1SEQUENCE VARIABILITY OF HUMAN CYTOMEGALOVIRUS UL144 OPEN READING FRAME IN LOW-PASSAGE CLINICAL ISOLATES显示文摘Objective To explore the relationship between human cytomegalovirus (HCMV) UL144 sequence variability and clinical disease. Methods HCMV UL144 open reading frame (ORF) was amplified by PCR assay in 72 lowpassage isolates [65 con-genitally infective children and 7 healthy children who were HCMV-DNA positive by quantitative PCR (qPCR)]. All positive PCR products were analyzed by heteroduplex mobility assay and single-stranded conformation polymorphism (HMA-SSCP) and 32 of them were sequenced. Results Fifty-five patient isolates and five healthy children isolates were HCMV-UL144 positive by PCR. Sequencing and HMA-SSCP analysis showed that significant strain-specific variability was present in the UL144 ORF. Phylogenetic analysis indicated that the nucleotide sequences could be separated into 3 major genotypes. Comparing between UL144 se-quences and the corresponding symptoms showed that genotype 2 did not exist in megacolon isolates. And genotype 1 and 3 were the major types among microcephaly and jaundice isolates respectively. Conclusions HCMV-UL144 existed in most of low passage isolates and sequences were hypervariable. 2Maghemite(γ-Fe_2O_3) nanoparticles enhance dissimilatory ferrihydrite reduction by Geobacter sulfurreducens: Impacts on iron mineralogical change and bacterial interactions显示文摘Microbially mediated bioreduction of iron oxyhydroxide plays an important role in the biogeochemical cycle of iron.Geobacter sulfurreducens is a representative dissimilatory ironreducing bacterium that assembles electrically conductive pili and cytochromes.The impact of supplementation withγ-Fe_2O_3 nanoparticles(NPs)(0.2 and 0.6 g)on the G.sulfurreducens-mediated reduction of ferrihydrite was investigated.In the overall performance of microbial ferrihydrite reduction mediated byγ-Fe_2O_3 NPs,stronger reduction was observed in the presence of direct contact withγ-Fe_2O_3 NPs than with indirect contact.Compared to the production of Fe(Ⅱ)derived from biotic modification with ferrihydrite alone,increases greater than 1.6-and 1.4-fold in the production of Fe(Ⅱ)were detected in the biotic modifications in which direct contact with 0.2 g and 0.6 gγ-Fe_2O_3 NPs,respectively,occurred.X-ray diffraction analysis indicated that magnetite was a unique representative iron mineral in ferrihydrite when active G.sulfurreducens cells were in direct contact withγ-Fe_2O_3 NPs.Because of the sorption of biogenic Fe(Ⅱ)ontoγ-Fe_2O_3 NPs instead of ferrihydrite,the addition ofγ-Fe_2O_3 NPs could also contribute to increased duration of ferrihydrite reduction by preventing ferrihydrite surface passivation.Additionally,electron microscopy analysis confirmed that the direct addition ofγ-Fe_2O_3 NPs stimulated the electrically conductive pili and cytochromes to stretch,facilitating long-range electron transfer between the cells and ferrihydrite.The obtained findings provide a more comprehensive understanding of the effects of iron oxide NPs on soil biogeochemistry.Zheng Chen Yaxian Zhang Qingliu Luo Liuying Wang Shurui Liu Yajuan Peng Haitao Wang Liang Shen Qingbiao Li Yuanpeng Wang 2019Journal of Environmental Sciences2019,31,4:5

4Evaluation of effect of hybrid bioartificial liver using end-stage liver disease model显示文摘AIM: To study the role of hybrid bioartificial liver (HBL) in clearing proinflammatory cytokines and endotoxin in patients with acute and sub-acute liver failure and the effects of HBL on systemic inflammatory syndrome (SIRS) and multiple organ dysfunction syndrome (MODS).METHODS: Five cases with severe liver failure (3 acute and 2 subacute) were treated with HBL. The clinical signs and symptoms, total bilirubin (TBIL), serum ammonia,endotoxin TNF-~, 1L-6 and prothrombin activity (PTA),cholinesterase (CHE) were recorded before, during and after treatment. The end-stage liver disease (MELD) was used for the study.RESULTS: Two patients were bridged for spontaneous recovery and 1 patient was bridged for OLT successfully.Another 2 patients died on d 8 and d 21. The spontaneous recovery rate was 30.0%. PTA and CHE in all patients were significantly increased (P<0.01), while the serum TBIL,endotoxin,TNF-α, IL-6 were decreased. MELD score (mean 43.6) predicted 100% deaths within 3 mo before treatment with HBL. After treatment with HBL, four out of 5 patients had decreased MELD scores (mean 36.6). The MELD score predicted 66% mortalities.CONCLUSION: The proinflammatory cytokines (TNFα, IL-6 and endotoxin)can be significantly removed by hybrid bioartificial liver and HBL appears to be effective in blocking SIRS and MODS in patients with acute and sub-acute liver failure. MELD is a reliable measure for predicting short-term mortality risk in patients with end-stage liver disease.
