| 1 | A novel selective autophagy receptor,CCDC50,delivers K63 polyubiquitination-activated RIG-I/MDA5 for degradation during viral infection显示文摘Autophagy is a con served process that delivers cytosolic substa nces to the lysosome for degradatio n,but its direct role in the regulation of antiviral inn ate immu nity remains poorly un derstood.Here,through high-throughput screeni ng,we discovered that CCDC50 functions as a previously unknown autophagy receptor that negatively regulates the type I interferon(IFN)signaling pathway initiated by RIG-I-like receptors(RLRs),the sensors for RNA viruses.The expression of CCDC50 is enhanced by viral infection,and CCDC50 specifically recognizes K63-polyubiquitinated RLRs,thus delivering the activated RIG-I/MDA5 for autophagic degradation.The associatio n of CCDC50 with phagophore membrane protei n LC3 is con firmed by crystal structure analysis.In contrast to other known autophagic cargo receptors that associate with either the LIR-docking site(LDS)or the UIMdocking site(UDS)of LC3,CCDC50 can bind to both LDS and UDS,representing a new type of cargo receptor.In mouse models with RNA virus infection,CCDC50 deficiency reduces the autophagic degradation of RIG-I/MDA5 and promotes type I IFN responses,resulting in enhaneed viral resistance and improved survival rates.These results reveal a new link between autophagy and antiviral innate immune resp on ses and provide additional in sights into the regulatory mecha nisms of RLR-mediated antiviral signaling. | Panpan Hou Kongxiang Yang Penghui Jia Lan Liu Yuxin Lin Zibo Li Jun Li Shuliang Chen Shuting Guo Ji'An Pan Junyu Wu Hong Peng Weijie Zeng Chunmei Li Yingfang Liu Deyin Guo | 2021 | Cell Research2021,31,1: | 5 |
| 2 | Rock damage and fracturing induced by high static stress and slightly dynamic disturbance with acoustic emission and digital image correlation techniques显示文摘A series of coupled static-dynamic loading tests is carried out in this study to understand the effect of slightly dynamic disturbance on the rocks under high static stress.The acoustic emission(AE)and digital image correlation(DIC)techniques are combined to quantitatively characterize the damage and fracturing behaviors of rocks.The effects of three influencing factors,i.e.initial static stress,disturbance amplitude,and disturbance frequency,on the damage and fracturing evolution are analyzed.The experimental results reveal the great differences in AE characteristics and fracturing behaviors of rocks under static loads and coupled static-dynamic loads.Both the Kaiser effect and Felicity effect are observed during the disturbance loading process.The crack initiation,stable and unstable propagation in the highly-stressed rocks can be triggered by cyclic disturbance loads,and more local tensile splitting cracks are found in the rocks subjected to coupled static-dynamic loads.The damage and fracturing evolution of rocks during cyclic disturbances can be divided into two stages,i.e.steady and accelerated stages,and the increase rate and proportion of each stage are greatly affected by these influencing factors.High initial static stress,low disturbance frequency,and high disturbance amplitude are considered to be adverse factors to the stability of the rocks,which would induce a high increase rate of the steady stage and a high proportion of the accelerated stage within the whole disturbance period.Based on the two-stage damage evolution trend,a linear-exponential damage model is employed to predict the instability of the rocks under coupled static-dynamic loads. | Shuting Miao Peng-Zhi Pan Petr Konicek Peiyang Yu Kunlun Liu | 2021 | Journal of Rock Mechanics and Geotechnical Engineering2021,13,5: | 2 |

| 11 | Spatial pattern of urban-rural integration in China and the impact of geography显示文摘Urban-rural integration (URI) is a global challenge that is highly related to inequalities, poverty, economic growth, and other Sustainable Development Goals (SDGs). Existing research has evaluated the extent of URI and explored its influencing factors, but urban-rural linkages are seldom incorporated in evaluation systems, and geographical factors are rarely recognized as the influencing factors. We construct a URI framework including regional economy, rural development, urban-rural linkage, and urban-rural gap. Based on a dataset consisting of 1,669 counties in China in 2020, we reveal the spatial pattern of URI and find a high correlation between the spatial pattern of URI and the relief degree of land surface (RDLS). Using structural equation modeling, we discover that topography has direct ( − 0.18, p < 0.001) and indirect ( − 0.17, p < 0.001) effects on URI. The indirect negative effects are mediated through the infrastructure, and the combination of localized advantages and modern technical conditions could mitigate the negative impact of topography. Finally, we identify 742 counties as lagging regions in URI, which can be clustered into eight types. | 12 | Shear band evolution and acoustic emission characteristics of sandstone containing non-persistent flaws显示文摘Direct shear tests were conducted on sandstone specimens under different constant normal stresses to study the coalescence of cracks between non-persistent flaws and the shear sliding characteristics of the shear-formed fault.Digital image correlation and acoustic emission(AE)techniques were used to monitor the evolution of shear bands at the rock bridge area and microcracking behaviors.The experimental results revealed that the shear stresses corresponding to the peak and sub-peak in the stressdisplacement curve are significantly affected by the normal stress.Strain localization bands emerged at both the tip of joints and the rock bridge,and their extension and interaction near the peak stress caused a surge in the AE hit rate and a significant decrease in the AE b value.Short and curvilinear strain bands were detected at low normal stress,while high normal stress generally led to more microcracking events and longer coplanar cracks at the rock bridge area.Furthermore,an increase in normal stress resulted in a higher AE count rate and more energetic AE events during friction sliding along the shearformed fault.It was observed that the elastic energy released during the crack coalescence at the prepeak stage was much greater than that released during friction sliding at the post-peak stage.More than 75%of AE events were located in the low-frequency band(0e100 kHz),and this proportion continued to rise with increasing normal stress.Moreover,more AE events of low AF value and high RA value were observed in specimens subjected to high normal stress,indicating that greater normal stress led to more microcracks of shear nature. | Shuting Miao Peng-Zhi Pan Chuanqing Zhang Lei Huo | 2024 | Journal of Rock Mechanics and Geotechnical Engineering2024,16,2: | 0 |
