1Genesis of sandstone-type uranium deposits along the northern margin of the Ordos Basin,China显示文摘Sandstone-type U mineral resources are among the important sources for nuclear energy.The U deposits in the Ordos Basin in China form part of the northern segment of the sandstone-hosted Central Asian Uranium MegaProvince.Two types of mineralizations are recognized in this basin:'phreatic permeable type'and'interlayer permeable type',both exhibiting features equivalent to roll-front subtypes.The'interlayer permeable type'is widely accepted as the dominant mineralization type for sandstone-type uranium deposits within large-scale basins,also designated as the'interlayer oxidation zone type',based on the horizontal color zoning model representing changing redox conditions.Here we synthesize data from several drill holes within the Ordos Basin,which suggest that major Mesozoic tectonic movements controlled the evolution of the sedimentary system in the basin.These tectonic movements contributed to the formation of three angular unconformities and four parallel unconformities as inferred from the stratigraphic relationships.In addition,other features such as vertical color zoning,paleo-channel controlled tabular or lentoid ore bodies(without roll-type)and a group interlayer horizontal zoning of altered minerals are also documented.Sequence stratigraphic analysis indicates that the Ordos Basin generally witnessed four cycles of water level variations during Mesozoic.During the variations,three high water level and three low water level events were recorded.Biological characteristics imply that the Ordos Basin went through multiple arid to humid climatic evolutions during Mesozoic.Combining the newly documented features with some novel concepts on the hydrodynamic mechanism for supergene ore-forming fluids,we propose a metallogenic model which invokes the importance of tectonic movements and water level fluctuations to explain the genesis of uranium deposits along the northern margin of the Ordos Basin.Ruoshi Jin Xueming Teng Xiaoguang Li Qinghong Si Wei Wang 2020Geoscience Frontiers2020,11,1:13
2Ratiometric delivery of doxorubicin and berberine by liposome enables superior therapeutic index than Doxil?显示文摘Although the appearance of Doxil alleviated the cardiotoxicity of DOX, the progression-free survival of patients was not prolonged compared with traditional medication regimens, and side effects such as hand-foot syndrome has occurred. In order to solve this dilemma, we have designed a novel co-delivery strategy to construct a co-loaded liposome of berberine(BER) and doxorubicin(DOX), which was called Lipo Be Do. The optimal synergistic ratio of the two drugs was screened by cell cytotoxicity experiments in vitro, and the optimal attenuation ratio was further determined by in vivo cardiac H&E staining pathological sections. The optimal combination treatment caused a robust increase in apoptotic cells of 4T1, as compared to drug alone treatment. The prepared co-loaded liposome, Lipo Be Do, had high encapsulation efficiency and good stability. The nanoliposome carrier controlled the biological fate of the drugs and maintained a pre-defined optimal ratio in vivo. The Lipo Be Do significantly inhibited tumor growth in 4T1 murine mammary carcinoma model compared with Doxil(P < 0.05), and completely overcame the myocardial rupture toxicity caused by Doxil in mice. 3Ecosystem health towards sustainability显示文摘Ecosystems are becoming damaged or degraded as a result of stresses especially associated with human activities.A healthy ecosystem is essential to provide the services that humans and the natural environment require and has tremendous social and economic value.Exploration of the definition of ecosystem health includes what constitutes health and what it means to be healthy.To evaluate ecosystem health,it is necessary to quantify ecosystem conditions using a variety of indicators.In this paper,the main principles and criteria for indicator selection,classification of indicators for different kinds of ecosystems,the most appropriate indicators for measuring ecosystem sustainability,and various methods and models for the assessment of ecosystem health are presented.Drivers,sustainability,and resilience are considered to be critical factors for ecosystem health and its assessment.Effective integration of ecological understanding with socioeconomic,biophysical,biogeochemical,and public-policy dimensions is still the primary challenge in this field,and devising workable strategies to achieve and maintain ecosystem health is a key future challenge.Yonglong Lu Ruoshi Wang Yueqing Zhang Hongqiao Su Pei Wang Alan Jenkins Rober C.Ferrier Mark Bailey Geoff Squire 2015Ecosystem Health and Sustainability2015,1,1:3

5METTL3-mediated m^(6)A RNA methylation regulates dorsal lingual epithelium homeostasis显示文摘The dorsal lingual epithelium,which is composed of taste buds and keratinocytes differentiated from K14^(+)basal cells,discriminates taste compounds and maintains the epithelial barrier.N6-methyladenosine(m^(6)A)is the most abundant mRNA modification in eukaryotic cells.How METTL3-mediated m^(6)A modification regulates K14^(+)basal cell fate during dorsal lingual epithelium formation and regeneration remains unclear.Here we show knockout of Mettl3 in K14^(+)cells reduced the taste buds and enhanced keratinocytes.Deletion of Mettl3 led to increased basal cell proliferation and decreased cell division in taste buds.Conditional Mettl3 knock-in mice showed little impact on taste buds or keratinization,but displayed increased proliferation of cells around taste buds in a protective manner during post-irradiation recovery.Mechanically,we revealed that the most frequent m^(6)A modifications were enriched in Hippo and Wnt signaling,and specific peaks were observed near the stop codons of Lats1 and FZD7.Our study elucidates that METTL3 is essential for taste bud formation and could promote the quantity recovery of taste bud after radiation.Qiuchan Xiong Caojie Liu Xin Zheng Xinyi Zhou Kexin Lei Xiaohan Zhang Qian Wang Weimin Lin Ruizhan Tong Ruoshi Xu Quan Yuan 2022International Journal of Oral Science2022,14,3:2

8Decoding early myelopoiesis from dynamics of core endogenous network显示文摘A decade ago mainstream molecular biologists regarded it impossible or biologically ill-motivated to understand the dynamics of complex biological phenomena, such as cancer genesis and progression, from a network perspective. Indeed, there are numerical difficulties even for those who were determined to explore along this direction. Undeterred, seven years ago a group of Chinese scientists started a program aiming to obtain quantitative connections between tumors and network dynamics. Many interesting results have been obtained. In this paper we wish to test such idea from a different angle: the connection between a normal biological process and the network dynamics. We have taken early myelopoiesis as our biological model. A standard roadmap for the cell-fate diversification during hematopoiesis has already been well established experimentally, yet little was known for its underpinning dynamical mechanisms. Compounding this difficulty there were additional experimental challenges, such as the seemingly conflicting hematopoietic roadmaps and the cell-fate inter-conversion events. With early myeloid cell-fate determination in mind, we constructed a core molecular endogenous network from well-documented gene regulation and signal transduction knowledge. Turning the network into a set of dynamical equations, we found computationally several structurally robust states. Those states nicely correspond to known cell phenotypes. We also found the states connecting those stable states.They reveal the developmental routes—how one stable state would most likely turn into another stable state. Such interconnected network among stable states enabled a natural organization of cell-fates into a multi-stable state landscape. Accordingly, both the myeloid cell phenotypes and the standard roadmap were explained mechanistically in a straightforward manner. Furthermore,recent challenging observations were also explained naturally. Moreover, the landscape visually enables a prediction of a pool of additional cell states and developmental routes, including the non-sequential and cross-branch transitions, which are testable by future experiments. 10METTL5 regulates cranial suture fusion via Wnt signaling显示文摘METTL5 is a methyltransferase that mediates eukaryotic 18S ribosomal RNA m^(6)A modification,and its mutations lead to intellectual disability,microcephaly,and facial dysmorphism in patients.However,the role of METTL5 in craniofacial development remains poorly understood.This study demonstrates that Mettl5 knockout mice exhibit poor ossification,widened cranial sutures,and a cleidocranial dysplasia-like phenotype.Deletion of Mettl5 leads to increased proliferation and decreased osteogenic differentiation of suture mesenchymal stem cells.Mechanistically,we find that Wnt signaling is significantly downregulated after Mettl5 knockout.Overall,we reveal an essential role of METTL5 in craniofacial development and osteogenic differentiation of suture mesenchymal stem cells,making METTL5 a potential diagnostic and therapeutic target for craniofacial developmental diseases.Kexin Lei Ruoshi Xu Qian Wang Qiuchan Xiong Xinyi Zhou Qiwen Li Dutmanee Seriwatanachai Shuibin Lin Chenchen Zhou Quan Yuan 2023Fundamental Research2023,3,3:0
