1Novel and potent inhibitors targeting DHODH are broad-spectrum antivirals against RNA viruses including newly-emerged coronavirus SARS-CoV-2显示文摘Emerging and re-emerging RNA viruses occasionally cause epidemics and pandemics worldwide,such as the on-going outbreak of the novel coronavirus SARS-CoV-2.Herein,we identified two potent inhibitors of human DHODH,S312 and S416,with favorable drug-likeness and pharmacokinetic profiles,which all showed broad-spectrum antiviral effects against various RNA viruses,including influenza A virus,Zika virus,Ebola virus,and particularly against SARS-CoV-2.Notably,S416 is reported to be the most potent inhibitor so far with an EC5o of 17 nmol/L and an SI value of 10,505.88 in infec-ted cells.Our results are the first to validate that DHODH is an attractive host target through high antiviral efficacy in vivo and low virus replication in DHODH knock-out cells.This work demonstrates that both S312/S416 and old drugs(Leflunomide/Teriflunomide)with dual actions of antiviral and immuno-regulation may have clinical potentials to cure SARS-CoV-2 or other RNA viruses circulating worldwide,no matter such viruses are mutated or not.Rui Xiong Leike Zhang Shiliang Li Yuan Sun Minyi Ding Yong Wang Yongliang Zhao Yan Wu Weijuan Shang Xiaming Jiang Jiwei Shan Zihao Shen Yi Tong Liuxin Xu Yu Chen Yingle Liu Gang Zou Dimitri Lavillete Zhenjiang Zhao Rui Wang Lili Zhu Gengfu Xiao Ke Lan Honglin Li Ke Xu 2020Protein & Cell2020,11,10:14
2Protonation induced high-T_c phases in iron-based superconductors evidenced by NMR and magnetization measurements显示文摘Chemical substitution during growth is a well-established method to manipulate electronic states of quantum materials, and leads to rich spectra of phase diagrams in cuprate and iron-based superconductors. Here we report a novel and generic strategy to achieve nonvolatile electron doping in series of(i.e.11 and 122 structures) Fe-based superconductors by ionic liquid gating induced protonation at room temperature. Accumulation of protons in bulk compounds induces superconductivity in the parent compounds, and enhances the Tclargely in some superconducting ones. Furthermore, the existence of proton in the lattice enables the first proton nuclear magnetic resonance(NMR) study to probe directly superconductivity. Using Fe S as a model system, our NMR study reveals an emergent high-Tcphase with no coherence peak which is hard to measure by NMR with other isotopes. 3Ultrafast optical spectroscopy evidence of pseudogap and electronphonon coupling in an iron-based superconductor KCa_(2)Fe_(4)As_(4)F_(2)显示文摘We use ultrafast optical spectroscopy to study the nonequilibrium quasiparticle relaxation dynamics of the iron-based superconductor KCa_(2)Fe_(4)A_(s4)F_(2)with T_(c)= 33.5 K. Our results reveal a possible pseudogap(△_(PG)=(2.4 ± 0.1) me V) below T*≈ 50 K but prior to the opening of a superconducting gap(△SC(0) ≈(4.3 ± 0.1) me V). Measurements under high pump fluence reveal two distinct, coherent phonon oscillations with 1.95 and 5.51 THz frequencies, respectively. 5Molecular phylogeny and species delimitation of Stachyuraceae: Advocating a herbarium specimen-based phylogenomic approach in resolving species boundaries显示文摘Species concept and delimitation are fundamental to taxonomic and evolutionary studies.Both inadequate informative sites in the molecular data and limited taxon sampling have often led to poor phylogenetic resolution and incorrect species delineation.Recently,the whole chloroplast genome sequences from extensive herbarium specimen samples have been shown to be ffective to amend the problem.Stachyuraceae are a small family consisting of only one genus Stachyurus of six to 16 species.However,species delimitation in Stachyurus has been highly controversial because of few and generally unstable morphological characters used for cassfcation.In this study,we sampled 69 individuals of seven species(each with at least three individual)covering the entire taxonomlc dlversity,geographlc range,and morphological varlation of Stachyurus from her barium specimens for genome wide plastid gene sequencing to address species delineation in the genus.We obtained high quality DNAS from specimens using a repently developed DNA reconstruction technique.We first assembled four whole chloroplast genome sequences.Based on the chloroplast genome and one nuclear ribosomal DNA sequence of Stachyunus,we designed primers for multiplex polymerase chain reaction and high throughput sequencing of 44 plastid loci for species of Stachyurus,Data of these chloroplast DNA and nuclear ribosomal DNA internal transcribed spacer sequences were used for phylogenetic analyses.The phylogenetic results showed that the Japanese species Stachyurus praecox Siebold&Zucc.was sister to the rest in China's Mainland,which indicated a typical Sino-Japanese distribution pattern.Based on diagnostic morphological characters,dlstinct distrbutional range,and monophyly of each clade,we redefined seven species for Stachyurus fllowving an integrative specles concept,and revised the taxonomy of the family based on previous reports and specimens,in particular the type specimens.Furthermore,our divergence time estimation results suggested that Stachyuraceae split from its sister group Crossosomataceae from the New World at ca.54.29 Mya,but extant species of Stachyuraceae started their diversifcation only recently at ca.6.85 Mya.Diversification time of Stachyurus in China's Mainland was estimated to be ca.4.45 Mya.This research has provided an example of using the herbarium specimen based phylogenomic approach in resolving species boundaries in a taxonomically dificult genus.Jun-Xia Su Cong-Cong Dong Yan-Ting Niu Li-Min Lu Chao xu Bing Liu ShiLiang Zhou An-Ming Lu Yu-Ping Zhu Jun Wen Zhi-Duan Chen 2020Journal of Systematics and Evolution2020,58,5:2
6Glial scar size, inhibitor concentration, and growth of regenerating axons after spinal cord transection显示文摘A mathematical model has been formulated in accordance with cell chemotaxis and relevant experimental data. A three-dimensional lattice Boltzmann method was used for numerical simulation. The present study observed the effects of glial scar size and inhibitor concentration on regenerative axonal growth following spinal cord transection. The simulation test comprised two parts: (1) when release rates of growth inhibitor and promoter were constant, the effects of glial scar size on axonal growth rate were analyzed, and concentrations of inhibitor and promoters located at the moving growth cones were recorded. (2) When the glial scar size was constant, the effects of inhibitor and promoter release rates on axonal growth rate were analyzed, and inhibitor and promoter concentrations at the moving growth cones were recorded. Results demonstrated that (1) a larger glial scar and a higher release rate of inhibitor resulted in a reduced axonal growth rate. (2) The axonal growth rate depended on the ratio of inhibitor to promoter concentrations at the growth cones. 7Comparative study on in vitro transformation of paralytic shellfish poisoning (PSP) toxins in different shellfish tissues显示文摘Dissected tissues of three shellfish species,the Chinese scallop,Chlamys farreri,Manila clam,Ruditapes philippinarum,and Razor shell,Solen strictu were evaluated for in vitro transformation of paralytic shellfish poisoning(PSP) toxins. Tissue homogenates were incubated with extraction from toxic algae Alexandrium minutum to determine toxin conversion. The effects of heating and addition of a natural reductant(glutathione) on toxin conversion were also assessed. The toxin profile was investigated through high performance liquid chromatography with fluorescence detection(HPLC-FLD) . The evident variations in the toxin content were observed only in Chinese scallop viscera homogenates. The concentration of GTX4 was reduced by 45%(approximately 0.8 μmol/dm 3 ) and 25%(approximately 1 μmol/dm 3 ) for GTX1,while GTX2 and GTX3 increased by six times(approximately 1 μmol/dm 3 ) and 3 times(approximately 0.3 μmol/dm 3 ) respectively. Simultaneously,the total toxicity decreased by 38% during the 48 h incubation period,the final toxicity was 20.4 nmol STXeq/g. Furthermore,heated Chinese scallop viscera homogenates samples were compared with non-heated samples. The concentration of the GTX4 and GTX1 was clearly 28%(approximately 0.53 μmol/dm 3 ) and 17%(approximately 0.69 μmol/dm 3 ) higher in heated samples,GTX2 and GTX3 were four times(0.66 μmol/dm 3 ) and two times(0.187 μmol/dm 3 ) lower respectively. GSH(+) Chinese scallop viscera homogenates samples were compared with GSH(-) samples,the concentration in the GTX4 and GTX1 was 9%(approximately 0.12 μmol/dm 3 ) and 11%(approximately 0.36 μmol/dm 3 ) lower respectively,GTX2 and GTX3 was 17%(approximately 0.14 μmol/dm 3 ) and 19%(approximately 0.006 μmol/dm 3 ) higher respectively. In contrast,there was a little change in the concentration of PSP toxins of Manila clam and Razor shell tissue homogenates. These observations on three shellfish tissues confirmed that there were species-specific differences in PSP toxins transformation. PSP toxins transformation was more pronounced in viscera tissue than in muscle tissue. 10DRY-SEASON VARIABILITY IN SUSPENDED SEDIMENT CONCENTRATIONS IN THE SOUTH PASSAGE OF THE CHANGJIANG ESTUARY显示文摘This paper reports the results of continuous monitoring of turbidity, water depth, salinity (using an Optical Backscatter Sensor (OBS)), and current velocity (using a Current meter (SLC9-2)) in the South Passage of the Changjiang Estuary over a spring–neap period in February 2003 (dry season). The turbidity measured via OBS was closely correlated with the suspended sediment concentration (SSC), which was highly variable. Over the study period, the SSC in the middle layer ranged from 110 to 1400 mg/l. The minimum SSC occurred during a late ebb tide, and the maximum SSC occurred during a late flood tide. On average, the SSC was 1.5 times higher during flood tide than during ebb tide. Vertically within the water column, SSC increased downward, with the ratio of SSC measured near the bed to that measured at the surface ranging from 1.90 to 18.3. 11Report on the Breeding of Dahen 799 Broilers显示文摘[Objectives]This study was conducted to improve the production efficiency of dark-shank partridge-like plumage high-quality broilers and ensure animal welfare,so as to meet the development needs of China’s yellow-feather broiler industry.[Methods]Sichuan Animal Science Academy and Sichuan Dahen Poultry Breeding Co.,Ltd.formed a joint breeding research team.The team bred Dahen 799 broiler complete set line with local chicken breeds as the main material and improving production efficiency,stabilizing meat quality and flavor and achieving autosexing of chicks as the main breeding goals by modern poultry breeding technologies,after 10 years and 8 generations.[Results]The Dahen 799 broiler complete set line was approved by National Commission of Livestock and Poultry Genetic Resources in December 2020.The 66-week-old egg production of the breed reached 189 eggs.For the commercial generation,cocks and hens showed 10-week-old weights of 2668 and 2269 g,respectively;the feed conversion ratio was 2.26-2.35;the survivability was 96.7%-97.2%;and the average intramuscular fat contents of cocks and hens were in the range of 1.8%-2.1%,and the inosinic acid contents were in the range of 2.3-3.2 mg/g.The production performance and meat flavor of Dahen 799 broilers were better than Dahen 699 broilers,Sichuan mountainous bone-black chickens and two other social breeds.Compared with Sichuan mountainous bone-black chickens,the egg production at the age of 66 weeks increased by 47 eggs;the average weight of cocks and hens of the commercial generation at 10 weeks increased by 45.21%;and the autosexing rate of chicks of the commercial generation reached 99.3%.[Conclusions]Dahen 799 broilers have the advantages of good production performance,high feed conversion ratio and excellent meat flavor and the characteristic of autosexing which other breeds do not have,and can fully meet the industrial development needs of dark shanks and partridge-like plumage broilers.Chaowu YANG Chunlin YU Mohan QIU Zengrong ZHANG Chenming HU Huarui DU Qingyun LI Xia XIONG Shiliang ZHU Siyang LIU Xiaosong JIANG 2022Agricultural Biotechnology2022,11,3:0
12Proximity-enabled covalent binding of IL-2 to IL-2Rα selectively activates regulatory T cells and suppresses autoimmunity显示文摘Interleukin-2(IL-2)is a pleiotropic cytokine that orchestrates bidirectional immune responses via regulatory T cells(Tregs)and effector cells,leading to paradoxical consequences.Here,we report a strategy that exploited genetic code expansion-guided incorporation of the latent bioreactive artificial amino acid fluorosulfate-L-tyrosine(FSY)into IL-2 for proximity-enabled covalent binding to IL-2Rαto selectively promote Treg activation.We found that FSY-bearing IL-2 variants,such as L72-FSY,covalently bound to IL-2Rαvia sulfur-fluoride exchange when in proximity,resulting in persistent recycling of IL-2 and selectively promoting the expansion of Tregs but not effector cells.Further assessment of L72-FSY-expanded Tregs demonstrated that L72-FSY maintained Tregs in a central memory phenotype without driving terminal differentiation,as demonstrated by simultaneously attenuated expression of lymphocyte activation gene-3(LAG-3)and enhanced expression of programmed cell death protein-1(PD-1).Subcutaneous administration of L72-FSY in murine models of pristane-induced lupus and graft-versus-host disease(GvHD)resulted in enhanced and sustained therapeutic efficacy compared with wild-type IL-2 treatment.The efficacy of L72-FSY was further improved by N-terminal PEGylation,which increased its circulatory retention for preferential and sustained effects.This proximity-enabled covalent binding strategy may accelerate the development of pleiotropic cytokines as a new class of immunomodulatory therapies.Bo Zhang Jiaqi Sun Yeshuang Yuan Dezhong Ji Yeting Sun Yudong Liu Shengjie Li Xingxing Zhu Xunyao Wu Jin Hu Qiu Xie Ling Wu Lulu Liu Boyang Cheng Yuanjie Zhang Lingjuan Jiang Lidan Zhao Fei Yu Wei Song Min Wang Yue Xu Shiliang Ma Yunyun Fei Lihe Zhang Demin Zhou Xuan Zhang 2023Signal Transduction and Targeted Therapy2023,8,2:0
13Design, synthesis, molecular modeling, and biological evaluation of acrylamide derivatives as potent inhibitors of human dihydroorotate dehydrogenase for the treatment of rheumatoid arthritis显示文摘Human dihydroorotate dehydrogenase(DHODH) is a viable target for the development of therapeutics to treat cancer and immunological diseases, such as rheumatoid arthritis(RA), psoriasis and multiple sclerosis(MS). Herein, a series of acrylamide-based novel DHODH inhibitors as potential RA treatment agents were designed and synthesized. 2-Acrylamidobenzoic acid analog 11 was identified as the lead compound for structureeactivity relationship(SAR) studies. The replacement of the phenyl group with naphthyl moieties improved inhibitory activity significantly to double-digit nanomolar range.Further structure optimization revealed that an acrylamide with small hydrophobic groups(Me, Cl or Br)at the 2-position was preferred. Moreover, adding a fluoro atom at the 5-position of the benzoic acid enhanced the potency. The optimization efforts led to potent compounds 42 and 53-55 with IC50 values of 41, 44, 32, and 42 nmol/L, respectively. 16Comparative Analysis of Slaughter Performance and Meat Flavor Traits of Quality Chicken under Cage Rearing and Free-range Farming Conditions显示文摘[Objective]The paper was to compare the slaughter performance and meat flavor of quality chicken under cage rearing and free-range farming conditions.[Method]A total of 4001-day-old cocks,S07 line cultivated by Sichuan Dahen Poultry Breeding Co.,Ltd.(bred for 4 succes-sive generations),were kept in cages until the end of 5 weeks of age,which were then divided into cage rearing and free-range farming groups.All cocks were fed with the same diet,and slaughter tests and meat quality analysis were carried out at the end of 10 and 22 weeks of age,respectively.[Result]The live weight,carcass weight and abdominal fat percentage in cage rearing group were significantly higher than those in free-range farming group(P<0.01),and the percentage of half-eviscerated yield and percentage of eviscerated yield in cage rearing group were significantly higher than those in free-range farming group(P<0.05),but there were no significant differences in dressing percentage,percentage of leg muscle and per-centage of breast muscle.The intramuscular fat(IMF)content and muscle fiber density of chicken in cage rearing group were significantly higher than those in free-range farming group(P<0.01),but there was no significant difference in inosinic acid(IMP)content.The content of IMF and IMF increased with the increase of feeding age,but the deposition rate of intramuscular fat was higher than that of IMF in late feeding period.[Conclusion]The study will provide reliable guidance for production and market consumption of high quality chicken.Yang Chaowu Yu Chunlin Qiu Mohan Zhang Zengrong Hu Chenming Xiong Xia Xia Bo Peng Han Song Xiaoyan Chen Jialei Zhu Shiliang Liu Siyang Du Huarui Li Qingyun 2022Animal Husbandry and Feed Science2022,14,1:0

18Discovery of a natural fluorescent probe targeting the Plasmodium falciparum cysteine protease falcipain-2显示文摘The Plasmodium falciparum cysteine protease falcipain-2(FP-2) is an attractive antimalarial target. Here, we discovered that the natural compound NP1024 is a nonpeptidic inhibitor of FP-2 with an IC50 value of 0.44 μmol L–1.
