1Expression of fragile histidine triad in primary hepatocellular carcinoma and its relation with cell proliferation and apoptosis显示文摘AIM: To evaluate the expression of fragile histidine triad (FHIT) gene protein, product of a candidate tumor suppressor,and to investigate the relationship between FHIT, cell apoptosis and proliferation, and pathological features of primary hepatocellular carcinoma (HCC).METHODS: Forty-seven HCC and ten normal liver specimens were collected during surgical operation between 2001and 2003. FHIT and proliferating cell nuclear antigen (PCNA)expression were detected by immunohistochemistry, and apoptotic level was evaluated by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) assay on the tissue sections.RESULTS: All normal liver tissues showed a strong expression of FHIT, whereas 28 of 47 (59.6%) carcinomas showed a significant loss or absence of FHIT expression (P = 0.001).The proportion of reduced FHIT expression in those carcinomas at stages Ⅲ-Ⅳ (70.6%) and in those with extrahepatic metastasis (86.7%) showed an increasing trend compared with those at stages Ⅰ-Ⅱ (30.8%, P= 0.013) and those without metastasis (46.9%, P = 0.010) respectively. Apoptotic incidence in advanced TNM stage carcinoma and those with positive FHIT expression was higher than that in early stage carcinoma (P = 0.030) and in those with negative FHIT expression (P = 0.044) respectively. The proliferating potential of hepatocellular carcinoma was associated with FHIT expression (P = 0.016) and the aggressive feature (P = 0.019). Kaplan-Meier analysis demonstrated that the survival time of these 47 patients correlated with TNM stage,FHIT expression and metastasis.CONCLUSION: There is marked loss or absence of FHIT expression, as well as abnormal apoptosis-proliferation balance in HCC. 2Expression and clinical significance of TAp73α, p53, PCNA and apoptosis in hepatocellular carcinoma显示文摘AIM: To study the prognostic role of TAp73α, p53, proliferating cell nuclear antigen (PCNA) and apoptosis inpatients with hepatocellular carcinoma (HCC) atter surgical tumor ablation.METHODS: Forty-seven human resected HCC tissues and 42 adjacent non-cancerous tissues were studied with 10 normal liver tissues as control group. TAp73α, p53, and PCNA were detected with Elivision immunohistochemistry.Terminal deoxynucleotidyl transferase (TdT)-mediatedd-UTP-biotin nick-end labeling (TUNEL) method wasused to detect the apoptosis cells. All clinical and pathological materials were analyzed by SPSS10.0statistical package.RESULTS: TAp73α overexpressed in HCC tissues (36.2%) when compared with adjacent non-cancerous tissues (2.38%, P<0.005) and normal liver tissues (0, P<0.01).Mutant type p53 (mt-p53) overexpressed in HCC tissues (38.3%) when contracted with adjacent non-cancerous tissues (16.7%, P<0.05) and normal liver tissues (0, P<0.01). Proliferation index (PI) level in HCC tissues was significantly higher than that in adjacent non-cancerous tissues (30.34%±4.46% vs 27.88%±5.89%, t, P= 0.028). Apoptosis index (AI) level in HCC tissues was higher than that in adjacent non-cancerous tissues (8.62%±2.28% vs 7.38%±2.61%, t, P = 0.019). Expression of TAp73α was associated with lymph node metastasis and mt-p53, with r = 0.407 and 0.265, respectively. Expression of mtp53 was associated with Edmondson's stage and AFP, with r = 0.295 and -0.357, respectively. In Kaplan-Meier univariant analysis, TAp73α, AFP, TNM stage, portal vein invasion, liver membrane invasion and HBsAg correlated with prognosis (log rank, P = 0.039, 0.012, 0.002, 0.000,0.014, 0.007, respectively). Multivariant Cox regression analysis showed that TAp73α, AFP, TNM stage, portal vein invasion, liver membrane invasion and age were independent factors of prognosis. CONCLUSION: These results suggest that TAp73α can be used as a prognostic indicator of patients with HCC undergoing surgical tumor ablation. 3Effect of initial material bulk density and easily-degraded organic matter content on temperature changes during composting of cucumber stalk显示文摘To inactivate the potentially pathogenic microorganisms and safely utilize vegetable waste compost, ultra-high temperatures(> 70°C) should be maintained during the composting without having an inhibitory effect on maturity. This study investigated the influence of bulk density(part 1) and easily-degraded organic matter content(EDOMC, part 2) on temperature evolution during vegetable waste composting: Part 1: corn straw with different particle sizes was used to achieve different bulk densities in the composting material(BD1–BD3); Part 2: partial or total substitution of the corn straw by corn starch was carried out to obtain different EDOMC(ED1–ED4). The composting experiments were conducted in a labscale reactor(1.75 kg material) and lasted for 30 d. Temperature and CO2 emission were recorded daily, and the organic matter, lignocellulose, microbial activity, germination index(GI) and C/N of the samples were measured at different stages. The highest temperature(65.7°C) in part 1 occurred in the treatment with the bulk density of 0.35 g/cm3, which also had the longest thermophilic phase. Bulk density was found to seriously influence the utilization efficiency of O2 and heat transfer through materials, rather than heat production from organic matter degradation. In experiment part 2, the highest temperature was obtained with EDOMC of 45%(71.4°C). 6Effect of loading rates on the characteristics of thermal damage for mudstone under different temperatures显示文摘The uniaxial compression tests for mudstone specimens are carried out with four different loading rates from room temperature to 400℃ by using the Rock Mechanics Servo-controlled Testing System MTS810 and high temperature furnace MTS652.02.The mechanical properties of mudstone with various loading rates are studied under different temperature conditions.The results show that when temperature increases from room temperature to 400℃ and loading rate is less than 0.03 mm/s,the peak strength of mudstone specimen decreases as loading rate increases,while the various peak strengths show significant differences when loading rate exceeds 0.03 mm/s.At room temperature,the elastic modulus decreases at the first time and then increases with loading rate rising.When the temperature is between200 and 400℃,the elastic modulus presents a decreasing trend with increasing loading rate.With increasing the loading rate,the number of fragments in mudstone becomes larger and even the powder is observed in mudstone with higher loading rate.Under high loading rate,the failure mode of mudstone specimens under different temperatures is mainly conical damage.Mao Rongrong Mao Xianbiao Zhang Lianying Liu Ruixue 2015International Journal of Mining Science and Technology2015,25,5:7
7DNA damage repair:historical perspectives,mechanistic pathways and clinical translation for targeted cancer therapy显示文摘Genomic instability is the hallmark of various cancers with the increasing accumulation of DNA damage.The application of radiotherapy and chemotherapy in cancer treatment is typically based on this property of cancers.However,the adverse effects including normal tissues injury are also accompanied by the radiotherapy and chemotherapy.Targeted cancer therapy has the potential to suppress cancer cells'DNA damage response through tailoring therapy to cancer patients lacking specific DNA damage response functions.Obviously,understanding the broader role of DNA damage repair in cancers has became a basic and attractive strategy for targeted cancer therapy,in particular,raising novel hypothesis or theory in this field on the basis of previous scientists'findings would be important for future promising druggable emerging targets.In this review,we first illustrate the timeline steps for the understanding the roles of DNA damage repair in the promotion of cancer and cancer therapy developed,then we summarize the mechanisms regarding DNA damage repair associated with targeted cancer therapy,highlighting the specific proteins behind targeting DNA damage repair that initiate functioning abnormally duo to extrinsic harm by environmental DNA damage factors,also,the DNA damage baseline drift leads to the harmful intrinsic targeted cancer therapy.In addition,clinical therapeutic drugs for DNA damage and repair including therapeutic effects,as well as the strategy and scheme of relative clinical trials were intensive discussed.Based on this background,we suggest two hypotheses,namely'environmental gear selection”to describe DNA damage repair pathway evolution,and'DNA damage baseline drift”,which may play a magnified role in mediating repair during cancer treatment.This two new hypothesis would shed new light on targeted cancer therapy,provide a much better or more comprehensive holistic view and also promote the development of new research direction and new overcoming strategies for patients.Ruixue Huang Ping-Kun Zhou 2021Signal Transduction and Targeted Therapy2021,6,8:6
8Geology and Isotope Geochemistry of the Yinchanggou-Qiluogou Pb-Zn Deposit,Sichuan Province,Southwest China显示文摘The Yinchanggou-Qiluogou Pb-Zn deposit,located in the western Yangtze Block,southwest China,is hosted by the Upper Sinian Dengying Formation dolostone.Ore bodies occur in the Qiluogou anticline and the NS-and NNW-trending faults.Sulfide ores mainly consist of sphalerite,pyrite,galena and calcite,with subordinate dolomite and quartz.Seventeen ore bodies have been discovered to date and they have a combined 1.0 million tons of sulfide ores with average grades of 2.27wt%Zn and 6.89wt%Pb.The δD_(H2O-SMOW) and δ^(18)O_(H2O-SMOW) values of fluid inclusions in quartz and calcite samples range from-68.9‰ to-48.7‰ and 7.3‰ to 15.9‰,respectively,suggesting that H_2O in the hydrothermal fluids sourced from metamorphic water.Calcite samples have δ^(13)C_(PDB) values ranging from-6.2‰ to-4.1‰ and δ^(18)O_(SMOW) values ranging from 15.1‰ to 17.4‰,indicating C and O in the hydrothermal fluids likely derived from a mixed source of metamorphic fluids and the host carbonates.The δ^(34)S_(CDT) values of sulfide minerals range from 5.5‰ to 20.3‰,suggesting that thermal chemical reduction of sulfate minerals in evaporates were the most probable source of S in the hydrothermal fluids.The ^(206)Pb/^(204)Pb,^(207)Pb/^(204)Pb and ^(208)Pb/^(204)Pb ratios of sulfide minerals fall in the range of 18.11 to 18.40,15.66 to 15.76 and 38.25 to 38.88,respectively.The Pb isotopic data of the studied deposit plot near the upper crust Pb evolution curve and overlap with the age-corrected Proterozoic basement rocks and the Upper Sinian Dengying Formation hosting dolostone.This indicates that the Pb originated from a mixed source of the basement metamorphic rocks and the ore-hosting carbonate rocks.The ore geology and C-H-O-S-Pb isotopic data suggest that the YinchanggouQiluogou deposit is an unusual carbonate-hosted,strata-bound and epigenetic deposit that derived ore-forming materials from a mixed source of the underlying Porterozoic basements and the Sinian hosting carbonates.LI Bo ZHOU Jiaxi LI Yingshu CHEN Aibing WANG Ruixue 2016Acta Geologica Sinica(English Edition)2016,90,5:5

11External use of Ruyanneixiao cream efficiently blocks precancerous mammary lesions by interfering with glycolysis induced by inhibition of hypoxia inducible factor-1α, hexokinase 2, phosphofructokinase, and pyruvate kinase M2 expression显示文摘OBJECTIVE:To investigate the effect of Ruyanneixiao cream(RYNX) on the expression of hypoxia inducible factor-1α(HIF-1α), hexokinase 2(HK2),phosphofructokinase(PFK), and pyruvate kinase M2(PKM2) mRNA and protein in MCF-10 AT cells and in an animal model of precancerous mammary lesions.METHODS:Following treatment of MCF-10 AT cells with RYNX, tamoxifen(TAM) and YC-1 for 48 h,HIF-1α, HK2, PFK, PKM2 mRNA and protein expression was analyzed.Fifty female SD rats were randomly divided into control, model, TAM, and highand low-dose RYNX groups, with 10 rats in each group.A precancerous mammary lesion model was established for all groups except the control group.High-and low-dose RYNX cream containing TAM was coated on the breasts of animals in the corresponding groups.The rat mammary tissue was removed in the 10 th week and HIF-1α, HK2, PFK,PKM2 mRNA and protein was analyzed.RESULTS:In vitro analyses demonstrated that, compared with the matrix group, HIF-1α, HK2, PFK,PKM2 mRNA and protein expression was significantly decreased in the RYNX group(P < 0.05).Compared with the YC-1 + RYNX group, HK2, PFK,and PKM2 protein expression was significantly reduced in the RYNX group.HIF-1α, HK2, PFK, and PKM2 protein expression was increased significantly in the model group(P < 0.05) compared with the control group, while HIF-1α, HK2, PFK, and PKM2 mRNA and protein expression was significantly decreased in both the high-and low-dose RYNX groups(P < 0.05), with the effect being greater in the high-dose group.CONCLUSION:RYNX can block precancerous breast lesions by decreasing the expression of HK2,PFK, and PKM2 mRNA and protein via inhibition of HIF-1α mRNA and protein overexpression in a dose-dependent manner.Li Xiaobo Ma Min Zhang Guijuan Ma Yi Liao Rui Chen Ruixue Yan Xianxin Bie Fengjie Huang Maojie Liang Shijie 2017Journal of Traditional Chinese Medicine2017,37,2:5
12Structural Changes of Wheat Straw Lignin during Formic Acid Treatment显示文摘The aromatic nature of lignin makes it a potential renewable source of chemicals and other valuable products.Isolation of lignin from lignocellulosic biomass using organic solvents enables the production of high-purity lignin.The use of formic acid in the organosolv pulping and fractionation process has been widely studied.Characterization of lignin is necessary to achieve valueadded applications of lignin.To simplify the isolation of formic acid-treated lignin,herein,milled wheat straw lignin(MWSL) was employed as an archetype for characterization of the structural changes of lignin during formic acid treatment.The results showed that the MWSL was GSH-type(comprising p-hydroxyphenyl(H),guaiacyl(G),and syringyl(S) monolignols) and underwent structural changes during formic acid treatment.Lignin was esterified during the formic acid treatment.The content of alkyl hydroxyl groups in lignin decreased upon formic acid treatment,corresponding to an increase of the number of double bond equivalents(DBE).Lignin units with active reaction sites were liable to slight condensation,which resulted in a moderate increase of the molecular weight.The molecular weight distribution of formic acid-treated MWSL(FMWSL) was wider than that of the MWSL,although the molecular weight of both species did not differ significantly.The β-O-4 linkage in lignin was partially cleaved during formic acid treatment,resulting in the production of new phenolic structures.This improved the solubility of lignin in the cooking liquor and its reactivity for downstream applications.RuiXue Dong XiuWen Mei Chang Ma MingFei Li YongMing Fan 2016Paper And Biomaterials2016,1,2:5
13Distinct immune signatures discriminate between asymptomatic and presymptomatic SARS-CoV-2^(pos)subjects显示文摘Increasing numbers of SARS-CoV-2-positive(SARS-CoV-2^(pos))subjects are detected at silent SARS-CoV-2 infection stage(SSIS).Yet,SSIS represents a poorly examined time-window wherein unknown immunity patterns may contribute to the fate determination towards persistently asymptomatic or overt disease.Here,we retrieved blood samples from 19 asymptomatic and 12 presymptomatic SARS-COV-2^(pos)subjects,47 age/gender-matched patients with mild or moderate COVID-19 and 27 normal subjects,and interrogated them with combined assays of 44-plex CyTOF,RNA-seq and Olink.Notably,both asymptomatic and presymptomatic subjects exhibited numerous readily detectable immunological alterations,while certain parameters including more severely decreased frequencies of CD107a^(low)classical monocytes,intermediate monocytes,non-classical monocytes and CD62L^(hi)CD8^(+)Tnaive cells,reduced plasma STC1 level but an increased frequency of CD4^(+)NKT cells combined to distinguish the latter.Intercorrelation analyses revealed a particular presymptomatic immunotype mainly manifesting as monocytic overactivation and differentiation blockage,a likely lymphocyte exhaustion and immunosuppression,yielding mechanistic insights into SSIS fate determination,which could potentially improve 5ARS-CoV-2 management.Shanhe Yu Caixia Di Shijun Chen Mingquan Guo Jiayang Yan Zhaoqin Zhu Li Liu Ruixue Feng Yinyin Xie Ruihong Zhang Juan Chen Mengxi Wang Dong Wei Hai Fang Tong Yin Jinyan Huang Saijuan Chen Hongzhou Lu Jiang Zhu Jieming Qu 2021Cell Research2021,31,11:4
14Macroscopic and microscopic fracture features of concrete used in coal mine under chlorine salt erosion显示文摘The microscopic morphology and pore structure characteristics of concrete with composite admixtures(fly ash and mineral powder) after chlorine salt erosion were analyzed via scanning electron microscopy(SEM) and mercury injection porosimetry(MIP), providing the basis for the design and maintenance of concrete shafts in coal mines. The above-mentioned characteristics were compared with the macroscopic characteristic of concrete fractures under uniaxial compression. The results show that the macroscopic fracture characteristics of concrete under uniaxial compression change from longitudinal split fracture and oblique section shear fracture to conjugate cant fracture, and the degree of breakage increases.Interface cracks, cement paste cracks, spherical surface cracks, and aggregate cracks appear in concrete under uniaxial compression. In the early stages of corrosion, the original cracks which are obvious are repaired. When the corrosion becomes more serious, cement paste cracks appear, and the number of harmful holes increases while the number of harmless holes decreases. This study also reveals the relationship between the macroscopic properties and microscopic structure of concrete under chloride salt erosion. 15Quantitative Prediction of Reduction in Large Pipe Setting Round Process显示文摘The control manner during the process to ensure the quality of pipe products mainly relies on the operator's experience, so it is very necessary to study the setting round process and obtain its spring-back law. The setting round process is shaping an oval section pipe into circular section, so it is difficult to provide a quantificational analysis for its spring-back process because of the curvature inequality of pipe section neutral layer. However, the spring-back law of the circle-oval process can be easily predicted. The experimental method is firstly used to establish the equivalent effect between the setting round process and the circle-oval process. The setting round process can be converted into the circle-oval process. There are two difficulties in the theoretical analysis for the circle-oval process: elastic-plastic bending problem of curved beam; statically indeterminate problem. A quantitative analytic method for the circle-oval process is presented on the basis of combination of the spring-back law of plane curved beam with the element dividing idea in finite element method. The ovality after unloading versus the relative reduction is plotted with analytical and experimental results respectively, which shows a fair agreement. Finally, the method of quantitative prediction of reduction for large pipe setting round is given based on the equivalent effect and the analytical results. Five pipes, which are needed to be set round, are used to carry out experiment so as to verify this method. The results of verification experiment indicates that, in the experimental range, the residual ovality are all under 0.35% after the once only setting round with the theoretical prediction reductions. It is much less than the 1% requirement of pipe standard. 18HIF-1 signaling: A key orchestrator of cancer radioresistance显示文摘Hypoxia is a big roadblock for cancer radiotherapy,in which the hypoxia-inducible factor(HIF-1)creates a microenvironment and cancer cells’intrinsic signaling networks conferring radioresistance to cancers.HIF-1 is a heterodimeric transcription factor HIF-1α/HIF-1β,that regulates the transcription of a broad range of downstream genes possessing an E-box-like hypoxia response element(HRE).The expression of HIF-1αis oxygensensitive while HIF-1βis constitutively expressed.In addition to hypoxia,ionizing radiation can also induce the expression of HIF-1α.The HIF-1 modulates a set of signaling pathways to cause profound effects on the response of cancer to radiotherapy,including radiation-induced DNA damage response(DDR),vasculogenesis and glucose metabolism reprograming,epithelial mesenchymal transition(EMT),etc.In this review,our aim is to summarize the current knowledge about the role and the related signal pathways of HIF-1 in association with the resistance of cancers to radiotherapy.Targeting HIF-1 and its signal pathways is a promising strategy for sensitization of cancers to radiotherapy.Ruixue Huang Ping-Kun Zhou 2020Radiation Medicine and Protection2020,1,1:3
19Integrative analysis of miRNA and mRNA profiles reveals that gga-miR-106-5p inhibits adipogenesis by targeting the KLF15 gene in chickens显示文摘Background:Excessive abdominal fat deposition in commercial broilers presents an obstacle to profitable meat quality,feed utilization,and reproduction.Abdominal fat deposition depends on the proliferation of preadipocytes and their maturation into adipocytes,which involves a cascade of regulatory molecules.Accumulating evidence has shown that microRNAs(miRNAs)serve as post-transcriptional regulators of adipogenic differentiation in mammals.However,the miRNA-mediated molecular mechanisms underlying abdominal fat deposition in chickens are still poorly understood.This study aimed to investigate the biological functions and regulatory mechanism of miRNAs in chicken abdominal adipogenesis.Results:We established a chicken model of abdominal adipocyte differentiation and analyzed miRNA and mRNA expression in abdominal adipocytes at different stages of differentiation(0,12,48,72,and 120 h).A total of 217 differentially expressed miRNAs(DE-miRNAs)and 3520 differentially expressed genes were identified.Target prediction of DE-miRNAs and functional enrichment analysis revealed that the differentially expressed targets were significantly enriched in lipid metabolism-related signaling pathways,including the PPAR signaling and MAPK signaling pathways.A candidate miRNA,gga-miR-106-5p,exhibited decreased expression during the proliferation and differentiation of abdominal preadipocytes and was downregulated in the abdominal adipose tissues of fat chickens compared to that of lean chickens.gga-miR-106-5p was found to inhibit the proliferation and adipogenic differentiation of chicken abdominal preadipocytes.A dual-luciferase reporter assay suggested that the KLF15 gene,which encodes a transcriptional factor,is a direct target of gga-miR-106-5p.gga-miR-106-5p suppressed the posttranscriptional activity of KLF15,which is an activator of abdominal preadipocyte proliferation and differentiation,as determined with gain-and loss-of-function experiments.Conclusions:gga-miR-106-5p functions as an inhibitor of abdominal adipogenesis by targeting the KLF15 gene in chickens.These findings not only improve our understanding of the specific functions of miRNAs in avian adipogenesis but also provide potential targets for the genetic improvement of excessive abdominal fat deposition in poultry.Weihua Tian Xin Hao Ruixue Nie Yao Ling Bo Zhang Hao Zhang Changxin Wu 2022Journal of Animal Science and Biotechnology2022,13,6:3
20Flood-illuminated adaptive optics ophthalmoscope with a single curved relay mirror显示文摘For decreasing light loss and diminishing the aberrations of the optical system,an open-loop adaptive optics(AO)system for retinal imaging in vivo is introduced.Taking advantage of the ability of young human eyes to accommodate,there was only one single curved mirror to make the pupil of the eye conjugate with the wavefront corrector and the wavefront sensor.A liquid crystal spatial light modulator(LC-SLM)was adopted as the wavefront corrector because the LC-SLM can be made in a small size to match the sensor.To reduce a pair of lenses or focusing mirrors,the wavefront corrector and sensor are positioned in the noncommon path.The system adopts open-loop control and the high-precision LC-SLM guarantees the effectiveness of the AO system.The designed field of view is 1°on the retina(about 300μm).The image quality was simulated with different mirror surface types,including circular,parabolic,and hyperbolic.A hyperbolic mirror with conic constant−1.07,which is close to−1,could best eliminate the aberrations.Theoretical analysis showed that the optical throughput of this system was at least 22.4%higher than that of a standard transmission AO system.In a practical experiment,a parabolic mirror was positioned in the optical path.Images of the cone photoreceptors and the capillary vessels were obtained successfully.This system simplifies the optical setup in comparison to the commonly used 4F systems while still guaranteeing the effectiveness of AO to correct the ocular aberrations.Ruixue Liu Yue Qi Xianliang Zheng Mingliang Xia Li Xuan 2013Photonics Research2013,1,3:3
