3Constructing retroviral vector carrying green fluorescent protein(GFP)and investigating the expression of GFP in primary rat myoblast显示文摘Objective:To construct green fluorescent protein(GFP)retroviral vector(pLgXSN),and to investigate the expression of GFP in primary rat myoblast.Methods:GFP cDNA was subcloned into the plasmid pLgXSN,and the recombinant vector was transfected into packaging cell PT67.G418 was used to select positive colony.Myoblasts were infected by a high-titer viral supernatant.The recombinant retroviral plasmid vector was identified by restriction endonuclease analysis and DNA sequence analysis.Confocal microscopy and flow cytometry were used to detect the expression of GFP.Results:The GFP cDNA sequence was identical to that of GenBank.Recombinant retroviral plasmid vector pLgGFPSN was constructed successfully.The titer of the packaged recombinant retrovirus was 1 × 106 cfu/ml.Bright green fluorescence of the transfected cells was observed under confocal microscope 48 h after transfection.The transfection rate was 33%.The effective expression of GFP in myoblast infected by recombinant retrovirus lasted for 6 weeks.Conclusion:GFP gene could be effectively and stably expressed in myoblast,which suggests that GFP could act as a marker for studies on myoblast.Shuling Rong Yongxin Lu Yuhua Liao Xiaolin Wang Xiaoqing Li Jiahua Zhang Yanli He 2006Journal of Nanjing Medical University2006,20,4:1
4Effects of caffeic acid phenethyl ester on proliferation of vascular smooth muscle cells in rats显示文摘Objective: To investigate the inhibitory effect of caffeic acid phenethyl ester(CAPE) on the proliferation of vascular smooth muscle cells (VSMC) activated by lipopolysaccharide (LPS) and to clarify its mechanism. Methods: VSMC activated by LPS(1 mg·L-1) were treated with CAPE at different concentrations. The inhibitory effects of CAPE on the proliferation of VSMC were determined by methabenzthiazuron(MTT) colorimetry. The effects of CAPE on the expression of proliferating cell nuclear antigen(PCNA) and Survivin protein in VSMC were evaluated by immunocytochemistry staining technique(SABC method). Cell cycle was analyzed by flow cytometry(FCM) with propidium iodide (PI) labeling method. The relative expression level of Survivin mRNA was measured with real-time quantified RT-PCR technique. Results: CAPE exerted significant inhibitory effects on proliferation of VSMC at concentrations ranging from 5 mg·L-1 to 80 mg·L-1, decreased the rate of cells positive for PCNA and Survivin protein and repressed the expression of Survivin mRNA in a dose- and time-dependent manner(P < 0.05). FCM analysis displayed that CAPE up-regulated the ratio of G0/G1 stages and reduced the percentage of VSMC in S stage(P < 0.05). 5Synthesis and Self-Assembly of Two 1,3-Alternate Thiacalix[4]arenes Derivatives Bearing Amide Groups显示文摘Two 1,3-alternate thiacalix[4]arene derivatives bearing amide groups, 1,3-alternate p-tert-butylthiacalix[4]arene tetraamide (4), and 1,3-alternate p-H-thiacalix[4]arene tetraamide (6) were prepared, and their crystal structures were determined by single-crystal X-ray diffraction method. The steric hindrances posed by tert-butyl groups play an important part in the synthesis and the self-assembly of the two compounds. Compound 6 was synthesized from the corresponding ester, which was obtained by the reaction of acid chloride with ammonia.
