| 1 | Achievements,challenges and global implications of China’s carbon neutral pledge显示文摘China has been committed to achieving carbon neutrality by 2060.China’s pledge of carbon neutrality will play an essential role in galvanising global climate action,which has been largely deferred by the Covid-19 pandemic.China’s carbon neutrality could reduce global warming by approximately 0.2–0.3°C and save around 1.8 million people from premature death due to air pollution.Along with domestic benefits,China’s pledge of carbon neutrality is a“game-changer”for global climate action and can inspire other large carbon emitters to contribute actively to mitigate carbon emissions,particularly countries along the Belt and Road Initiative(BRI)routes.In order to achieve carbon neutrality by 2060,it is necessary to decarbonise all sectors in China,including energy,industry,transportation,construction,and agriculture.However,this transition will be very challenging,because major technological breakthroughs and large-scale investments are required.Strong policies and implementation plans are essential,including sustainable demand,decarbonizing electricity,electrification,fuel switching,and negative emissions.In particular,if China can peak carbon emissions earlier,it can lower the costs of the carbon neutral transition and make it easier to do so over a longer time horizon.China’s pledge of carbon neutrality by 2060 and recent pledges at the 26th UN Climate Change Conference of the Parties(COP26)are significant contributions and critical steps for global climate action.However,countries worldwide need to achieve carbon neutrality to keep the global temperature from growing beyond the level that will cause catastrophic damages globally. | Hong Yang Xianjin Huang Jianlin Hu Julian R.Thompson Roger J.Flower | 2022 | Frontiers of Environmental Science & Engineering2022,16,8: | 2 |

| 4 | Osteocyte Egln1/Phd2 links oxygen sensing and biomineralization via FGF23显示文摘Osteocytes act within a hypoxic environment to control key steps in bone formation.FGF23,a critical phosphate-regulating hormone,is stimulated by low oxygen/iron in acute and chronic diseases,however the molecular mechanisms directing this process remain unclear.Our goal was to identify the osteocyte factors responsible for FGF23 production driven by changes in oxygen/iron utilization.Hypoxia-inducible factor-prolyl hydroxylase inhibitors(HIF-PHI)which stabilize HIF transcription factors,increased Fgf23 in normal mice,as well as in osteocyte-like cells;in mice with conditional osteocyte Fgf23 deletion,circulating i FGF23 was suppressed.An inducible MSC cell line(‘MPC2’)underwent FG-4592 treatment and ATACseq/RNAseq,and demonstrated that differentiated osteocytes significantly increased HIF genomic accessibility versus progenitor cells.Integrative genomics also revealed increased prolyl hydroxylase Egln1(Phd2)chromatin accessibility and expression,which was positively associated with osteocyte differentiation.In mice with chronic kidney disease(CKD),Phd1-3 enzymes were suppressed,consistent with FGF23 upregulation in this model.Conditional loss of Phd2 from osteocytes in vivo resulted in upregulated Fgf23,in line with our findings that the MPC2 cell line lacking Phd2(CRISPR Phd2-KO cells)constitutively activated Fgf23 that was abolished by HIF1αblockade.In vitro,Phd2-KO cells lost iron-mediated suppression of Fgf23 and this activity was not compensated for by Phd1 or-3.In sum,osteocytes become adapted to oxygen/iron sensing during differentiation and are directly sensitive to bioavailable iron.Further,Phd2 is a critical mediator of osteocyte FGF23 production,thus our collective studies may provide new therapeutic targets for skeletal diseases involving disturbed oxygen/iron sensing. | Megan L.Noonan Pu Ni Emmanuel Solis Yamil G.Marambio Rafiou Agoro Xiaona Chu Yue Wang Hongyu Gao Xiaoling Xuei Erica L.Clinkenbeard Guanglong Jiang Sheng Liu Steve Stegen Geert Carmeliet William R.Thompson Yunlong Liu Jun Wan Kenneth E.White | 2023 | Bone Research2023,11,1: | 1 |
