| 1 | Effect of Nimesulide on proliferation and apoptosis of human hepatoma SMMC-7721 cells显示文摘AIM: Cyclooxygenase-2 (COX-2) has been suggested to beassociated with carcinogenesis. We sought to investigatethe effect of the selective COX-2 inhibitor, Nimesulide onproliferation and apoptosis of SMMC-7721 human hepatomacells.METHODS: This study was carried out on the culture ofhepatic carcinoma SMMC-7721 cell line. Variousconcentrations of Nimesulide (0、200 μmol/L、300 μmol/L、400μmol/L) were added and incubated. Cell proliferation wasdetected with MTT colorimetric assay, cell apoptosis byelectron microscopy, flow cytometry and TUNEL.RESULTS: Nimesulide could significantly inhibit SMMC-7721cells proliferation dose-dependent and in a dependentmanner compared with that of the control group. Theduration lowest inhibition rate produced by Nimesulide inSMMC-7721 cells was 19.06 %, the highest inhibition ratewas 58.49 %. After incubation with Nimesulide for 72 h, themost highest apoptosis rate and apoptosis index of SMMC-7721 cells comparing with those of the control were 21. 20 %+1.62% vs2.24% +0.26% and21.23+ 1.78 vs2.01+0.23(p<0.05).CONCLUSION: The selective COX-2 inhibitor, Nimesulide caninhibit the proliferation of SMMC-7721 cells and increaseapoptssis rate and apoptosis index of SMMC-7721 cells. | 2 | Changes of plasma D(-) -lactate, diamine oxidase and endotoxin in patients with liver cirrhosis显示文摘BACKGROUND: Plasma D(-)-lactate and diamine oxidase (DAO) can reflect patients’ intestinal mucosal condition. We evaluated the changes of plasma D (-)-lactate, DAO and endotoxin activities and their significance in patients with liver cirrhosis. METHODS: Fifty liver cirrhosis patients were enrolled into experimental group and 30 healthy people into control group. The plasma levels of D(-)-lactate, DAO and endo- toxin were detected spectrophotographically. RESULTS: The level of D(-)-lactate was significantly high- er in the experimental group than that in the control group (P<0.01). Significant differences of D (-)-lactate levels were observed in Child-Pugh subgroups of the experimen- tal group (P <0. 01). The level of DAO was significantly higher in the experimental group than that in the control group (P <0.01), but the level of DAO in Child-Pugh sub- group C was significantly lower than that in Child-Pugh subgroup B (P<0.01). The level of endotoxin was signifi- cantly increased in the experimental group except Child Pugh subgroup A (P<0.01). The plasma levels of D(-) lactate, DAO and endotoxin were positively correlated with each other (P<0.01). CONCLUSIONS: The data suggest that both plasma D(-) lactate and DAO activity are sensitive markers for early diagnosis of gut failure and endotoxemia in patients with liver cirrhosis. | 3 | Re-Os isotope dating of molybdenites in the Huang-shaping Pb-Zn-W-Mo polymetallic deposit, Hunan Province, South China and its geological significance显示文摘The large-scale Huangshaping Pb-Zn-W-Mo polymetallic deposit is located in the central Nanling min- eralization zone, South China. Six molybdenite samples from the Huangshaping deposit were selected for Re-Os isotope measurement in order to define the mineralization age of the deposit. It yields a Re-Os isochron age of 154.8±1.9 Ma (2σ ), which is in accordance with the Re-Os model ages of 150.9― 156.9 Ma. This age is about 7 Ma younger than their host granite porphyry, which was dated as 161.6±1.1 Ma by zircon U-Pb method using LA-ICPMS. All these ages demonstrate that the Huang- shaping granite and related Pb-Zn-W-Mo deposit occurred in the middle Yanshanian period, when many other granitoid and related ore deposits emplaced and formed, e.g. the Qitianling granite and Furong tin deposit, the Qianlishan granite and giant Shizhuyuan W-Sn-Mo-Bi deposit and Jinchuantang Sn-Bi deposit in the nearby area. They constitute the main part of the magmatic-metallogenic belt of southern Hunan, and represent the large-scale metallogeny in middle Yanshanian in the area. | 4 | Metallogenesis related to Mesozoic Granitoids in the Nanling Range, South China and Their Geodynamic Settings显示文摘Affected by the compressive stress from the South-Central (Indo-China) Peninsula, the Indosinianorogenesis, characterized by collision, thrust and uplifting, took place inside the South China Plate during 250?230 Ma.The ages of the Indosinian granitoids in the Nanling Range and vicinity areas are mostly 240?205 Ma, indicating that theywere emplaced in both late collision and post-collision geodynamic environments. No important granite-relatedmetallogenesis occurred in this duration. A post-orogenic setting started at the beginning of the Yanshanian Period, whichcontrolled large-scale granitic magmatism and related metallogenesis. This paper makes the first attempt to divide theYanshanian Period into three sub-periods, i.e. the early, middle and late Yanshanian Periods, based mainly on the featuresof magmatism, especially granitoids and related metallogenesis and their geodynamic environments. The magmaticassociation of the Early Yanshanian (about 185?170 Ma) comprises four categories of magmatism, i.e. basalt, bimodalvolcanics, A-type granite and intraplate high-K calc-alkaline (HKCA) magmatism, which indicates an extension-thinningof lithosphere and upwelling of mantle material to a relative small and local extent. Pb-Zn, Cu and Au mineralizationsassociated with HKCA magmatism represents the first high tide of Mesozoic metallogenesis in the Nanling Range area.During the middle Yanshanian, the lithosphere was subjected to more extensive and intensive extending and thinning, andhence mantle upwelling and basaltic magma underplating caused a great amount of crust remelting granitoids. This periodcan be further divided into two stages. The first stage (170?150 Ma) is represented by large-scale emplacement of crustremelting granites with local tungsten mineralization at its end. The second stage (150?140 Ma) is the most important timeof large-scale mineralizations of non-ferrous and rare metals, e.g. W, Sn, Nb-Ta, Bi, Mo, Be, in the Nanling Range area.The late Yanshanian (140?65 Ma) was generally characterized by full extension and breakup of the lithosphere of SouthChina. However, owing to the influence of the Pacific Plate movement, the eastern part of South China was predominatedby subduction-related compression, which resulted in magmatism of calc-alkaline and shoshonite series and relatedmetallogeneses of Au, Ag, Pb-Zn, Cu and (Mo, Sn), followed by extension in its late stage. | 5 | Zircon U-Pb dating confirms existence of a Caledonian scheelite-bearing aplitic vein in the Penggongmiao granite batholith, South Hunan显示文摘Granite at Penggongmiao is a large batholith in the Nanling Range, with an outcrop area of over 900 km 2 . There are many scheelite-quartz veins around the granite. LA-ICP-MS U-Pb dating was carried out for zircons from the granite. The middlecoarse-grained biotite granite has U-Pb ages of 435 to 436 Ma. Ages of 426.5±2.5 Ma were obtained for aplitic dyke cross-cutting the granite. The scheelite of magmatic origin in the aplite dyke was identified from petrographic investigation. This demonstrates that W-bearing granites of Early Paleozoic (corresponding to the Caledonian orogensis in the traditional sense) occur in the Nanling Range. This finding has important implications for the ore-forming potential of Paleozoic granites and on the extent of Mesozoic mineralization. Thus it merits performing an intensive study of Paleozoic granites in South China. | 6 | Role of VEGF and CD44v6 in differentiating benign from malignant ascites显示文摘AIM: To detect the vascular endothelial growth factor (VEGF)and soluble splice variant 6 of CD44 (sCD44v6) levels in ascites and to explore their role in differentiating benign from malignant ascites.METHODS: Cirrhotic ascites (n=36), tuberculosis ascites (n=8) and malignant ascites (n=23) were collected and studied. Concentrations of soluble VEGF and sCD44v6 in various kinds of ascites (n=67) were measured using a sandwich enzyme-linked immunoadsorbent assay.RESULTS: VEGF and sCD44v6 levels in malignant ascites were 640.74±264.81 pg/ml and 89.22±38.20 ng/ml,respectively, both of which were significantly higher than those in cirrhotic ascites and tuberculous ascites (q=18.98,11.89 and q=8.92, 5.09; P<0.01). However, the levels of VEGF and sCD44v6 in cirrhotic and tuberculous ascites had no significant difference (q=0.48, 0.75; P>0.05).Furthermore, VEGF levels in malignant ascites in patients with ovarian cancer were higher than those with gastric and colon cancer (q=5.03, 6.79; P<0.01, respectively). But differences of VEGF levels between gastric and colon cancer were not significant (q=1.90, P>0.05). Whereas, sCD44v6 levels in malignant ascites from patients with ovadan, gastric and colon cancer had no significant difference (q=0.06, 0.91,0.35;P>0.05, respectirely). | 10 | He-Ar isotope geochemistry of the Yaoling-Meiziwo tungsten deposit,North Guangdong Province:Constraints on Yanshanian crust-mantle interaction and metallogenesis in SE China显示文摘Isotopic abundances and ratios of He and Ar found in inclusion fluids in pyrites formed in the Yaoling-Meiziwo tungsten miner-alization epoch show that the concentration of 4He varies widely,from 1.54×10-7 cm3 STP/g to 2609×10-7 cm3 STP/g.3He is 0.759×10-12 cm3 STP/g-3.463×10-12 cm3 STP/g.3He/4He is 0.0043-4.362 Ra,varying from crustal to mantle values.The concen-tration of 40Ar ranges from 0.624×10-7 cm3 STP/g to 8.89×10-7 cm3 STP/g.The 40Ar/36Ar varies extensively,from 330 to 2952,between atmospheric and crustal or mantle radiogenic values.Mantle-derived He is present in ore-forming fluids and the calcu-lated average proportion of the mantle He is 22%;the maximum is 67%.Our research results show that mantle-derived fluids play a significant role in tungsten mineralization.The fractionation of He and Ar indicate that there was 4He-enriched air-saturated water(MSAW) in the ore-forming fluid.The ore-forming fluid was a mixture of mantle fluid,crustal magmatic fluid and MSAW.The occurrence of a mantle component in ore-forming fluid indicates the large-scale W and Sn mineralization,including Yaol-ing-Meiziwo,in southeastern China was the result of crust and mantle interaction.The underplating or intrusion of voluminous basaltic magma formed by partial melting of the upper mantle provided the necessary heat to cause partial melting of the crust and the generation of voluminous S-type granitic magmas.Crustal magmatic fluid and mantle fluid with high 3He/4He were released from magma crystallization and fractionation,mixed with the circulating modified air-saturated water,and filled the extensional tectonic fractures,leading to the formation of world-class W and Sn deposits in southeastern China. | ZHAI Wei SUN XiaoMing WU YunShan SUN YanYan HUA RenMin YE XianRen | 2012 | Chinese Science Bulletin2012,57,10: | 16 |
| 11 | Detection of type Ⅳ collagenase activity in malignant ascites显示文摘AIM: Type IV collagenase participates in invasion and metastasis of cancer cells. Malignant asdtes is a manifestation of advanced malignant disease that is associated with invasion and metastasis of the peritoneal cavity. Thus, it is reasonable to hypothesize that type IV collagenase is linked to malignant ascites. The purpose of our study was to detect type IV collagenase activity in malignant ascites so as to provide the scientific basis for clinic diagnosis and treatment of malignant ascites. METHODS: Cirrhotic ascites (n=36), tuberculous ascites (n=8) and malignant ascites (n=-23) from patients with gastric cancer (n=6), colon cancer (n=5), ovarian cancer (n=8) and other cancers (n=4), including 2 hepatocellular cancers, 1 pancreatic cancer, 1 primary peritoneal carcinoma were collected by paracentesis. The ascites were made cellfree by centrifugation and stored frozen at -70℃ before determination. Type IV collagenase activity was determined by gelatin zymography. RESULTS: The activity of matrix metalloproteinases-2 and -9 could not be detected in ascites of hepatic cirrhosis and tuberculous peritonitis but could be detected in 20 and 18 out of 23 malignant ascites respectively. The positive rate of type IV collagenase (MMP-2, 87.0% and MMP-9, 78.3%) was higher than that by routine ascites tests (P<0.01) in malignant ascites. Furthermore, the activity of MMP-2 was higher than that of MMP-9 (P=0.022<0.05). CONCLUSION: Type IV collagenase is positive in malignant ascites. Detection of type IV collagenase activity is useful in qualitative diagnosis of ascites. The purpose is to develop further an exact relationship between SVMs and the statistical learning theory (SLT). As a representative, the standard C-support vector classification (C-SVC) is considered here. More precisely, we show that the decision function obtained by C-SVC is just one of the decision functions obtained by solving the optimization problem derived directly from the structural risk minimization principle. The kinetic mechanism of hydrogen absorption is not affected by initial hydrogen pressure. Temperature does not influence the rate of hydrogen absorption obviously. In the prior and later period of hydrogen absorption the rate-controlling step is chemical reaction and hydrogen diffusion in the hydride phase respectively for MLNi3.8(Co,Mn,Al)1.2 alloy. | 14 | Clinical and endoscopic analysis of gastric Dieulafoy’s lesion显示文摘AIM:To investigate the incidence,location,clinical presentation,diagnosis and effectiveness of endoscopic treatment of gastric Dieulafoy's lesion(DL)in China. METHODS:All patients who received emergency upper gastrointestinal(GI)endoscopy due to gastric DL from February 2000 to August 2008 at GI endoscopy center of Renmin Hospital of Wuhan University were included in this study.The clinical presentation,medical history,location and characteristics of DL methods and effectiveness of therapy of patients with DL were retrospectively analysed by chart reviews.Long-term follow-up data were collected at outpatient clinics or telephone interviews. RESULTS:Fifteen patients were diagnosized with DL,which account for 1.04%of the source of bleed- ing in acute non-variceal upper GI bleeding.Common comorbidities were found in one patient with hypertension and diabetic mellitus.Hemoclip or combined therapy with hemoclip produced primary hemostasis in 92.8%(13/14) of patients. | 15 | Expression of TFF2 and Helicobacter pylori infection in carcinogenesis of gastric mucosa显示文摘AIM: To investigate the expression of TFF2 and Helicobacter pyloriinfection in carcinogenesis of gastric mucosa.METHODS: The expression of TFF2 was immunohistochemically analyzed in paraffin-embedded samples from 119 patients with endoscopic biopsy and subtotal gastrectomy specimens of gastric mucosal lesions, including 16 cases of chronic superficial gastritis (CSG), 20 chronic atrophic gastritis (CAG),35 intestinal metaplasia (IN), 23 gastric epithelial dysplasia (GED) and 25 gastric carcinoma (CA), and Helicobacter pylori infection was detected by Warthin-Starry staining.RESULTS: 1:TFF2 was located in the cytoplasm of gastrk mucous neck cell. The expression of TFF2 was 100 %,100 %, 0, 56.5 % and 0 in CSGs, CAGs, INs, GEDs and CAs, respectively. 2: The value of TFF2 positive cell density in CSG with Helicobacter pyloriinfection was higher than that without Helicobacter pyloriinfection. (52.89±7.27vs46.49±13.04, P>0.05); But the value of TFF2 positive cell density in CAG and GED with Helicobacter pyloriinfection was significantly lower than that without Helicobacter pylori infection (18.17±4.09 vs 37.93±13.80, P<0.01 and 14.44±9.32 vs 24.84±10.22, P<0.05).CONCLUSION: Increase of TFF2 expression in CSG is perhaps associated with the protective mechanism after gastric mucosal injury. Decrease of TFF2 expression in CAG possibly attributes to the decrease in the number of gastric gland cell expressing TFF2. Re-expression of TFF2 in gastric epithelial dysplasia implies that TFF2 possibly contributes to the initiation of gastric carcinoma. | 16 | Effects of HBV gene variations on disease development and antiviral therapy for patients with chronic hepatitis B显示文摘Viral variation may change pathogenicity, escape immunity, lead to persistence infection, and cause drug resistance against antiviral therapy. This study was undertaken to investigate the effects of HBV gene variation on the progression of disease and on the efficacy of antiviral therapy for patients with chronic hepatitis B(CHB). METHODS:Hepatitis B virus (HBV) gene mutational sites were detected using gene chip in selected hepatitis B patients. RESULTS:In the patients HBeAg did not show serologic conversion or HBeAg(-)/anti-HBe(+), but their HBV DNA remained positive 24 weeks after α-interferon therapy, which was associated with mutations of nt1896, nt1814, nt1762 and nt1764. In the patients, that HBV DNA levels decreased or were undetectable, but rebounded later after antiviral therapy by lamivudine was associated with mutations of aa528 and(or) aa552(i.e.YMDD mutation), which resulted in lamivudine-resistance. YMDD mutation was prone to occur 52 weeks after lamivudine therapy in some chronic hepatitis B patients (26.4%). Nt1896 mutation was common in most chronic hepatitis B patients (68.5%). Chronic severe hepatitis, cirrhosis, and primary liver carcinoma were related to the mutations of nt1896, nt1762 and nt1764. CONCLUSIONS:HBV gene mutations could aggravate patient’s condition and affect the efficacy of antiviral therapy. | 17 | Preventive effect of hydrotalcite on gastric mucosal injury in rats induced by taurocholate显示文摘AIM: To study the preventive effect of hydrotalcite on gastric mucosal injury in rat induced by taurocholate, and to investigate the relationship between the protective mechanism of hydrotalcite and the expression of trefoil factor family 2 (TFF2) mRNA and c-fos protein.METHODS: Forty five male Wistar rats were randomly divided into hydrotalcite group, ranitidine group and control group. Gastric mucosal injury was induced by introgastric acidified taurocholate. OD value of TFF2 mRNA expression in gastric mucous cells was determined by hybridization and computer image analysis system. OD value of c-fos protein expression in gastric mucous cells was measured by immunohistochemistry and computer image analysis system.RESULTS: The gross mucosal injury index in hydrotalcite group was significantly lower than that in ranitidine group and control group (8.60±2.20 vs 16.32±4.27, 29.53±5.39;P<0.05, P<0.01). The expression level of TFF2 mRNA in hydrotalcite group was markedly higher than that in ranitidine group and control group (0.56±0.09 vs 0.30±0.05, 0.28±0.03,P<0.05). | 19 | Potential role of retina as a biomarker for progression of Parkinson's disease显示文摘Optical coherence tomography (OCT) noninvasively quantifies the thickness of the retinal nerve fiber layer (RNFL). OCT has been studied in several neuro-ophthalmic conditions, including Parkinson's disease (PD). Recent studies suggest that the quantitative analysis of RNFL can be precisely and noninvasively done by OCT scans and the results suggest that the thickness of RNFL is significantly decreased in patients with PD compared with age-matched controls and the foveal retinal thickness correlates with disease severity in PD. In this article, the application of OCT imaging of the retina in PD was reviewed. Literature survey of PubMed was carried out using the search terms of 'Optical Coherence Tomography' combined with 'Parkinson's Disease' and 'retinal nerve fiber layer' (without restriction to the year of publication). Some related articles were also included. The search was completed in Jul. 2011 and revised and updated as necessary. | 20 | Factors impacting nanoindentation testing results of the cuticle of dung beetle Copris ochus Motschulsky显示文摘The cuticle of dung beetle is a layered composite material in micro- or nano-scale. Dung beetle can fly, walk and dig. It can shovel and compact dung of mammals into balls. It use foreleg to walk, midleg and hindleg to hold and impel dung ball. Its two foreleges as digging legs are developed. The factors impacting the nanoindentation testing results of the femur cuticle of forelegs of dung beetle Copris ochus Motschulsky were examined. The nanomechanical test instrument used for the tests was Hysitron nanomechanical system. The results shown that the holding time and loading time are important factors im- pacting the accuracy of such indentation properties as reduced modulus (Er) and the harness ( H ) of the femur cuticle of the forelegs of dung beetle Copris ochus Motschulsky in nanoscale. There exists a threshold holding time of 20 s for the reduced modulus of the femur cuticle. The tests of nanoindentation creep property and the regression analysis of relationship between the depth increment at the maximum load and the time further confirmed the correction of the above threshold holding time. There exist visco-elastic-plastic behaviour and creep phenomenon in the femur cuticle during indenting. Its creep property during the holding procedure at maximum load can be regressed by a general logarithmic equation.
