1连接肽在融合蛋白设计中的选择及应用显示文摘融合蛋白的设计中,经常需要用链接肽将不同功能的蛋白进行连接融合。连接肽的设计和选择对融合蛋白的表达、稳定性及功能活性至关重要。文章将常见的两种连接肽分为柔性(Flexibility)和刚性(Rigidity)进行比较说明。其中柔性连接肽即成柔软线性的一类易弯折的氨基酸序列,常见为(GGGGS)。重复序列(n=3—4);刚性连接肽即成稳固螺旋结构的一类长度固定的氨基酸序列,常见为(EAAAK)。重复序列(n=2~5)。设计连接肽时应考虑长度、氨基酸成分、亲水性、构象、柔韧性及电荷等因素。连接肽中的氨基酸组成对柔性连接肽影响较大,而氨基酸排列影响着刚性连接肽的结构。文章结合近年来融合蛋白设计中两种最常用连接肽的相关报道进行阐述,为连接肽的研究提供参考。于健 Sarra Setrerrahmane 徐寒梅 2016药物生物技术2016,23,3:13
2Trends in insulin resistance:insights into mechanisms and therapeutic strategy显示文摘The centenary of insulin discovery represents an important opportunity to transform diabetes from a fatal diagnosis into a medically manageable chronic condition.Insulin is a key peptide hormone and mediates the systemic glucose metabolism in different tissues.Insulin resistance(IR)is a disordered biological response for insulin stimulation through the disruption of different molecular pathways in target tissues.Acquired conditions and genetic factors have been implicated in IR.Recent genetic and biochemical studies suggest that the dysregulated metabolic mediators released by adipose tissue including adipokines,cytokines,chemokines,excess lipids and toxic lipid metabolites promote IR in other tissues.IR is associated with several groups of abnormal syndromes that include obesity,diabetes,metabolic dysfunction-associated fatty liver disease(MAFLD),cardiovascular disease,polycystic ovary syndrome(PCOS),and other abnormalities.Although no medication is specifically approved to treat IR,we summarized the lifestyle changes and pharmacological medications that have been used as efficient intervention to improve insulin sensitivity.Ultimately,the systematic discussion of complex mechanism will help to identify potential new targets and treat the closely associated metabolic syndrome of IR.Mengwei Li Xiaowei Chi Ying Wang Sarra Setrerrahmane Wenwei Xie Hanmei Xu 2022Signal Transduction and Targeted Therapy2022,7,8:5
3Centranthera grandiflore alleviates alcohol-induced oxidative stress and cell apoptosis显示文摘Alcohol liver disease(ALD)has become a global threat to human health.It is associated with a wide range of liver diseases including alcohol fatty liver,steatosis,fibrosis and cirrhosis,and finally leads to liver cancer and even death.Centranthera grandiflora is a traditional Chinese medicinal herb commonly used to treat ALD,but no research about its mechanism is available.This study evaluated the hepatoprotective effect and mechanism of C.grandiflora against alcohol-induced liver injury in mice.We found that the ethanol extracts of C.grandiflora(CgW)alleviated the alcohol-induced liver injury,enhanced the levels of antioxidant enzymes,and reduced the amount of lipid peroxides.CgW also affected cell apoptosis by inhibiting the activity of Bax,cleaved-caspase 3 and cleaved-caspase 9,and increasing the activity of Bcl-2.In conclusion,the results showed that CgW can effectively improve ALD through alleviating oxidative stress and inhibiting cell apoptosis for the first time.This study suggested that C.grandiflora is a promising herbal medicine for ALD treatment.GAO Xin-Mei DONG Wen-Han XIA Chun-Lei MA Zhuo-Yi WANG Ying SARRA Setrerrahmane XU Han-Mei QI Wei-Yan 2022Chinese Journal of Natural Medicines2022,20,8:2
4Intein-mediated purification system: mechanism and applications显示文摘The incorporation of self-cleaving protein elements into a variety of fusion-based purification systems; has been an important development in the area of recombinant protein purification. The self-cleaving capability of these tags has recently been combined with additional purification tags to generate novel and convenient protein purification methods.
