1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2Infection with novel coronavirus(SARS-CoV-2)causes pneumonia in Rhesus macaques显示文摘The 2019 novel coronavirus(SARS-CoV-2)outbreak is a major challenge for public health.SARS-CoV-2 infection in human has a broad clinical spectrum ranging from mild to severe cases,with a mortality rate of^6.4%worldwide(based on World Health Organization daily situation report).However,the dynamics of viral infection,replication and shedding are poorly understood.Here,we show that Rhesus macaques are susceptible to the infection by SARS-CoV-2.After intratracheal inoculation,the first peak of viral RNA was observed in oropharyngeal swabs one day post infection(1 d.p.i.),mainly from the input of the inoculation,while the second peak occurred at 5 d.p.i.,which reflected on-site replication in the respiratory tract.Histopathological observation shows that SARS-CoV-2 infection can cause interstitial pneumonia in animals,characterized by hyperemia and edema,and infiltration of monocytes and lymphocytes in alveoli.We also identified SARS-CoV-2 RNA in respiratory tract tissues,including trachea,bronchus and lung;and viruses were also re-isolated from oropharyngeal swabs,bronchus and lung,respectively.Furthermore,we demonstrated that neutralizing antibodies generated from the primary infection could protect the Rhesus macaques from a second-round challenge by SARS-CoV-2.The non-human primate model that we established here provides a valuable platform to study SARS-CoV-2 pathogenesis and to evaluate candidate vaccines and therapeutics.Chao Shan Yan-Feng Yao Xing-Lou Yang Yi-Wu Zhou Ge Gao Yun Peng Lian Yang Xue Hu Jin Xiong Ren-Di Jiang Hua-Jun Zhang Xiao-Xiao Gao Cheng Peng Juan Min Ying Chen Hao-Rui Si Jia Wu Peng Zhou Yan-Yi Wang Hong-Ping Wei Wei Pang Zheng-Fei Hu Long-Bao Lv Yong-Tang Zheng Zheng-Li Shi Zhi-Ming Yuan 2020Cell Research2020,30,8:19
3Palaeogeomorphology and its control on the development of sequence stratigraphy and depositional systems of the Early Silurian in the Tarim Basin显示文摘The Silurian in the Tarim Basin was deposited on the basement deformed by the Caledonian tectonic movements at the end of the Late Ordovician. The development and distribution of sedimentary sequences of the Early Silurian have been clearly controlled by the palaeogeomorphology of the Late Ordovician. Based on unconformity characteristics and distribution of erosion, several zones can be differentiated including a high uplifted erosion zone, a transitional slope zone and a depression zone. The central and west Tabei Uplift zones show high angular unconformity and intense erosion. The Tarim Basin in the late Ordovician shows characteristics of higher in the west, lower in the east while higher in the south, lower in the north. The Early Silurian mainly developed transgressive and highstand systems tracts on the whole, while the lowstand systems tract only developed partly below the slope break. The palaeogeomorphology controlled the clastic source supply and deposit distribution. Braided delta system and tidal flat–estuary system were deposited. The duration of uplifting of the Tazhong paleouplift was longer than that of the Tabei paleo-uplift, and deposition was later. This led to the lower and middle members of the Kepingtage Formation missing in that area. As a large-scale transgression occurred during the deposition period of the upper member of the Kepingtage Formation, sediment from the west of the basin was transported and deposited by tides and waves, forming tidal–marine debris systems above the uplift. Proximal alluvial fan and fan delta coarse clastic deposits developed in proximal uplift zone in the east and southeast of the basin, and braided delta put forward to the transitional zone between the edge of uplift and the sea. Large-scale tidal channel, sub-distributary channel and mouth bar of the delta front can form favorable reservoirs, and they are primary targets for oil and gas exploration. 4Mir-30d increases intracellular survival of Helicobacter pylori through inhibition of autophagy pathway显示文摘AIM: To determine if mir-30 d inhibits the autophagy response to Helicobacter pylori(H. pylori) invasion and increases H. pylori intracellular survival.METHODS: The expression of mir-30 d was detected by quantitative polymerase chain reaction(PCR), and autophagy level was examined by transmission electron microscopy, western blot, and GFP-LC3 puncta assay in human AGS cells and GES-1 cells. Luciferase reporter assay was applied to confirm the specificity of mir-30 d regulation on the expression of several core molecules involved in autophagy pathway. The expression of multiple core proteins were analyzed at both the m RNA and protein level, and the intracellular survival of H. pylori after different treatments was detected by gentamicin protection assay.RESULTS: Autophagy level was increased in AGS and GES-1 cells in response to H. pylori infection, which was accompanied by upregulation of mir-30 d expression(P < 0.05, vs no H. pylori infection). In the two gastric epithelial cell lines, mimic mir-30 d was found to repress the autophagy process, whereas mir-30 d inhibitor increased autophagy responseto H. pylori invasion. mir-30 d mimic decreased the luciferase activity of wild type reporter plasmids carrying the 3′ untranslated region(UTR) of all five tested genes(ATG2B, ATG5, ATG12, BECN1, and BNIP3L), whereas it had no effect on the mutant reporter plasmids. These five genes are core genes of autophagy pathway, and their expression was reduced significantly after mir-30 d mimic transfection(P < 0.05, vs control cells without mir-30 d mimic treatment). 5Anlotinib has good efficacy and low toxicity: a phase Ⅱ study of anlotinib in pre-treated HER-2 negative metastatic breast cancer显示文摘Objective:Anlotinib is a novel tyrosine kinase inhibitor blocking angiogenesis.This study was performed to assess the efficacy and safety of anlotinib in patients with metastatic breast cancer.Methods:Patients with HER2-negative breast cancer,who were pre-treated with anthracycline or taxanes in a neoadjuvant,adjuvant,or metastatic setting,and had treatment failure after at least one prior chemotherapy regimen in the metastatic setting were enrolled.Anlotinib was administered at 12 mg daily for 14 days in a 21-day cycle until disease progression or unacceptable toxicity occurred.Simultaneously,5–10 m L of venous blood was collected to perform circulating tumor DNA(ct DNA)testing every 2 treatment cycles.The primary endpoint was the objective response rate(ORR).Secondary endpoints included the disease control rate(DCR),progression-free survival(PFS),overall survival,safety,and biomarkers.Results:Twenty-six eligible patients were enrolled,with a median age of 56(30–75)years.The median follow-up time was 10.5 months.The ORR was 15.4%,the DCR was 80.8%,and the median PFS was 5.22 months(95%confidence interval 2.86–6.24).Fourteen(53.8%)patients survived for more than 10 months.The changes in the detectable ct DNA variant allele frequency were consistent with the tumor response.The most common treatment-related adverse events were hypertension(57.7%),thyroidstimulating hormone elevation(34.6%),and hand-foot syndrome(23.1%).Conclusion:Anlotinib showed objective efficacy with tolerable toxicity in heavily pre-treated,metastatic HER2-negative breast cancer.The dynamic changes in the ct DNA variant allele fraction may be predictive of the tumor response.Nanlin Hu Yiran Si Jian Yue Tingting Sun Xue Wang Zhuqing Jia Songlin Gao Qiao Li Yang Shao Jiayu Wang Yang Luo Fei Ma Binghe Xu Peng Yuan 2021Cancer Biology & Medicine2021,18,3:11
6Paris chinensis dioscin induces G2/M cell cycle arrest and apoptosis in human gastric cancer SGC-7901 cells显示文摘AIM:To investigate the anti-tumor effects of Paris chinensis dioscin(PCD)and mechanisms regarding cell cycle regulation and apoptosis in human gastric cancer SGC-7901 cells.METHODS:Cell viability was analyzed by the 3-(4,5-di-methylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide assay.Cell apoptosis was evaluated by flow cytometry and laser scanning confocal microscope(LSCM)using Annexin-V/propidium iodide(PI)staining,and the cell cycle was evaluated using PI staining with flow cytom-etry.Intracellular calcium ions were detected under fluorescence microscope.The expression of cell cycle and apoptosis-related proteins cyclin B1,CDK1,cytochrome C and caspase-3 was measured by immunohistochemical staining.RESULTS:PCD had an anti-proliferation effect on human gastric cancer SGC-7901 cells in a dose-and time-de-pendent manner.After treatment of SGC-7901 cells with PCD,apoptosis appeared in SGC-7901 cells.Morpho-logical changes typical of apoptosis were also observed with LSCM by Annexin V/PI staining,and the cell number of the G0/G1 phase was decreased,while the number of cells in the G2/M phase was increased.Cell cycle-related proteins,such as cyclin B1 and CDK1,were all down-regulated,but caspase-3 and cytochrome C were up-regulated.Moreover,intracellular calcium accumulation occurred in PCD-treated cells.CONCLUSION:G2/M phase arrest and apoptosis induced by PCD are associated with the inhibition of CDK-activating kinase activity and the activation of Ca2+-related mitochondrion pathway in SGC-7901 cells.Lin-Lin Gao Fu-Rong Li Peng Jiao Ming-Feng Yang Xiao-Jun Zhou Yan-Hong Si Wen-Jian Jiang Ting-Ting Zheng 2011World Journal of Gastroenterology2011,17,39:11
7Study of HIV-1 Drug Resistance in Patients Receiving Free Antiretroviral Therapy in China显示文摘To investigate the prevalence of drug-resistance mutations,resistance to antiretroviral drugs,and the subsequent virological response to therapy in treatment-naive and antiretroviral-treated patients infected with HIV/AIDS in Henan,China,a total of 431 plasma samples were collected in Queshan county between 2003 and 2004,from patients undergoing the antiretroviral regimen Zidovudine + Didanosine + Nevirapine(Azt+Ddi+Nvp).Personal information was collected by face to face interview.Viral load and genotypic drug resistance were tested.Drug resistance mutation data were obtained by analyzing patient-derived sequences through the HIVdb Program(http://gffzz38c50ed9119344cfhupqx6pc9qon66w69.ffgz.tsg.suse.edu.cn).Overall,38.5% of treatment-naive patients had undetectable plasma viral load(VL),the rate significantly increased to 61.9% in 0 to 6 months treatment patients(mean 3 months)(P<0.005)but again significantly decrease to 38.6% in 6 to 12 months treatment patients(mean 9 months)(P<0.001)and 40.0% in patients receiving more than 12 months treatment(mean 16 months)(P<0.005).The prevalence of drug resistance in patients who had a detectable VL and available sequences were 7.0%,48.6%,70.8%,72.3% in treatment-na?ve,0 to 6 months treatment,6 to 12 months treatment,and treatment for greater than 12 months patients,respectively.No mutation associated with resistance to Protease inhibitor(PI)was detected in this study.Nucleoside RT inhibitor(NRTI)mutations always emerged after non-nucleoside RT inhibitor(NNRTI)mutations,and were only found in patients treated for more than 6 months,with a frequency less than 5%,with the exception of mutation T215Y(12.8%,6/47)which occurred in patients treated for more than 12 months.NNRTI mutations emerged quickly after therapy begun,and increased significantly in patients treated for more than 6 months(P<0.005),and the most frequent mutations were K103N,V106A,Y181C,G190A.There had been optimal viral suppression in patients undergoing treatment for less than 6 months in Queshan,Henan.The drug resistance strains were highly prevalent in antiretroviral-treated patients,and increased with the continuation of therapy,with many patients encountering virological failure after 6 months therapy.Xin-ping LIP Hui XING Zhe WANG Xue-feng SI Lian-en WANG Hua CHENG Wei-guo CUI Shu-lin JIANG Ling-jie LIAO Hai-wei ZHOU Jiang-hong HUANG Hong PENG Peng-fei MA Yi-ming SHAO 2007中国病毒学2007,22,3:9
8Using the concept of ecological groundwater level to evaluate shallow groundwater resources in hyperarid desert regions显示文摘This paper,based on the analysis and calculation of the groundwater resources in an arid region from 1980 to 2001,put forward the concept of ecological groundwater level threshold for either salinity control or the determination of ecological warning.The surveys suggest that soil moisture and soil salinity are the most important environmental factors in determining the distribution and changes in vegetation.The groundwater level threshold of ecological warning can be determined by using a network of groundwater depth observation sites that monitor the environmental moisture gradient as reflected by plant physiological characteristics.According to long-term field observations within the Ejin oases,the groundwater level threshold for salinity control varied between 0.5 m and 1.5 m,and the ecological warning threshold varied between 3.5 m and 4.0 m.The quantity of groundwater resources(renewable water resources,ecological water resources,and exploitable water resources) in arid areas can be calculated from regional groundwater level information,without localized hydrogeological data.The concept of groundwater level threshold of ecological warning was established according to water development and water resources supply,and available groundwater resources were calculated.The concept not only enriches and broadens the content of groundwater studies,but also helps in predicting the prospects for water resources development.Qi FENG JiaZhong PENG JianGuo LI HaiYang XI JianHua SI 2012Journal of Arid Land2012,4,4:9
9Review on the NO removal from flue gas by oxidation methods显示文摘Due to the increasingly strict emission standards of NOx on various industries,many traditional flue gas treatment methods have been gradually improved.Except for selective catalytic reduction(SCR)and selective non-catalytic reduction(SNCR)methods to remove NOx from flue gas,theoxidation method is paying more attention to NOx removal now because of the potential to simultaneously remove multiple pollutants from flue gas.This paper summarizes the efficiency,reaction conditions,effect factors,and reaction mechanism of NO oxidation from the aspects of liquid-phase oxidation,gas-phase oxidation,plasma technology,and catalytic oxidation.The effects of free radicals and active components of catalysts on NO oxidation and the combination of various oxidation methods are discussed in detail.The advantages and disadvantages of different oxidation methods are summarized,and the suggestions for future research on NO oxidation are put forward at the end.The review on the NO removal by oxidation methods can provide new ideas for future studies on the NO removal from flue gas.Meng Si Boxiong Shen George Adwek Lifu Xiong Lijun Liu Peng Yuan Hongpei Gao Cai Liang Qihai Guo 2021Journal of Environmental Sciences2021,33,3:8

11Projected impacts of climate change on protected birds and nature reserves in China显示文摘Knowledge about climate change impacts on species distribution at national scale is critical to biodiversity conservation and design of management programs.Although China is a biodiversity hot spot in the world,potential influence of climate change on Chinese protected birds is rarely studied. Here, we assess the impact of climate change on 108 protected bird species and nature reserves using species distribution modeling at a relatively fine spatial resolution(1 km) for the first time. We found that a large proportion of protected species would have potential suitable habitat shrink and northward range shift by 77–90 km in response to projected future climate change in 2080. Southeastern China would suffer from losing climate suitability, whereas the climate conditions in Qinghai–Tibet Plateau and northeastern China were projected to become suitable for more protected species. Onaverage, each protected area in China would experience a decline of suitable climate for 3–4 species by 2080. Climate change will modify which species each protected area will be suitable for. Our results showed that the risk of extinction for Chinese protected birds would be high, even in the moderate climate change scenario. 13A semi-physical sediment yield model for estimation of suspended sediment in loess region显示文摘Sediment yield is a complex function of many environmental factors including climate,hydrology,vegetation,basin topography,soil types,and land cover.We present a new semi-physical watershed sediment yield model for the estimation of suspended sediment in loess region.This model is composed by three modules in slope,gully,and stream phases.For slope sediment yield,a balance equation is established based on the concept of hydraulic erosion capacity and soil erosion resistance capacity.According to the statistical analysis of watershed characteristics,we use an exponential curve to approximately describe the spatial variability of watershed soil erosion resistance capacity.In gully phase,the relationship between gully sediment concentration and flow velocity is established based on the Bagnold' stream power function.In the stream phase,we assume a linear dependence of the sediment volume in the reach on the weighted sediment input and output.The proposed sediment yield model is operated in conjunction with a conceptual hydrologic model,and is tested over 16 regions including testing grounds,and small,medium and large watersheds in the loess plateau region in the mid-reach of Yellow River.Our results indicate that the model is reasonable in structure and is able to provide a good simulation of sediment generation and transportation processes at both flood event scale and inter-annual time scale.The proposed model is generally applicable to the watersheds with soil texture similar to that of the loess plateau region in the Yellow River basin in China.Wei Si Weimin Bao Peng Jiang Liping Zhao Simin Qu 2017International Journal of Sediment Research2017,32,1:7

15An integrated ore prospecting model for the Nyasirori gold deposit in Tanzania显示文摘The Nyasirori gold deposit,located in the middle-western end of the Musoma-Mara Archean greenstone belt in Tanzania,is a tectonic altered rock type gold deposit controlled by shear tectonic zone.This work conducted high-precision ground magnetic measurements to delineate fault structures and favorable prospecting targets,utilized induced polarization(IP)intermediate gradient to roughly determine the distribution and extension of the tectonic altered zone and gold ore(mineralized)bodies,and further carried out IP sounding and magnetotelluric sounding to locate the tectonic altered zone and gold ore(mineralized)bodies.The anomalous gradient belt of the combination of positive and negative micromagnetic measurements reflects the detail of shallow surface tectonic alteration zone and gold mineralization body.Micromagnetic profile anomalies indicate the spatial location and occurrence of concealed tectonic alteration zone and gold(mineralized)ore bodies.Soil geochemical measurements indicate that the ore-forming element Au correlates well with As and Sb,and As and Sb anomalies have a good indication to gold orebodies.Based on the multi-source geological-geophysical-geochemical information of the Nyasirori gold deposit,this work established an integrated prospecting model and proposed a set of geophysical and geochemical methods for optimizing prospecting targets.Yang-sen Yuan Shui-ping Li Jun Peng Jian-tao Si Hua Cheng Jin Sun Jian-zheng Wei Jiang-bo Shao 2019China Geology2019,2,4:6
16Agglutinin isolated from Arisema heterophyllum Blume induces apoptosis and autophagy in A549 cells through inhibiting PI3K/Akt pathway and inducing ER stress显示文摘Arisaema heterophyllum Blume is one of the three medicinal plants known as traditional Chinese medicine Rhizoma Arisaematis(RA). RA has been popularly used to treat patients with convulsions, inflammation, and cancer for a long time. However, the underlying mechanisms for RA effects are still unclear. The present study was designed to determine the cytotoxicity of agglutinin isolated from Arisema heterophyllum Blume(AHA) and explore the possible mechanisms in human non-small-cell lung cancer A549 cells. AHA with purity up to 95% was isolated and purified from Arisaema heterophyllum Blume using hydrophobic interaction chromatography. AHA dose-dependently inhibited the proliferation of A549 cells and induced G_1 phase cell cycle arrest. AHA induced apoptosis by up-regulating pro-apoptotic Bax, decreasing anti-apoptotic Bcl-2, and activating caspase-9 and caspase-3. In A549 cells treated with AHA, the PI3K/Akt pathway was inhibited. Furthermore, AHA induced increase in the levels of ER stress markers such as phosphorylated eukaryotic initiation factor 2α(p-eIF2α), C/EBP-homologous protein(CHOP), inositol-requiring enzyme 1α(IRE1α), and phosphorylated c-Jun NH_2-terminal kinase(p-JNK). AHA also induced autophagy in A549 cells. Staining of acidic vesicular organelles(AVOs) and increase in the levels of LC3II and ATG7 were observed in AHA-treated cells. These findings suggested that AHA might be one of the active components with anti-cancer effects in Arisaema heterophyllum Blume. 17Modulation of expression of P16 and Her2 in rat breast tissues of mammary hyperplasia model by external use of Rupifang Extract显示文摘OBJECTIVE:To observe the effect of Rupifang Extract in external use on expression of proto-oncogenes her2 and tumor suppression genes p16 in rat breast tissues of mammary hyperplasia model.To explore the mechanisms of Rupifang Extract in external use for preventing and treating mammary hyperplasia.METHODS:Thirty virginal female Wistar rats were randomized into 5 groups,6 in each,A:blank control group;B:model group;C:the low dose group of Rupifang;D:the middle dose group of Rupifang;and E:The high dose group of Rupifang.The mammary hyperplasia rat models were produced by injecting estradiol benzoate and progesterone and irritating by tail nipping.Drug intervention was also launched during the model formation.After 30 days,the expression of her2 and p16 in breast tissues of rats in each group were detected by the SP immunohistochemical method.RESULTS:Compared with Blank control group,the expression of her2 in breast tissues in Model group was higher,and the expression of p16 was lower(P<0.05 or P<0.01).After intervention with Rupifang Extract,compared with Model group,the expression of her2 in breast tissues in Rupifang groups was lower,and the expression of p16 higher(P<0.05 or P<0.01).CONCLUSION:The mechanisms of Rupifang Extract in external application for preventing and treating mammary hyperplasia may be reducing the expression of proto-oncogenes her2 and increasing the expression of tumor suppression genes p16.Guijuan Zhang Dehui Li Hui Guo Rui Liao Bizhu Tan Suyi Zhang Yubin Liu Min Ma Xiaoting Zeng Zhuolong Peng Si Huang Yinghui Zheng Yi Ma 2012Journal of Traditional Chinese Medicine2012,32,4:6
18Surveys in Areas of High Risk of Iodine Deficiency and Iodine Excess in China, 2012-2014: Current Status and Examination of the Relationship between Urinary Iodine Concentration and Goiter Prevalence in Children Aged 8-10 Years显示文摘Objective We aimed to evaluate goiter prevalence and iodine nutritional status in areas with high levels of water iodine;to monitor the prevalence of iodine deficiency disorders(IDD)in areas at high risk of IDD;and to compare the prevalence of goiter and urine iodine(UI)concentrations between children living in the two areas.Methods Based on surveillance from 2012-2014,we analyzed the concentration of UI and prevalence of goiter in 8-10-year-old children from 12 high-risk IDD provinces,and from 8 provinces and municipalities with excessive water iodine.We calculated goiter prevalence for each UI level according to World Health Organization(WHO)standards and constructed predictive prevalence curves.Results The goiter prevalence and median UI of children from areas with high water iodine were not optimal,being above the WHO standards(5%and 100-199μg/L,respectively),whereas those in high-risk areas fell within the standard.UI and goiter prevalence exhibited a U-shaped relationship in high-risk endemic areas and a parabolic relationship in areas of iodine excess.Conclusion Iodine surplus in high-iodine areas leads to high goiter prevalence and UI.However,in high-risk areas,UI was optimal and goiter prevalence met the national criteria for IDD elimination.CUI Si Lu LIU Peng SU Xiao Hui LIU Shou Jun 2017Biomedical and Environmental Sciences2017,30,2:6
19Improving the quantification of waterfowl migration with remote sensing and bird tracking显示文摘Accurately quantifying waterfowl migration patterns is pertinent to monitor ecosystem health and control bird-borne infectious diseases. In this review, we summarize the current understanding of the environmental mechanisms that drive waterfowl migration and then investigate the effect of intra- and inter-annual change in food supply and temperature(e.g., climate change) on their migration patterns. Recent advances in remote sensing and animal tracking techniques make it possible to monitor these environmental factors over a wide range of scales and record bird movements in detail. The synergy of these techniques will facilitate substantial progress in our understanding of the environmental drivers of bird migration. We identify prospects for future studies to test existing hypotheses and develop models integrating up-todate knowledge, high-resolution remote sensing data and high-accuracy bird tracking data.
