Hepatocellular Carcinoma-Cause,Treatment and Metastasis显示文摘In the recent decades, the incidence of hepatocellular carcinoma (HCC) has been found to be increasing in males in some countries. In China, HCC ranked second of cancer mortality since 1990s. Hepatitis B and C viruses (HBV and HCV) and dietary aflatoxin intake remain the major causative factors of HCC. Surgery plays a major role in the treatment of HCC, particularly for small HCC. Downstaging unresectable huge HCC to smaller HCC and followed by resection will probably be a new approach for further study. Liver transplantation is indicated for small HCC, however, some issues remain to be solved.Different modes of 'regional cancer therapy for HCC' have been tried. Systemic chemotherapy has been disappointing in the past but the future can be promising.Biotherapy, such as cytokines, differentiation inducers,anti-angiogenic agents, gene therapy and tumor vaccine will probably play a role, particularly in the prevention of tumor recurrence. HCC invasiveness is currently the major target of study. Tremendous works have been done at the molecular level, which will provide clues for biomarker of HCC progressionas well as targets for intervention. | Zhao-You Tang Liver Cancer Institute & Zhongshan Hospital of Fudan University Professor of Surgery Chairman.Liver Cancer Institute of Fudan University(previous Liver Cancer Institute of Shanghai Medical University)136 Yixueyuan Road,Zhongshan Hospital,Shanghai 200032,China. | 2001 | World Journal of Gastroenterology2001,7,4: | 213

Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats. The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression, immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats. However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. Colchicine has verylimited therapeutic effect on hepatic fibrosis, furthermore, its toxicity and side effects areobvious. | Qing He Nie Yong Qian Cheng Yu Mei Xie Yong Xing Zhou Yi Zhan Cao The Center of Infectious Disease Diagnosis and Treatment of PLA,Tangdu Hospital,Forth Military Medical University,Xi’an 710038,Shaanxi Province,ChinaDr,Qing He Nie graduated from Qinghai Medical College as a doctor in 1983,got master degree at Beijing 302 Army Hospital in 1993,got doctor degree at the Third Military Medical University in 1998,engaged in postdoctoral research at the Fourth Military Medical University from 1998 to 2000,now an associate professor, specialized in clinical and experimental research of infectious diseases, had more than 90 papers published, coauthor of ten books, first author of one book. | 2001 | World Journal of Gastroenterology2001,7,3: | 73
Mathematical Models for Combined Refraction-Diffraction of Waves on Non-Uniform Current and Depth显示文摘Two mathematical models for combined refraction-diffraction of regular and irregular waves on non-uniform current in water of slowly varying topography are presented in this paper. Model I is derived by wave theory and variational principle separately. It has two kinds of expressions including the dissipation term. Model n is based on the energy conservation equation with energy flux through the wave crest lines in orthogonal curvilinear coordinates and the wave kinematic conservation equation. The analysis and comparison and special cases of these two models are also given. | Hong Guangwen Professor, Research Institute of Coastal and Ocean Engineering, Hohai University, 1 Xikang Road, Nanjing 210024 | 1996 | China Ocean Engineering1996,11,4: | 35

High-Order Models of Nonlinear and Dispersive Wave in Water of Varying Depth with Arbitrary Sloping Bottom显示文摘High-order models with a dissipative term for nonlinear and dispersive wave in water of va-rying depth with an arbitrary sloping bottom are presented in this article.First, the formal derivations toany high order of μ(=h/λ,depth to deep-water wave length ratio)and ε(=α/h,wave amplitude todepth ratio)for velocity potential, particle velocity vector, pressure and the Boussinesq-type equations forsurface elevation η and horizontal velocity vector U at any given level in water are given. Then, the exactexplicit expressions to the fourth order of μ are derived. Finally, the linear solutions of η,U,C(phase ce-lerity)and C_g(group velocity)for a constant water depth are obtained.Compared with the Airy theory, excellent results can be found even for a water depth as large as the wave legnth. The present high-ordermodels are applicable to nonlinear regular and irregular waves in water of any varying depth(from shal-low to deep)and bottom slope(from mild to steep). | Hong Guangwen Professor, Coastal and Ocean Engineering Research Institute, Hohai University, Nanjing 210024, P. | 7 | The New Form of Time-Dependent Mild Slope Equation for Random Waves显示文摘The mild-slope equation derived by Berkhoff (1972), has widely been used in the numerical calculation of refraction and diffraction of regular waves. However, it is well known that the random sea waves has a significant effect in the refraction and diffraction problems. In this paper, a new form of time-dependent mild slope equation for irregular waves was derived with Fade approximation and Kubo’s time series concept. The equation was simplified using WKB method, and simple and practical irregular mild slope equation was obtained. Results of numerical calculations are compared with those of laboratory experiments. | Zhang Yonggang , Li Yucheng Teng Bin Doctor Degree Candidate, The National Key Laboratory of Coastal and Offshore Engineering, Dalian University of Technology, Dalian 116023Professor, The National,Key Laboratory of Coastal and Offshore Engineering, Dalian University of Technology, Dalian 116023Associate Professor, The National Key Laboratory of Coastal and Offshore Engineering, Dalian University of Technology, Dalian 116023 | 1995 | China Ocean Engineering1995,10,4: | 21
Antihepatoma effect of alpha-fetoprotein antisense phosphorothioate oligodeoxyribonucleotides in vitro and in mice显示文摘AIM To evaluate antihepatoma effect ofantisense phosphorothioate oligodeo-xyribonucleotides (S-ODNs) targeted to alpha-fetoprotein (AFP) genes in vitro and in nudemice.METHODS AFP gene expression was examinedby immunocytochemical method or enzyme-linked immunosorbent assay. Effect of S-ODNson SMMC-7721 human hepatoma cell growth invitro was determined using microculturetetrazolium assay. In vivo antitumor activitiesof S-ODNs were monitored by measuring tumorweight differences in treated and control micebearing SMMC-7721 xenografts. Induction of cellapoptosis was evaluated by fluorescence-activated cell sorter (FACS) analysis.RESULTS Antisense S-ODN treatment led toreduced AFP gene expression. Specificantisense S-ODNs, but not control S-ODNs,inhibited the growth of heaptoma cells in vitro. In vivo. only antisense S-ODNs exhibitedobvious antitumor activities. FACS analysisrevealed that the growth inhibition by antisenseS. ODNs was associated with their cell apoptosisinduction.CONCLUSION Antisense S-ODNs targeted toAFP genes inhibit the growth of human hepatomacells and solid hepatoma, which is related totheir cell apoptosis induction. | Xing Wang Wang~1 Jin Hui Yuan~1 Ru Gang Zhang~1 Li Xia Guo~1 Yong Xie~2 Hong Xie~1 ~1Department of Biotherapy,Shanghai Institute of Cell Biology,Chinese Academy of Sciences,Shanghai 200031,China ~2Department of Biology,Hong Kong University of Science and Technology,ChinaDr.Xing Wang Wang earned Ph.D.from Shanghai Institute of Materia Medical,Chinese Academy of Sciences in 1997.Now a professor at Shanghai Institute of Cell Biology,Chinese Academy of Sciences. | 2001 | World Journal of Gastroenterology2001,7,3: | 21
Effects of aminoguanidine on nitric oxide production induced by inflammatory cytokines and endotoxin in cultured rat hepatocytes显示文摘AIM To study the effects of aminoguanidine(AG) and two L-arginine analogues Nω-nitro-L-arginine methyl ester (L-NAME) and Nω-nitro-L-arginine (L-NNA) on nitric oxide (NO) productioninduced by cytokines (TNF-α, IL-11β, and IFN-γ)and bacterial lipopolysaccharide (LPS) mixture(CM) in the cultured rat hepatocytes, andexamine their mechanisms action.METHODS Rat hepatocytes were incubatedwith AG, L-NAME, L-NNA, Actinomycin D (ActD)and dexamethasene in a medium containing CM(LPS plus TNF-α, IL-1β, and IFN-γ) for 24 h. NOproduction in the cultured supernatant wasmeasured with the Griese reaction. IntracellularcGMP level was detected with radioimmunoasey.RESULTS NO production was markedlyblocked by AG and L-NAME in a dose-dependentmanner under inflammatory stimuli conditiontriggered by CM in vitro. The rate of themaximum inhibitory effects of L-NAME (38.9%)was less potent than that obtained with AG(53.7%, P<0.05). There was no significantdifference between the inhibitory effects of AGand two L-arginine analogues on intracellularcGMP accumulation in rat cultured hepatocytes.Non-specific NOS expression inhibitordexamethasone ( DEX ) and iNOS mRNAtranscriptional inhibitor ActD also significantlyinhibited CM-induced NO production. AG(0.1mmol.L-1) and ActD (0.2ng@Lt) wereequipotent in decreasing NO production inducedby inflammatory stimuli in vitro, and botheffects were more potent than that induced bynon-selectivity NOS activity inhibitor L-NAME(0. 1 mmol@ L- 1) under similar stimuli conditions(P | Guo Liang Zhang Ye Hong Wang Hui Ling Teng Zhi Bin Lin Department of Pharmacology,School of Basic Medical Sciences,Beijing University,Beijiog 100083,ChinaDr.Guo Liang Zhang graduated from Xinxiang Medical College in 1982,got Ph.D.at Nagoya City University Medical School,Japan in 1994,finished postdoctoral research at Beijing Medical Univcrsity in 1996,now an associate professor of pharmacology, specialized in hepatic pharmacology, having 15 papers published. | 2001 | World Journal of Gastroenterology2001,7,3: | 20
Orthotopic transplantation model of human gastrointestinal cancer and detection of micrometastases显示文摘AIM To establish a relevant animal model ofhuman gastrointestinal cancer, which can beused for repetitive investigations, so as toimprove our understanding and management ofcarcinogenesis and cancer metastasis.METHODS Intact tissues of human colorectaland pancreatic cancers were transplanted innude mice. The biological characteristics of theoriginal and the corresponding transplantedtumors were investigated by HE staining, PASstaining and immunostaining. The metastases inthe livers and lungs of nude mice wereinvestigated by immunostaining withbiotinylated mab KL-1 and by RT-PCR using CK20specific primers.RESULTS There were totally 9 of 16 surgicalspecimens growing in nude mice subcutaneouslyand/.or orthotopically (4 of 6 colorectal and 5 of10 pancreatic cancer). Tumor cell content of thespecimens and freezing of tissue specimens areimportant factors influencing the growth oftransplanted tumor. In the group of fresh tumortissues with greater than 50% tumor cellcontent, the success rate of the transplantationwas 100% (3 cases of pancreatic cancer and 3cases of colorectal cancer). The orthotopicallytransplanted tumors resemble the original tumormorphologically and biologically, including TAAexpression such as CEA byimmunohistochemistry, and CEA level in theserum of mice. Ki-67 labeling index and theexpression of TAA especially K-ras, 17-lA andRA-96, are associated with the potential of tumorgrowth in nude mice. Micrometastases in thelungs and livers of tumor bearing mice can bedetected by immunostaining with biotinylatedmab KL-1 and CK20-specific RT-PCR.CONCLUSION An orthotopic transplantationmodel for human colon and pancreatic cancer innude mice has been set up. We have alsoestablished sensitive detection methods withCK-immunohistochemistry and CK20-RT-PCR tostudy xenotransplanted human cancer and itsmetastatic cancer cells in the liver and lung ofnude mice. This study may be helpful inunderstanding the mechanism of cancermetastasis and in developing new diagnosticmethods and therapeutic strategies formetastases including micrometastases. | Jun Hui Cui~1 Uwe Krueger~2 Doris Henne-Bruns~2 Bemd Kremer~2 Holger Kalthoff~2 ~1Department of General Surgery,First Affiliated Hospital,College of Medicine,Zhejiang University,Hangzhou 310003,Zhejiang Province,China ~2Department of General Surgery,Christian-Albrechts-University,Kiel,GermanyDr.Jun Hui Cui graduated from Zhejiang Medical University in 1984,earned master degree in 1990,studied in the Surgical Department of Kiel University and worked in the Lab of Molecular Oncology of Kiel University from 1994-1997achieved M.D.from Kiel University.Germany, now associate professor of surgery, specialized in colorectal oncology.Adviser of graduated student for master degree, having 20 publications published in key Chinese or English journals. | 2001 | World Journal of Gastroenterology2001,7,3: | 19
Virulence and potential pathogenicity of coccoid Helicobacter pylori induced by antibiotics显示文摘AIM To explore the virulence and the potential pathogenicity of coccoid Helicobacter pylori (H. pylori) transformed from spiral form by exposure to antibiotic.METHODS Three strains of H. pylori, isolated from gastric biopsy specimens of confirmed peptic ulcer, were converted from spiral into coccoid from by exposure to metronidazole. Both spiral and coccoid form of H. pylori were tested for the urease activity, the adherence to Hep-2 cells and the vacuolating cytotoxicity to Hela cells, and the differences of the protein were analysed by SDS-PAGE and Western blot,The mutation of the genes including ureA, ureB,hpaA; vacA and cagA, related with virulence, was detected by means of PCR and PCR-SSCP.RESULTS In the coccoid H. pylori, the urease activity, the adherence to Hep-2 cells and the vacuolating cytotoxicity to Hela cells alldecreased. In strain F44, the rate and index of adherence reduced from 70.0% ± 5.3% to 33% ±5.1% and from 2.6 ±0.4 to 0.96 ±0.3 (P<0.01),respectively. The invasion of coccoid H. pylori into Hep-2 cell could be seen under electronmicroscope. SDS-PAGE showed that the content of the protein with the molecular weight over Mr74 000 decreased, and the hybriditional signal in band Mr 125 000 weakened, while the band Mr 110000 and Mr63000 strengthened in coccoid H. pylori as shown in Western blot. The results of PCR were all positive, and PCR-SSCP indicated that there may exist the point mutation in gene hpaA or vacA.CONCLUSION The virulence and the proteins with molecular weight over Mr74 000 in coccoid H. pylori decrease, but no deletion exists in amplification fragments from ureA, ureB, hpaA,vacA and cagA genes, suggesting that coccoid H. pylori may have potential pathogenicity. | Fei Fei She1 Dong Hui Su1 Jian Yin Lin2 Lin Ying Zhou3 1Department of Microbiology, Fujian Medical University. | 12 | Reliability-Based Design for Jacket Platform Under Extreme Loads显示文摘In this paper, reliability analysis for the offshore jacket platform with the interaction of structure- pile- soil under extreme environmental loads is carried out. The inherent uncertainties of the environmental load, foundation soil, platform itself, and calculating models are evaluated. The action of extreme loads on the offshore platform is modeled as a function of extreme wave height. The system capacity of the whole platform is determined by nonlinear pushover analysis, and the relevant probability property is obtained by the simulation method. The reliability model for the whole jacket platform is described as the relationship between the load and resistance based on the offshore design codes. The reliability of whole platform is calculated by the analytical method and the importance sampling method on the basis of a case study for a tripod jacket platform. | Jin Weiliang Associate Professor, Dr. -Eng., Zhejiang University, Hangzhou 310027 | 1996 | China Ocean Engineering1996,11,2: | 17

Emergency assessment of seismic landslide susceptibility: a case study of the 2008 Wenchuan earthquake affected area显示文摘The 8.0 Mw Wenchuan earthquake triggered widespread and large scale landslides in mountainous regions. An approach was used to map and assess landslide susceptibility in a given area.A numerical rating system was applied to fi ve factors that contribute to slope instability.Factors such as lithology, topography, streams and faults have an important influence as event-controlling factors for landslide susceptibility assessment.A f inal map is provided to show areas of low, medium, and high landslide susceptibility.Areas identifi ed as having high landslide susceptibility were located in the central, northeastern, and far south regions of the study area. The assessment results will help decision makers to select safe sites for emergency placement of refuges and plan for future reconstruction. The maps may also be used as a basis for landslide risk management in the study area. | Tang Chuan,Zhu Jing and Liang Jingtao State Key Laboratory of Geo-Hazard Prevention,Chengdu University of Technology,Chengdu 610059,China Professor Associate Professor Graduate Student | 2009 | Earthquake Engineering and Engineering Vibration2009,8,2: | 16
Diffraction of plane SV waves by a cavity in poroelastic half-space显示文摘This paper presents an indirect boundary integration equation method for diffraction of plane SV waves by a 2-D cavity in a poroelastic half-space.The Green’s functions of compressive and shear wave sources are derived based on Biot’s theory. The scattered waves are constructed using fi ctitious wave sources close to the boundary of the cavity, and their magnitudes are determined by the boundary conditions. Verifi cation of the accuracy is performed by: (1) checking the satisfaction extent of the boundary conditions, (2) comparing the degenerated solutions of a single-phased case with wellknown solutions, and (3) examining the numerical stability of the solutions. The nature of diffraction of plane SV waves around a cavity in a poroelastic half-space is investigated by numerical examples. | Liang Jianwen and Liu Zhongxian Department of Civil Engineering, Tianjin University, Tianjin 300072, China Professor PhD Candidate | 2009 | Earthquake Engineering and Engineering Vibration2009,8,1: | 15
The Bedload Movement in the Changjiang Estuary显示文摘- Sandwaves in the Changjiang estuary were measured with a shallow sediment profiler and an echosounder from 1978 to 1988. The data, together with grain size and bedform of sediment indicates that the bedload movement by rolling and saltation is of great significance to sediment transport and is the principal factor responsible for sandwave and sandbody development in the estuary. The sandwaves were found well-developed, which is related to the tidal range and the velocity of ebb current. However, the further growth is restricted by strong flood current prevailing in the estuary. Because of the significant bedload, the sandbodies shift obviously and frequently, and sometimes the exchange of position occurs between the sandbodies and tidal channels. As a result, ships are regularly forced to change their navigation course. | Li Jiufa, Shi Weirong, Shen Huanting Xu Haigen and Doeke Eisma Associate Professor, Institute of Estuarine & Coastal Research (1ECR), East China Normal University, Shanghai200062 Professor, IECR, East China Normal University, Shanghai 200062 Professor, Netherlands Institute for Sea Research, P. | 18 | Building damage in Dujiangyan during Wenchuan Earthquake显示文摘A field damage survey of 1,005 buildings damaged by the Wenchuan Earthquake in Dujiangyan City was carried out and the resulting data was analyzed using the statistical method. It is shown that buildings that were seismically designed achieved the desired seismic fortification target; they sustained less damage than the non-seismically designed buildings. Among the seismically designed buildings investigated, RC frame buildings performed the best in terms of seismic resistance. Masonry buildings with a ground story of RC frame structure were the second best, and masonry buildings performed the worst. Considering building height, multistory buildings sustained more severe damage than high-rise buildings and 2- and 3-story buildings. Compared to residential buildings, public buildings, such as schools and hospitals, suffered more severe damage. | Zhang Minzheng and Jin Yingjie Institute of Engineering Mechanics, China Earthquake Administration, Harbin 150080, China Professor Graduate Student | 2008 | Earthquake Engineering and Engineering Vibration2008,7,3: | 13
| 19 | Experimental Research on Sewage Outfall Diffusers显示文摘The influence of diffuser parameters, including the riser spacing, port number in a riser, injection angle, port arrangement, etc., on the surface initial dilution is experimentally investigated. The relative density difference between the effluent and the sea water in the model is the same as that in the prototype, and the effect of the cross current is simulated by an inverse model technique. Based on the result analysis, the arrangement with more ports in a riser and larger riser spacing is suggested to save construction cost. The relationship between the Reynolds number based on the port diameter and velocity, and the surface initial dilution is also explored, and the critical Reynolds number is proposed. | Jiang Chuanfeng , Yan Zhongmin , Zhou Chuntian and Liu Mingming Professor, Res.
