| 1 | Epalrestat protects against diabetic peripheral neuropathy by alleviating oxidative stress and inhibiting polyol pathway显示文摘Epalrestat is a noncompetitive and reversible aldose reductase inhibitor used for the treatment of diabetic neuropathy. This study assumed that epalrestat had a protective effect on diabetic peripheral nerve injury by suppressing the expression of aldose reductase in peripheral nerves of diabetes mellitus rats. The high-fat and high-carbohydrate model rats were established by intraperitoneal injection of streptozotocin. Peripheral neuropathy occurred in these rats after sustaining high blood glucose for 8 weeks. At 12 weeks after streptozotocin injection, rats were intragastrically administered epalrestat 100 mg/kg daily for 6 weeks. Transmission electron microscope revealed that the injuries to myelinated nerve fibers, non-myelinated nerve fibers and Schwann cells of rat sciatic nerves had reduced compared to rats without epalrestat administuation. Western blot assay and immunohistochemical results demonstrated that after intervention with epalrestat, the activities of antioxidant enzymes such as superoxide dismutase, catalase and glutathione peroxidase gradually increased, but aldose reductase protein expression gradually diminished. | 2 | Identifying paleoflood deposits archived in Zhongba Site,the Three Gorges reservoir region of the YangtzeRiver,China显示文摘Based on the principle that the present is the key to the past , detailed analyses, such as AMS 14C dating, grain size, component and morphology of heavy minerals, micro-morphology of zircon, Rb/Sr, magnetic susceptibility and total organic carbon (TOC), were conducted to identify paleoflood sediments archived in Zhongba Site. The results indicate that the plaeoflood sediments bear great similarities with modern flood sediments in the following aspects: (1) probability cumulative curves mainly show a pattern of 3―4 segments; (2) grain-size distribution of suspended matter ranges between 3 and 10Ф; (3) the sediments are well-sorted, most of which are suspended matter (>50%); (4) the same species, quantity and morphology of heavy minerals; (5) scanning electronic microscope images show that shapes of zircon are mainly oval and nearly spheral, rounded due to long-distance transport; (6) higher Rb/Sr values (0.55―0.66)than those of sediments from cultural layers (0.03―0.26); (7) magnetic susceptibility values (133.73―433.05 10-6m3/kg) are lower than those of sediments from cultural layers (959.25―2442.44 10?6 m3/kg); (8) TOC (0.14%―0.33%) are lower than those of sediments from cultural lay-ers (1.13%―2.95%). | 3 | High-resolution geochemistry records of climate changes since late-glacial from Dajiuhu peat in Shennongjia Mountains,Central China显示文摘A peat core with depths of 297 cm was obtained from the Dajiuhu Basin(31°29′27″N,109°59′45″E,1760 m)in Hubei Province,Central China.10 AMS ages provide a time control and reveal that this core spans the past about 16.0 kaBP(calibrated age)(1 4C age:13.3 kaBP).Multi-proxy indexes analysis of geochemistry shows the following character of climate and environmental changes since about 16 kaBP:(1) The climate during the late-glacial period was cold and wet as a whole,but fluctuated continually.11.4- 12.6 cal.kaBP,12.6-15.2 cal.kaBP and 15.2-16 kaBP were corresponding respectively to the Younger Drays,Bφlling-Allerφd Warm Period and the Oldest Drays.(2)Inheriting the some climate characteristics of the late-glacial,the climate during the early-Holocene was wet and temperature increased gradually,during which an obvious dry event around 10.6 cal.kaB appeared.(3)The climate during the mid-Holocene was genarally warm and wet.During 9.2-7.5 kaBP,temperature increased gradually, precipitation was less comparatively and the 8.2 kaBP cold event which might be representative in the globe was reflected markedly.Then,multi-proxy records were relatively stable during 6.7-4.2 kaBP, which shows the best water and thermal condition in the Holocene Optimum.(4)Around 4.2 kaBP,the climate and environment transform from warm and wet to cool and dry,which may result in the collapse of the Neolithic Culture and midwifery the civilization of Xia Dynasty in this region.After 0.9 kaBP,the climate turned cool and wet.Climate and environmental changes archived in Dajiuhu peat respond to the global changes since the late-glacial period and can be contrasted to the changes recorded in other high-resolution archives from the East Asia Monsoon region,which take on the variety model that the monsoon strengthened abruptly after the late-glacial,was strong during the early Holocene,subse- quently declined and became weak after the middle Holocene with dry climate.According to our analysis,the driving mechanism should be the response of solar radiation changes in the East Asia Monsoon region at middle latitudes. | MA ChunMei ZHU Cheng ZHENG ChaoGui WU ChunLin GUAN Yong ZHAO ZhiPing HUANG LinYan HUANG Run | 2008 | Chinese Science Bulletin2008,53,S1: | 19 |
| 4 | ^(226)Ra evidence for the ecosystem shift over the past 40 years in the North Pacific Subtropical Gyre显示文摘Surface seawater was collected for ^(226)Ra measurement in the North Pacific Subtropical Gyre from July to October, 1999 and October to December, 2003. Combined with the historical data reported for this sea area, a declined trend of surface ^(226)Ra concentrations was observed since 1960s, indicating the ecosystem shift in response to global warming. On one side, the enhanced stratification of the upper water column resulting from global warming reduced the ^(226)Ra input from the depth, on the other, the temporal increase of biological production resulting from the climate-related ecosystem structure change strengthened the ^(226)Ra removal from the surface ocean. Both the physical and biological processes resulted in the decrease of surface ^(226)Ra concentrations in the North Pacific Subtropical Gyre. The temporal trend of surface ^(226)Ra concentrations was consistent with the trends of chlorophyll a, silicate, phosphate and primary production previously reported. | 5 | Transformation and migration of phosphorus in excess sludge reduction pretreatment by alkaline ferrate oxidation combined with anaerobic digestion显示文摘Recently,more and more attention has been paid to the strong oxidation ability of newly prepared potassium ferrate(NAPF) in sludge reduction process,but less attention has been paid to the change of phosphorus in this process.The feasibility of phosphorus migration and transformation during excess sludge reduction pretre atment using NAPF pre-oxidation combined with anaerobic digestion was investigated.After 70 mg/g suspended solids NAPF pretreatment and 16 days anaerobic digestion,the solid-phase volatile suspended solids decreased by 44.2%,and much organic matter had been released into the liquid-phase and then degraded during digestion by indigenous microorganisms.As the sludge pre-oxidation process was performed,solid-phase organic phosphorus and chemically combined phosphorus also released into the liquid-phase as PO4^3-,peaking at 100 mg/L.During anaerobic digestion,the Fe3+in the liquid-phase was gradually reduced to Fe2+,and then formed Fe^2+-PO4^3- compound crystals and re-migrated to the solid-phase.The concentration of PO4^3- decreased to 17.08± 1.1 mg/L in the liquid-phase after anaerobic digestion.Finally,the phosphorus in the Fe^2+-PO4^3- compound accounts for 80% of the total phosphorus in the solid-phase.A large number of vivianite crystals in sludge were observed.Therefore,this technology not only effectively reduces sludge,but also increases the proportion of PO43-in the sludge in the form of Vivianite. | Xiang Li Yan Yuan Yong Huang Chao-ran Guo Run Jin Ting-ting Liu Jun Ma | 2020 | Journal of Environmental Sciences2020,32,6: | 4 |
| 6 | Influence of particle composition on thorium scavenging in the marginal China seas显示文摘Thorium-234 and particle composition (organic matter, biogenic silica, carbonate and lithogenic component) were examined in the East China Sea (ECS) and the northern South China Sea (NSCS) in order to constrain the particle types scavenging thorium isotopes. Good positive correlations be-tween particulate organic matter (POM) or carbonate and thorium-234 in suspended particulate matter (SPM) indicates that POM and carbonate are efficient to scavenge thorium-234. No relation-ship between biogenic silica and thorium-234 suggests that geochemical behavior of thorium-234 may be not influenced by biogenic silica. A simple model was used to evaluate the affinity of thorium-234 to different particle components. The results show that POM is the most efficient scavenger for thorium-234 in the ECS and the NSCS, followed by carbonate. The authors’ re-sults lend support to the utility of thorium-234 as a proxy of POC and carbonate in the upper layer. | 8 | Programmable activation of Bombyx gene expression using CRISPR/dCas9 fusion systems显示文摘The recently developed clustered regularly interspaced short palindromic repeats (CRISPR)-based techniques have made it possible to reprogram target gene expression without cloning complementary DNA or disturbing genomic sequence in mammalian cells and several multicellular organisms. We previously showed that CRISPR-associated protein 9 (Cas9) and CRISPR from Prevotella and Francisella 1 (Cpfl) could induce target mutations, deletions, inversions, and duplications both singly and multiplex in silkworm, Bombyx mori. However, it remains unknown whether the CRISPR activation (CRISPRa) system can be used in B. mori. In this study, we investigated the CRISPRa system, in which a nuclease dead Streptococcus pyogenes Cas9 (SpCas9) is fused to two transcription activation domains, including VP64 (a tetramer of the herpes simplex VP 16 transcriptional activator domain), and VPR (a tripartite activator, composed of VP64, p65, and Rta). The results showed that both dCas9-VP64 and dCas9-VPR systems could be used in B. mori cells, of which the latter showed significantly higher activity. The dCas9-VPR system showed considerable activity on all five tested target genes, and further analysis revealed that the up-regulation of genes was negatively correlated to their basal expression level. We also observed that this system could be used to upregulate a range of target genes. A comprehensive toolbox for multiplex genome editing has been developed for the silkworm,Bombyx mori, a lepidopteran model insect of economic importance. However, as previous methods mainly relied on delivery of transient Cas9/guide RNA(gRNA), they could not be used in loss-of-function studies of essential genes. Here, we report a simple and versatile tissue-specific genome editing strategy.We perform a proof-of-principle demonstration by establishing and crossing two transgenic B. mori lines,one expressing Cas9 protein in the posterior silk glands(PSGs) and the other constitutively expressing BmlaminA/C(BmLMN) gRNA. All BmLMN alleles in the PSG cells were edited precisely at the target genome region, resulting in diverse mutations. mRNA expression of BmLMN was reduced by up to 75%,and only very low levels of BmLaminA/C protein were detected. Knockout of BmLMN produced obvious defects in gland cell development and cocoon production. | 17 | Effect of powdery mildew on interleaf microbial communities and leaf antioxidant enzyme systems显示文摘Chinese peony(Paeonia lactiflora Pall.)is both medicinally and aesthetically beneficial.Powdery mildew is a common fungal disease that seriously jeopardizes the value of numerous species,including peonies as a crop.In order to provide a basis for the prevention and treatment of peony powdery mildew,we examined the microbial diversity,the malondialdehyde(MDA)concentrations and antioxidant enzyme activities of peony leaves infected with three levels of powdery mildew to determine any modifications to the leaf's antioxidant enzyme systems and microbial community structure following the onset of disease.The results show that the MDA content rose as the degree of infection became worse.Antioxidant enzyme activity rose and then declined.Following the initiation of powdery mildew,fungal community diversity decreased,whereas there was not any appreciable change in bacterial communities according to microbial diversity sequencing.The relative abundance of more than half of fungal species decreased,with the bacterial genera displaying both abundant and diminished communities with less pronounced alterations in their community structure after the disease spread.Significant different taxa that were critical to the organization of each microbiome were found.Correlation analysis showed that the relative abundance of powdery mildew pathogenic fungal genus Erysiphe was correlated with those of 11 fungal genera and one bacterial genus.Among them,Aureobasidium,Neosetophoma and Sclerostagonospora showed significant positive correlations with Erysiphe and MDA. | Xinbo Ma Zhanbin Wang Run Liu Yibing Jiang | 2023 | Journal of Forestry Research2023,34,5: | 0 |

| 19 | Insights into the coupling of upper ocean-benthic carbon dynamics in the western Arctic Ocean from an isotopic(^(13)C, ^(234)Th) perspective显示文摘The coupling of upper ocean-benthic carbon dynamics in the ice-free western Arctic Ocean(the Chukchi Sea and the Canada Basin) was evaluated during the late July–early September 2003 using natural stable(13C) and radioactive(238U-234Th) isotope tracers. POC export flux estimated from 234Th/238 U disequilibria and dissolved CO2 concentration([CO2(aq)]) pointed out that the strengthened biological pump in the Chukchi Shelf have significantly lowered [CO2(aq)] and altered the magnitude of isotopic(12C/13C) fractionation during carbon fixation in the surface ocean. Further, d13 C signatures of surface sediments(d13Csed) are positively correlated to those of weighted d13 CPOC in upper ocean(d13Csed =13.64+1.56×d13CPOC, r2=0.73, p<0.01), suggesting that the POC isotopic signals from upper ocean have been recorded in the sediments, partly due to the rapid export of particles as evidenced by low residence times of the highly particle-reactive 234 Th from the upper water column. | 20 | Nitrogen uptake regime regulated by ice melting during austral summer in the Prydz Bay, Antarctica显示文摘Using a combination of stable isotope(15N) and radionuclide(226Ra) analyses, we examine possible controls on the interactions between melting ice and the uptake of nitrogen in the Prydz Bay during the 2006 austral summer.We find that specific rates of uptake for nitrate and ammonium correlate positively to their concentrations, thus suggesting a substrate effect. In the study area, we observe that regions along open, oceanic water have high fratios(nitrate uptake/nitrate+ammonium uptake), while areas near the Amery Ice Shelf have significantly low fratios. Further analysis reveals a negative correlation between the f-ratio and the melt water fraction, thus implying that the melting of ice plays an essential role in regulating pelagic N dynamics in the Southern Ocean(SO). Stratification, produced by melting ice, should profoundly affect the efficiency of the SO’s biological pump and consequently affect the concentration of CO2 in the atmosphere. Results presented in this study add information to an already significant base of understanding of the controls on pelagic C and N dynamics in the SO.
