1Stem cell-based therapy for human diseases显示文摘Recent advancements in stem cell technology open a new door for patients suffering from diseases and disorders that have yet to be treated.Stem cell-based therapy,including human pluripotent stem cells(hPsCs)and multipotent mesenchymal stem cells(MSCs),has recently emerged as a key player in regenerative medicine.hPSCs are defined as self-renewable cell types conferring the ability to differentiate into various cellular phenotypes of the human body,including three germ layers.MsCs are multipotent progenitor cells possessing self-renewal ability(limited in vitro)and differentiation potential into mesenchymal lineages,according to the International Society for Cell and Gene Therapy(ISCT).This review provides an update on recent clinical applications using either hPSCs or MSCs derived from bone marrow(BM),adipose tissue(AT),or the umbilical cord(UC)for the treatment of human diseases,including neurological disorders,pulmonary dysfunctions,metabolic/endocrine-related diseases,reproductive disorders,skin burns,and cardiovascular conditions.Moreover,we discuss our own clinical trial experiences on targeted therapies using MsCs in a clinical setting,and we propose and discuss the MSC tissue origin concept and how MSC origin may contribute to the role of MSCs in downstream applications,with the ultimate objective of facilitating translational research in regenerative medicine into clinical applications.The mechanisms discussed here support the proposed hypothesis that BM-MSCs are potentially good candidates for brain and spinal cord injury treatment,AT-MSCs are potentially good candidates for reproductive disorder treatment and skin regeneration,and UC-MsCs are potentially good candidates for pulmonary disease and acute respiratory distress svndrometreatment.Duc M.Hoang Phuong T.Pham Trung Q.Bach Anh T.L.Ngo Quyen T.Nguyen Trang T.K.Phan Giang H.Nguyen Phuong T.T.Le Van T.Hoang Nicholas R.Forsyth Michael Heke Liem Thanh Nguyen 2022Signal Transduction and Targeted Therapy2022,7,9:7

3Reinforcement Learning Based Controller Synthesis for Flexible Aircraft Wings显示文摘Aeroelastic study of flight vehicles has been a subject of great interest and research in the last several years. Aileron reversal and flutter related problems are due in part to the elasticity of a typical airplane. Structural dynamics of an aircraft wing due to its aeroelastic nature are characterized by partial differential equations. Controller design for these systems is very complex as compared to lumped parameter systems defined by ordinary differential equations. In this paper, a stabilizing statefeedback controller design approach is presented for the heave dynamics of a wing-fuselage model. In this study, a continuous actuator in the spatial domain is assumed. A control methodology is developed by combining the technique of “proper orthogonal decomposition” and approximate dynamic programming. The proper orthogonal decomposition technique is used to obtain a low-order nonlinear lumped parameter model of the infinite dimensional system. Then a near optimal controller is designed using the single-network-adaptive-critic technique. Furthermore,to add robustness to the nominal single-network-adaptive-critic controller against matched uncertainties, an identifier based adaptive controller is proposed. 4Recommendations for the use of the acetaminophen hepatotoxicity model for mechanistic studies and how to avoid common pitfalls显示文摘Acetaminophen(APAP)is a widely used analgesic and antipyretic drug,which is safe at therapeutic doses but can cause severe liver injury and even liver failure after overdoses.The mouse model of APAP hepatotoxicity recapitulates closely the human pathophysiology.As a result,this clinically relevant model is frequently used to study mechanisms of drug-induced liver injury and even more so to test potential therapeutic interventions.However,the complexity of the model requires a thorough understanding of the pathophysiology to obtain valid results and mechanistic information that is translatable to the clinic.However,many studies using this model are flawed,which jeopardizes the scientific and clinical relevance.The purpose of this review is to provide a framework of the model where mechanistically sound and clinically relevant data can be obtained.The discussion provides insight into the injury mechanisms and how to study it including the critical roles of drug metabolism,mitochondrial dysfunction,necrotic cell death,autophagy and the sterile inflammatory response.In addition,the most frequently made mistakes when using this model are discussed.Thus,considering these recommendations when studying APAP hepatotoxicity will facilitate the discovery of more clinically relevant interventions.Hartmut Jaeschke Olamide B.Adelusi Jephte Y.Akakpo Nga T.Nguyen Giselle Sanchez-Guerrero David S.Umbaugh Wen-Xing Ding Anup Ramachandran 2021Acta Pharmaceutica Sinica B2021,11,12:3
5Bi-Objective Optimal Control Modification Adaptive Control for Systems with Input Uncertainty显示文摘This paper presents a new model-reference adaptive control method based on a bi-objective optimal control formulation for systems with input uncertainty. A parallel predictor model is constructed to relate the predictor error to the estimation error of the control effectiveness matrix. In this work, we develop an optimal control modification adaptive control approach that seeks to minimize a bi-objective linear quadratic cost function of both the tracking error norm and the predictor error norm simultaneously. The resulting adaptive laws for the parametric uncertainty and control effectiveness uncertainty are dependent on both the tracking error and the predictor error, while the adaptive laws for the feedback gain and command feedforward gain are only dependent on the tracking error. The optimal control modification term provides robustness to the adaptive laws naturally from the optimal control framework. 6Qualification of Soybean Responses to Flooding Stress Using UAV-Based Imagery and Deep Learning显示文摘Soybean is sensitive to flooding stress that may result in poor seed quality and significant yield reduction.Soybean production under flooding could be sustained by developing flood-tolerant cultivars through breeding programs.Conventionally,soybean tolerance to flooding in field conditions is evaluated by visually rating the shoot injury/damage due to flooding stress,which is labor-intensive and subjective to human error.Recent developments of field high-throughput phenotyping technology have shown great potential in measuring crop traits and detecting crop responses to abiotic and biotic stresses.The goal of this study was to investigate the potential in estimating flood-induced soybean injuries using UAV-based image features collected at different flight heights.The flooding injury score(FIS)of 724 soybean breeding plots was taken visually by breeders when soybean showed obvious injury symptoms.Aerial images were taken on the same day using a five-band multispectral and an infrared(IR)thermal camera at 20,50,and 80 m above ground.Five image features,i.e.,canopy temperature,normalized difference vegetation index,canopy area,width,and length,were extracted from the images at three flight heights.A deep learning model was used to classify the soybean breeding plots to five FIS ratings based on the extracted image features.Results show that the image features were significantly different at three flight heights.The best classification performance was obtained by the model developed using image features at 20 m with 0.9 for the five-level FIS.The results indicate that the proposed method is very promising in estimating FIS for soybean breeding.Jing Zhou Huawei Mou Jianfeng Zhou Md Liakat Ali Heng Ye Pengyin Chen Henry T.Nguyen 2021Plant Phenomics2021,3,1:2
7Use of light-emitting diode(LED)lamps in combination with metal halide(MH)lamps reduce fuel consumption in the Vietnamese purse seine fishery显示文摘The use of high-power lights during night-time purse seining is common in Vietnam.Typically,metal halide(MH)lamps are used in the commercial fishery to attract fish,however these lights require more energy,have a shorter lifespan,and lower chromatic performance than light emitting diode(LED)lamps.This study examined catch efficiency and fuel consumption when using LED lamps in combination with reduced numbers of MH lamps(10.24 kW),compared to conventional lighting(28.6 kW),used during purse seining off the coast of Ninh Thuan province,Vietnam.The economic performance associated with using LED lamps in this fishery was also assessed.We found no significant differences in catch rates between the different light treatments,however fuel consumption was significantly reduced.Fuel consumption per nightly trip using LED with MH lamps was 70.8 l(11.1 l/h)compared to114 l(17.45 l/hr)using MH lamps alone,an estimated 37.9%reduction in fuel consumption.An investment in LED lamps by a fishing enterprise will require additional initial costs,however our analysis revealed the financial break-event point can be reached after approximately 101 nightly trips when the fuel price is at the 2015 level of USD$0.74 per l.Fishing enterprises can increase their profitability,and reduce CO2 emissions,by using LED lamps in the Vietnamese purse seine fishery.Khanh Q.Nguyen Phu D.Tran Luong T.Nguyen Phuong V.To Corey J.Morris 2021Aquaculture and Fisheries2021,6,4:2
8Mitochondrial damage and biogenesis in acetaminophen-induced liver injury显示文摘Liver injury and acute liver failure caused by acetaminophen(APAP)overdose is the clinically most important drug toxicity in Western countries.Mechanistic investigations have revealed a central role of mitochondria in the pathophysiology.Excess formation of the reactive metabolite N-acetyl-p-benzoquinone imine(NAPQI)after an overdose leads to hepatic glutathione depletion,mitochondrial protein adducts formation and an initial oxidant stress,which triggers the activation of mitogen activated protein(MAP)kinase cascade ultimately leading to c-jun N-terminal kinase(JNK)phosphorylation.Phospho-JNK translocates to the mitochondria and amplifies the oxidative and nitrosative stress eventually causing the mitochondrial membrane permeability transition pore opening and cessation of adenosine triphosphate(ATP)synthesis.In addition,mitochondrial matrix swelling ruptures the outer membrane and releases endonucleases,which cause nuclear deoxyribonucleic acid(DNA)fragmentation.Together,the nuclear DNA damage and the extensive mitochondrial dysfunction result in necrotic cell death.However,the procell death signaling events are counteracted by adaptive responses such as autophagy and mitochondrial biogenesis.The improved mechanistic insight into the pathophysiology leads to better understanding of the mechanisms of action of the existing antidote N-acetylcysteine and justifies the clinical testing of novel therapeutics such as 4-methylpyrazole and calmangafodipir.Hartmut Jaeschke Luqi Duan Nga T.Nguyen Anup Ramachandran 2019Liver Research2019,3,3:2

10Temperature-sensitive polymeric nanogels encapsulating withβ-cyclodextrin and ICG complex for high-resolution deep-tissue ultrasound-switchable fluorescence imaging显示文摘One of the thorny problems currently impeding the applications of the fluorescence imaging technique is the poor spatial resolution in deep tissue.Ultrasound-switchable fluorescence(USF)imaging is a novel imaging tool that has recently been explored to possibly surmount the above-mentioned bottleneck.Herein,αβ-cyclodextrin/indocyanine green(ICG)complex-encapsulated poly(N-isopropylacrylamide)(PNIPAM)nanogel was synthesized and studied for ex vivo/in vivo deep tissue/high-resolution near infrared USF(NIR-USF)imaging.To be specific,our results revealed that the average diameter of the as-prepared nanogels was significantly decreased to-32 nm from-335 nm compared to the reported ICG-PNIPAM nanoparticles.Additionally,the excitation/emission characteristics of the ICG itself in present nanogels were almost completely retained,and the resultant nanogel exhibited high physiological stability and positive biocompatibility.In particular,the signal-to-noise ratio of the USF image for the PNIPAM/P-cyclodextrin/ICG nanogel(33.01±2.42 dB)was prominently higher than that of the ICG-PNIPAM nanoparticles(18.73±0.33 dB)in 1.5-cm-thick chicken breast tissues.The NIR-USF imaging in 3.5-cm-thick chicken breast tissues was achieved using this new probe.The e x v iv o NIR-USF imaging of the mouse liver was also successfully obtained.Animal experiments showed that the present nanogels were able to be effectively accumulated into U87 tumor-bearing mice via enhanced permeability and retention effects,and the high-resolution NIR-USF imaging of in v ivo tumor was efficiently acquired.The metabolism and in vivo biodistribution of the nanogels were evaluated.Overall,the results suggest that the current nanogel is a highly promising NIR-USF probe for deep tissue and high-resolution USF imaging.Ruilin Liu Tingfeng Yao Yang Liu Shuai Yu Liqin Ren Yi Hong Kytai T.Nguyen Baohong Yuan 2020Nano Research2020,13,4:1

14甘蔗生物能系统的温室气体减排潜能显示文摘与化石能源相比,生物能源系统一个最主要的优势是其温室气体(GHG)排放量较低。通过发电和生产乙醇,把蔗糖厂转变成生物能源厂,可以减缓气候变暖的趋势。本论文基于泰国制糖企业的现状,提出有利于减少温室气体排放量的有效措施:提高利用甘蔗废弃物(如蔗渣和蔗梢)的发电效率;同时在酒精厂将生产酒精后的废液转化成能量,也有利于温室气体的减排;最佳减排方案是用富余的蔗糖生产乙醇,与泰国目前制糖工业相比可大大减少CO2的排放量,每年减少CO2总量可达一千四百万吨。但假定甘蔗种植地来源于草地的转换,即使应用了以上措施,在土地转变过程中产生的碳债务也需要4.5—7年的时间才能被偿还。(碳债务是指由可耕地和森林转变为生产生物燃料而引起的间接排放,比起因生物燃料所得到的利益为负值,所以被形象地称为碳债务)。Thu Lan T.Nguyen Shabbir H.Gheewala Masayuki Sagisaka 王淋靓 2011广西蔗糖2011,,3:1

18Supertransport of excitons in atomically thin organic semiconductors at the 2D quantum limit显示文摘Long-range and fast transport of coherent excitons is important for the development of high-speed excitonic circuits and quantum computing applications.However,most of these coherent excitons have only been observed in some low-dimensional semiconductors when coupled with cavities,as there are large inhomogeneous broadening and dephasing effects on the transport of excitons in their native states in materials.Here,by confining coherent excitons at the 2D quantum limit,we first observed molecular aggregation-enabled‘supertransport’of excitons in atomically thin two-dimensional(2D)organic semiconductors between coherent states,with a measured high effective exciton diffusion coefficient of ~346.9 cm^(2)/s at room temperature.This value is one to several orders of magnitude higher than the values reported for other organic molecular aggregates and low-dimensional inorganic materials.Without coupling to any optical cavities,the monolayer pentacene sample,a very clean 2D quantum system(~1.2 nm thick)with high crystallinity(J-type aggregation)and minimal interfacial states,showed superradiant emission from Frenkel excitons,which was experimentally confirmed by the temperature-dependent photoluminescence(PL)emission,highly enhanced radiative decay rate,significantly narrowed PL peak width and strongly directional in-plane emission.The coherence in monolayer pentacene samples was observed to be delocalised over~135 molecules,which is significantly larger than the values(a few molecules)observed for other organic thin films.In addition,the supertransport of excitons in monolayer pentacene samples showed highly anisotropic behaviour.Our results pave the way for the development of future high-speed excitonic circuits,fast OLEDs,and other optoelectronic devices.Ankur Sharma Linglong Zhang Jonathan O.Tollerud Miheng Dong Yi Zhu Robert Halbich Tobias Vogl Kun Liang Hieu T.Nguyen Fan Wang Shilpa Sanwlani Stuart K.Earl Daniel Macdonald Ping Koy Lam Jeffrey A.Davis Yuerui Lu 2020Light(Science & Applications)2020,9,1:1
