2A new cell death program regulated by toll-like receptor 9 through p38 mitogen-activated protein kinase signaling pathway in a neonatal rat model with sepsis associated encephalopathy显示文摘Background:Sepsis,a serious condition with high mortality,usually causes sepsis associated encephalopathy(SAE)that involves neuronal cell death.However,the cell death programs involved and their underlying mechanisms are not clear.This study aimed to explore the regulatory mechanisms of different cell death programs in SAE.Methods:A neonatal rat model of SAE was established by cecal ligation and perforation.Survival rate and vital signs(mean arterial pressure and heart rate)were monitored,nerve reflexes were evaluated,and cortical pathological changes were observed by hematoxylin and eosin staining.The expression of pyroptosis,apoptosis,and necroptosis(PANoptosis)-related proteins,mitogen-activated protein kinase(MAPK),and its upstream regulator toll-like receptor 9(TLR9)were detected.The expression of TLR9 in neurons was observed by immunofluorescence staining.The ultrastructure of neurons was observed by transmission electron microscope.Results:First,PANoptosis was found in cortical nerve cells of the SAE rats.Meanwhile,the subunits of MAPKs,p38 MAPK,Jun N-terminal kinase,and extracellular signal-regulated kinase(ERK)were activated.After pharmacologically inhibiting each of the subunits,only p38 MAPK was found to be associated with PANoptosis.Furthermore,blocking the p38 MAPK signaling pathway activated necroptosis but inhibited apoptosis and pyroptosis.When necroptosis was pharmacologically inhibited,apoptosis and pyroptosis were reactivated.Finally,we found that the expression of TLR9,a regulator of MAPKs,was significantly increased in this model.After down-regulation of TLR9,p38 MAPK,and ERK signaling pathways were inhibited,which led to the inhibition of PANoptosis.Further analysis found that down-regulation of TLR9 improved the survival rate and reduced the pathological changes in SAE rats.Conclusions:Our study showed that the programs comprising PANoptosis are activated simultaneously in SAE rats.TLR9 activated PANoptosis through the p38 MAPK signaling pathway.TLR9 may work as a potential target for SAE treatment.Ruixi Zhou Junjie Ying Xia Qiu Luting Yu Yan Yue Qian Liu Jing Shi Xihong Li Yi Qu Dezhi Mu 2022Chinese Medical Journal2022,,12:3
3Dynamical analysis of submesoscale fronts associated with wind-forced offshore jet in the western South China Sea显示文摘This study investigates the submesoscale fronts and their dynamic effects on the mean flow due to frontal instabilities in the wind-driven summer offshore jet of the western South China Sea(WSCS),using satellite observations,a 500 m-resolution numerical simulation,and diagnostic analysis.Both satellite measurements and simulation results show that the submesoscale fronts occupying a typical lateral scale of O(~10)km are characterized with one order of Rossby(Ro)and Richardson(Ri)numbers in the WSCS.This result implies that both geostrophic and ageostrophic motions feature in these submesoscale fronts.The diagnostic results indicate that a net cross-frontal Ekman transport driven by down-front wind forcing effectively advects cold water over warm water.By this way,the weakened local stratification and strong lateral buoyancy gradients are conducive to a negative Ertel potential vorticity(PV)and triggering frontal symmetric instability(SI)at the submesoscale density front.The cross-front ageostrophic secondary circulation caused by frontal instabilities is found to drive an enhanced vertical velocity reaching O(100)m/d.Additionally,the estimate of the down-front wind forcing the Ekman buoyancy flux(EBF)is found to be scaled with the geostrophic shear production(GSP)and buoyancy flux(BFLUX),which are the two primary energy sources for submesoscale turbulence.The large values of GSP and BFLUX at the fronts suggest an efficient downscale energy transfer from larger-scale geostrophic flows to the submesoscale turbulence owing to down-front wind forcing and frontal instabilities.In this content,submesoscale fronts and their instabilities substantially enhance the local vertical exchanges and geostrophic energy cascade towards smaller-scale.These active submesoscale processes associated density fronts and filaments likely provide new physical interpretations for the filamentary high chlorophyll concentration and frontal downscale energy transfer in the WSCS.Xiaolong Huang Zhiyou Jing Ruixi Zheng Haijin Cao 2020Acta Oceanologica Sinica2020,39,11:2
4The small GTPase RABA2a recruits SNARE proteins to regulate the secretory pathway in parallel with the exocyst complex in Arabidopsis显示文摘Delivery of proteins to the plasma membrane occurs via secretion,which requires tethering,docking,priming,and fusion of vesicles.In yeast and mammalian cells,an evolutionarily conserved RAB GTPase activation cascade functions together with the exocyst and SNARE proteins to coordinate vesicle transport with fusion at the plasma membrane.However,it is unclear whether this is the case in plants.In this study,we show that the small GTPase RABA2a recruits and interacts with the VAMP721/722-SYP121-SNAP33 SNARE ternary complex for membrane fusion.Through immunoprecipitation coupled with mass spectrometry analysis followed by the validatation with a series of biochemical assays,we identified the SNARE proteins VAMP721 and SYP121 as the interactors and downstream effectors of RABA2a.Further expreiments showed that RABA2a interacts with all members of the SNARE complex in its GTP-bound form and modulates the assembly of the VAMP721/722-SYP121-SNAP33 SNARE ternary complex.Intriguingly,we did not observe the interaction of the exocyst subunits with either RABA2a or theSNARE proteins in several different experiments.Neither RABA2a inactivation affects the subcellular localization or assembly of the exocystnor the exocyst subunit mutant exo84b shows the disrupted RABA2a-SNARE association or SNARE assembly,suggesting that the RABA2a-SNARE-and exocyst-mediated secretory pathways are largely independent.Consistently,our live imaging experiments reveal that the two sets of proteins follow non-overlapping trafficking routes,and genetic and cell biologyanalyses indicate that the two pathways select different cargos.Finally,we demonstrate that the plant-specific RABA2a-SNARE pathway is essential for the maintenance of potassium homeostasis in Arabisopsis seedlings.Collectively,our findings imply that higher plants might have generated different endomembrane sorting pathways during evolution and may enable the highly conserved endomembrane proteins to participate in plant-specific trafficking mechanisms for adaptation to the changing environment.Lei Pang Zhiming Ma Xi Zhang Yuanzhi Huang Ruili Li Yansong Miao Ruixi Li 2022Molecular Plant2022,15,3:2
5Timing of formation of the Hongdonggou Pb-Zn polymetallic ore deposit,Henan Province,China:Evidence from Rb-Sr isotopic dating of sphalerites显示文摘The Hongdonggou Pb-Zn polymetallic ore deposit,located in the southwestern part of the Luanchuan Mo-W-Pb-Zn-Ag polymetallic ore mineralization in Henan Province,China,is an important part of the East Qinling metallogenic belt.The orebodies in the deposit,which are vein,bedded and lenticular,are mainly hosted in the syenite porphyry,and formed within the carbonate and clastic rocks of the Yuku and Qiumugou formations partially.The genesis of the deposit has previously been argued to be of hydrothermal-vein type or of skarn-hydrothermal type.In this study,we report the results of Rb-Sr isotopic dating based on sphalerites from the main orebody of the Hongdonggou Pb-Zn polymetallic ore deposit,which yield an isochron age of 135.7 ± 3.2 Ma,constraining the timing of mineralization as early Cretaceous.The age is close to those reported for the Pb-Zn deposits in the Luanchuan ore belt.The(^(87)Sr/^(86)Sr),values of the sphalerites(0.71127 + 0.00010) are lower than that of terrigenous silicates(0.720) and higher than the mantle(0.707),suggesting that the metallogenic components were mainly derived through crust-mantle mixing.Combining the results from this study with those from previous work,we propose that the Hongdonggou Pb-Zn polymetallic ore deposit is a hydrothermal-vein deposit associated with the early Cretaceous tectonothermal event,and the mineralization is controlled by NWand near EW-trending faults in the Luanchuan Mo-W-Pb-Zn-Ag polymetallic ore concentration belt.Fan Yang Gongwen Wang Huawen Cao Ruixi Li Li Tang Yufeng Huang Hao Zhang Fei Xue Wenjuan Jia Nana Guo 2017Geoscience Frontiers2017,8,3:2

15Resource prediction and assessment based on 3D/4D big data modeling and deep integration in key ore districts of North China显示文摘The North China district has been subjected to significant research with regard to the ore-forming dynamics,processes,and quantitative forecasting of gold deposits;it accounts for the highest number of gold reserves and annual products in China.Based on the top-level design of geoscience theory and the method adopted by the National Key R&D Project(deep process and metallogenic mechanism of North China Craton(NCC)metallogenic system),this paper systematically collects and constructs the geoscience data(district,camp,and deposit scales)in four key gold districts of North China(Jiaojia-Sanshandao,Southern Zhaoping,Wulong,and Qingchengzi).The settings associated with the geological dynamics of gold deposits were quantitatively and synthetically analyzed,namely:NCC destruction,metallogenic events,genetic models,and exploration models.Three-dimensional(3D)and four-dimensional(4D)geological modeling was performed using the big data on the districts,while the district-scale 3D exploration criteria were integrated to construct a quantitative exploration model.Among them,FLAC3D modelling and the Geo Cube software(version 3.0)were used to implement the numerical simulation of the 3D geological models and the constraints of the fluid saturation parameters of the Jiaojia fault to reconstruct the 4D fault structure models of the Jiaojia fault(with a depth of 5000 m).Using Geo Cube3.0,multiple integration modules(general weights of evidence(Wof E),Boost Wof E,Fuzzy Wof E,Logistic Regression,Information Entropy,and Random Forest)and exploration criteria were integrated,while the C-V fractal classification of A,B and C targets in four districts was carried out.The research results are summarized in the following four areas:(1)Four gold districts in the study area have more than three targets(the depth is 3000 m),and the class A,B and C targets exhibit a good spatial correlation with gold bodies that are controlled by mining engineering at depths greater than 1000 m.(2)The Boost Wof E method was used to identify the target optimization in 3D spaces(at depths of 3000–5000 m)of the Jiaojia-Sanshandao,Southern Zhaoping,and Wulong districts.(3)The general Wof E method is based on the Bayesian theory in 3D space and provides robust integration and target optimization that are suitable for the Jiaojia-Sanshandao and Southern Zhaoping districts in the Jiaodong area;it can also be applied to the Wulong district in the Liaodong area using a quantitative genetic model and an exploration model.Random forest is a multi-objective integration and target optimization method for 3D spaces,and it is suitable for the complex exploration model in the Qingchengzi district of the Liaodong area.The genetic model and exploration criteria associated with the exploration model of the Qingchengzi district were constrained by the common characteristics of the gold fault structure,magmatic rock emplacement in North China,and the strata fold and interlayer detachment structure.(4)Based on the gold reserves and the 3D block unit model of the Sanshandao gold deposit in the Jiaojia-Sanshandao district,the gold contents of the 3D block units in class A and B targets of the ore concentration were estimated to be 65.5%and 25.1%,respectively.The total Au resources of the optimized targets below a depth of 3000 m were 3908 t(including 1700 t reserves),and the total Au resources of the targets at depths from 3000 to 5000 m were 936 t.The study shows that the deep gold deposits in the four gold districts of North China exhibit a strong'transport-deposition'spatial correlation with potential targets.These'transport-deposition'spatial models represent the tectonic-magmatic-hydrothermal activities of the metallogenic system associated with the NCC destruction events and indicate the Au enrichment zones.Gongwen WANG Zhiqiang ZHANG Ruixi LI Junjian LI Deming SHA Qingdong ZENG Zhenshan PANG Dapeng LI Leilei HUANG 2021Science China Earth Sciences2021,64,9:1

17Methanol electro-oxidation to formate on iron-substituted lanthanum cobaltite perovskite oxides显示文摘Electrochemically producing formate by oxidizing methanol is a promising way to add value to methanol.Noble metal-based electrocatalysts,which have been extensively studied for the methanol oxidation reaction,can catalyze the complete oxidation of methanol to carbon dioxide,but not the mild oxidation to formate.As a result,exploring efficient and earth-abundant electrocatalysts for formate production from methanol is of interest.Herein,we present the electro-oxidation of methanol to formate,catalyzed by iron-substituted lanthanum cobaltite(LaCo_(1-x)Fe_(x)O_(3)).The Fe/Co ratio in the oxides greatly influences the activity and selectivity.This effect is attributed to the higher affinity of Fe and Co to the two reactants:CH3OH and OH,respectively.Because a balance between these affinities is favored,LaCo_(0.5)Fe_(0.5)O_(3) shows the highest formate production rate,at 24.5 mmol h^(-1) g_(oxide)^(-1),and a relatively high Faradaic efficiency of 44.4%in a series of(LaCo_(1-x)Fe_(x)O_(3))samples(x=0.00,0.25,0.50,0.75,1.00)at 1.6 V versus a reversible hydrogen electrode.Fanxu Meng Chencheng Dai Zheng Liu Songzhu Luo Jingjie Ge Yan Duan Gao Chen Chao Wei Riccardo Ruixi Chen Jiarui Wang Daniel Mandler Zhichuan J.Xu 2022eScience2022,2,1:1
