1MicroRNA in TLR signaling and endotoxin tolerance显示文摘Toll-like receptors(TLRs)in innate immune cells are the prime cellular sensors for microbial components.TLR activation leads to the production of proinflammatory mediators and thus TLR signaling must be properly regulated by various mechanisms to maintain homeostasis.TLR4-ligand lipopolysaccharide(LPS)-induced tolerance or cross-tolerance is one such mechanism,and it plays an important role in innate immunity.Tolerance is established and sustained by the activity of the microRNA miR-146a,which is known to target key elements of the myeloid differentiation factor 88(MyD88)signaling pathway,including IL-1 receptor-associated kinase(IRAK1),IRAK2 and tumor-necrosis factor(TNF)receptor-associated factor 6(TRAF6).In this review,we comprehensively examine the TLR signaling involved in innate immunity,with special focus on LPS-induced tolerance.The function of TLR ligand-induced microRNAs,including miR-146a,miR-155 and miR-132,in regulating inflammatory mediators,and their impact on the immune system and human diseases,are discussed.Modulation of these microRNAs may affect TLR pathway activation and help to develop therapeutics against inflammatory diseases.Md A Nahid Minoru Satoh Edward KL Chan 2011Cellular & Molecular Immunology2011,8,5:34

5Activation of JAK-STAT pathway is required for platelet-derived growth factor-induced proliferation of pancreatic stellate cells显示文摘AIM: To clarify the role of Janus kinase-signal transducersand activators of transcription (JAK-STAT) pathway in platelet-derived growth factor (PDGF) induced proliferation in activated pancreatic stellate cells (PSCs).METHODS: PSCs were isolated from rat pancreas tissue, and used in their culture-activated, myofibroblast-like phenotype. STAT-specific binding activity was assessed by electrophoretic mobility shift assay. Activation of Src, JAK2,STAT1, STAT3, and ERK was determined by Western blotting using anti-phosphospecific antibodies. Cell proliferation was assessed by measuring the incorporation of 5-bromo-2′deoxyuridine.RESULTS: PDGF-BB induced STAT-specific binding activity, and activation of Src, JAK2, STAT1, STAT3, and ERK. Ethanol and acetaldehyde at clinically relevant concentrations decreased basal activation of JAK2 and STAT3. PDGFinduced activation of STAT1 and STAT3 was inhibited bya Src inhibitor PP1 and a JAK2 inhibitor AG490, whereasPDGF-induced activation of ERK was inhibited by PP1, and not by AG490. PDGF-induced proliferation was inhibited by PP1 and AG490 as well as by STAT3 antisense oligonucleotide.CONCLUSION: PDGF-BB activated JAK2-STAT pathwayvia Src-dependent mechanism. 6Bacteremia in hemodialysis patients显示文摘Infection is a common complication and is the second leading cause of death in hemodialysis patients. The risk of bacteremia in hemodialysis patients is 26-fold higher than in the general population, and 1/2-3/4 of the causative organisms of bacteremia in hemodialysis patients are Gram-positive bacteria. The ratio of resistant bacteria in hemodialysis patients compared to the general population is unclear. Several reports have indicated that hemodialysis patients have a higher risk of methicillin-resistant Staphylococcus aureus infection. The most common site of infection causing bacteremia is internal prostheses; the use of a hemodialysis catheter is the most important risk factor for bacteremia. Although antibiotic lock of hemodialysis catheters and topical antibiotic ointment can reduce catheter-related blood stream infection(CRBSI), their use should be limited to necessary cases because of the emergence of resistant organisms. Systemic antibiotic administration and catheter removal is recommended for treating CRBSI, although a study indicated the advantages of antibiotic lock and guidewire exchange of catheters over systemic antibiotic therapy. An infection control bundle recommended by the Center for Disease Control and Prevention succeeded in reducing bacteremia in hemodialysis patients with either a catheter or arteriovenous fistula. 7用ORP作为氧化沟同步硝化反硝化控制参数显示文摘同步硝化反硝化(simultaneous nitrification and denitrification,简称SND)是氧化沟工艺实现优良脱氮效果的主要原因,为了较好实现SND,采用ORP作为氧化沟工艺SND的控制参数.采用缺氧-厌氧-氧化沟模型对市政污水进行了生物脱氮研究.UORP在-30~30mV,NH4^+和NO3^-的含量均比较低,发生了较好的SND;UORP在30mV以上,出水的总无机氮(TIN)中95%以上为NO3^-,该状况下硝化效果良好,UORP在-30mV以下,硝化不充分,出水的TIN中78%以上为NH4^+;UORP在-30~30mV,TN的去除率在88%以上,SND作用去除的NO3^-占总的NO3^-去除的99%以上;氧化沟中的NH4^+和NO3^-之比的对数与UORP有着很好的相关性,相关系数R=-0.97.侯红勋 陈伦强 王淑莹 Takashi Mino Hiroyasu Satoh 2006北京工业大学学报2006,32,12:13
8早胃的癌症用的程度的诊断灵活光谱成像颜色改进显示文摘 The demarcation line between the cancerous lesion and the surrounding area could be easily recognized with flexible spectral imaging color enhancement (FICE) system compared with conventional white light images. The characteristic f inding of depressed-type early gastric cancer (EGC) in most cases was revealed as reddish lesions distinct from the surrounding yellowish non-cancerous area without magnification. Conventional endoscopic images provide little information regarding depressed lesions located in the tangential line, but FICE produces higher color contrast of such cancers. Histological f indings in depressed area with reddish col- or changes show a high density of glandular structure and an apparently irregular microvessel in intervening parts between crypts, resulting in the higher color con- trast of FICE image between cancer and surrounding area. Some depressed cancers are shown as whitish lesion by conventional endoscopy. FICE also can pro- duce higher color contrast between whitish cancerous lesions and surrounding atrophic mucosa. For nearly flat cancer, FICE can produce an irregular structuralpattern of cancer distinct from that of the surrounding mucosa, leading to a clear demarcation. Most elevated-type EGCs are detected easily as yellowish lesions with clearly contrasting demarcation. In some cases, a partially reddish change is accompanied on the tumor surface similar to depressed type cancer. In addition, the FICE system is quite useful for the detection of minute gastric cancer, even without magnif ication. 9Endothelin-1 stimulates contraction and migration of rat pancreatic stellate cells显示文摘AIM: To examine the ability of ET-1 to affect the cell functions of PSCs and the underlying molecular mechanisms.METHODS: PSCs were isolated from the pancreas of male Wistar rats after perfusion with collagenase, and cells between passages two and five were used. Expression of ET-1 and ET receptors was assessed by reverse transcription-PCR and immunostaining. Phosphorylation of myosin regulatory light chain (MLC), extracellular-signal regulated kinase (ERK), and Akt was examined by Western blotting. Contraction of PSCs was assessed on hydrated collagen lattices. Cell migration was examined using modified Boyden chambers. Cell proliferation was assessed by measuring the incorporation of 5-bromo-2'-deoxyuridine.RESULTS: Culture-activated PSCs expressed ETA and ETB receptors, and ET-1. ET-1 induced phosphorylation of MLC and ERK, but not Akt. ET-1 induced contraction and migration,but did not alter proliferation of PSCs. ET-1-induced contraction was inhibited by an ETA receptor antagonist BQ-123 and an ETB receptor antagonist BQ-788, whereas migration was inhibited by BQ-788 but not by BQ-123. A Rho kinase inhibitor Y-27632 abolished both contraction and migration.CONCLUSION: ET-1 induced contraction and migration of PSCs through ET receptors and activation of Rho-Rho kinase. 10Establishment and characterization of a rat pancreatic stellate cell line by spontaneous immortalization显示文摘AIM: Activated pancreatic stellate cells (PSCs) have been implicated in the pathogenesis of pancreatic fibrosis and inflammation. Primary PSCs can be subcultured only several times because of their limited growth potential. A continuous cell line may therefore be valuable in studying molecular mechanisms of these pancreatic disorders. The aim of this study was to establish a cell line of rat PSCs by spontaneous immortalization.METHODS: PSCs were isolated from the pancreas of male Wistar rats, and conventional subcultivation was performed repeatedly. Telomerase activity was measured using the telomere repeat amplification protocol. Activation of transcription factors was assessed by electrophoretic mobility shift assay.Activation of mitogen-activated protein (MAP) kinases was examined by Western blotting using anti-phosphospecific antibodies. Expression of cytokine-induced neutrophil chemoattractant-1 was determined by enzyme immunoassay.RESULTS: Conventional subcultivation yielded actively growing cells. One clone was obtained after limiting dilution,and designated as SIPS. This cell line has been passaged repeatedly more than 2 years, and is thus likely immortalized.SIPS cells retained morphological characteristics of primary,culture-activated PSCs. SIPS expressed α-smooth muscle actin, glial acidic fibrillary protein, vimentin, desmin, type Ⅰ collagen, fibronectin, and prolyl hydroxylases. Telomerase activity and p53 expression were negative. Proliferation of SIPS cells was serum-dependent, and stimulated with platelet-derived growth factor-BB through the activation of extracellular signal-regulated kinase. 13小尺度森林火灾的渐近幂律分布显示文摘森林火灾系统是一个自组织临界性系统,它的表现之一就是火灾的尺度分布满足幂律规律。然而实际数据分析表明,在尺度最大和最小的两个区域,火灾分布与幂律规律存在较大的差异。对小尺度火灾的偏离现象,研究者通常猜测是由于小面积火灾记录数据遗漏较多造成的,目前尚没有一个理论上的依据,而小尺度火灾的分布规律对大尺度火灾的预测起着关键作用。为了解释这种偏离现象,该文将火灾看作一个生灭过程,并对火灾蔓延和熄灭的机理进行了分析,最终得到了森林火灾分布与火灾尺度的定量依赖关系及其机理。将该文的结果与日本近年来全国范围内的森林火灾统计数据进行了对比,表明该文得到的结果与真实数据得到较好符合,并且成功解释了小尺度森林火灾分布与幂律关系的偏离现象。该研究可望对深入认识火灾系统的自组织临界性和防治火灾具有一定的实际意义。宋卫国 刘广义 于彦飞 K Satoh 2003火灾科学2003,12,2:9
14Distribution of late gadolinium enhancement in various types of cardiomyopathies:Significance in differential diagnosis, clinical features and prognosis显示文摘The recent development of cardiac magnetic resonance(CMR)techniques has allowed detailed analyses of cardiac function and tissue characterization with high spatial resolution.We review characteristic CMR features in ischemic and non-ischemic cardiomyopathies(ICM and NICM),especially in terms of the location and distribution of late gadolinium enhancement(LGE).CMR in ICM shows segmental wall motion abnormalities or wall thinning in a particular coronary arterial territory,and the subendocardial or transmural LGE.LGE in NICM generally does not correspond to any particular coronary artery distribution and is located mostly in the mid-wall to subepicardial layer.The analysis of LGE distribution is valuable to differentiate NICM with diffusely impaired systolic function,including dilated cardiomyopathy,end-stage hypertrophic cardiomyopathy(HCM),cardiac sarcoidosis,and myocarditis,and those with diffuse left ventricular(LV)hypertrophy including HCM,cardiac amyloidosis and Anderson-Fabry disease.A transient low signal intensity LGE in regions of severe LV dysfunction is a particular feature of stress cardiomyopathy.In arrhythmogenic right ventricular cardiomyopathy/dysplasia,an enhancement of right ventricular(RV)wall with functional and morphological changes of RV becomes apparent.Finally,the analyses of LGE distribution have potentials to predict cardiac outcomes and response to treatments.Hiroshi Satoh Makoto Sano Kenichiro Suwa Takeji Saitoh Mamoru Nobuhara Masao Saotome Tsuyoshi Urushida Hideki Katoh Hideharu Hayashi 2014World Journal of Cardiology2014,6,7:8
15Green tea polyphenol epigallocatechin-3-gallate blocks PDGF-induced proliferation and migration of rat pancreatic stellate cells显示文摘AIM: To clarify the effects of epigallocatechin-3-gallate (EGCG) on the platelet-derived growth factor (PDGF)-BBinduced proliferation and migration of pancreatic stellate cells (PSCs).METHODS: PSCs were isolated from rat pancreas tissue and used in their culture-activated, myofibroblast-like phenotype. Cell proliferation was assessed by measuring the incorporation of 5-bromo-2'-deoxyuridine. Cell migration was assessed using modified Boyden chambers. CyclinD1, p21waf1, and p27kip1 expression and phosphorylation of PDGF β-receptor, extracellular signal-regulated kinase, and Akt were examined by Western blotting. Activation of phosphatidylinositol 3-kinase was examined by kinase assay using phosphatidylinositol as a substrate. Cell cycle was assessed by flow cytometry after staining with propidium iodide. RESULTS: EGCG at non-cytotoxic concentrations inhibited PDGF-induced proliferation and migration. This effect was associated with the inhibition of cell cycle progression beyondthe G1 phase, decreased cyclin D1 and increased p27Kip1 expression. 19Granular cell tumor of the pancreas:A case report and review of literature显示文摘Granular cell tumors,also called Abrikossof's tumors,were originally described by Abrikossof A in 1926.The f irst case of a pancreatic granular cell tumor was described in 1975 and only 6 cases have been reported.We describe a case of granular cell tumor in the pancreas showing pancreatic duct obstruction.Because imaging studies showed f indings compatible with those of pancreatic carcinoma,the patient underwent distal pancreatectomy.Histological examination showed that the tumor consisted of a nested growth of large tumor cells with ample granular cytoplasm and small round nuclei.The tumor cells expressed S-100 protein and were stained with neuron-specific enolase and periodic acid-Schiff,but were negative for desmin,vimentin,and cytokeratin.The resected tumor was diagnosed as a granular cell tumor.To our knowledge,this is the seventh case of Granular cell tumor of the pancreas to be reported.Atsushi Kanno Kennichi Satoh Morihisa Hirota Shin Hamada Jun Umino Hiromichi Itoh Atsushi Masamune Shinichi Egawa Fuyuhiko Motoi Michiaki Unno Kazuyuki Ishida Tooru Shimosegawa 2010World Journal of Gastrointestinal Oncology2010,2,2:5
20城市污水SBR法短程生物脱氮系统硝化菌群的定量分析显示文摘为了研究实时控制对短程生物脱氮中试系统内硝化菌群结构组成的影响,在考察系统常温条件下处理城市污水时短程硝化效果的同时,采用FISH、PCR-DGGE和PCR-克隆序列(Cloning Sequencing)分子生物学方法对SBR中试系统(有效容积为54 m^3)硝化菌群中的氨氧化菌(AOB)和亚硝酸氧化菌(NOB)进行定性与定量化分析.FISH结果表明,在短程脱氮系统中,AOB相比于NOB已成为明显的优势菌群,占总菌群的3%~12%;且没有检测出NOB.PCR-DGGE结果表明,SBR短程脱氮中试系统中的AOB均以Nitrosomonas-like为主.污泥样品的PCR-Cloning-Sequencing结果表明,所有的克隆相似于Nitrosomonas,其中60%以上的克隆相似于Nitro- somonas europaea.杨庆 彭永臻 曾薇 杨岸明 Hiroyasu Satoh Takashi Mino 2007北京工业大学学报2007,33,8:5
