| 1 | The Epitope Study on the SARS-CoV Nucleocapsid Protein显示文摘The nucleocapsid protein (N protein) has been found to be an antigenic protein in a number of coronaviruses. Whether the N protein in severe acute respiratory syndrome-associated coronavirus (SARS-CoV) is antigenic remains to be elucidated. Using Western blot and Enzyme-linked Immunosorbent Assay (ELISA),the recombinant N proteins and the synthesized peptides derived from the N protein were screened in sera from SARS patients. All patient sera in this study displayed strong positive immunoreactivities against the recombinant N proteins,whereas normal sera gave negative immunoresponses to these proteins, indicating that the N protein of SARS-CoV is an antigenic protein. Furthermore, the epitope sites in the N protein were determined by competition experiments, in which the recombinant proteins or the synthesized peptides competed against the SARS-CoV proteins to bind to the antibodies raised in SARS sera. One epitope site located at the C-terminus was confirmed as the most antigenic region in this protein. A detailed screening of peptide with ELISA demonstrated that the amino sequence from Codons 371 to 407 was the epitope site at the C-terminus of the N protein. | 3 | Historical Evolution of Mariculture in China During Past 40 Years and Its Impacts on Eco-environment显示文摘The rapid development of mariculture in China in recent decades has satisfied people's demand of seafood, and has made a great contribution to economic development. However, mariculture has also caused some negative impacts on the eco-environment. By statistically analyzing national data(as well as data for four main provinces) regarding the mariculture area and cultivation species in China since reform and opening up, the historical evolution of mariculture during the past 40 years was analyzed, the driving factors related to policy, market and technology innovation were discussed, and the potential impacts of mariculture on the coastal eco-environment were also illustrated. The statistical results indicated that the maricultural area increased radically during the past 40 years, from 1979(116.47 × 10~3 ha) to 2012(2205.65 × 10~3 ha), with an increase of nearly 20-fold. Shandong Province represented the fastest-growing region, with an increase of 31-fold. Moreover, the cultivated species had gradually become diversified. Initially, shellfish and algae were major species, and then it developed to various species including fish, and currently shellfish and crustaceans were the major species. The development of mariculture was driven by government policies, market economy and technology innovation. Rapid development of mariculture also caused significantly adverse impacts on the eco-environment of the coastal regions. For example, the sea reclamation for mariculture decreased the area and function of coastal wetlands and the contaminants originating from mariculture destroyed the coastal aquatic environment. Fortunately, the implementation of ‘13 th Five-Year Fisheries Planning' is expected to help both improve the seafood quality and reduce the contamination in coastal aquatic environment. | 4 | Nlrc3-like is required for microglia maintenance in zebrafish显示文摘Microglia are tissue-resident macrophages residing in the central nervous system (CNS) and play critical roles in removing cellular debris and infectious agents as well as regulating neurogenesis and neuronal activities. Yet, the molecular basis underlying the establishment of microglia pool and the maintenance of their homeostasis in the CNS remain largely undefined. Here we report the identification and characterization of a mutant zebrafish, which harbors a point mutation in the nucleotide-binding oligomerization domain (NOD) like receptor gene nlrc3-like, resulting in the loss of microglia in a temperature sensitive manner. Temperature shift assay reveals that the late onset of nlrc3-like deficiency leads to excessive microglia cell death. Further analysis shows that the excessive microglia death in nlrc3-like deficient mutants is attributed, at least in part, to aberrant activation of canonical inflammasome pathway. | 5 | Occurrence and impact of polychlorinated dibenzo-p-dioxins/dibenzofurans in the air and soil around a municipal solid waste incinerator显示文摘To assess the influence of polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans(PCDD/Fs) on the environment in the vicinity of municipal solid waste incinerators(MSWIs), we determined the levels of PCDD/Fs in air and soil samples collected around a MSWI, which is the largest in China. The International Toxicity Equivalency Quantity(I-TEQ) concentrations of PCDD/Fs in air samples were from 0.0300 to 1.03 pg I-TEQ/m^3(0.445–13.6 pg/m^3), with an average of 0.237 pg I-TEQ/m^3, while in soil samples they ranged from 0.520 to 3.40 pg I-TEQ/g(2.41–88.7 pg/g) with an average of1.49 pg I-TEQ/g. The concentrations of PCDD/Fs in air and soil samples were comparable to other areas, and Pe CDFs were the dominant contributors, which was different from stack gas homologue patterns. Multivariate statistical analysis showed that PCDD/Fs emission from the MSWI did not directly affect the profiles of PCDD/Fs in air and soils, so that vehicles and unidentified emission sources should be considered. The daily inhalation levels of PCDD/Fs for children(0.0110 to 0.392 pg I-TEQ/(kg·day) and adults(0.00600 to 0.221 pg I-TEQ/(kg·day) near the MSWI were lower than the tolerable daily intake of 1.00 to 4.00 pg WHO-TEQ/(kg·day), but in winter the values were higher than in summer. | 8 | A Strategy for Searching Antigenic Regions in the SARS-CoV Spike Protein显示文摘In the face of the worldwide threat of severe acute respiratory syndrome (SARS)to human life, some of the most urgent challenges are to develop fast and accurate analytical methods for early diagnosis of this disease as well as to create a safe anti-viral vaccine for prevention. To these ends, we investigated the antigenicity of the spike protein (S protein), a major structural protein in the SARS-coronavirus (SARS-CoV). Based upon the theoretical analysis for hydrophobicity of the S protein, 18 peptides were synthesized. Using Enzyme-Linked Immunosorbent Assay (ELISA), these peptides were screened in the sera from SARS patients. According to these results, two fragments of the S gene were amplified by PCR and cloned into pET-32a. Both S fragments were expressed in the BL-21 strain and further purified with an affinity chromatography. These recombinant S fragments were confirmed to have positive cross-reactions with SARS sera, either by Western blot or by ELISA. Our results demonstrated that the potential epitope regions were located at Codons 469-882 in the S protein, and one epitope site was located at Codons 599-620. | 11 | Colorful liquid metal printed electronics显示文摘Liquid metal based printed electronics was a newly emerging frontier in recent years. However, restricted by the single silver-white appearance of the liquid metal(LM), the colors of currently available printed electronics were rather limited. Here,a new conceptual LM based colorful printed electronics was proposed where electrical wires and circuits with numerous colors can be made via a straightforward, efficient and accurate printing procedure. Firstly, the LM was printed on the substrate to construct a conductive wire. Then it was frozen to a solid. Subsequently, colorful pigments were coated on the originally printed liquid metal conductive wires, which finally were packaged with PDMS. Such multicolored conductive wire exhibits excellent conductivity, and good temperature resistance(do not fade at high temperature). Further, the adhesion mechanism of the mineral pigments on the liquid metal layer was disclosed. And the pigment layer was discovered to well protect the LM from the outside environments, and enhance the durability of the LM conductive wire at the same time. | 12 | Spatial-temporal heterogeneity and determinants of HIV prevalence in the Mano River Union countries显示文摘Background:Utilizing population-based survey data in epidemiological research with a spatial perspective can integrate valuable context into the dynamics of HIV prevalence in West Africa.However,the situation in the Mano River Union(MRU)countries is largely unknown.This research aims to perform an ecological study to determine the HIV prevalence patterns in MRU.Methods:We analyzed Demographic and Health Survey(DHS)and AIDS Indicator Survey(AIS)data on HIV prevalence in MRU from 2005 to 2020.We examined the country-specifc,regional-specifc and sex-specifc ratios of respondents to profle the spatial–temporal heterogeneity of HIV prevalence and determine HIV hot spots.We employed Geodetector to measure the spatial stratifed heterogeneity(SSH)of HIV prevalence for adult women and men.We assessed the comprehensive correct knowledge(CCK)about HIV/AIDS and HIV testing uptake by employing the Least Absolute Shrinkage and Selection Operator(LASSO)regression to predict which combinations of CCKs can scale up the ratio of HIV testing uptake with sex-specifc needs.Results:In our analysis,we leveraged data for 158,408 respondents from 11 surveys in the MRU.From 2005–2015,Cote d’Ivoire was the hot spot for HIV prevalence with a Gi_Bin score of 3,Z-Score 8.0–10.1 and P<0.001.From 2016 to 2020,Guinea and Sierra Leone were hot spots for HIV prevalence with a Gi_Bin score of 2,Z-Score of 3.17 and P<0.01.The SSH confrmed the signifcant diferences in HIV prevalence at the national level strata,with a higher level for Cote d’Ivoire compared to other countries in both sexes with q-values of 0.61 and 0.40,respectively.Our LASSO model predicted diferent combinations of CCKs with sex-specifc needs to improve HIV testing uptake.Conclusions:The spatial distribution of HIV prevalence in the MRU is skewed and the CCK about HIV/AIDS and HIV testing uptake are far below the threshold target set by UNAIDS for ending the epidemic in the sub-region.Geodetector detected statistically signifcant SSH within and between countries in the MRU.Our LASSO model predicted that diferent emphases should be implemented when popularizing the CCK about HIV/AIDS for adult women and men. | Idrissa Laybohr Kamara Liang Wang Yaxin Guo Shuting Huo Yuanyuan Guo Chengdong Xu Yilan Liao William J.Liu Wei Ma George F.Gao | 2022 | Infectious Diseases of Poverty2022,11,6: | 0 |
| 13 | Transcriptomic and genetic approaches reveal that low-light-induced disease susceptibility is related to cellular oxidative stress in tomato显示文摘The impact of low light intensities on plant disease outbreaks represents a major challenge for global crop security,as it frequently results in significant yield losses.However,the underlying mechanisms of the effect of low light on plant defense are still poorly understood.Here,using an RNA-seq approach,we found that the susceptibility of tomato to Pseudomonas syringae pv.tomato DC3000(Pst DC3000)under low light was associated with the oxidation-reduction process.Low light conditions exacerbated Pst DC3000-induced reactive oxygen species(ROS)accumulation and protein oxidation.Analysis of gene expression and enzyme activity of ascorbate peroxidase 2(APX2)and other antioxidant enzymes revealed that these defense responses were significantly induced by Pst DC3000 inoculation under normal light,whereas these genes and their associated enzyme activities were not responsive to pathogen inocula-tion under low light.Additionally,the reduced ascorbate to dehydroascorbate(AsA/DHA)ratio was lower under low light compared with normal light conditions upon Pst DC3000 inoculation.Furthermore,the apx2 mutants generated by a CRISPR-Cas9 gene-editing approach were more susceptible to Pst DC3000 under low light conditions.Notably,this increased susceptibility could be significantly reduced by exogenous AsA treatment.Collectively,our findings suggest that low-light-induced disease susceptibility is associated with increased cellular oxidative stress in tomato plants.This study sheds light on the intricate relationship between light conditions,oxidative stress,and plant defense responses,and may pave the way for improved crop protection strategies in low light environments. | Qian Luo Jiao Wang Ping Wang Xiao Liang Jianxin Li Changqi Wu Hanmo Fang Shuting Ding Shujun Shao Kai Shi | 2023 | Horticulture Research2023,10,10: | 0 |
