| 1 | Single-cell transcriptomic atlas of primate cardiopulmonary aging显示文摘Aging is a major risk factor for many diseases,especially in highly prevalent cardiopulmonary comorbidities and infectious diseases including Coronavirus Disease 2019(COVID-19).Resolving cellular and molecular mechanisms associated with aging in higher mammals is therefore urgently needed.Here,we created young and old non-human primate single-nucleus/cell transcriptomic atlases of lung,heart and artery,the top tissues targeted by SARS-CoV-2.Analysis of cell type-specific aging-associated transcriptional changes revealed increased systemic inflammation and compromised virus defense as a hallmark of cardiopulmonary aging.With age,expression of the SARS-CoV-2 receptor angiotensin-converting enzyme 2(ACE2)was increased in the pulmonary alveolar epithelial barrier,cardiomyocytes,and vascular endothelial cells.We found that interleukin 7(IL7)accumulated in aged cardiopulmonary tissues and induced ACE2 expression in human vascular endothelial cells in an NF-κB-dependent manner.Furthermore,treatment with vitamin C blocked IL7-induced ACE2 expression.Altogether,our findings depict the first transcriptomic atlas of the aged primate cardiopulmonary system and provide vital insights into age-linked susceptibility to SARS-CoV-2,suggesting that geroprotective strategies may reduce COVID-19 severity in the elderly. | Shuai Ma Shuhui Sun Jiaming Li Yanling Fan Jing Qu Liang Sun Si Wang Yiyuan Zhang Shanshan Yang Zunpeng Liu Zeming Wu Sheng Zhang Qiaoran Wang Aihua Zheng Shuguang Duo Yang Yu Juan Carlos Izpisua Belmonte Piu Chan Qi Zhou Moshi Song Weiqi Zhang Guang-Hui Liu | 2021 | Cell Research2021,31,4: | 10 |
| 2 | Flow Control Effect of Spanwise Distributed Pulsed Arc Discharge Plasma Actuation on Supersonic Compressor Cascade Flow显示文摘To achieve efficient control of supersonic compressor cascade flow,a type of spanwise distributed pulsed arc discharge plasma actuation(PADPA)was designed.To simulate the influences of PADPA on the flow field,a phenomenological model was established.Then,the flow control effects of PADPA on supersonic compressor cascade flow were researched numerically.The results show that under low static pressure ratio condition,the compressive wave induced by PADPA reduced the intensity of the passage shock wave,which eventually reduced shock wave loss.It was also found that PADPA produced an adverse pressure gradient(pre-compression effect)around the actuation location,which reduced the strength of the high adverse pressure gradient induced by the passage shock wave.The airflow on both sides of the actuation location was accelerated by PADPA owing to the spanwise distributed layout.Thus,it improved the ability of the boundary layer to resist the effect of the adverse pressure gradient and reduced the separation zone.Consequently,the total pressure loss was reduced by 6.8%.Under high pressure ratio condition,the effect of PADPA on the suction side controlling the large separation of the boundary layer was insignificant.The total pressure loss also increased slightly. | SHENG Jiaming WU Yun ZHANG Haideng WANG Yizhou TANG Mengxiao | 2022 | Journal of Thermal Science2022,31,5: | 1 |

| 5 | Genome-wide identification of the MATE gene family and functional characterization of PbrMATE9 related to anthocyanin in pear显示文摘Plant multidrug and toxic compound extrusion(MATE) genes play an important role in the process of detoxification, plant morphogenesis, and anthocyanin accumulation. However, whether the MATE gene family functions in pear peel coloration is still unknown. To evaluate and identify the MATE gene family members which are involving in anthocyanin accumulation and coloration in pear. In this study, 85 MATE genes were identified in the reference pear genome of ‘Dangshansuli’ through genome-wide identification. Based on gene structure and phylogenetic tree analysis, the MATE family was divided into five subfamilies. RNA sequencing and quantitative real-time polymerase chain reaction(qRTPCR) indicated that the expression patterns of PbrMATEs were tissue-specific. 28.24%(24) of PbrMATE genes were expressed in the fruits, and44.71%(38) of PbrMATE genes were expressed in the leaves. Additionally, we found that the expression levels of PbrMATE9, PbrMATE26,PbrMATE50, and PbrMATE69 in debagged fruits with red peel were significantly higher than those in bagged fruits without red peel, according to our bagging/debagging treatment of ‘Mantianhong’. The expression pattern of PbrMATE9 was consistent with the variation trend in anthocyanin content, suggesting that it might play an important role in anthocyanin accumulation in response to light exposure. Subcellular localization showed that PbrMATE9 was a membrane protein. More strikingly, the transient overexpression of PbrMATE9 promoted anthocyanin accumulation in the peel of pear, and the expression of structural genes(PbrCHI, PbrANS, PbrDFR, and PbrUFGT) in the anthocyanin biosynthesis pathway also increased significantly. Through co-expression network analysis, the transcription factors were identified, such as WRKY, COL,GATA, and BBX, which might be involved in the regulation of PbrMATE9. | 6 | Traffic signal coordination control for arterials with dedicated CAV lanes显示文摘Purpose–This study aims to make full use of the advantages of connected and autonomous vehicles(CAVs)and dedicated CAV lanes to ensure all CAVs can pass intersections without stopping.Design/methodology/approach–The authors developed a signal coordination model for arteries with dedicated CAV lanes by using mixed integer linear programming.CAV non-stop constraints are proposed to adapt to the characteristics of CAVs.As it is a continuous problem,various situations that CAVs arrive at intersections are analyzed.The rules are discovered to simplify the problem by discretization method.Findings–A case study is conducted via SUMO traffic simulation program.The results show that the efficiency of CAVs can be improved significantly both in high-volume scenario and medium-volume scenario with the plan optimized by the model proposed in this paper.At the same time,the progression efficiency of regular vehicles is not affected significantly.It is indicated that full-scale benefits of dedicated CAV lanes can only be achieved with signal coordination plans considering CAV characteristics.Originality/value–To the best of the authors’knowledge,this is the first research that develops a signal coordination model for arteries with dedicated CAV lanes. | Liang Xu Sheng Jin Bolin Li Jiaming Wu | 2022 | Journal of Intelligent and Connected Vehicles2022,5,2: | 0 |
