Construction of robust coupling interface between MoS2 and nitrogen doped graphene for high performance sodium ion batteries显示文摘As a layered inorganic material,MoS2 has recently attracted intensive attention as anode for sodium ion batteries(SIBs).However, this anode is plagued with low electronic conductivity, serious volume expansion and sluggish kinetics, resulting in capacity fading and poor rate performance.Herein,we develop an interface engineering strategy to substantially enhance the sodium storage performance of MoS2 by incorporating layered MoS2 into three dimensional N-doped graphene scaffold. The strong coupling-interface between MoS2 and N-doped graphene scaffold can not only stabilize the MoS2 structure during sodium insertion/extraction processes, but also provide plenty of anchor sites for additional surface sodium storage. The 3D MoS2@N-doped graphene composite as anode for SIBs performs an outstanding specific capacity of 667.3 mA h g^-1 at 0.2 A g^-1,a prolonged stability with a capacity retention of 94.4%after 140cycles and excellent rate capability of 445 mA h g^-1 even at a high rate of 10 A g^-1.We combined experiment and theoretical simulation to further disclose the interaction between MoS2 and N-doped graphene, adsorption and diffusion of sodium on the composite and the corresponding sodium storage mechanism. This study opens a new door to develop high performance SIBs by introducing the interface engineering technique. | Yun Qiao Jiawei Wu Xiaoguang Cheng Yudong Pang Zhansheng Lu Xiangdong Lou Qingling Li Jin Zhao Shuting Yang Yang Liu | 2020 | Journal of Energy Chemistry2020,29,9: | 3
Small interfering RNA delivery to the neurons near the amyloid plaques for improved treatment of Alzheimer’s disease显示文摘Gene therapy represents a promising treatment for the Alzheimer’s disease(AD). However, gene delivery specific to brain lesions through systemic administration remains big challenge. In our previous work, we have developed an siRNA nanocomplex able to be specifically delivered to the amyloid plaques through surface modification with both CGN peptide for the blood–brain barrier(BBB)penetration and QSH peptide for β-amyloid binding. But, whether the as-designed nanocomplex could indeed improve the gene accumulation in the impaired neuron cells and ameliorate AD-associated symptoms remains further study. Herein, we prepared the nanocomplexes with an siRNA against β-site amyloid precursor protein-cleaving enzyme 1(BACE1), the rate-limiting enzyme of Aβ production, as the therapeutic siRNA of AD. The nanocomplexes exhibited high distribution in the Aβ deposits-enriched hippocampus, especially in the neurons near the amyloid plaques after intravenous administration. In APP/PS1 transgenic mice, the nanocomplexes down-regulated BACE1 in both mRNA and protein levels,as well as Aβ and amyloid plaques to the level of wild-type mice. Moreover, the nanocomplexes significantly increased the level of synaptophysin and rescued memory loss of the AD transgenic mice without hematological or histological toxicity. Taken together, this work presented direct evidences that the design of precise gene delivery to the AD lesions markedly improves the therapeutic outcome. | Qian Guo Xiaoyao Zheng Peng Yang Xiaoying Pang Kang Qian Pengzhen Wang Shuting Xu Dongyu Sheng Liuchang Wang Jinxu Cao Wei Lu Qizhi Zhang Xinguo Jiang | 2019 | Acta Pharmaceutica Sinica B2019,9,3: | 2
Exosomes secreted from cardiomyocytes suppress the sensitivity of tumor ferroptosis in ischemic heart failure显示文摘Heart failure(HF)patients in general have a higher risk of developing cancer.Several animal studies have indicated that cardiac remodeling and HF remarkably accelerate tumor progression, highlighting a cause-and-effect relationship between these two disease entities.Targeting ferroptosis,a prevailing form of non-apoptotic cell death, has been considered a promising therapeutic strategy for human cancers.Exosomes critically contribute to proximal and distant organ-organ communications and play crucial roles in regulating diseases in a paracrine manner. However, whether exosomes control the sensitivity of cancer to ferroptosis via regulating the cardiomyocyte-tumor cell crosstalk in ischemic HF has not yet been explored. Here,we demonstrate that myocardial infarction(MI)decreased the sensitivity of cancer cells to the canonical ferroptosis activator erastin or imidazole ketone erastin in a mouse model of xenograft tumor.Post-MI plasma exosomes potently blunted the sensitivity of tumor cells to ferroptosis inducers both in vitro in mouse Lewis lung carcinoma cell line LLC and osteosarcoma cell line K7M2 and in vivo with xenograft tumorigenesis model. The expression of miR-22-3p in cardiomyocytes and plasma-exosomes was significantly upregulated in the failing hearts of mice with chronic MI and of HF patients as well.Incubation of tumor cells with the exosomes isolated from post-MI mouse plasma or overexpression of miR-22-3p alone abrogated erastin-induced ferroptotic cell death in vitro.Cardiomyocyte-enriched miR-22-3p was packaged in exosomes and transferred into tumor cells.Inhibition of cardiomyocyte-specific miR-22-3p by AAV9 sponge increased the sensitivity of cancer cells to ferroptosis.ACSL4,a pro-ferroptotic gene, was experimentally established as a target of miR-22-3p in tumor cells.Taken together, our findings uncovered for the first time that MI suppresses erastin-induced ferroptosis through releasing miR-22-3p-enriched exosomes derived from cardiomyocytes. Therefore, targeting exosome-mediated cardiomyocyte/tumor pathological communication may offer a novel approach for the ferroptosis-based antitumor therapy. | Ye Yuan Zhongting Mei Zhezhe Qu Guanghui Li Shuting Yu Yingqi Liu Kuiwu Liu Zhihua Shen Jiaying Pu Yanquan Wang Changhao Wang Zhiyong Sun Qian Liu Xiaochen Pang Ao Wang Zijing Ren Tong Wang Ying Liu Jinhuan Hong Jiajie Xie Xin Li Zhonghua Wang Weijie Du Baofeng Yang | 2023 | Signal Transduction and Targeted Therapy2023,8,4: | 2
Low-bandgap conjugated polymers based on benzodipyrrolidone with reliable unipolar electron mobility exceeding 1 cm^(2) V^(-1)s^(-1)显示文摘The employment of an intrinsic quinoidal building block, benzodipyrrolidone(BDP),on constructing conjugated polymers(PBDP-2F and PBDP-2CN)with high electron mobility and unipolar transport characteristic in polyethylenimine ethoxylated(PEIE)modified organic field-effect transistors(OFETs)is reported. The intrinsic quinoidal characteristic and excellent coplanarity of BDP can lower the lowest unoccupied molecular orbital(LUMO)levels and improve ordered interchain packing of the resulting polymers in solid states, which are favorable for electron-injection and transport.By using PEIE as the interlayer to block the hole injection, unipolar n-type transport characteristics with high electron mobility of 0.58 and 1.01 cm^(2) V^(-1)s^(-1)were achieved by the OFETs based on PBDP-2F and PBDP-2CN,respectively.More importantly, the extracted mobilities are highly reliable with the reliability factor of above 80%.To the best of our knowledge,PBDP-2CN is the very first quinoid-based conjugated polymer with reliable electron mobility exceeding 1 cm^(2) V^(-1)s^(-1).This work represents a significant step in exploring intrinsic quinoidal CPs for application in n-channel OFETs and logic complementary circuits. | Mingqun Yang Tian Du Xuxia Zhao Xuelong Huang Langheng Pan Shuting Pang Haoran Tang Zhongxiang Peng Long Ye Yunfeng Deng Mingliang Sun Chunhui Duan Fei Huang Yong Cao | 2021 | Science China Chemistry2021,64,7: | 1
Manipulations of micro/nanoparticles using gigahertz acoustic streaming tweezers显示文摘Contactless acoustic manipulation of micro/nanoscale particles has attracted considerable attention owing to its near independence of the physical and chemical properties of the targets, making it universally applicable to almost all biological systems.Thin-film bulk acoustic wave(BAW)resonators operating at gigahertz(GHz)frequencies have been demonstrated to generate localized high-speed microvortices through acoustic streaming effects.Benefitting from the strong drag forces of the high-speed vortices,BAW-enabled GHz acoustic streaming tweezers(AST)have been applied to the trapping and enrichment of particles ranging in size from micrometers to less than 100 nm.However, the behavior of particles in such 3D microvortex systems is still largely unknown. In this work, the particle behavior(trapping, enrichment, and separation)in GHz AST is studied by theoretical analyses,3D simulations, and microparticle tracking experiments. It is found that the particle motion in the vortices is determined mainly by the balance between the acoustic streaming drag force and the acoustic radiation force. This work can provide basic design principles for AST-based lab-on-a-chip systems for a variety of applications. | Hang Wu Zifan Tang Rui You Shuting Pan Wenpeng Liu Hongxiang Zhang Tiechuan Li Yang Yang Chongling Sun Wei Pang Xuexin Duan | 2022 | Nanotechnology and Precision Engineering2022,5,2: | 1
High open-circuit voltage organic solar cells enabled by a difluorobenzoxadiazole-based conjugated polymer donor显示文摘A new polymer donor based on 3,3′-difluoro-2,2′-bithiophene(2F2T) and difluorobenzoxadiazole(ffBX), named 2F2T-ffBX, is designed and synthesized. The organic solar cell(OSC) based on 2F2T-ffBX donor and [6,6]-phenyl-C60-butyl acid methyl ester([60]PCBM) acceptor exhibits a high efficiency of 7.3% with a high open-circuit voltage(Voc) of 1.03 V. When blended with perylenediimide-based acceptor(PDI6), the corresponding OSC shows a higher Voc of 1.19 V with a low energy loss of 0.50 e V but a much lower efficiency of 2.0%. The detailed analyses including charge generation, transport, recombination properties, and morphology were performed to understand the performance of corresponding devices. | Ruiwen Zhang Junyi Wang Xi Liu Shuting Pang Chunhui Duan Fei Huang Yong Cao | 2019 | Science China Chemistry2019,62,7: | 0
Mettl13 protects against cardiac contractile dysfunction by negatively regulating C-Cbl-mediated ubiquitination of SERCA2a in ischemic heart failure显示文摘Ischemic heart failure(HF)remains a leading cause of morbidity and mortality.Maintaining homeostasis of cardiac function and preventing cardiac remodeling deterioration are critical to halting HF progression.Methyltransferase-like protein 13(Mettl13)has been shown to regulate protein translation efficiency by acting as a protein lysine methyltransferase, but its role in cardiac pathology remains unexplored. This study aims to characterize the roles and mechanisms of Mettl13 in cardiac contractile function and HF.We found that Mettl13 was downregulated in the failing hearts of mice post-myocardial infarction(MI)and in a cellular model of oxidative stress.Cardiomyocyte-specific overexpression of Mettl13 mediated by AAV9-Mettl13 attenuated cardiac contractile dysfunction and fibrosis in response to MI,while silencing of Mettl13 impaired cardiac function in normal mice.Moreover,Mettl13 overexpression abrogated the reduction in cell shortening,Ca^(2+)transient amplitude and SERCA2a protein levels in the cardiomyocytes of adult mice with MI.Conversely, knockdown of Mettl13 impaired the contractility of cardiomyocytes, and decreased Ca^(2+)transient amplitude and SERCA2a protein expression in vivo and in vitro.Mechanistically,Mettl13 impaired the stability of c-Cbl by inducing lysine methylation of c-Cbl,which in turn inhibited ubiquitination-dependent degradation of SERCA2a.Furthermore,the inhibitory effects of knocking down Mettl13 on SERCA2a protein expression and Ca^(2+)transients were partially rescued by silencing c-Cbl in H_(2)O_(2)-treated cardiomyocytes. In conclusion, our study uncovers a novel mechanism that involves the Mettl13/c-Cbl/SERCA2a axis in regulating cardiac contractile function and remodeling, and identifies Mettl13 as a novel therapeutic target for ischemic HF. | Shuting Yu ZhiYong Sun Xiuzhu Wang Tiantian Ju Changhao Wang Yingqi Liu Zhezhe Qu KuiWu Liu Zhongting Mei Na Li Meixi Lu Fan Wu Min Huang Xiaochen Pang Yingqiong Jia Ying Li Yaozhi Zhang Shunkang Dou Jianhao Jiang Xin Li Baofeng Yang Weijie Du | 2023 | Science China(Life Sciences)2023,66,12: | 0
