| 2 | Deformation mechanism and microstructures on polycrystalline aluminum induced by high-current pulsed electron beam显示文摘In this paper, we present an experimental investigation of deformation twinning in polycrystalline aluminum exposed to high-current pulsed electron beam (HCPEB) irradiation. The residual tensile stress with about 10 2 MPa was introduced in the irradiated surface layer. The feature characteristic irradiated with various numbers of pulses was investigated. The formation of a large number of twin bands on the surface irradiated with multiple pulses was determined. The experimental observations indicated that the deformation twinning was indeed triggered during HCPEB irradiation. It is suggested that high value of stress and strain rate induced by rapid heating and cooling due to HCPEB irradiation may cause the shifting of whole atomic planes simultaneously. | 7 | A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. | 8 | Genome-wide profiling of histone H3 lysine 27 trimethylation and its modification in response to chilling stress in grapevine leaves显示文摘Histone H3 lysine 27 trimethylation(H3K27me3) is a histone modification associated with transcriptional repression. However, insights into the genome-wide pattern of H3K27me3 in grapevines are limited. Here, anti-H3K27 chromatin immunoprecipitation(ChIP), high-throughput sequencing, and transcriptome analysis were performed using leaves of Vitis amurensis. The leaves were treated at 4°C for 2 h and 24 h and used to investigate changes in H3K27me3 under chilling treatment. The results show that H3K27me3 is well-distributed both in gene regions(-50%) and in the intergenic region(-50%) in the grapevine genome(Vitis vinifera ‘Pinot Noir PN40024'). H3K27me3 was found to be localized in8 368 annotated gene regions in all detected samples(leaves at normal temperature and under chilling treatments) and mainly enriched in gene bodies with the adjacent promoter and downstream areas. The short-term chilling treatments(4°C for 2 h) induced 2 793 gains and 305losses in H3K27me3 modification. Subsequently, 97.3% of the alterations were restored to original levels after 24 h treatment. The ChIP-qPCR for five differential peaks showed similar results to the data for ChIP-seq, indicating that the chilling-induced H3K27me3 modification is reliable.Integrative analysis of transcriptome and ChIP-seq results showed that the expression of H3K27me3 target genes was significantly lower than those of non-target genes, indicating transcriptional repression of H3K27me3 in grapevine leaves. Furthermore, histone methylation alterations were detected in 82 genes and were related to either repression or activation of their expression during chilling stress. | 15 | A de novo missense mutation in MPP2 confers an increased risk of Vogt–Koyanagi–Harada disease as shown by trio-based whole-exome sequencing显示文摘Vogt–Koyanagi–Harada(VKH)disease is a leading cause of blindness in young and middle-aged people.However,the etiology of VKH disease remains unclear.Here,we performed the first trio-based whole-exome sequencing study,which enrolled 25 VKH patients and 50 controls,followed by a study of 2081 VKH patients from a Han Chinese population to uncover detrimental mutations.A total of 15 de novo mutations in VKH patients were identified,with one of the most important being the membrane palmitoylated protein 2(MPP2)p.K315N(MPP2-N315)mutation.The MPP2-N315 mutation was highly deleterious according to bioinformatic predictions.Additionally,this mutation appears rare,being absent from the 1000 Genome Project and Genome Aggregation Database,and it is highly conserved in 10 species,including humans and mice.Subsequent studies showed that pathological phenotypes and retinal vascular leakage were aggravated in MPP2-N315 mutation knock-in or MPP2-N315 adeno-associated virus-treated mice with experimental autoimmune uveitis(EAU).In vitro,we used clustered regularly interspaced short palindromic repeats(CRISPR‒Cas9)gene editing technology to delete intrinsic MPP2 before overexpressing wild-type MPP2 or MPP2-N315.Levels of cytokines,such as IL-1β,IL-17E,and vascular endothelial growth factor A,were increased,and barrier function was destroyed in the MPP2-N315 mutant ARPE19 cells.Mechanistically,the MPP2-N315 mutation had a stronger ability to directly bind to ANXA2 than MPP2-K315,as shown by LC‒MS/MS and Co-IP,and resulted in activation of the ERK3/IL-17E pathway.Overall,our results demonstrated that the MPP2-K315N mutation may increase susceptibility to VKH disease. | Xianyang Liu Jiayu Meng Xingyun Liao Yusen Liu Qian Zhou Zongren Xu Shuming Yin Qingfeng Cao Guannan Su Siyuan He Wanqian Li Xiaotang Wang Guoqing Wang Dali Li Peizeng Yang Shengping Hou | 2023 | Cellular & Molecular Immunology2023,20,11: | 0 |
| 16 | Surface charge property governing co-transport of illite colloids and Eu(Ⅲ) in saturated porous media显示文摘The transport of colloids and radionuclides is sophisticated because of the variety of charge properties between colloidal particles and host subsurface media, which causes great difficulty in establishing a reliable model of radionuclides migration by taking the colloid phase into consideration. In this work,the co-transport of illite colloids(IC) and Eu(Ⅲ) in the quartz sand and iron-coated sand porous media was investigated by column experiments to address the predominant mechanism of charge properties on co-transport. Results showed that Eu(Ⅲ) transport was driven by the illite colloids and electrostatic interaction was critical in governing the co-transport patterns. The promotion of Eu(Ⅲ) transport by IC was attenuated in the iron-coated sand systems;more IC-Eu(Ⅲ) complexes were retained uniformly in the column. The pore throat shrinkage caused by electrostatic attachment between aggregated IC and iron oxides exacerbated the physical straining and size exclusion effect of IC-Eu(Ⅲ) complexes. An aggravated irreversible retention of IC-Eu(Ⅲ) was detected in iron-coated sand column due to the electrostatic attraction of IC-Eu(Ⅲ) to host media. | 20 | Modular zonal fluid sampling and pressure testing technology for production well显示文摘To accurately obtain development dynamic data such as zonal pressure and fluid parameters of each oil layer in the late development stage of a high water-cut old oilfield, a modular zonal sampling and testing technology with the characteristics of modularization, full electronic control and rapidity was proposed and developed. Lab testing and on-site testing was carried out. The modular zonal sampling and testing system is composed of 10 functional modules, namely ground control system, downhole power supply module, drainage pump, electronically controlled anchor, electronically controlled packer, electronically controlled sampler, magnetic positioning sub, terminal sub, adapter cable, and quick connector. Indoor tests have confirmed that the performance parameters of each module meet the design requirements. The downhole function modules of the system can withstand pressures up to 35 MPa and temperatures up to 85 ℃. The rubber cylinder of the electronically controlled packer can withstand a pressure difference of more than 10 MPa. The electronically controlled anchor has an anchoring force of greater than 6.9 t, and can be forcibly detached in the event of an accident. The discharge pump has a displacement of 0.8 m;/d and a head of 500 m. The electronically controlled sampler can meet the requirement of taking 500 mL of sample in each of the 3 chambers. Field tests in Jilin Oilfield show that the system can realize rapid isolation and self-check of isolation of a certain production interval downhole, as well as layer-by layer pressure build-up test. The drainage pump can be used to discharge the mixed liquid between the upper and lower packers and near the wellbore to obtain real fluid samples of the tested formation interval.
