2Increased function of the TRPV1 channel in small sensory neurons after local inflammation or in vitro exposure to the pro-inflammatory cytokine GRO/KC显示文摘Objective Inflammation at the level of the sensory dorsal root ganglia (DRGs) leads to robust mechanical pain behavior and the local inflammation has direct excitatory effects on sensory neurons including small, primarily nociceptive, neurons. These neurons express the transient receptor potential vanilloid-1 (TRPV1) channel, which integrates multiple signals of pain and inflammation. The aim of this study was to characterize the regulation of the TRPV1 channel by local DRG inflammation and by growth-related oncogene (GRO/KC, systemic name: CXCL1), a cytokine known to be upregulated in inflamed DRGs. Methods Activation of the TRPV1 receptor with capsaicin was studied with patch clamp methods in acutely isolated small-diameter rat sensory neurons in primary culture. In vivo, behavioral effects of TRPV1 and GRO/KC were examined by paw injections. Results Neurons isolated from lumbar DRGs 3 days after local inflammation showed enhanced TRPV1 function: tachyphylaxis (the decline in response to repeated applications of capsaicin) was significantly reduced. A similar effect on tachyphylaxis was observed in neurons pre-treated for 4 h in vitro with GRO/KC. This effect was blocked by H-89, a protein kinase A inhibitor. Consistent with the in vitro results, in vivo behavioral responses to paw injection of capsaicin were enhanced and prolonged by pre-injecting the paw with GRO/KC 4 h before the capsaicin injection. GRO/KC paw injections alone did not elicit pain behaviors. Conclusion Function of the TRPV1 channel is enhanced by DRG inflammation and these effects are preserved in vitro during short-term culture. The effects (decreased tachyphylaxis) are mimicked by incubation with GRO/KC, which has previously been found to be strongly upregulated in this and other pain models.Fei Dong Yi-Ru Du Wenrui Xie Judith A. 3Recent evidence for activity-dependent initiation of sympathetic sprouting and neuropathic pain显示文摘Traumatic injury or inflammatory irritation of the peripheral nervous system often leads to persistent pathophysiological pain states.It has been well-documented that,after peripheral nerve injury or inflammation,functional and anatomical alterations sweep over the entire peripheral nervous system including the peripheral nerve endings,the injured or inflamed afferent fibers,the dorsal root ganglion(DRG),and the central afferent terminals in the spinal cord.Among all the changes,ectopic discharge or spontaneous activity of primary sensory neurons is of great clinical interest,as such discharges doubtless contribute to the develop-ment of pathological pain states such as neuropathic pain.Two key sources of abnormal spontaneous activity have been identified following peripheral nerve injury:the injured afferent fibers(neuroma) leading to the DRG,and the DRG somata.The purpose of this review is to provide a global account of the abnormal spontaneous activity in various animal models of pain.Particular attention is focused on the consequence of peripheral nerve injury and localized inflammation.Further,mechanisms involved in the generation of spontaneous activity are also reviewed;evidence of spontaneous activity in contributing to abnormal sympathetic sprouting in the axotomized DRG and to the initiation of neuropathic pain based on new findings from our research group are discussed.An improved understanding of the causes of spontaneous activity and the origins of neuropathic pain should facilitate the development of novel strategies for effective treatment of pathological pain.Jun-Ming ZHANG Judith A. 5肝移植治疗原发性胆汁性肝硬化的疗效分析显示文摘目的 观察原发性胆汁性肝硬化(PBC)患者和移植肝的实际生存时间,肝移植后死亡的原因及肝移植后的复发率。 方法 根据QLTS的数据库中的随访资料,回顾性分析52例共接受54次肝移植的PBC患者。52例PBC肝移植患者中,平均年龄(5 3.2±6.7)岁,平均随访时间是(5 5.4±11.3)个月。分析术前的临床特征、移植后生存情况和死亡原因,采用欧洲模式计算未接受肝移植患者和接受肝移植的患者的生存率。 结果 PBC肝移植患者的1、5、1 0年的实际生存率分别为8 8.4%、80.1%和76.9%,未接受肝移植患者实际生存率分别为80.9%、6 5.4%和19.8%。PBC患者移植后的生存率比未接受肝移植的患者(欧洲模型)预期生存率高。有6例患者经肝活检后证实PBC复发,平均复发时间(34.1±10.2)个月。肝移植术后死亡的原因是多器官功能衰竭、腹腔内出血、肾功能衰竭、败血症及心血管疾病。 结论 肝移植可提高P B C患者的生存率,延长其生存时间。许崇恩 Stephen V.Lynch Glenda A.Balderson Jonathan Fawcett Russell W Strong Shinn Yeong 2004中华肝脏病杂志2004,12,9:6
6Inflammatory Changes in Paravertebral Sympathetic Ganglia in Two Rat Pain Models显示文摘Injury to peripheral nerves can lead to neuropathic pain, along with well-studied effects on sensory neurons, including hyperexcitability, abnormal spontaneous activity, and neuroinflammation in the sensory ganglia. Neuropathic pain can be enhanced by sympathetic activity. Peripheral nerve injury may also damage sympathetic axons or expose them to an inflammatory environment. In this study, we examined the lumbar sympathetic ganglion responses to two rat pain models: ligation of the L5 spinal nerve, and local inflammation of the L5 dorsal root ganglion(DRG), which does not involve axotomy.Both models resulted in neuroinflammatory changes in the sympathetic ganglia, as indicated by macrophage responses, satellite glia activation, and increased numbers of T cells, along with very modest increases in sympathetic neuron excitability(but not spontaneous activity) measured in ex vivo recordings. The spinal nerve ligation model generally caused larger responses than DRG inflammation.Plasticity of the sympathetic system should be recognized in studies of sympathetic effects on pain.Ai-Ling Li Jing-Dong Zhang Wenrui Xie Judith A. 13Evaluating the Environmental Health Effect of Bamboo-Derived Volatile Organic Compounds through Analysis the Metabolic Indices of the Disorder Animal Model显示文摘Objective To identify the bamboo VOCs(volatile organic compounds) effect on animal physiological indices, which associated with human health. Methods GC/MS was used to analyze the volatile organic compounds from Moso bamboo(Phyllostachys heterocyla cv. pubescens). The effect of VOCs on environmental health was evaluated by analyzing the metabolic indices of the type 2 diabetic mouse model. Results Spectra of VOC generated by GC/MS were blasted against an in-house MS library confirming the identification of 33 major components that were manually validated. The relative constituent compounds as a percentage of total VOCs determined were alcohols(34.63%), followed by ether(22.02%), aldehyde(15.84%), ketone(11.47%), ester(4.98%), terpenoid(4.38%), and acids(3.83%). Further experimentation established that the metabolic incidence of the disease can be improved if treated with vanillin, leaf alcohol, β-ionone and methyl salicylate. The effects of these VOCs on type 2 diabetes were evident in the blood lipid and blood glucose levels. Conclusion Our model suggests that VOCs can potentially control the metabolic indices in type 2 diabetes mice.
