1MicroRNA in vivo precipitation identifies miR-151-3p as a computational unpredictable miRNA to target Stat3 and inhibits innate IL-6 production显示文摘MicroRNAs(miRNAs)function as important regulators in the immune response and inflammation.Several approaches have been reported to computationally predict miRNAs and their potential targets.However,there are still many miRNA–target interactions that are unpredictable by using the current computational algorithms.We established a miRNA in vivo precipitation method(miRIP)to identify unpredictable miRNAs with definite targets in these cells.Because Stat3 is a well-known transcription factor involved in innate immunity and inflammation,we utilized the miRIP method to identify miRNAs that bind Stat3 mRNA in macrophages.Among the captured miRNAs,miR-151-3p was confirmed to interact with Stat3 mRNA 3′-UTR and downregulate the Stat3 protein levels.LPS stimulation decreased miR-151-3p expression,thereby increasing IL-6 production.Therefore,we found that miR-151-3p inhibited LPS-induced IL-6 production by targeting Stat3.These data further confirmed miRIP as an efficient method to identify unpredictable miRNAs and explore miRNAs-mediated regulation in innate immunity and inflammation.Xiang Liu Xiaoping Su Sheng Xu Huamin Wang Dan Han Jiangxue Li Mingyan Huang Xuetao Cao 2018Cellular & Molecular Immunology2018,15,2:8
2Phenformin alone or combined with gefitinib inhibits bladder cancer via AMPK and EGFR pathways显示文摘Background:In previous studies,we have shown that the combination of metformin and gefitinib inhibits the growth of bladder cancer cells.Here we examined whether the metformin analogue phenformin,either used alone or in combination with gefitinib,could inhibit growth of bladder cancer cells.Methods:The growth-inhibitory effects of phenformin and gefitinib were tested in one murine and two human bladder cancer cell lines using MTT and clonogenic assays.Effects on cell migration were assessed in a wound healing assay.Synergistic action between the two drugs was assessed using CompuSyn software.The potential involvement of AMPK and EGFR pathways in the effects of phenformin and gefitinib was explored using Western blotting.Results:In MTT and clonogenic assays,phenformin was>10-fold more potent than metformin in inhibiting bladder cancer cell growth.Phenformin also potently inhibited cell migration in wound healing assays,and promoted apop-tosis.AMPK signaling was activated;EGFR signaling was inhibited.Phenformin was synergistic with gefitinib,with the combination of drugs showing much stronger anticancer activity and apoptotic activation than phenformin alone.Conclusions:Phenformin shows potential as an effective drug against bladder cancer,either alone or in combination with gefitinib.Yanjun Huang Sichun Zhou Caimei He Jun Deng Ting Tao Qiongli Su Kwame Oteng Darko Mei Peng Xiaoping Yang 2018Cancer Communications2018,38,1:5
3Depositional characteristics and petroleum geological significance of wetland显示文摘Wetland is a new type of sedimentary facies proposed for the first time in this paper.It is a type of sedimentary environment,transitional between land facies and water(sea or lake) facies.In this paper,wetland is redefined as 'a type of sedimentary environments whose ground surface is flat and wet throughout the year,which may be covered with very shallow water(less than 2 m deep),and is covered with lush plant growth'.Wetland is reclassified into two types,swamp wetland(swamp in brief) and wet plain wetland(wet plain in brief).Swamp deposits are coal,while wet plain deposits are dark colored mudstones or silty mudstones rich in plant fossils and carbon debris.The deposits of wet plain are different from those of lake and floodplain in the abundance of plant fossils,color,resistivity well logging curve,and other sedimentary characteristics.In boreholes,resistivity logging can be used to distinguish between wet plain mudstones and lake mudstones.Understanding the sedimentary characteristics of wet plain wetland can help to identify wet plain deposits which were formerly classified as floodplain or lake deposits.This will help to reconstruct the palaeogeography and to understand the history of basin and climate evolution accurately,and is especially important in evaluation of the hydrocarbon generating potential of a basin.With the Jurassic Sangonghe Formation of the Yanqi Basin as an example,the identification characteristics of wetland are described.Jin Zhenkui Gu Junfeng Su Nina Wang Zhaofeng Huang Xiaoping 2009Petroleum Science2009,6,4:5
4Numerical Investigation on Porous Media Quenching Behaviors of Premixed Deflagrating Flame using RANS/LES Model显示文摘To understand the mechanism of premixed flame quenching by porous media,a zonal hybrid RANS/LES model was employed,in which the LES flow solver was used to resolve the large turbulent structures within the non-porous region,while RANS was applied to porous media zone.The predicted results were compared with previous experimental data.And it was evident that the premixed flame propagation rates and structure as well as quenching behaviors were reproduced by this numerical approach with a better accuracy.Due to the inherently higher heat transfer coefficients between the solid and gas phases in porous media,the gas phase temperature has been decreased rapidly.However,upstream obstacles can cause the flame propagating faster and thus reduce the axial gas temperature gradients,resulting in the invalidity of the operation of premixed flame quenching.By comparison with the case without upstream obstacle,the values of reaction rate attained in the case with three pairs of obstacles are higher,which makes a positive impact on the flame passing through the porous medium.In addition,the porous media with greater pore density has an excellent flame quenching property mainly owing to both the larger volumetric heat transfer and higher quenching temperature.WEN Xiaoping SU Tengfei LIU Zhigang XIE Maozhao WANG Fahui LIU Zhichao 2019Journal of Thermal Science2019,28,4:4
5Selective catalytic hydrogenation of naphthalene to tetralin over a NiMo/Al2O3 catalyst显示文摘The selective catalytic hydrogenation of naphthalene to high-value tetralin was systematically investigated.A series of Al2O3 catalysts containing different active metals(Co,Mo,Ni,W)were prepared by incipient wetness impregnation.The effects of different active metals forms(oxidation,reduction,sulfuration)and reaction conditions on naphthalene hydrogenation were investigated and the catalysts were characterized by XRD,XPS,BET,NH3-TPD and SEM.Especially,Ni-Mo/Al2O3 was first used in this reactive system.The results show that the oxidative4%Ni O-20%MoO3/Al2O3 is the best catalyst for the preparation of tetralin.The conversion of naphthalene and the selectivity of tetralin can reach 95.62%and 99.75%respectively at 200℃,8 h and 6 MPa.Compared with reduced and sulfureted 4%Ni O-20%MoO3/Al2O3 catalysts,oxidative 4%Ni O-20%MoO3/Al2O3 has a well dispersed and uniform monolayer of the active metals,larger pore volume and size,and larger total acidity.NiO-MoO3/Al2O3 has a synergistic effect between NiO activity and MoO3 selectivity.Xiaoping Su Pu An Junwen Gao RuchengWang Yujuan Zhang Xi Li Yangkun Zhao Yongqi Liu Xiaoxun Ma Ming Sun 2020Chinese Journal of Chemical Engineering2020,28,10:4
6Abundant Production of Reactive Water Radical Cations under Ambient Conditions显示文摘Water radical cations,the crucial intermediates in many aqueous reactions and biochemical processes,are difficult to investigate experimentally due to their short lifetime andlowabundance.Herein,ahomemade device based on energy-tunable discharge was employed to deposit suitable amounts of energy to atmospheric pressure pure water vapor for abundant production of water radical cations,stabilized as(H_(2)O)n^(+·)(n=2-5)with a maximal abundance of≥8.3×10^(6) cps for(H_(2)O)_(2)^(+·),characterized by mass spectrometry(MS).The abundance of water radical cations was optimized by adjusting the experimental parameters such as the discharge voltage(2.5 kV),temperature of the MS inlet(140℃),carrier gas flow(20mL/min),and the distance between the discharge tip and MS inlet(12 mm).The ambient formation of water radical cationswas further confirmedby thehigh reactivity of as-prepared water radical cations,which reacted with benzene,ethyl acetate,and dimethyl disulfide instantly,showing rich chemistry with ionic and radical characteristics.Moreover,the computations usingCCSD(T)//MP2 method and density functional theory confirmed that the O-O single-electron bound dimer(B),as well as the hydronium hydroxyl radical complex(A),accounted for the unusual chemistry of the water radical cations,providing a facile approach to access the high reactivity ofwater radical cations under the ambient conditions.Meng Wang Xiao-Fei Gao Rui Su Peng He Yuan-Yuan Cheng Ke Li Dongbo Mi Xiaoping Zhang Xinglei Zhang Huanwen Chen R.Graham Cooks 2022CCS Chemistry2022,4,4:3
7Quantifying 3D cell-matrix interactions during mitosis and the effect of anticancer drugs on the interactions显示文摘The mechanical force between cells and the extracellular microenvironment is crucial to many physiological processes such as cancer metastasis and stem cell differentiation. Mitosis plays an essential role in all these processes and thus an in-depth understanding of forces during mitosis gains insight into disease diagnosis and disease treatment. Here, we develop a traction force microscope method based on monolayer fluorescent beads for measuring the weak traction force (tens of Pa) of mitotic cells in three dimensions. We quantify traction forces of human ovarian granulosa (KGN) cells exerted on the extracellular matrix throughout the entire cell cycle in three dimensions. Our measurements reveal how forces vary during the cell cycle, especially during cell division. Furthermore, we study the effect of paclitaxel (PTX) and nocodazole (NDZ) on mitotic KGN cells through the measurement of traction forces. Our results show that mitotic cells with high concentrations of PTX exert a larger force than those with high concentrations of NDZ, which proved to be caused by changes in the structure and number of microtubules. These findings reveal the key functions of microtubule in generating traction forces during cell mitosis and explain how dividing cells regulate themselves in response to anti-mitosis drugs. 8Influence of gas flow on thermal field and stress during growth of sapphire single crystal using Kyropoulos method显示文摘The professional modeling software package CrysVUn was employed to study the process of a large sapphire single crystal growth using Kyropoulos method. The influence of gas pressure on thermal field, solid-liquid interface shape, gas velocity field and von Mises stress were studied for the first time. It is found that the root of the seed melt when gas pressure equals to one atmosphere or more than one atmosphere, especially during the seeding period, this result is consistent with the experimental observation, and this paper presents three ways to solve this problem. The temperature gradient and stress decreases significantly as the gas pressure increases. The convexity of the solid-liquid interface slightly increases when the gas pressure increases. 9Serological Investigation of Laboratory-Confirmed and Suspected Ebola Virus Disease Patients During the Late Phase of the Ebola Outbreak in Sierra Leone显示文摘This study aimed to investigate the serological characteristics of Ebola virus(EBOV) infection during the late phase of the Ebola outbreak in Sierra Leone. In total, 877 blood samples from 694 suspected Ebola virus disease(EVD) cases assessed from March to December 2015, were analyzed via real-time reverse transcription polymerase chain reaction(RT-PCR) for viral RNA and enzyme-linked immunosorbent assay(ELISA) and Luminex to detect antibodies against EBOV. Viral load and EBOV-specific IgM/IgG titers displayed a declining trend during March to December 2015. Viral RNA load decreased rapidly at earlier stages after disease onset, while EBOV-specific IgM and IgG still persisted in 58.1%(18/31) and 93.5%(29/31) of the confirmed EVD patients and in 3.8%(25/663) and 17.8%(118/663) of the RNA-negative suspected patients in the later phase, respectively. 10Intramedullary schwannoma of the upper cervical spinal cord:a case study of identification in pathologic autopsy显示文摘Intramedullary schwannoma of the upper cervical spinal cord is rarely reported in forensic medicine.We herein report a case involving a patient who died of compression from an intramedullary schwannoma in the upper cervical spinal cord.A 30-year-old man initially presented with a five-day history of pain in the left chest that progressed to weakening in the left arm.Although the patient was treated with analgesic poultices,he developed inspiratory dyspnoea and died while working the next day without having undergone any medical imaging examination or surgical treatment.Anatomical and histopathological examinations revealed an intramedullary schwannoma in the left cervical spinal cord(C_(3)-C_(5))underneath the spinal nerve root.The cause of death might have been asphyxia secondary to the tumour,which interfered with the nerve function in the respiratory muscles.This finding suggests that an autopsy is essential for pathologists and medicolegists to comprehensively undertake their due obligation to obtain“the first evidence”,especially when there is a lack of directly related evidence.As part of the central nervous system,the spinal cord could be systematically included in a routine pathological autopsy in some cases.Xianxian Li Guangtao Xu Ruibing Su Junyao Lv Xiaoping Lai Xiaojun Yu 2017Forensic Sciences Research2017,2,1:2

14苯乙双胍单药或联合吉非替尼通过AMPK和EGFR通路抑制膀胱癌显示文摘背景与目的在前期研究中,我们发现二甲双胍和吉非替尼联合使用可抑制膀胱癌细胞生长。本文旨在研究二甲双胍类似物——苯乙双胍单药或联合吉非替尼能否抑制膀胱癌细胞生长。方法在1种鼠源和2种人源膀胱癌细胞系中,采用MTT和克隆形成实验检测苯乙双胍和吉非替尼对细胞生长的抑制作用。采用伤口愈合实验检测细胞迁移情况。使用CompuSyn软件评估2种药物之间的协同作用。采用蛋白免疫印迹实验分析苯乙双胍和吉非替尼对AMPK和EGFR通路的潜在作用。结果 MTT和克隆形成实验结果显示,苯乙双胍抑制膀胱癌细胞生长的作用是二甲双胍的10倍以上。伤口愈合实验结果显示,苯乙双胍可有效抑制细胞迁移,并促进凋亡。AMPK信号被激活,EGFR信号被抑制。苯乙双胍与吉非替尼可协同作用,二者联合使用比苯乙双胍单药表现出更强的抑癌活性和促凋亡活性。结论苯乙双胍单药或联合吉非替尼均可能成为治疗膀胱癌的有效药物。Yanjun Huang Sichun Zhou Caimei He Jun Deng Ting Tao Qiongli Su Kwame Oteng Darko Mei Peng Xiaoping Yang 2019癌症2019,38,5:1

19Study on dislocation multiplication in the head of Germanium single crystal显示文摘The high strength, radiation hardness and cost-effectiveness make Germanium the substrate of choice for high-efficiency multi-junction solar cells for space applications. Numerical modeling and large-scale simulation are important and indispensable tools in the analysis and development of crystal growth process. In this study, germanium single crystals with low dislocation density were produced by Czochralski method by applying the necking technique. Chemical etching pits method was used to measure the dislocation density, and a professional modeling software CrysVUn was used to obtain the thermal-stress distribution. The results show that the thermal-stress of the sample with diameter of 15 mm is nearly equal to that of other samples, so the thermal-stress does not influence the dislocation multiplication. 20A low-energy ion spectrometer with half-space entrance for three-axis stabilized spacecraft显示文摘A low-energy ion spectrometer(LEIS) for use aboard three-axis stabilized spacecraft has been developed to measure ion energy per charge distribution in three-dimensional space with good energy-, angular-and temporal-resolutions. For the standard top-hat electrostatic analyzer used widely in space plasma detection, three-axis stabilized spacecraft makes it difficult to obtain complete coverage of all possible ion arrival directions. We have designed angular scanning deflectors supplementing to a cylindrically symmetric top-hat electrostatic analyzer to provide a half-space field of view as 360°×90°(–45°–+45°), and fabricated the LEIS flight model for detecting magnetospheric ions in geosynchronous orbit. The performance of this payload has been evaluated in detail by a series of simulation and environmental tests, and the payload has also been calibrated through laboratory experiments using a low-energy ion source.
