| 1 | A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. | 4 | Characterization of a Novel Anti-DR5 Monoclonal Antibody WD1 with the Potential to Induce Tumor Cell Apoptosis显示文摘TNF-related apoptosis-inducing ligand (TRAIL) is a TNF family member capable of inducing apoptosis. Death receptor 5 (DR 5) is a key receptor of TRAIL and plays an important role in TRAIL-induced apoptosis. To prepare monoclonal antibodies (mAbs) against DR5,cDNA encoding soluble DR5 (sDR5) was firstly amplified by reverse transcriptase-polymerase chain reaction (RT-PCR) with specific primers,and then inserted into a prokaryotic expression vector pET-30a. The recombinant plasmid was expressed in Escherichia coli strain BL21 (DE3),and sDR5 was purified by nickel affinity chromatography. As an antigen,sDR5 was used to immunize mice. Hybridomas secreting antibodies against sDR5 were identified. One positive clone was selected to produce antibody,WD1. ELISA and immunofluorescence demonstrated that WD1 could bind recombinant sDR5 and membrane-bound DR5 (mDR5) on Jurkat and Molt-4 cells. ATPLite assays showed that Jurkat and Molt-4 cells were sensitive to the antibody in a dose dependent manner. The Annexin V/PI assays and Giemsa's staining both showed that WD1 could induce Jurkat cell apoptosis efficiently. Transient transfection of 293T cells and indirect immunofluorescence assay demonstrated that mAb (WD1) couldn't cross-react with DR4. Our findings indicated that the novel antibody,WD1 could act as a direct agonist,bind DR5 characteristically,and initiate efficient apoptotic signaling and tumor regression. | 5 | Influence factors of two dimensional position map on photomultiplier detector block designed by quadrant sharing technique显示文摘The position decoding accuracy and the spatial resolution of positron emission tomography detectors are greatly influenced by the performance of the two-dimensional position map,including the gain uniformity of photomultiplier tube (PMT),the baseline offset of the PMT signals and the accuracy of analogue to digital converter (ADC).In this work,a PMT-quadrant sharing detector was designed.Two data acquisition platforms are employed to conduct the influence factors on the two-dimensional position map performances,one was that the waveforms of the PMT signals were scanned by the sequence acquisition mode based on the oscilloscope of LeCroy waveRunner 204MXi-A,and another was a self-developed high speed ADC data acquisition module.Results show that the event decoding positions were concentrated on the PMT with higher gain,the position map was distorted at the baseline offset of signal,and the cross-line artifacts were caused by the insufficient ADC sampling bit for a larger size position map.All the parameters need be adjusted properly to stabilize a real system,and the flexible oscilloscope platform can be used to design the detector block and the other platform with high ADC accuracy.Likely,the electrical circuit with a proper ADC accuracy adjusts the PMT gains and baseline offsets. | WEI Qingyang WANG Shi Ma Tianyu MA Fei DAI Tiantian XU Tianpeng WU Zhaoxia JIN Yongjie LIU Yaqiang | 2011 | Nuclear Science and Techniques2011,22,4: | 3 |
| 6 | GATE simulation based feasibility studies of in-beam PET monitoring in ^(12)C beam cancer therapy显示文摘In comparison with conventional radiotherapy techniques,12C beam therapy has its significant advantage in cancer treatment because the radiation dose are mostly concentrated near the Bragg peak region and damage to normal tissues along the beam path is thus greatly reduced.In-beam PET provides a way to monitor dose distribution inside human body since several kinds of positron-emitting nuclei are produced through the interaction between 12C beam and body matters.In this work,we study the quantitative relationship between the spatial location of the Bragg peak and the spatial distribution of positrons produced by positron-emitting nuclei.Monte Carlo package GATE is used to simulate the interactions between the incident 12C beam of different energies(337.5,270.0 and 195.0 MeV/u) and various target matters(water,muscle and spine bone).Several data post-processing operations are performed on the simulated positron-emitting nuclei distribution data to mimic the impacts of positron generation and finite spatial resolution of a typical PET imaging system.Simulation results are compared to published experimental data for verification.In all the simulation cases,we find that 10C and 11C are two dominant positron-emitting nuclei,and there exists a significant correlation between the spatial distributions of deposited energy and positrons.Therefore,we conclude that it is possible to determine the location of Bragg peak with 1 mm accuracy using current PET imaging systems by detecting the falling edge of the positron distribution map in depth direction. | WU Jing LIU Yaqiang MA Tianyu WEI Qingyang WANG Shi CHENG Jianping | 2010 | Nuclear Science and Techniques2010,21,5: | 2 |

| 9 | Regulating off-centering distortion maximizes photoluminescence in halide perovskites显示文摘Metal halide perovskites possess unique atomic and electronic configurations that endow them with high defect tolerance and enable high-performance photovoltaics and optoelectronics.Perovskite light-emitting diodes have achieved an external quantum efficiency of over 20%.Despite tremendous progress,fundamental questions remain,such as how structural distortion affects the optical properties.Addressing their relationships is considerably challenging due to the scarcity of effective diagnostic tools during structural and property tuning as well as the limited tunability achievable by conventional methods.Here,using pressure and chemical methods to regulate the metal off-centering distortion,we demonstrate the giant tunability of photoluminescence(PL)in both the intensity(>20 times)and wavelength(>180 nm/GPa)in the highly distorted halide perovskites[CH_(3)NH_(3)GeI_(3),HC(NH_(2))2GeI_(3),and Cs Ge I_(3)].Using advanced in situ high-pressure probes and first-principles calculations,we quantitatively reveal a universal relationship whereby regulating the level of off-centering distortion towards 0.2 leads to the best PL performance in the halide perovskites.By applying this principle,intense PL can still be induced by substituting CH_(3)NH_(3)^(+)with Cs^(+)to control the distortion in(CH_(3)NH_(3))_(1-x)Cs_(x)GeI_(3),where the chemical substitution plays a similar role as external pressure.The compression of a fully substituted sample of CsGe I_(3)further tunes the distortion to the optimal value at 0.7 GPa,which maximizes the emission with a10-fold enhancement.This work not only demonstrates a quantitative relationship between structural distortion and PL property of the halide perovskites but also illustrates the use of knowledge gained from high-pressure research to achieve the desired properties by ambient methods. | Xujie Lv Constantinos Stoumpos Qingyang Hu Xuedan Ma Dongzhou Zhang Songhao Guo Justin Hoffman Kejun Bu Xiaofeng Guo Yingqi Wang Cheng Ji Haijie Chen Hongwu Xu Quanxi Jia Wenge Yang Mercouri G.Kanatzidis Ho-Kwang Mao | 2021 | National Science Review2021,8,9: | 1 |
| 10 | Real-time,in situ probing of gamma radiation damage with packaged integrated photonic chips显示文摘Integrated photonics is poised to become a mainstream solution for high-speed data communications and sensing in harsh radiation environments,such as outer space,high-energy physics facilities,nuclear power plants,and test fusion reactors.Understanding the impact of radiation damage in optical materials and devices is thus a prerequisite to building radiation-hard photonic systems for these applications.In this paper,we report real-time,in situ analysis of radiation damage in integrated photonic devices.The devices,integrated with an optical fiber array package and a baseline-correction temperature sensor,can be remotely interrogated while exposed to ionizing radiation over a long period without compromising their structural and optical integrity.We also introduce a method to deconvolve the radiation damage responses from different constituent materials in a device.The approach was implemented to quantify gamma radiation damage and post-radiation relaxation behavior of SiO2-cladded SiC photonic devices.Our findings suggest that densification induced by Compton scattering displacement defects is the primary mechanism for the observed index change in SiC.Additionally,post-radiation relaxation in amorphous SiC does not restore the original pre-irradiated structural state of the material.Our results further point to the potential of realizing radiation-hard photonic device designs taking advantage of the opposite signs of radiation-induced index changes in SiC and SiO2. | QINGYANG DU JEROME MICHON BINGZHAO LI DEREK KITA DANHAO MA HAIJIE ZUO SHAOLIANG YU TIAN GU ANURADHA AGARWAL MO LI JUEJUN HU | 2020 | Photonics Research2020,8,2: | 1 |

| 12 | Intact regulation of G1/S transition renders esophageal squamous cell carcinoma sensitive to PI3Kαinhibitors显示文摘Phosphatidylinositol 3-kinase alpha(PI3Kα)inhibitors are currently evaluated for the therapy of esophageal squamous cell carcinoma(ESCC).It is of great importance to identify potential biomarkers to predict or monitor the efficacy of PI3Kαinhibitors in an aim to improve the clinical responsive rate in ESCC.Here,ESCC PDXs with CCND1 amplification were found to be more sensitive to CYH33,a novel PI3Kα-selective inhibitor currently in clinical trials for the treatment of advanced solid tumors including ESCC.Elevated level of cyclin D1,p21 and Rb was found in CYH33-sensitive ESCC cells compared to those in resistant cells.CYH33 significantly arrested sensitive cells but not resistant cells at G1 phase,which was associated with accumulation of p21 and suppression of Rb phosphorylation by CDK4/6 and CDK2.Hypo-phosphorylation of Rb attenuated the transcriptional activation of SKP2 by E2F1,which in turn hindered SKP2-mediated degradation of p21 and reinforced accumulation of p21.Moreover,CDK4/6 inhibitors sensitized resistant ESCC cells and PDXs to CYH33.These findings provided mechanistic rationale to evaluate PI3Kαinhibitors in ESCC patients harboring amplified CCND1 and the combined regimen with CDK4/6 inhibitors in ESCC with proficient Rb. | Xu Zhang Yuxiang Wang Xi Zhang Yanyan Shen Kang Yang Qingyang Ma Yuemei Qiao Jiajie Shi Yi Wang Lan Xu Biyu Yang Gaoxiang Ge Landian Hu Xiangyin Kong Chunhao Yang Yi Chen Jian Ding Linghua Meng | 2023 | Signal Transduction and Targeted Therapy2023,8,5: | 0 |

| 14 | Effects of edge on graphene plasmons as revealed by infrared nanoimaging显示文摘We used scattering-type scanning near-field optical microscopy(s-SNOM)to investigate the plasmonic properties of edges in well-defined graphene nanostructures,including sharp tapers,nanoribbons and nanogaps,which were all fabricated via the growth-etching chemical vapor deposition(GECVD)method.The obtained near-field images revealed the localized plasmon modes along the graphene nanoribbon;these modes strongly depended on the size of the graphene pattern,the angle of the tapered graphene and the infrared excitation wavelength.These interesting plasmon modes were verified by numerical simulations and explained by the reflection,and interference of electromagnetic waves at the graphene–SiO_(2) edge.The constructive interference at the graphene nanogap caused by charge accumulation was demonstrated for the first time.Using the infrared nanoimaging technique,greater plasmon broadening was observed in the zigzag edge than in the armchair edge.Our study suggests that graphene edges should be separated by an effective working distance to avoid the overlapping of localized plasmon modes,which is very important for the design of graphene-based plasmonic circuits and devices. | Qingyang Xu Teng Ma Mohammad Danesh Bannur Nanjunda Shivananju Sheng Gan Jingchao Song Cheng-Wei Qiu Hui-Ming Cheng Wencai Ren Qiaoliang Bao | 2016 | Light(Science & Applications)2016,5,1: | 0 |
