| 1 | Geological Features and Reservoiring Mode of Shale Gas Reservoirs in Longmaxi Formation of the Jiaoshiba Area显示文摘This study is based on the sedimentation conditions, organic geochemistry, storage spaces, physical properties, lithology and gas content of the shale gas reservoirs in Longmaxi Formation of the Jiaoshiba area and the gas accumulation mode is summarized and then compared with that in northern America. The shale gas reservoirs in the Longmaxi Formation in Jiaoshiba have good geological conditions, great thickness of quality shales, high organic content, high gas content, good physical properties, suitable depth, good preservation conditions and good reservoir types. The quality shales at the bottom of the deep shelf are the main target interval for shale gas exploration and development. | 3 | Expression of the Phycoerythrin Gene of Gracilaria lemaneiformis (Rhodophyta) in E. coli and Evaluation of the Bioactivity of Recombinant PE显示文摘Phycoerythrin (PE) is one of the most important proteins involved in light capturing during photosynthesis in red algae. Its potential biological activities had gained wide concerns. In the present study, tumor cytotoxic and hydroxyl radical assay were preformed to detect the bioactivity of recombinant PE. Recombinant plasmids pGEX-PE and pBGL were transformed into E.coli BL21 to make two recombinant strains BEX (pGEX-PE) and BGL (pBGL). PE expressing in BEX (pGEX-PE) was validated by SDS-PAGE and Western blotting analysis. SDS-PAGE analysis indicated that the PE-GST fusion protein was mostly inclusion bod- ies. Specific expression of PE was confirmed by Western blotting analysis. The recombinant E. coli BEX (pGEX-PE) cells were col- lected and sonicated. The supernatants were reserved for the tumor cytotoxic experiments. The result of tumor cytotoxic assay indi- cated that the supernatants containing PE had the activity of inhibiting the growth of Hela cells and with the increase of protein con- centration, the inhibiting rate increased from 37.31% to 63.26%, which showed significant difference from the control. Hydroxyl radical scavenging effect was tested with supernatants of BEX (pGEX-PE) and BGL (pBGL) cell lysates treated with sonication and heating. For the sonication samples, the scavenging rates of the supernatants of BEX (pGEX-PE) and BGL (pBGL) cell lysates were significantly higher than the negative control BL21(pGEX-4T) (P<0.02), and the scavenging rates increased slowly following the increase of the protein content. For the heating samples, except for the 0.2 mg mL-1 BGL (pBGL) products, the scavenging effects of the supernatants of BEX (pGEX-PE) and BGL (pBGL) cell lysates were stronger than that of negative control BL21(pGEX-4T). However, the effect intensity was not positively correlated with the increase of the protein concentration. Though a partially de- creased hydroxyl radical scavenging activity was led by heating, the biological activity was still retained and conspicuous. | 4 | Prognostic value of clinicopathological characteristics in patients with pancreatic cancer显示文摘Background:The purpose of this study was to analyze me effects of all clinical characteristics on the overall survival time,in order to provide a basis for determining the prognostic factor of patients with pancreatic cancer.Methods:A total of 103 pancreatic cancer patients were admitted to the Department of Radiotherapy and Chemotherapy of the Ruijin Hospital,Shanghai Jiaotong University School of Medicine,between January 2002 and December 2012.There were 68 men and 35 women;the median age was 62 years.Diagnoses of pancreatic cancer in all patients were confirmed by histopathology,cytology,or clinical diagnosis.The Kaplan-Meier method was performed to calculate the overall survival rate.The log-rank method was used to examine the univariate analysis.The Cox regression model was performed for multivariate analysis.Results:The median survival time was 293 days,the 1-,2-,and 3-year survival rates were 27.18%,5.83%,and 1.94%,respectively.Cox regression analysis revealed that age(P=0.015),Karnofsky performance status(PS)(P=0.002),surgical types(P<0.001),and platelet counts(P<0.001) were independent prognostic factors affecting the overall survival of patients with pancreatic cancer.Conclusions:Pancreatic cancer had a poor prognosis,the general physical condition,age,the availability of radical surgery,and platelet counts were factors influencing the overall survival of patients with pancreatic cancer. | Mei Geng Haoping Xu Ruobing Ren Qing Qu Chengfang Shangguan Junwei Wu Jinsong Jiang Hao Li Weiguo Cao | 2015 | Chinese Journal of Cancer Research2015,27,5: | 4 |
| 5 | Temporal lobe epilepsy animal model established by stereotaxic microinjection of kainic acid显示文摘BACKGROUND: Kainic acid can be used to induce a model of epilepsy by systemic injection, such as intraperitoneal or subcutaneous injection. Individual rats have different responses to kainic acid, therefore high doses of drug are required and the success rate of model induction is low. It is necessary to develop an improved method to establish a temporal lobe epilepsy (TLE) animal model. OBJECTIVE: To explore an economic, stable and efficient method of establishing a TLE animal model. DESIGN, TIME AND SETTING: A completely randomized, controlled study. The experiments were performed in the Cellular Function Laboratory of the Physiology Department, Anhui Medical University from March to July 2007. MATERIALS: Twenty adult male Wistar rats, weighing 230–260 g, were provided by the Experimental Animal Centre of Nanjing Medical University. Kainic acid was purchased from Sigma in USA. Type SN-2 stereotaxic apparatus was made by Narishge in Japan. METHODS: Wistar rats were randomly divided into a kainic acid (KA) group (n = 12) and a normal saline (NS) group (n = 8). For intrahippocampal microinjection, a burr hole was drilled in the skull at the following stereotaxic coordinates: anteroposterior (AP) 4.1 mm caudal to bregma; lateral (ML) 4.2 mm right lateral to the midline. Rats in the KA group were injected with 2.5 μL KA (0.4 g/L) into the center of the CA3 region, while in the NS group the same volume of NS was injected into the same site. MAIN OUTCOME MEASURES: Both groups were monitored under a video capture system for 12 weeks to record spontaneous seizures. Intracranial eletroencepholograph (IEEG) recordings in vivo were performed after the behavioral observations. After the IEEG recordings, hippocampi were processed into coronal sections. Nissl and Timm stainings were then performed to observe and confirm pathology. RESULTS: Twenty rats were involved in the final analysis. Behavioral observations: the earliest spontaneous onset of epilepsy appeared 2 weeks after injection of KA. Eight rats had spontaneous onset of epilepsy 3–12 weeks after treatment. None of rats in the NS group had spontaneous onset of epilepsy. IEEG recordings: Epileptic-form waves, such as sharp waves and spike waves, were calculated by artificial analysis. The number of epileptic-form waves in the KA group increased significantly compared to those of the NS group (P < 0.01). Morphology results: In the KA group, Nissl staining and Timm staining revealed typical pathology in the hippocampal temporosphenoid lobe. In the NS group, no pathology was observed. | 6 | Mode transition induced by back-breakdown of the gliding arc and its influence factors显示文摘Gliding arc is a mostly used non-equilibrium plasma generation method whose characteristic is affected by the backbreakdown phenomenon.Mode transition induced by back-breakdown of the gliding arc is studied in this work and effects of gas flow rate,applied voltage,electrode parameters on mode transition were studied.Experimental results show that there are two typical modes during the development of the gliding arc.Mode A comprises periodicitical development of the arc from the minimum gap breakdown to the longest extinguishment,while the arc in mode B continues to produce a back-breakdown at the ends of the electrodes.As the flow rate descends,the gliding arc gradually changes from mode A to B.It is the decrease of the arc velocity caused by lower flow rate leads to the occurrence of back-breakdown,which generates mode B.Smaller electrode opening angle,shorter length and wider minimum gap reduce the gliding speed,so that arc is more likely to enter mode B.As the applied voltage is increased,enhancing of the electric field strength on the breakdown path of the back-breakdown and thickening of the arc's diameter allow the gliding arc to enter mode B at a higher arc speed. | Ruobing Zhang Haochen Huang Tianshu Yang | 2020 | High Voltage2020,5,3: | 3 |
| 7 | Sphingosine-1-phosphate,a novel TREM2 ligand,promotes microglial phagocytosis to protect against ischemic brain injury显示文摘The mechanism of sphingosine-1-phosphate(S1P)-mediated phagocytosis remains unknown.Here,we found that S1P or FTY720(an analog of S1P)promoted microglial phagocytosis in stroke independent of S1PRs.First,we used computer simulation of molecular docking to predict that S1P might be a ligand for triggering receptor expressed on myeloid cells 2(TREM2).Next,microscale thermophoresis(MST),surface plasmon resonance(SPR)and liquid chromatography–tandem mass spectrometry(LC–MS/MS)were performed to reveal that S1P was a novel TREM2 ligand.Then,we confirmed the pro-phagocytosis of S1P targeting in Trem2-Dap12 transfected CHO cells and TREM2 knockdown microglia.Point mutation analysis showed that D104 was the critical binding residue.Trem2^(−/−)mice were used to demonstrate the role of S1P-induced phagocytosis targeting on TREM2 in protecting against ischemic brain injury.Finally,further studies revealed that apolipoprotein E(APOE)loaded with S1P was released by microglia and bound to apoptotic neurons via LDL receptor related protein 1B(LRP1B)and thereby induced microglia to phagocytose apoptotic neurons.Overall,the present work reveals for the first time that S1P acts as a novel endogenous ligand of TREM2 to effectively promote microglial phagocytosis.Our findings provide a new lead compound for developing immunomodulator targeting on TREM2. | Tengfei Xue Juan Ji Yuqin Sun Xinxin Huang Zhenyu Cai Jin Yang Wei Guo Ruobing Guo Hong Cheng Xiulan Sun | 2022 | Acta Pharmaceutica Sinica B2022,12,4: | 3 |

| 9 | Ultrasound-guided percutaneous microwave ablationfor small liver cancers adjacent to large vessels:long-term outcomes and strategies显示文摘Objective The aim of the study was to evaluate the long-term efficacy and safety of percutaneous microwave ablation(MWA) for small hepatic cancers adjacent to large vessels and to investigate the treatment strategies.Methods From March 2009 to July 2015,a total of 86 patients with 94 tumors underwent ultrasound(US)-guided percutaneous MWA,with pathologically proven or clinically diagnosed liver cancers measuring ≤ 3 cm in diameter and located ≤ 10 mm from a major vessel(n = 94).Regular follow-up after MWA was performed to assess treatment efficacy and perioperative complications.Results The complete ablation rate at 1 month after MWA was 93.3%(84/90).The 6-,9-,12-,24-,36-,48-,60-,72-,and 84-month local recurrence rates were 2.4%,2.4%,3.7%,6.6%,8.4%,8.4%,8.4%,8.4%,and 8.4%,respectively.There were no major complications.The perioperative special complication rate was 5.32%(5/94),including 3 cases of moderate liver function damage and 2 cases of limited sub-capsular hematoma.Conclusion Percutaneous MWA for small hepatic cancers adjacent to large vessels is feasible,effective,and safe with an acceptable rate of complications.The key point is to strictly follow operative indications and adopt proper treatment strategies. | Ruobing Liu Kaiyan Li Hongchang Luo Wei Zhang Tingting Zhang Meng Gao Wenhui Zha Xinwu Cui Youbin Deng | 2017 | Oncology and Translational Medicine2017,3,2: | 3 |
