/1194/04(H5N1)-NS1和A/HongKong/486/97(H5N1)-NS1重组蛋白基础上,用A/Viet-nam/1194/04(H5N1)-NS1蛋白免疫BALB/c小鼠,取其脾细胞与小鼠骨髓瘤细胞进行融合,间接ELISA筛选阳性的杂交瘤细胞,并结合免疫荧光和免疫印迹对抗体的特异性进行鉴定,通过竞争抑制实验对单抗识别的抗原位点进行分析。结果共获得19株能识别4个H5N1-NS1蛋白不同抗原位点的mAb,亚类测定显示,5株为IgG2a、1株为IgG2b,另外13株为IgG1。这些mAb均与A/Vietnam/1194/04(H5N1)-NS1和A/HongKong/486/97(H5N1)-NS1重组蛋白特异性结合,免疫荧光检测均与A型流感病毒(H1N1和H3N2)有交叉反应,而与B型流感病毒无交叉现象。表明成功获得特异性针对H5N1-NS1蛋白的mAb,为进一步研究禽流感病毒NS1蛋白的结构与功能奠定基础。丁细霞 Hoi-wah Tsoi 陈伟俊 丘立文 王压娣 廖志勇 袁国勇 车小燕 2007现代免疫学2007,27,6:4

3Review of current progress in hole-transporting materials for perovskite solar cells显示文摘Recent advancements in perovskites’ application as a solar energy harvester have been astonishing. The power conversion efficiency(PCE) of perovskite solar cells(PSCs) is currently reaching parity(>25 percent), an accomplishment attained over past decades. PSCs are seen as perovskites sandwiched between an electron transporting material(ETM) and a hole transporting material(HTM). As a primary component of PSCs, HTM has been shown to have a considerable effect on solar energy harvesting, carrier extraction and transport, crystallization of perovskite, stability, and price. In PSCs, it is still necessary to use a HTM.While perovskites are capable of conducting holes, they are present in trace amounts, necessitating the use of an HTM layer for efficient charge extraction. In this review, we provide an understanding of the significant forms of HTM accessible(inorganic, polymeric and small molecule-based HTMs), to motivate further research and development of such materials. 4Early-onset primary torsional dystonia in a 4-generation Chinese family with a mutation in the DYT1 gene显示文摘Primary torsion dystonia (PTD) is a clinically and genetically heterogeneous movement disorder. At least thirteen different types of dystonia can be distinguished on a genetic basis.1 The DYT1 gene was first mapped by Ozelius et al in 1989.2 (Kramer et) al3 linked the same locus to PTD in 12 Ashkenazi Jewish families in 1990. 7Oncogene GAEC1 regulates CAPN10 expression which predicts survival in esophageal squamous cell carcinoma显示文摘AIM: To identify the downstream regulated genes of GAEC1 oncogene in esophageal squamous cell carcinoma and their clinicopathological significance. METHODS: The anti-proliferative effect of knocking down the expression of GAEC1 oncogene was stud-ied by using the RNA interference (RNAi) approach through transfecting the GAEC1 -overexpressed esophageal carcinoma cell line KYSE150 with the pSilencer vector cloned with a GAEC1 -targeted sequence, followed by MTS cell proliferation assay and cell cycle analysis using flow cytometry. RNA was then extracted from the parental, pSilencer-GAEC1 -targeted sequence transfected and pSilencer negative control vector transfected KYSE150 cells for further analysis of different patterns in gene expression. Genes differentially expressed with suppressed GAEC1 expression were then determined using Human Genome U133 Plus 2.0 cDNA microarray analysis by comparing with the parental cells and normalized with the pSilencer negative control vector transfected cells. The most prominently regulated genes were then studied by immunohistochemical staining using tissue microarrays to determine their clinicopathological correlations in esophageal squamous cell carcinoma by statistical analyses. RESULTS: The RNAi approach of knocking down gene expression showed the effective suppression of GAEC1 expression in esophageal squamous cell carcinoma cell line KYSE150 that resulted in the inhibition of cell proliferation and increase of apoptotic population. cDNA microarray analysis for identifying differentially expressed genes detected the greatest levels of downregulation of calpain 10 (CAPN10 ) and upregulation of trinucleotide repeat containing 6C (TNRC6C ) transcripts when GAEC1 expression was suppressed. At the tissue level, the high level expression of calpain 10 protein was significantly associated with longer patient survival (month) of esophageal squamous cell carcinoma compared to the patients with low level of calpain 10 expression (37.73 ± 16.33 vs 12.62 ± 12.44, P = 0.032). No significant correction was observed among the TNRC6C protein expression level and the clinocopathologcial features of esophageal squamous cell carcinoma. CONCLUSION: GAEC1 regulates the expression of CAPN10 and TNRC6C downstream. 8Mechanical and biological properties of Ti–(0–25 wt%)Nb alloys for biomedical implants application显示文摘Binary titanium–niobium(Ti–Nb)alloys have recently been attracted due to low Young’s moduli and non-toxic properties.This study explores the influence of low Nb content(0–25 wt%)on the comprehensive parameters of tensile stress–strain relationships(ultimate strength(rUTS),yield strength(r0.2)and elastic modulus(E)),surfaces properties(Vickers microhardness,surface roughness(Ra),water contact angle(WCA),X-ray diffraction(XRD)and scanning electron microscopy(SEM)),corrosion resistance(in artificial saliva and lactic acid)and biological properties(cytotoxicity and alkaline phosphatase activity of MC3T3-E1 pre-osteoblasts)of Ti–xNb alloys(x紏5,10,15,20 and 25 wt%),with using commercially pure grade 2 titanium(cp-Ti)as control.XRD results shown that all the Ti–xNb alloys comprised atb Ti alloy phases,such that the b phase increased correspondingly with the increased amount of Nb in the alloy,as well as the reduction of E(69–87 GPa).Except Ti–5Nb,all other Ti–xNb alloys showed a significantly higher hardness,increased rUTS and r0.2,and decreased WCA compared with cp-Ti.No corrosion was detected on Ti–xNb alloys and cp-Ti in artificial saliva and lactic acid solutions.The cytotoxicity of Ti–xNb alloys was comparable to that of cp-Ti in MC3T3-E1 pre-osteoblasts without interference from differentiation behaviour,but the proliferation rate of the Ti–5Nb alloy was lower than other groups.In overall,binary Ti–(10–25 wt%)Nb alloys are promising candidate for orthopaedic and dental implants due to their improved mechanical properties and comparable biological performance,while Ti–5Nb should be used with caution.Yuqing Zhang Danni Sun Jun Cheng James Kit Hon Tsoi Jiang Chen 2020Regenerative Biomaterials2020,7,1:2
