1Impact of a single bout of high-intensity interval exercise and short-term interval training on interleukin-6, FNDC5, and METRNL mRNA expression in human skeletal muscle显示文摘Background: Exercise promotes numerous phenotypic adaptations in skeletal muscle that contribute to improved function and metabolic capacity. An emerging body of evidence suggests that skeletal muscle also releases a myriad of factors during exercise, termed 'myokines'. The purpose of this study was to examine the effects of high-intensity interval training(HIIT) on the acute regulation of the mRNA expression of several myokines, including the prototypical myokine interleukin-6(IL-6), and recently identified myokines fibronectin type III domain-containing protein 5(FNDC5)(irisin) and meteorin-like protein(METRNL).Methods: Both before and after a 20-day period of twice-daily high-volume HIIT, 9 healthy males(20.5 ± 1.5 years performed a standardized bout of high-intensity interval exercise(HIIE; 5 × 4 min at ~80% pretraining peak power output) with skeletal muscle biopsy samples(vastus lateralis) obtained at rest, immediately following exercise, and at 3 h recovery.Results: Before training, a single bout of HIIE increased IL-6(p < 0.05) and METRNL(p < 0.05) mRNA expression measured at 3 h recovery when compared to rest. Following 20 days of HIIT, IL-6 and FNDC5 mRNA were increased at 3 h recovery from the standardized HIIE bout when compared to rest(both p < 0.05). Resting METRNL and FNDC5 mRNA expression were higher following training(p < 0.05), and there was an overall increase in FNDC5 mRNA post-training(main effect of training, p < 0.05).Conclusion: In human skeletal muscle(1) an acute bout of HIIE can induce upregulation of skeletal muscle IL-6 mRNA both before and after a period of intensified HIIT;(2) Resting and overall FNDC5 mRNA expression is increased by 20 days of HIIT; and(3) METRNL mRNA expression is responsive to both acute HIIE and short-term intense HIIT. 3Rituximab for troublesome cases of childhood nephrotic syndrome显示文摘Nephrotic syndrome(NS) is the most common glomerular disease of childhood. Steroid-dependent and steroid-resistant nephrotic syndrome present challenges in their pharmaceutical management; patients may need several immunosuppressive medication for optimum control, each of which medication has its own safety profile. Rituximab(RTX) is a monoclonal antibody that targets B cells and has been used successfully for management of lymphoma and rheumatoid arthritis. Recent clinical studies showed that rituximab may be an efficacious and safe alternative for the treatment of complicated nephrotic syndrome. In this review article, we aim to review the efficacy and safety of RTX therapy in nephrotic syndrome. We reviewed the literature pertaining to this topic by searching for relevant studies on Pub Med and Medline using specific keywords. The initial search yielded 452 articles. These articles were then examined to ensure their relevance to the topic of research. We focused on multicenter randomized controlled trials with relatively large numbers of patients. A total of 29 articles were finally identified and will be summarized in this review. The majority of clinical studies of RTX in complicated pediatric NS showed that rituximab is effective in approximately 80% of patients with steroid-dependent NS, as it decreases the number of relapses and steroid dosage. However, RTX is less effective at achieving remission in steroid-resistant NS. RTX use was generally safe, and most side effects were transient and infusion-related. 5Identification of key gene networks controlling organic acid and sugar metabolism during watermelon fruit development by integrating metabolic phenotypes and gene expression profiles显示文摘The organoleptic qualities of watermelon fruit are defined by the sugar and organic acid contents,which undergo considerable variations during development and maturation.The molecular mechanisms underlying these variations remain unclear.In this study,we used transcriptome profiles to investigate the coexpression patterns of gene networks associated with sugar and organic acid metabolism.We identified 3 gene networks/modules containing 2443 genes highly correlated with sugars and organic acids.Within these modules,based on intramodular significance and Reverse Transcription Quantitative polymerase chain reaction(RT-qPCR),we identified 7 genes involved in the metabolism of sugars and organic acids.Among these genes,Cla97C01G000640,Cla97C05G087120 and Cla97C01G018840(r^(2)=0.83 with glucose content)were identified as sugar transporters(SWEET,EDR6 and STP)and Cla97C03G064990(r^(2)=0.92 with sucrose content)was identified as a sucrose synthase from information available for other crops.Similarly,Cla97C07G128420,Cla97C03G068240 and Cla97C01G008870,having strong correlations with malic(r^(2)=0.75)and citric acid(r^(2)=0.85),were annotated as malate and citrate transporters(ALMT7,CS,and ICDH).The expression profiles of these 7 genes in diverse watermelon genotypes revealed consistent patterns of expression variation in various types of watermelon.These findings add significantly to our existing knowledge of sugar and organic acid metabolism in watermelon.Muhammad Jawad Umer Luqman Bin Safdar Haileslassie Gebremeskel Shengjie Zhao Pingli Yuan Hongju Zhu M.O.Kaseb Muhammad Anees Xuqiang Lu Nan He Chengsheng Gong Wenge Liu 2020Horticulture Research2020,7,1:4
6Effect of seedling age and water depth on morphological and physiological aspects of transplanted rice under high temperature显示文摘To study the effect of high temperature, rice seedlings 20, 30, 40 and 50 d were kept at 5, 10, 15 and 20 cm water depth in a water pool. Meteorological findings indicated that water temperature varied up to 10 cm but became stable below this depth. Deep water inflicted higher tiller mortality, minimal increase in dry weight of aerial parts and leaf area, decrease in root length, and decrease in root dry weight especially at 20 cm water depth and produced an unbalanced T/R ratio (top versus root dry weight). However, deep water tended to increase plant length. These parameters, however, excel in shallow water. Older seedlings, with the exception of root dry weight, could not perform well compared to young seedlings in all physiological and morphological aspects. 7Growth and yield of rice as affected by transplanting dates and seedlings per hill under high temperature of Dera Ismail Khan, Pakistan显示文摘Studies were initiated for two consecutive years to find out the effect of time of transplanting and seedlings hill?1 on the productivity of rice in Dera Ismail Khan district of North West Frontier Province (NWFP), Pakistan. The experiment was laid out in a randomized complete block design with split plot arrangements. Main plots consisted of four transplanting dates viz. 20th and 27th of June and 4th and 11th of July while sub-plots contained 1, 2, 3 or 4 seedlings hill?1. Among transplanting dates, June 20th planted crop gave highest paddy yield and net return with 1 seedling hill–1. It explains that the use of more seedlings hill?1 not only adds to cost but is also a mere wastage of natural resources. 8Mutation of the PHYTOENE DESATURASE 3 gene causes yellowish-white petals in Brassica napus显示文摘Oilseed rape (Brassica napus) with yellow flowers is an attractive ornamental landscape plant during the flowering period,and the development of different petal colors has become a breeding objective.Although yellowish flower color is a common variant observed in field-grown oilseed rape,the genetics behind this variation remains unclear.We obtained a yellowish-white flower (ywf) mutant from Zhongshuang 9 (ZS9) by ethyl methanesulfonate mutagenesis (EMS) treatment.Compared with ZS9,ywf exhibited a lower carotenoid content with a reduced and defective chromoplast ultrastructure in the petals.Genetic analysis revealed that the yellowish-white trait was controlled by a single recessive gene.Using bulked-segregant analysis sequencing (BSA-seq) and kompetitive allele-specific PCR(KASP),we performed map-based cloning of the ywf locus on chromosome A08 and found that ywf harbored a C-to-T substitution in the coding region,resulting in a premature translation termination.YWF,encoding phytoene desaturase 3 (PDS3),was highly expressed in oilseed rape petals and involved in carotenoid biosynthesis.Pathway enrichment analysis of the transcriptome profiles from ZS9 and ywf indicated the carotenoid biosynthesis pathway to be highly enriched.Further analyses of differentially expressed genes and carotenoid components revealed that the truncated Bna A08.PDS3 resulted in decreased carotenoid biosynthesis in the mutant.These results contribute to an understanding of the carotenoid biosynthesis pathway and manipulation of flower-color variation in B.napus.Chuanji Zhao Luqman Bin Safdar Meili Xie Meijuan Shi Zhixue Dong Li Yang Xiaohui Cheng Yueying Liu Zetao Bai Yang Xiang Chaobo Tong Junyan Huang Lijiang Liu Shengyi Liu 2021The Crop Journal2021,9,5:3
9Effect of Establishment Methods and Weed Management Practices on Some Growth Attributes of Rice显示文摘Studies were carried out for two years to evaluate the effect of methods of sowing and weed control practices on the productivity of transplanted and direct wet-seeded rice in Dera Ismail Khan, NWFP, Pakistan. The experiment was laid out in a randomized complete block design with a split plot arrangement. The planting techniques viz. transplanting and direct seeding were maintained in main plots while weed control practices included the use of granular herbicide Sunstar 15WG (ethoxy sulfuron), Machete 60EC (butachlor), conventional hand weeding, and the weedy check (control) were assigned to the sub-plots. Data were recorded on weed parameters like weed density and dry weed biomass 60 and 90 days after sowing (DAS); agronomic parameters including plant population, number of panicles and paddy yield and physiological parameters like leaf area index and net assimilation rate 45 and 90 DAS. The planting methods and weed management significantly influenced most of the parameters studied. The data revealed that the paddy yield and its components were significantly higher in the transplanted method than that in direct-seeded method, while the weed density and biomass were lower in the transplanted plots than the direct-seeded plots.
