1Cytokine- and chemokine-induced inflammatory colorectal tumor microenvironment: Emerging avenue for targeted therapy显示文摘Colorectal cancer(CRC)is a predominant life-threatening cancer,with liver and peritoneal metastases as the primary causes of death.Intestinal inflammation,a known CRC risk factor,nurtures a local inflammatory environment enriched with tumor cells,endothelial cells,immune cells,cancer-associated fibroblasts,immunosuppressive cells,and secretory growth factors.The complex interactions of aberrantly expressed cytokines,chemokines,growth factors,and matrix-remodeling enzymes promote CRC pathogenesis and evoke systemic responses that affect disease outcomes.Mounting evidence suggests that these cytokines and chemokines play a role in the progression of CRC through immunosuppression and modulation of the tumor microenvironment,which is partly achieved by the recruitment of immunosuppressive cells.These cells impart features such as cancer stem cell-like properties,drug resistance,invasion,and formation of the premetastatic niche in distant organs,promoting metastasis and aggressive CRC growth.A deeper understanding of the cytokineand chemokine-mediated signaling networks that link tumor progression and metastasis will provide insights into the mechanistic details of disease aggressiveness and facilitate the development of novel therapeutics for CRC.Here,we summarized the current knowledge of cytokine-and chemokine-mediated crosstalk in the inflammatory tumor microenvironment,which drives immunosuppression,resistance to therapeutics,and metastasis during CRC progression.We also outlined the potential of this crosstalk as a novel therapeutic target for CRC.The major cytokine/chemokine pathways involved in cancer immunotherapy are also discussed in this review.Ajaz A.Bhat Sabah Nisar Mayank Singh Bazella Ashraf Tariq Masoodi Chandra P.Prasad Atul Sharma Selma Maacha Thasni Karedath Sheema Hashem Syed Besina Yasin Puneet Bagga Ravinder Reddy Michael P.Frennaux Shahab Uddin Punita Dhawan Mohammad Haris Muzafar A.Macha 2022Cancer Communications2022,42,8:8
2Development of polyvinylpyrrolidone/paclitaxel self-assemblies for breast cancer显示文摘The goal of this investigation was to develop and demonstrate a polymer/paclitaxel selfassembly(PTX-SA) formulation. Polymer/PTX-SAs were screened based on smaller size of formulation using dynamic light scattering analysis. Additionally, fluorescence microscopy and flow cytometry studies exhibited that polyvinylpyrrolidone(PVP)-based PTX-SAs(PVP/PTX-SAs) had superior cellular internalization capability in MCF7 and MDA-MB-231 breast cancer cells. The optimized PVP/PTXSAs exhibited less toxicity to human red blood cells indicating a suitable formulation for reducing systemic toxicity. The formation of PVP and PTX self-assemblies was confirmed using fluorescence quenching and transmission electron microscopy which indicated that the PVP/PTX-SAs were spherical in shape with an average size range of 53.81 nm as detected by transmission electron microscopy(TEM).FTIR spectral analysis demonstrates incorporation of polymer and paclitaxel functional groups in PVP/PTX-SAs. Both proliferation(MTS) and clonogenic(colony formation) assays were used to validate superior anticancer activity of PVP/PTX-SAs in breast cancer cells over paclitaxel. Such superior anticancer activity was also demonstrated by downregulation of the expression of pro-survival protein(Bcl-x L), upregulation of apoptosis-associated proteins(Bid, Bax, cleaved caspase 7, and cleaved PARP)and β-tubulin stabilization. 5Predicting the distribution of the Asian tapir in Peninsular Malaysia using maximum entropy modeling显示文摘In 2008,the IUCN threat status of the Asian tapir(Tapirus indicus)was reclasified from‘vulnerable'to‘en-dangered'.The latest distribution map from the IUCN Red List suggests that the tapirs'native range is becom-ing increasingly fragmented in Peninsular Malaysia,but distribution data collected by local researchers sug-gest a more extensive geographical range.Here,we compile a database of 1261 tapir occurrence records within Peninsular Malaysia,and demonstrate that this species,indeed,has a much broader geographical range than the IUCN range map suggests.However,extreme spatial and temporal bias in these records limits their utility for conservation planning.Therefore,we used maximum entropy(MaxEnt)modeling to elucidate the potential ex-tent of the Asian tapir's occurrence in Peninsular Malaysia while accounting for bias in existing distribution data.Our MaxEnt model predicted that the Asian tapir has a wider geographic range than our fine-scale data and the IUCN range map both suggest.Approximately 37%of Peninsular Malaysia contains potentially suit-able tapir habitats.Our results justify a revision to the Asian tapir's extent of occurrence in the IUCN Red List.Furthermore,our modeling demonstrated that selectively logged forests encompass 45%of potentially suitable tapir habitats,underscoring the importance of these habitats for the conservation of this species in Peninsular Malaysia.Gopalasamy Reuben CLEMENTS D.Mark RAYAN Sheema Abdul AZIZ Kae KAWANISHI Carl TRAEHOLT David MAGINTAN Muhammad Fadlli Abdul YAZI Reid TINGLEY 2012Integrative Zoology2012,7,4:1

10Organic micro-pollutant removal in liquid-phase using carbonized silk cotton hull显示文摘Phenolic compounds constitute one of the major pollutants in the modern world. Although many physical and chemical treatment technologies for their removal exist, most of them are economically not feasible. The present study was aimed at using silk cotton hull, a potent agricultural waste as an adsobent for removal of 2,4-dichlorophenol (2,4-DCP), which was used as a model phenolic compound. The process parameters were investigated and optimized conditions were determined. The equilibrium time was found to be 60 and 80 min for 10 and 20 mg/L and 100 min for 30 and 40 mg/L 2,4-DCP concentrations, respectively. Among the kinetic models applied, pseudo-second order model fitted well. The maximum adsorption capacity was 16.0 mg/g by Langmuir isotherm. Acidic pH was found favorable for the adsorption of 2,4-DCP. Studies on pH effect and desorption seemed to show that chemisorption played a major role in the adsorption process. In thermodynamic study, the change in entropy (△S 0) and heat of adsorption (△H0) of silk cotton hull carbon (SCHC) was estimated as 14.01 J/(mol·K) and 3.04 kJ/mol, respectively.
