| 1 | A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. | 3 | Impact of China’s Air Pollution Prevention and Control Action Plan on PM2.5 chemical composition over eastern China显示文摘China promulgated the Air Pollution Prevention and Control Action Plan(the Action Plan)in 2013 and developed stringent control measures to mitigate fine particulate matter(PM2.5) pollution.Here,we investigated the PM2.5 chemical composition changes over eastern China associated with the Action Plan during 2013-2017 using satellite-based PM2.5 chemical composition data derived using CMAQ simulations and satellite inputs.The PM2.5 concentrations decreased considerably during this time as a result of the reductions in all chemical species in PM2.5.The population-weighted mean concentrations over eastern China decreased from 11.1 to 6.7μgm-3 for SO42-,13.8-13.1μgm-3 for NO3-,7.4-5.8μgm-3 for NH4+,9.9-8.4μgm-3 for OM,4.6-3.8μg m-3 for BC and 12.9-9.6μg m-3 for other species in PM2.5.SO42-had the largest reduction of 40%,while NO3-had the lowest reduction of 5%,resulting in a greater fraction of NO3-and a smaller fraction of SO42-in PM2.5.Among the three key regions,Beijing-Tianjin-Hebei had the largest reduction in PM2.5 and its chemical compositions.The decrease in SO42-concentrations was in line with the reduction of SO2 emissions,and the major driver of the SO2 emission reductions was the industrial sector.The decrease in NO3 concentrations was limited because the decrease in SO2 emissions and the stable NH3 emissions facilitated the formation of NO3-from HNO3,which partially offset the reduction in NOx emissions driven by the power sector.To mitigate PM2.5 pollution more effectively,future efforts are needed to reduce NH3 emissions. | Guannan GENG Qingyang XIAO Yixuan ZHENG Dan TONG Yuxuan ZHANG Xiaoye ZHANG Qiang ZHANG Kebin HE Yang LIU | 2019 | Science China Earth Sciences2019,62,12: | 16 |

| 5 | Characterization of a Novel Anti-DR5 Monoclonal Antibody WD1 with the Potential to Induce Tumor Cell Apoptosis显示文摘TNF-related apoptosis-inducing ligand (TRAIL) is a TNF family member capable of inducing apoptosis. Death receptor 5 (DR 5) is a key receptor of TRAIL and plays an important role in TRAIL-induced apoptosis. To prepare monoclonal antibodies (mAbs) against DR5,cDNA encoding soluble DR5 (sDR5) was firstly amplified by reverse transcriptase-polymerase chain reaction (RT-PCR) with specific primers,and then inserted into a prokaryotic expression vector pET-30a. The recombinant plasmid was expressed in Escherichia coli strain BL21 (DE3),and sDR5 was purified by nickel affinity chromatography. As an antigen,sDR5 was used to immunize mice. Hybridomas secreting antibodies against sDR5 were identified. One positive clone was selected to produce antibody,WD1. ELISA and immunofluorescence demonstrated that WD1 could bind recombinant sDR5 and membrane-bound DR5 (mDR5) on Jurkat and Molt-4 cells. ATPLite assays showed that Jurkat and Molt-4 cells were sensitive to the antibody in a dose dependent manner. The Annexin V/PI assays and Giemsa's staining both showed that WD1 could induce Jurkat cell apoptosis efficiently. Transient transfection of 293T cells and indirect immunofluorescence assay demonstrated that mAb (WD1) couldn't cross-react with DR4. Our findings indicated that the novel antibody,WD1 could act as a direct agonist,bind DR5 characteristically,and initiate efficient apoptotic signaling and tumor regression. | 9 | Identification and evaluation of shale oil micromigration and its petroleum geological significance显示文摘Taking the Lower Permian Fengcheng Formation shale in Mahu Sag of Junggar Basin,NW China,as an example,core observation,test analysis,geological analysis and numerical simulation were applied to identify the shale oil micro-migration phenomenon.The hydrocarbon micro-migration in shale oil was quantitatively evaluated and verified by a self-created hydrocarbon expulsion potential method,and the petroleum geological significance of shale oil micro-migration evaluation was determined.Results show that significant micro-migration can be recognized between the organic-rich lamina and organic-poor lamina.The organic-rich lamina has strong hydrocarbon generation ability.The heavy components of hydrocarbon preferentially retained by kerogen swelling or adsorption,while the light components of hydrocarbon were migrated and accumulated to the interbedded felsic or carbonate organic-poor laminae as free oil.About 69% of the Fengcheng Formation shale samples in Well MY1 exhibit hydrocarbon charging phenomenon,while 31% of those exhibit hydrocarbon expulsion phenomenon.The reliability of the micro-migration evaluation results was verified by combining the group components based on the geochromatography effect,two-dimension nuclear magnetic resonance analysis,and the geochemical behavior of inorganic manganese elements in the process of hydrocarbon migration.Micro-migration is a bridge connecting the hydrocarbon accumulation elements in shale formations,which reflects the whole process of shale oil generation,expulsion and accumulation,and controls the content and composition of shale oil.The identification and evaluation of shale oil micro-migration will provide new perspectives for dynamically differential enrichment mechanism of shale oil and establishing a“multi-peak model in oil generation”of shale. | HU Tao JIANG Fujie PANG Xiongqi LIU Yuan WU Guanyun ZHOU Kuo XIAO Huiyi JIANG Zhenxue LI Maowen JIANG Shu HUANG Liliang CHEN Dongxia MENG Qingyang | 2024 | Petroleum Exploration and Development2024,51,1: | 0 |
| 10 | MEIC-global-CO_(2):A new global CO_(2)emission inventory with highly-resolved source category and sub-country information显示文摘CO_(2)emission inventory provides fundamental data for climate research and emission mitigation.Currently,most global CO_(2)emission inventories were developed with energy statistics from International Energy Agency(IEA)and were available at country level with limited source categories.Here,as the first step toward a high-resolution and dynamic updated global CO_(2)emission database,we developed a data-driven approach to construct seamless and highly-resolved energy consumption data cubes for 208 countries/territories,797 sub-country administrative divisions in 29 countries,42 fuel types,and 52 sectors,with the fusion of activity data from 24 international statistics and 65 regional/local statistics.Global CO_(2)emissions from fossil fuel combustion and cement production in 1970–2021 were then estimated with highly-resolved source category(1,484 of total)and sub-country information(797 of total).Specifically,73%of global CO_(2)emissions in 2021 were estimated with sub-country information,providing considerably improved spatial resolution for global CO_(2)emission accounting.With the support of detailed information,the dynamics of global CO_(2)emissions across sectors and fuel types were presented,representing the evolution of global economy and progress of climate mitigation.Remarkable differences of sectoral contribution were found across sub-country administrative divisions within a given country,revealing the uneven distribution of energy and economic structure among different regions.Our estimates were generally consistent with existing databases at aggregated level for global total or large emitters,while large discrepancies were observed for middle and small emitters.Our database,named the Multiresolution Emission Inventory model for Climate and air pollution research(MEIC)is publicly available through http://gffzz6c575fb1d0a54d36hqnfnk9bwbf696knw.ffgz.tsg.suse.edu.cn with highly-resolved information and timely update,which provides an independent carbon emission accounting data source for climate research. | Ruochong XU Dan TONG Qingyang XIAO Xinying QIN Cuihong CHEN Liu YAN Jing CHENG Can CUI Hanwen HU Wenyu LIU Xizhe YAN Huaxuan WANG Xiaodong LIU Guannan GENG Yu LEI Dabo GUAN Kebin HE Qiang ZHANG | 2024 | Science China Earth Sciences2024,67,2: | 0 |
