1Malignant peritoneal mesothelioma显示文摘Malignant mesothelioma is a highly aggressive neoplasm.The incidence of malignant mesothelioma is increasing worldwide.Diffuse malignant peritoneal mesothelioma(DMPM) represents one-fourth of all mesotheliomas.Association of asbestos exposure with DMPM has been observed,especially in males.The great majority of patients present with abdominal pain and distension,caused by accumulation of tumors and ascitic ? uid.In the past,DMPM was considered a pre-terminal condition;therefore attracted little attention.Patients invariably died from their disease within a year.Recently,several prospective trials have demonstrated a median survival of 40 to 90 mo and 5-year survival of 30% to 60% after combined treatment using cytoreductive surgery and perioperative intraperitoneal chemotherapy.This remarkable improvement in survival has prompted new search into the medical science related to DMPM,a disease previously ignored as uninteresting.This review article focuses on the key advances in the epidemiology,diagnosis,staging,treatments and prognosis of DMPM that have occurred in the past decade.Stine Munkholm-Larsen Christopher Q Cao Tristan D Yan 2009World Journal of Gastrointestinal Surgery2009,1,1:19

4Osteocyte dysfunction promotes osteoarthritis through MMP13-dependent suppression of subchondral bone homeostasis显示文摘Osteoarthritis(OA),long considered a primary disorder of articular cartilage,is commonly associated with subchondral bone sclerosis.However,the cellular mechanisms responsible for changes to subchondral bone in OA,and the extent to which these changes are drivers of or a secondary reaction to cartilage degeneration,remain unclear.In knee joints from human patients with end-stage OA,we found evidence of profound defects in osteocyte function.Suppression of osteocyte perilacunar/canalicular remodeling(PLR)was most severe in the medial compartment of OA subchondral bone,with lower protease expression,diminished canalicular networks,and disorganized and hypermineralized extracellular matrix.As a step toward evaluating the causality of PLR suppression in OA,we ablated the PLR enzyme MMP13 in osteocytes while leaving chondrocytic MMP13 intact,using Cre recombinase driven by the 9.6-kb DMP1 promoter.Not only did osteocytic MMP13 deficiency suppress PLR in cortical and subchondral bone,but it also compromised cartilage.Even in the absence of injury,osteocytic MMP13 deficiency was sufficient to reduce cartilage proteoglycan content,change chondrocyte production of collagen II,aggrecan,and MMP13,and increase the incidence of cartilage lesions,consistent with early OA.Thus,in humans and mice,defects in PLR coincide with cartilage defects.Osteocyte-derived MMP13 emerges as a critical regulator of cartilage homeostasis,likely via its effects on PLR.Together,these findings implicate osteocytes in bone-cartilage crosstalk in the joint and suggest a causal role for suppressed perilacunar/canalicular remodeling in osteoarthritis.Courtney MMazur Jonathon JWoo Cristal SYee Aaron JFields Claire Acevedo Karsyn NBailey Serra Kaya Tristan WFowler Jeffrey CLotz Alexis Dang Alfred CKuo Thomas PVail Tamara Alliston 2019Bone Research2019,7,4:10
5High-fidelity multimode fibre-based endoscopy for deep brain in vivo imaging显示文摘Progress in neuroscience relies on new techniques for investigating the complex dynamics of neuronal networks.An ongoing challenge is to achieve minimally invasive and high-resolution observations of neuronal activity in vivo inside deep brain areas.Recently introduced methods for holographic control of light propagation in complex media enable the use of a hair-thin multimode optical fibre as an ultranarrow imaging tool.Compared to endoscopes based on graded-index lenses or fibre bundles,this new approach offers a footprint reduction exceeding an order of magnitude,combined with a significant enhancement in resolution.We designed a compact and high-speed system for fluorescent imaging at the tip of a fibre,achieving a resolution of 1.18±0.04μm across a 50-μm field of view,yielding 7-kilopixel images at a rate of 3.5 frames/s.Furthermore,we demonstrate in vivo observations of cell bodies and processes of inhibitory neurons within deep layers of the visual cortex and hippocampus of anaesthetised mice.This study paves the way for modern microscopy to be applied deep inside tissues of living animal models while exerting a minimal impact on their structural and functional properties.Sergey Turtaev Ivo T.Leite Tristan Altwegg-Boussac Janelle M.P.Pakan Nathalie L.Rochefort Tomáš Čižmár 2018Light(Science & Applications)2018,7,1:8
6Maize grain concentrations and above-ground shoot acquisition of micronutrients as affected by intercropping with turnip, faba bean, chickpea, and soybean显示文摘Most research on micronutrients in maize has focused on maize grown as a monocrop. The aim of this study was to determine the effects of intercropping on the concentrations of micronutrients in maize grain and their acquisition via the shoot. We conducted field experiments to investigate the effects of intercropping with turnip (Brassica campestris L.), faba bean (Vicia faba L.), chickpea (Cicer arietinum L.), and soybean (Glycine max L.) on the iron (Fe), manganese (Mn), copper (Cu) and zinc (Zn) concentrations in the grain and their acquisition via the above-ground shoots of maize (Zea mays L.). Compared with monocropped maize grain, the grain of maize intercropped with legumes showed lower concentrations of Fe, Mn, Cu, and Zn and lower values of their corresponding harvest indexes. The micronutrient concentrations and harvest indexes in grain of maize intercropped with turnip were the same as those in monocropped maize grain. Intercropping stimulated the above-ground maize shoot acquisition of Fe, Mn, Cu and Zn, when averaged over different phosphorus (P) application rates. To our knowledge, this is the first report on the effects of intercropping on micronutrient concentrations in maize grain and on micronutrients acquisition via maize shoots (straw+grain). The maize grain Fe and Cu concentrations, but not Mn and Zn concentrations, were negatively correlated with maize grain yields. The concentrations of Fe, Mn, Cu, and Zn in maize grain were positively correlated with their corresponding harvest indexes. The decreased Fe, Mn, Cu, and Zn concentrations in grain of maize intercropped with legumes were attributed to reduced translocation of Fe, Mn, Cu, and Zn from vegetative tissues to grains. This may also be related to the delayed senescence of maize plants intercropped with legumes. We conclude that turnip/maize intercropping is beneficial to obtain high maize grain yield without decreased concentrations of Fe, Mn, Cu, and Zn in the grain. 10Practical assessment of rock damage due to blasting显示文摘Blasting is the most cost effective methodology to break rock for mining or civil engineering applications.A good production blast will break only the rock that is needed to be removed,leaving the host rock with minimal damage.The control of rock damage due to blasting is very important when it comes to mine or construction design,safety,and cost.Damage to the host rock due to a production blast could result in failures,overbreak and unstable ground.Knowing how far the fractures generated by a production blast will go into the host rock is a valuable tool for engineers to design a safe highwall while keeping the actual excavation close to the design.Currently,there are several methods available to predict damage due to blasting.The accuracy of many of these methods is questionable,and in most cases,the methodologies over predict the results.This often leads to inefficient mines and poor construction works.When the current methodologies are reviewed,each one presents sound approaches,but in many cases they also lack consideration of other variables that,according to the authors,need to be included when predicting blast damage.This paper presents a practical methodology to assess the rock damage from blasting by combining other methodologies.The proposed method allows consideration of more variables when compared to available methods,resulting in a more accurate rock damage assessment.The method uses the estimation of the generated levels of peak particle velocity with the distance from a production blast presented by Persson and Holmberg,the peak particle velocity damage ranges proposed by Forsyth and the relationship between the static compressive strength and dynamic compressive strength of rocks from Liu.The new methodology was validated using the data published in a large-scale study performed in granite by Siskind.Jhon Silva Tristan Worsey Braden Lusk 2019International Journal of Mining Science and Technology2019,29,3:5
11Cell signalling pathways underlying induced pluripotent stem cell reprogramming显示文摘Induced pluripotent stem(i PS) cells, somatic cells reprogrammed to the pluripotent state by forced expression of defined factors, represent a uniquely valuable resource for research and regenerative medicine. However, this methodology remains inefficient due to incomplete mechanistic understanding of the reprogramming process. In recent years, various groups have endeavoured to interrogate the cell signalling that governs the reprogramming process, including LIF/STAT3, BMP, PI3 K, FGF2, Wnt, TGFβ and MAPK pathways, with the aim of increasing our understanding and identifying new mechanisms of improving safety, reproducibility and efficiency. This has led to a unified model of reprogramming that consists of 3 stages: initiation, maturation and stabilisation. Initiation of reprogramming occurs in almost all cells that receive the reprogramming transgenes; most commonly Oct4, Sox2, Klf4 and c Myc, and involves a phenotypic mesenchymal-to-epithelial transition. The initiation stage is also characterised by increased proliferation and a metabolic switch from oxidative phosphorylation to glycolysis. The maturation stage is considered the major bottleneck within the process, resulting in very few 'stabilisation competent' cells progressing to the final stabilisation phase. To reach this stage in both mouse and human cells, pre-i PS cells must activate endogenous expression of the core circuitry of pluripotency, comprising Oct4, Sox2, and Nanog, and thus reach a state of transgene independence. By the stabilisation stage, i PS cells generally use the same signalling networks that govern pluripotency in embryonic stem cells. These pathways differ between mouse and human cells although recent work has demonstrated that this is context dependent. 16Manipulating Bloch surface waves in 2D: a platform concept-based flat lens显示文摘At the end of the 1970s,it was confirmed that dielectric multilayers can sustain Bloch surface waves(BSWs).However,BSWs were not widely studied until more recently.Taking advantage of their high-quality factor,sensing applications have focused on BSWs.Thus far,no work has been performed to manipulate and control the natural surface propagations in terms of defined functions with two-dimensional(2D)components,targeting the realization of a 2D system.In this study,we demonstrate that 2D photonic components can be implemented by coating an in-plane shaped ultrathin(l/15)polymer layer on the dielectric multilayer.The presence of the polymer modifies the local effective refractive index,enabling direct manipulation of the BSW.By locally shaping the geometries of the 2D components,the BSW can be deflected,diffracted,focused and coupled with 2D freedom.Enabling BSW manipulation in 2D,the dielectric multilayer can play a new role as a robust platform for 2D optics,which can pave the way for integration in photonic chips.Multiheterodyne near-field measurements are used to study light propagation through micro-and nano-optical components.We demonstrate that a lens-shaped polymer layer can be considered as a 2D component based on the agreement between near-field measurements and theoretical calculations.Both the focal shift and the resolution of a 2D BSW lens are measured for the first time.The proposed platform enables the design of 2D all-optical integrated systems,which have numerous potential applications,including molecular sensing and photonic circuits.Libo Yu Elsie Barakat Tristan Sfez Lubos Hvozdara Joab Di Francesco Hans Peter Herzig 2014Light(Science & Applications)2014,3,1:3

20Molecular targets for modulating the protein translation vital to proteostasis and neuron degeneration in Parkinson’s disease显示文摘Parkinson’s disease(PD)is the most common neurodegenerative movement disorder,which is characterized by the progressive loss of dopaminergic neurons in the Substantia Nigra pars compacta concomitant with Lewy body formation in affected brain areas.The detailed pathogenic mechanisms underlying the selective loss of dopaminergic neurons in PD are unclear,and no drugs or treatments have been developed to alleviate progressive dopaminergic neuron degeneration in PD.However,the formation ofα-synuclein-positive protein aggregates in Lewy body has been identified as a common pathological feature of PD,possibly stemming from the consequence of protein misfolding and dysfunctional proteostasis.Proteostasis is the mechanism for maintaining protein homeostasis via modulation of protein translation,enhancement of chaperone capacity and the prompt clearance of misfolded protein by the ubiquitin proteasome system and autophagy.Deregulated protein translation and impaired capacities of chaperone or protein degradation can disturb proteostasis processes,leading to pathological protein aggregation and neurodegeneration in PD.In recent years,multiple molecular targets in the modulation of protein translation vital to proteostasis and dopaminergic neuron degeneration have been identified.The potential pathophysiological and therapeutic significance of these molecular targets to neurodegeneration in PD is highlighted.Zhi Dong Zhou Thevapriya Selvaratnam Ji Chao Tristan Lee Yin Xia Chao Eng-King Tan 2019Translational Neurodegeneration2019,8,1:2
