1Bmi-1:At the crossroads of physiological and pathological biology显示文摘Bmi-1 is a member of the Polycomb repressor complex 1 that mediates gene silencing by regulating chromatin structure and is indispensable for self-renewal of both normal and cancer stem cells.Despite three decades of research that have elucidated the transcriptional regulation,post-translational modifications and functions of Bmi-1 in regulating the DNA damage response,cellular bioenergetics,and pathologies,the entire potential of a protein with such varied functions remains to be realized.This review attempts to synthesize the current knowledge on Bmi-1 with an emphasis on its role in both normal physiology and cancer.Additionally,since cancer stem cells are emerging as a new paradigm for therapy resistance,the role of Bmi-1 in this perspective is also highlighted.The wide spectrum of malignancies that implicate Bmi-1 as a signature for stemness and oncogenesis also make it a suitable candidate for therapy.Nonetheless,new approaches are vitally needed to further characterize physiological roles of Bmi-1 with the long-term goal of using Bmi-1 as a prognostic marker and a therapeutic target.Resham Bhattacharya Soumyajit Banerjee Mustafi Mark Street Anindya Dey Shailendra Kumar Dhar Dwivedi 2015Genes & Diseases2015,2,3:15
2Hepatitis B virus therapy:What's the future holding for us?显示文摘Hepatitis B is one of the leading causes of liver cancer worldwide and unfortunately the number of people affected with hepatitis B virus(HBV) infection is still on the rise. Although the HBV has been known to cause fatal illness since decades but the population effected by this lethal virus have still only a few options for its management. The major treatment strategies include interferons and nucleos(t)ide analogues. These agents have so far produced unsatisfactory results in terms of complete virus eradication. Interferons cannot be used for long term therapy because of their potential side effects. Prolong treatment with nucleos(t)ide analogues has also been reported to cause serious side effects besides the increasing resistance by the virus. The need for new innovative solutions for treatment of HBV has been realized by global research institutes and pharmaceutical industry. Present review focuses in detail on the new ideas that are being transformed into therapeutic tools for use as future therapies in HBV infection. Modern drug designing and screening methods have made the drug discovery process shorter and more reliable. HBV therapeutics will take a new turn in coming years owing to these intelligent drug designing and screening methods. 3Carbon Dioxide and Nitrous Oxide Emissions from Naturally Occurring Sulfate-Based Saline Soils at Different Moisture Contents显示文摘Soil salinization may negatively affect microbial processes related to carbon dioxide(CO_2) and nitrous oxide(N_2O) emissions. A short-term laboratory incubation experiment was conducted to investigate the effects of soil electrical conductivity(EC) and moisture content on CO_2 and N_2O emissions from sulfate-based natural saline soils. Three separate 100-m long transects were established along the salinity gradient on a salt-affected agricultural field at Mooreton, North Dakota, USA. Surface soils were collected from four equally spaced sampling positions within each transect, at the depths of 0–15 and 15–30 cm. In the laboratory, artificial soil cores were formed combining soils from both the depths in each transect, and incubated at 60% and 90% water-filled pore space(WFPS) at 25?C. The measured depth-weighted EC of the saturated paste extract(EC_e) across the sampling positions ranged from 0.43 to 4.65 dS m^(-1). Potential nitrogen(N) mineralization rate and CO_2 emissions decreased with increasing soil EC_e, but the relative decline in soil CO_2 emissions with increasing ECe was smaller at 60% WFPS than at 90% WFPS. At 60% WFPS, soil N_2O emissions decreased from 133 μg N_2O-N kg^(-1) soil at EC_e < 0.50 dS m^(-1) to 72 μg N_2O-N kg^(-1) soil at EC_e = 4.65 dS m^(-1). In contrast, at 90% WFPS,soil N_2O emissions increased from 262 μg N_2O-N kg^(-1) soil at EC_e = 0.81 dS m^(-1) to 849 μg N_2O-N kg^(-1) soil at EC_e = 4.65 dS m^(-1), suggesting that N_2O emissions were linked to both soil ECe and moisture content. 4Saline-Sodic Soils:Potential Sources of Nitrous Oxide and Carbon Dioxide Emissions?显示文摘Increasing salt-affected agricultural land due to low precipitation,high surface evaporation,irrigation with saline water,and poor cultural practices has triggered the interest to understand the influence of salt on nitrous oxide(N_2O) and carbon dioxide(CO_2)emissions from soil.Three soils with varying electrical conductivity of saturated paste extract(EC_e)(0.44-7.20 dS m^(-1)) and sodium adsorption ratio of saturated paste extract(SARe)(1.0-27.7),two saline-sodic soils(S2 and S3) and a non-saline,non-sodic soil(S1),were incubated at moisture levels of 40%,60%,and 80%water-filled pore space(WFPS) for 30 d,with or without nitrogen(N)fertilizer addition(urea at 525 μg g^(-1) soil).Evolving CO_2 and N2 O were estimated by analyzing the collected gas samples during the incubation period.Across all moisture and N levels,the cumulative N_2O emissions increased significantly by 39.8%and 42.4%in S2 and S3,respectively,compared to S1.The cumulative CO_2 emission from the three soils did not differ significantly as a result of the complex interactions of salinity and sodicity.Moisture had no significant effect on N_2O emissions,but cumulative CO_2 emissions increased significantly with an increase in moisture.Addition of N significantly increased cumulative N_2O and CO_2 emissions.These showed that saline-sodic soils can be a significant contributor of N_2O to the environment compared to non-saline,non-sodic soils.The application of N fertilizer,irrigation,and precipitation may potentially increase greenhouse gas(N2O and CO_2) releases from saline-sodic soils.Upasana GHOSH Resham THAPA Thomas DESUTTER HE Yangbo Amitava CHATTERJEE 2017Pedosphere2017,27,1:5

18The Compulsion of Foreign Employment: Socio-Economic Circumstances of Developing Country显示文摘Foreign employment is a burning issue for developing countries like Nepal. Nearly thousand Nepalese depart forforeign employment everyday. This paper is focused on exploring the reasons of foreign employment. Both primaryand secondary facts are used in this paper. Primary facts are collected through the case study and observationmethod. The purposive sampling method is used to select the cases. The cases are located in the Pokharametropolitan city of Nepal. Most people have gone abroad after marriage because of the financial burden and theresponsibility of earning money. Lack of cash, superstition, traditional beliefs, political and family conflicts,inter-caste marriages, and family structure have led to foreign employment.
