| 1 | Single-cell transcriptome analysis of the novel coronavirus (SARS-CoV-2) associated gene ACE2 expression in normal and non-obstructive azoospermia (NOA) human male testes显示文摘Being infected by SARS-CoV-2 may cause damage to multiple organs in patients, such as the lung, liver and heart. Angiotensin-converting enzyme 2(ACE2), reported as a SARS-CoV-2 receptor, is also expressed in human male testes. This suggests a potential risk in human male reproductive system. However, the characteristics of ACE2-positive cells and the expression of other SARS-CoV-2 process-related genes are still worthy of further investigation. Here, we performed singlecell RNA seq(scRNA-seq) analysis on 853 male embryo primordial germ cells(PGCs) and 2,854 normal testis cells to assess the effects of the SARS-CoV-2 virus on the male reproductive system from embryonic stage to adulthood. We also collected and constructed the scRNA-seq library on 228 Sertoli cells from three non-obstructive azoospermia(NOA) patients to assess the effects at disease state. We found that ACE2 expressing cells existed in almost all testis cell types and Sertoli cells had highest expression level and positive cells ratio. Moreover, ACE2 was also expressed in human male PGCs. In adulthood, the level of ACE2 expression decreased with the increase of age. We also found that ACE2 positive cells had high expressions of stress response and immune activation-related genes. Interestingly, some potential SARS-CoV-2 process-related genes such as TMPRSS2, BSG, CTSL and CTSB had different expression patterns in the same cell type. Furthermore, ACE2 expression level in NOA donors’ Sertoli cells was significantly decreased. | 2 | Understanding the improved performance of sulfur-doped interconnected carbon microspheres for Na-ion storage显示文摘As one of the low-cost energy storage systems,Na-ion batteries(NIBs)have received tremendous attention.However,the performance of current anode materials still cannot meet the requirements of NIBs.In our work,we obtain sulfur-doped interconnected carbon microspheres(S-CSs)via a simple hydrothermal method and subsequent sulfurizing treatment.Our S-CSs exhibit an ultrahigh reversible capacity of 520 mAh g^(-1) at 100 mA g^(-1) after 50 cycles and an excellent rate capability of 257 mAh g^(-1),even at a high current density of 2 A g^(-1).The density functional theory calculations demonstrate that sulfur doping in carbon favors the adsorption of Na atom during the sodiation process,which is accountable for the performance enhancement.Furthermore,we also utilize operando Raman spectroscopy to analyze the electrochemical reaction of our S-CSs,which further highlights the sulfur doping in improving Na-ion storage performance. | Xinran Yuan Siming Chen Jinliang Li Junpeng Xie Genghua Yan Botian Liu Xibo Li Rui Li Likun Pan Wenjie Mai | 2021 | Carbon Energy2021,3,4: | 3 |

| 4 | Correlation between the physical parameters and the electrochemical performance of a silicon anode in lithium-ion batteries显示文摘Lithium-ion battery anode used as silicon particles were obtained from different major suppliers,and they were characterized by different spectroscopic techniques and evaluated by electrochemical experiments.Correlations between the key physical parameters and electrochemical properties of the silicon particles were investigated.Silicon particle size,surface oxygen content,-OH content and physical appearance are found to strongly influence the electrochemical properties of the Si anode.The particle size of 100 nm has great promise for the practical application of Si nanoparticles in the lithium-ion battery industry.An inverse correlation between the oxygen content and the reversible capacity or first coulombic efficiency was obtained.The-OH content by surface treatment contributes to enhanced cycling stability by the improved affinity between the Si particle and the water-soluble binder.Spherical Si particles perform better compared to irregular particles,and agglomeration dramatically decreases the cycling stability of the Si anode.Among the investigated Si particles,the sample that exhibited a reversible capacity of more than 2500 mAh g^(-1),a first coulombic efficiency of 89.26%and an excellent cycling stability,has great potential for use in the battery industry. | Guobin Zhu Yan Wang Siming Yang Qunting Qu Honghe Zheng | 2019 | Journal of Materiomics2019,5,2: | 2 |
| 5 | A loss-of-function mutant allele of a glycosyl hydrolase gene has been co-opted for seed weight control during soybean domestication显示文摘The resultant DNA from loss-of-function mutation can be recruited in biological evolution and development.Here,we present such a rare and potential case of“to gain by loss”as a neomorphic mutation during soybean domestication for increasing seed weight.Using a population derived from a chromosome segment substitution line of Glycine max(SN14)and Glycine soja(ZYD06),a quantitative trait locus(QTL)of 100-seed weight(q HSW)was mapped on chromosome 11,corresponding to a truncatedβ-1,3-glucosidase(βGlu)gene.The novel gene hsw results from a 14-bp deletion,causing a frameshift mutation and a premature stop codon in theβGlu.In contrast to HSW,the hsw completely lostβGlu activity and function but acquired a novel function to promote cell expansion,thus increasing seed weight.Overexpressing hsw instead of HSW produced large soybean seeds,and surprisingly,truncating hsw via gene editing further increased the seed size.We further found that the core 21-aa peptide of hsw and its variants acted as a promoter of seed size.Transcriptomic variation in these transgenic soybean lines substantiated the integration hsw into cell and seed size control.Moreover,the hsw allele underwent selection and expansion during soybean domestication and improvement.Our work cloned a likely domesticated QTL controlling soybean seed weight,revealed a novel genetic variation and mechanism in soybean domestication,and provided new insight into crop domestication and breeding,and plant evolution. | Siming Wei Bin Yong Hongwei Jiang Zhenghong An Yan Wang Bingbing Li Ce Yang Weiwei Zhu Qingshan Chen Chaoying He | 2023 | Journal of Integrative Plant Biology2023,65,11: | 1 |
| 6 | Features Associated with Recurrence of Idiopathic Orbital Inflammatory Pseudotumor显示文摘Purpose: To explore factors which lead to recurrence of idiopathic orbital inflam-matory pseudotumor (IOIP). Methods: Idiopathic orbital inflammatory pseudotumor in 209 cases between Jan 1, 1978 and Dec 31, 1999 in our hospital was evaluated retrospectively. The comparison of clinical and pathological parameters between patients with at least one episode of recurrence and those with no recurrence at all was performed and analyzed using logistic regression method. Results: Follow-up results (with a mean follow-up time of 3.4 years, ranging from 0.5 year to 21.0 years) showed that the recurrence rate of IOIP was 41%. Sex and proptosis were associated with the recurrence of IOIP. Male gender was more likely to relapse than female counterparts, with the male being 52% and female being 25%. The severer the proptosis is, the higher the rate of recurrence. Among the clinical subtypes of IOIP, the rate of recurrence (17%) in cases with dacryoadinitis was the lowest, followed by anterior local orbital mass (44%), posterior orbital mass (54%), myositis (75%) and diffuse subtype (100%). However, the clinical subtypes did not show significant relationship with the recurrence of IOIP. | 8 | Capacity of human umbilical cord-derived mesenchymal stem cells to differentiate into sweat gland-like cells:a preclinical study显示文摘Human umbilical cord-derived mesenchymal stem cells(hUC-MSCs)possess various advantageous properties,including self-renewal,extended proliferation potential,multi-lineage differentiation potential and capacity for differentiating into sweat gland-like cells in certain conditions.However,little is known about the effect of clinical-grade culture conditions on these properties and on the differentiative potential of hUC-MSCs.In this study,we sought to investigate the properties of hUC-MSCs expanded with animal serum free culture media(ASFCM)in order to determine their potential for differentiation into sweat gland-like cells.We found that primary cultures of hUC-MSCs could be established with ASFCM.Moreover,cells cultured in ASFCM showed vigorous proliferation comparable to those of cells grown in classical culture conditions containing fetal bovine serum(FBS).Morphology of hUC-MSCs cultured in ASFCM was comparable to those of cells grown under classical culture conditions,and hUC-MSCs grown in both of the two culture conditions tested showed the typical antigen profile of MSCs—positive for CD29,CD44,CD90,and CD105,and negative for CD34 and CD45,as expected.Chromosomal aberration assay revealed that the cells were stable after long-term culture under both culture conditions.Like normal cultured MSCs,hUC-MSCs induced under ASFCM conditions exhibited expression of the same markers(CEA,CK14 and CK19)and developmental genes(EDA and EDAR)that are characteristic of normal sweat gland cells.Taken together,our findings indicate that the classical culture medium used to differentiate hUC-MSCs into sweat gland-like cells can be replaced safely by ASFCM for clinical purposes. | Siming Yang Kui Ma Changjiang Feng Yan Wu Yao Wang Sha Huang Xiaobing Fu | 2013 | Frontiers of Medicine2013,7,3: | 0 |
