| 1 | Human bronchial epithelial cell injuries induced by fine particulate matter from sandstorm and non-sandstorm periods:Association with particle constituents显示文摘Epidemiological studies have demonstrated the exacerbation of respiratory diseases following sandstorm-derived particulate matter(PM) exposure.The presence of anthropogenic and biological agents on the sandstorm PM and the escalation of PM < 2.5 μm(PM2.5)pollution in China have led to serious concerns regarding the health effects of PM2.5during Asian sandstorms.We investigated how changes in PM2.5composition,as the weather transitioned towards a sandstorm,affected human airway epithelial cells.Six PM2.5samples covering two sandstorm events and their respective background and transition periods were collected in Baotou,an industrial city near the Gobi Desert in China.PM samples from all three periods had mild cytotoxicity in human bronchial epithelial cell line BEAS-2B,which was positively correlated with the contents of polycyclic aromatic hydrocarbons and several metals.All PM samples potently increased the release of interleukin-6(IL-6) and interleukin-8(IL-8).Endotoxin in all samples contributed significantly to the IL-6 response,with only a minor effect on IL-8.Cr was positively correlated with both IL-6 and IL-8 release,while Si was only associated with the increase of IL-6.Our study suggests that local agricultural and industrial surroundings in addition to the sandstorm play important roles in the respiratory effects of sandstorm-derived PM. | Bin Wang Ning Li Furong Deng Nicholas Buglak George Park Shu Su Aiguo Ren Guofeng Shen Shu Tao Xinbiao Guo | 2016 | Journal of Environmental Sciences2016,28,9: | 9 |

| 3 | Effect of cooling strategies on performance and mechanism of helical milling of CFRP/Ti-6Al-4V stacks显示文摘Hole-making for Carbon Fiber Reinforced Plastics(CFRP)/Ti-6Al-4V stacks is crucial for the assembling strength of aircraft structure parts.This work carried out experimental work for helical milling(HM)of the stacks with sustainable cooling/lubrication(dry,MQL and cryogenic)conditions.Cutting forces and temperatures at the CFRP layer,Ti-6Al-4V layer and the interface of stacks were obtained by a developed measuring system.The temperatures in CFRP machining at cryogenic condition varied from-167℃to-94℃,which were much lower than those at dry and MQL conditions.The maximum temperature near the interface of stacks for the ninth hole was higher than 240℃due to heat conduction from Ti-6Al-4V layer.The hole quality,hole diameter and tool wear mechanism at different cooling/lubrication conditions were presented and discussed.MQL condition generated mainly extrusion fracture for the fibers,due to the reduced friction effect compared with dry condition.MQL was helpful to reduce the feed mark at the hole surface of Ti-6Al-4V alloy.The flank wear of cutting edge at MQL condition was better than those at dry and cryogenic conditions.Cryogenic cooling contributed to better CFRP surface with smaller delamination and hole entrance damage due to the increased resin strength and fiber brittleness.The damage near the entrance of CFRP were analyzed by the contact state of cutting edges and fibers.Additionally,hole diameters near the exit of CFPR layer were larger than other test positions.This work provided feasible processes for improving hole quality and tool life in hole-making of CFRP/Ti-6Al-4V stacks. | Jiaying GE Guang CHEN Yongxiang SU Yunhe ZOU Chengzu REN Xuda QIN Guofeng WANG | 2022 | Chinese Journal of Aeronautics2022,35,2: | 4 |
| 4 | Development of Human in vitro Brain-blood Barrier Model from Induced Pluripotent Stem Cell-derived Endothelial Cells to Predict the in vivo Permeability of Drugs显示文摘An in vitro blood-brain barrier(BBB) model is critical for enabling rapid screening of the BBB permeability of the drugs targeting on the central nervous system.Though many models have been developed, their reproducibility and renewability remain a challenge. Furthermore, drug transport data from many of the models do not correlate well with the data for in vivo BBB drug transport.Induced-pluripotent stem cell(i PSC) technology provides reproducible cell resources for in vitro BBB modeling.Here, we generated a human in vitro BBB model by differentiating the human i PSC(hi PSC) line GM25256 into brain endothelial-type cells. The model displayed BBB characteristics including tight junction proteins(ZO-1,claudin-5, and occludin) and endothelial markers(von Willebrand factor and Ulex), as well as high transendothelial electrical resistance(TEER)(1560 X.cm2±230 X.cm2) and c-GTPase activity. Co-culture with primary rat astrocytes significantly increased the TEER of the model(2970 X.cm2 to 4185 X.cm2). RNAseq analysis confirmed the expression of key BBB-related genes in the hi PSC-derived endothelial cells in comparison with primary human brain microvascular endothelial cells,including P-glycoprotein(Pgp) and breast cancer resistant protein(BCRP). Drug transport assays for nine CNS compounds showed that the permeability of non-Pgp/BCRP and Pgp/BCRP substrates across the model was strongly correlated with rodent in situ brain perfusion data for these compounds(R2= 0.982 and R2= 0.9973,respectively), demonstrating the functionality of the drug transporters in the model. | 6 | Effect of a single weak lithological structure on the height of a collapsing roof in a deep soft rock roadway显示文摘Besides the cross sections of roadways and the tendency and obliquity of roadway axes, the major controlling factors affecting the height of a collapsing roof include the weak lithological structure of surrounding rocks. This thesis analyzes the effect of two single and weak lithological structures of both sides and the roof on the height of a collapsing roof in a deep soft rock road-way. Using the two-dimensional UDEC3.1 software, a numerical simulation was carried out on the models of weak lithological structures of both sides of a roadway and of two weak lithological structures of roof of different depths. We reconstruct the overall processes from a break-away layer, bending, subsidence and the cracking of a collapsing roof. We also illustrate the distribution characteristics of displacement fields in the surrounding rock after the roof collapse in a deep soft rock roadway. The results of our numerical simulations indicate that the form of a roof collapse is side-expanding when the roadway is a weak structure at both sides.
