| 1 | 中国年轻乳腺癌发病危险因素——多中心十年回顾性研究显示文摘[目的]研究年轻乳腺癌与非年轻乳腺癌患者的临床病理特征差异,探寻年轻女性罹患乳腺癌的危险因素。[方法]数据来自于一项以医院为基础的10年(1999-2008)多中心回顾性研究。对年轻组(〈35岁)与非年轻组(≥35岁)患者的肿瘤大小、淋巴结转移状况和人口学信息进行统计分析。[结果]研究共纳入4211例患者。其中年轻乳腺癌患者(〈35岁)295例(7.06%)。年轻组与非年轻组患者在临床病理特征方面仅有肿瘤最大径(31.91mm vs.27.44mm,P=0.009)和淋巴结转移发生率(62.31%vs.49.01%,P〈0.001)存在统计学差异。此外,年轻乳腺癌患者的生产次数少(Ptrend〈0.001)、哺乳比例低(P〈0.001)。年轻患者中未婚者(P〈0.001)、使用口服避孕药者(P〈0.001)、饮酒者(P=0.006)均较非年轻组的比例高。年轻乳腺癌患者具有较高的脑力劳动比例(P=0.005)和较高的教育背景(Ptrend〈0.001)。[结论 ]年轻乳腺癌患者在临床病理特征以及人口学信息等方面与非年轻患者存在差异。其中一些在年轻乳腺癌患者中出现频率高的变量提示了年轻患者罹患乳腺癌的危险因素。 | 张柏林 张倩 Priya G.Sivasubramaniam 高纪东 方仪 王翔 张保宁 张斌 唐中华 谢小明 杨红建 何建军 李卉 李佳圆 范金虎 乔友林 | 2015 | 中国肿瘤2015,24,12: | 31 |

| 5 | Active learning for accelerated design of layered materials显示文摘Hetero-structures made from vertically stacked monolayers of transition metal dichalcogenides hold great potential for optoelectronic and thermoelectric devices.Discovery of the optimal layered material for specific applications necessitates the estimation of key material properties,such as electronic band structure and thermal transport coefficients.However,screening of material properties via brute force ab initio calculations of the entire material structure space exceeds the limits of current computing resources.Moreover,the functional dependence of material properties on the structures is often complicated,making simplistic statistical procedures for prediction difficult to employ without large amounts of data collection.Here,we present a Gaussian process regression model,which predicts material properties of an input hetero-structure,as well as an active learning model based on Bayesian optimization,which can efficiently discover the optimal hetero-structure using a minimal number of ab initio calculations.The electronic band gap,conduction/valence band dispersions,and thermoelectric performance are used as representative material properties for prediction and optimization.The Materials Project platform is used for electronic structure computation,while the BoltzTraP code is used to compute thermoelectric properties.Bayesian optimization is shown to significantly reduce the computational cost of discovering the optimal structure when compared with finding an optimal structure by building a regression model to predict material properties.The models can be used for predictions with respect to any material property and our software,including data preparation code based on the Python Materials Genomics(PyMatGen)library as well as python-based machine learning code,is available open source. | Lindsay Bassman Pankaj Rajak Rajiv K.Kalia Aiichiro Nakano Fei Sha Jifeng Sun David J.Singh Muratahan Aykol Patrick Huck Kristin Persson Priya Vashishta | 2018 | npj Computational Materials2018,,1: | 10 |
| 6 | Differential changes in intrinsic innervation and interstitial cells of Cajal in small bowel atresia in newborns显示文摘AIM: To investigate morphological changes of the enteric nervous system (ENS) and the interstitial cells of Cajal (ICCs) in small bowel atresia.METHODS: Resected small bowel specimens from affected patients (n = 7) were divided into three parts (proximal, atretic, distal). Standard histology and enzyme immunohistochemistry anti-S100, anti-protein gene product (PGP) 9.5, anti-neurofilament (NF), antic-kit-receptor (CD117) was carried out on conventional paraffin sections of the proximal and distal part. RESULTS: The neuronal and glial markers (PGP 9.5, NF, S-100) were expressed in hypertrophied ganglia and nerve fibres within the myenteric and submucosal plexuses. Furthermore, the submucous plexus contained typical giant ganglia. The innervation pattern of the proximal bowel resembled intestinal neuronal dysplasia. The density of myenteric ICCs was clearly reduced in the proximal bowel, whereas a moderate number of muscular ICCs were found. The anti-CD117 immunore- action revealed additional numerous mast cells. | 7 | Genomic change in hepatitis B virus associated with development of hepatocellular carcinoma显示文摘AIM: To determine the genomic changes in hepatitis B virus(HBV) and evaluate their role in the development of hepatocellular carcinoma(HCC) in patients chronically infected with genotype C HBV.METHODS: Two hundred and forty chronic hepatitis B(CHB) patients were subjected and followed for a median of 105 mo. HCC was diagnosed in accordance with AASLD guidelines. The whole X, S, basal core promoter(BCP), and precore regions of HBV were sequenced using the direct sequencing method.RESULTS: All of the subjects were infected with genotype C HBV. Out of 240 CHB patients, 25(10%) had C1653 T and 33(14%) had T1753 V mutation in X region; 157(65%) had A1762T/G1764 A mutations in BCP region, 50(21%) had G1896 A mutation in precore region and 67(28%) had pre-S deletions. HCC occurred in 6 patients(3%). The prevalence of T1753 V mutation was significantly higher in patients who developed HCC than in those without HCC. Salinity-induced ionic and osmotic stresses affect stomata closure and gaseous exchange, and reduce transpiration and the rate of carbon assimilation, and hence decrease plant yield. Understanding the response of rice plants toward salinity stress at the genetic level and developing salt-tolerant varieties are the vital mandates for its effective management. This review described the present status of salt-tolerance achieved in rice by various mechanisms including the ion homeostasis(Na^+/H^+, OsNHX antiporters), compatible organic solutes(glycine betaine and proline), antioxidative genes(OsECS, OsVTE1, OsAPX and OsMSRA4.1), salt responsive regulatory elements(transcription factors, cis-acting elements and miRNAs) and genes ecoding protein kinases(MAPKs, SAPKs and STRKs). Further, the future perspective of developing salt-tolerant varieties lies in exploring halotolerant gene homologs from rice varieties, especially the landraces. Genetic diversity among rice landraces can serve as a valuable resource for future studies toward variety improvement through breeding and genome editing. | 13 | Plecanatide and dolcanatide, novel guanylate cyclase-C agonists, ameliorate gastrointestinal inflammation in experimental models of murine colitis显示文摘AIM: To evaluate the effect of orally administeredplecanatide or dolcanatide, analogs of uroguanylin, on amelioration of colitis in murine models.METHODS: The cyclic guanosine monophosphate(cG MP) stimulatory potency of plecanatide and dolcanatide was measured using a human colon carcinoma T84 cellbased assay. For animal studies all test agents were formulated in phosphate buffered saline. Sulfasalazine or 5-amino salicylic acid(5-ASA) served as positive controls. Effect of oral treatment with test agents on amelioration of acute colitis induced either by dextran sulfate sodium(DSS) in drinking water or by rectal instillation of trinitrobenzene sulfonic(TNBS) acid, was examined in BALB/c and/or BDF1 mice. Additionally, the effect of orally administered plecanatide on the spontaneous colitis in T-cell receptor alpha knockout(TCRα-/-) mice was also examined. Amelioration of colitis was assessed by monitoring severity of colitis, disease activity index and by histopathology. Frozen colon tissues were used to measure myeloperoxidase activity.RESULTS: Plecanatide and dolcanatide are structurally related analogs of uroguanylin, which is an endogenous ligand of guanylate cyclase-C(GC-C). As expected from the agonists of GC-C, both plecanatide and dolcanatide exhibited potent cG MP-stimulatory activity in T84 cells. Once-daily treatment by oral gavage with either of these analogs(0.05-0.5 mg/kg) ameliorated colitis in both DSS and TNBS-induced models of acute colitis, as assessed by body weight, reduction in colitis severity(P < 0.05) and disease activity index(P < 0.05). Amelioration of colitis by either of the drug candidates was comparable to that achieved by orally administered sulfasalazine or 5-ASA. Plecanatide also effectively ameliorated colitis in TCRα-/- mice, a model of spontaneous colitis. As dolcanatide exhibited higher resistance to proteolysis in simulated gastric and intestinal juices, it was selected for further studies. Connections also exist between the two conditions, including mutual maternal risk factors for the conditions and an increased prevalence of LGA offspring amongst pregnancies affected by GDM. Thus, it is important to elucidate potential shared underlying mechanisms of both LGA and GDM. One potential mechanistic link relates to macronutrient metabolism. Indeed, derangement of carbohydrate and lipid metabolism is present in GDM, and maternal biomarkers of glucose and lipid control are associated with LGA neonates in such pregnancies. The aim of this paper is therefore to reflect on the existing nutritional guidelines for GDM in light of our understanding of the pathophysiological mechanisms of GDM and LGA offspring. Lifestyle modification is first line treatment for GDM, and while there is some promise that nutritional interventions may favourably impact outcomes, there is a lack of definitive evidence that changing the macronutrient composition of the diet reduces the incidence of either GDM or LGA offspring. | 16 | Fast and sensitive LC–MS/MS method for the simultaneous determination of lisinopril and hydrochlorothiazide in human plasma显示文摘A sensitive and rapid liquid chromatography-tandem mass spectrometry(LC–MS/MS) method has been developed for the simultaneous determination of lisinopril(LIS) and hydrochlorothiazide(HCTZ) in human plasma using their labeled internal standards(ISs). Sample pre-treatment involved solid phase extraction on Waters Oasis HLB cartridges using 100 μL of plasma, followed by liquid chromatography on Hypersil Gold C_(18)(50 mm×3.0 mm, 5 μm) column. The analytes were eluted within 2.0 min using acetonitrile-5.0 m M ammonium formate, p H 4.5(85:15, v/v) as the mobile phase. The analytes and ISs were analyzed in the negative ionization mode and quantified using multiple reaction monitoring. The method showed excellent linearity over the concentration range of 0.50–250.0 ng/m L for both the analytes. The intra-batch and inter-batch precision(% CV) was ≤5.26% and their extraction recoveries were in the range of 96.6%–103.1%. Matrix effect evaluated in terms of IS-normalized matrix factors ranged from 0.97 to 1.03 for both the analytes. | 17 | Oral treatment with plecanatide or dolcanatide attenuates visceral hypersensitivity via activation of guanylate cyclase-C in rat models显示文摘AIM To investigate the effects of plecanatide and dolcanatide on maintenance of paracellular permeability, integrity of tight junctions and on suppression of visceral hypersensitivity. METHODS Transport of fluorescein isothiocyanate(FITC)-dextran was measured to assess permeability across cell monolayers and rat colon tissues. Effects of plecanatide and dolcanatide on the integrity of tight junctions in Caco-2 and T84 monolayers and on the expression and localization of occludin and zonula occludens-1(ZO-1) were examined by immunofluorescence microscopy. Anti-nociceptive activity of these agonists was evaluated in trinitrobenzene sulfonic acid(TNBS)-induced inflammatory as well as in non-inflammatory partial restraint stress(PRS) rat models. Statistical significance between the treatment groups in the permeability studies were evaluated using unpaired t-tests.RESULTS Treatment of T84 and Caco-2 monolayers with lipopolysaccharide(LPS) rapidly increased permeability, which was effectively suppressed when monolayers were also treated with plecanatide or dolcanatide. Similarly, when T84 and Caco-2 monolayers were treated with LPS, cell surface localization of tight junction proteins occludin and ZO-1 was severely disrupted. When cell monolayers were treated with LPS in the presence of plecanatide or dolcanatide, occludin and ZO-1 were localized at the cell surface of adjoining cells, similar to that observed for vehicle treated cells. Treatment of cell monolayers with plecanatide or dolcanatide without LPS did not alter permeability, integrity of tight junctions and cell surface localization of either of the tight junction proteins. In rat visceral hypersensitivity models, both agonists suppressed the TNBS-induced increase in abdominal contractions in response to colorectal distension without affecting the colonic wall elasticity, and both agonists also reduced colonic hypersensitivity in the PRS model. CONCLUSION Our results suggest that activation of GC-C signaling might be involved in maintenance of barrier function, possibly through regulating normal localization of tight junction proteins. Consistent with these findings, plecanatide and dolcanatide showed potent antinociceptive activity in rat visceral hypersensitivity models. | 18 | Contemporary approach to diagnosis and classification of renal cell carcinoma with mixed histologic features显示文摘Renal cell carcinoma (RCC) is an important contributor to cancer-specific mortality worldwide. Targeted agents that inhibit key subtype-specific signaling pathways have improved survival times and have recently become part of the standard of care for this disease. Accurately diagnosing and classifying RCC on the basis of tumor histology is thus critical. RCC has been traditionally divided into clear-cell and non-clearcell categories, with papillary RCC forming the most common subtype of non-clear-cell RCC. Renal neoplasms with overlapping histologies, such as tumors with mixed clear-cell and papillary features and hybrid renal oncocytic tumors, are increasingly seen in contemporary practice and present a diagnostic challenge with important therapeutic implications. | 20 | Multiobjective genetic training and uncertainty quantification of reactive force fields显示文摘The ReaxFF reactive force-field approach has significantly extended the applicability of reactive molecular dynamics simulations to a wide range of material properties and processes.ReaxFF parameters are commonly trained to fit a predefined set of quantummechanical data,but it remains uncertain how accurately the quantities of interest are described when applied to complex chemical reactions.Here,we present a dynamic approach based on multiobjective genetic algorithm for the training of ReaxFF parameters and uncertainty quantification of simulated quantities of interest.ReaxFF parameters are trained by directly fitting reactive molecular dynamics trajectories against quantum molecular dynamics trajectories on the fly,where the Pareto optimal front for the multiple quantities of interest provides an ensemble of ReaxFF models for uncertainty quantification.Our in situ multiobjective genetic algorithm workflow achieves scalability by eliminating the file I/O bottleneck using interprocess communications.The in situ multiobjective genetic algorithm workflow has been applied to high-temperature sulfidation of MoO_(3) by H_(2)S precursor,which is an essential reaction step for chemical vapor deposition synthesis of MoS_(2) layers.Our work suggests a new reactive molecular dynamics simulation approach for far-from-equilibrium chemical processes,which quantitatively reproduces quantum molecular dynamics simulations while providing error bars. | Ankit Mishra Sungwook Hong Pankaj Rajak Chunyang Sheng Ken-ichi Nomura Rajiv K.Kalia Aiichiro Nakano Priya Vashishta | 2018 | npj Computational Materials2018,,1: | 3 |
