1Gut microbiota drives macrophage-dependent self-renewal of intestinal stem cells via niche enteric serotonergic neurons显示文摘Lgr5+intestinal stem cells(ISCs)reside within specialized niches at the crypt base and harbor self-renewal and differentiation capacities.ISCs in the crypt base are sustained by their surrounding niche for precise modulation of self-renewal and differentiation.However,how intestinal cells in the crypt niche and microbiota in enteric cavity coordinately regulate ISC stemness remains unclear.Here,we show that ISCs are regulated by microbiota and niche enteric serotonergic neurons.The gut microbiota metabolite valeric acid promotes Tph2 expression in enteric serotonergic neurons via blocking the recruitment of the NuRD complex onto Tph2 promoter.5-hydroxytryptamine(5-HT)in turn activates PGE2 production in a PGE2+macrophage subset through its receptors HTR2A/3 A;and PGE2 via binding its receptors EP1/EP4,promotes Wnt/β-catenin signaling in ISCs to promote their self-renewal.Our findings illustrate a complex crosstalk among microbiota,intestinal nerve cells,intestinal immune cells and ISCs,revealing a new layer of ISC regulation by niche cells and microbiota.Pingping Zhu Tiankun Lu Jiayi Wu Dongdong Fan Benyu Liu Xiaoxiao Zhu Hui Guo Ying Du Feng Liu Yong Tian Zusen Fan 2022Cell Research2022,32,6:8
2Experimental investigations on the fluid flow mechanism in porous media of enhanced oil recovery by alkli/surfactant/polymer flooding显示文摘The fluid flow mechanism in porous media of enhanced oil recovery by Alkli/ Surfactant/ Polymer (ASP) flooding is investigated by measuring production performance, pressure distribution and saturation distribution through installing differential pressure transducers and saturation measuring probes in a physical model of vertical heterogeneous reservoir. The fluid flow variation in porous media is the main reason of enhanced oil recovery of ASP flooding. The pressure field and saturation field are nonlinearly coupled together and the interaction between them results in the fluid flow variation in the reservoir. In a vertical heterogeneous reservoir, the ASP agents initially flow in the high permeability layer, and fluid changes the flow direction toward the low and the middle permeability layers because the resistance in the high permeability layer is increased under the physical and chemical action of adsorption, retention and emulsion. 3Circular RNA circZbtb20 maintains ILC3 homeostasis and function via Alkbh5-dependent m6A demethylation of Nr4a1 mRNA显示文摘Group 3 innate lymphoid cells(ILC3s)play critical roles in innate immunity and gut homeostasis.However,how ILC3 homeostasis is regulated remains elusive.Here,we identified a novel circular RNA,circZbtb20,that is highly expressed in ILC3s and required for their maintenance and function.CircZbtb20 deletion causes reduced ILC3 numbers,increasing susceptibility to C.rodentium infection.Mechanistically,circZbtb20 enhances the interaction of Alkbh5 with Nr4a1 mRNA,leading to ablation of the m6A modification of Nr4a1 mRNA to promote its stability.Nr4a1 initiates Notch2 signaling activation,which contributes to the maintenance of ILC3 homeostasis.Deletion of Alkbh5 or Nr4a1 also impairs ILC3 homeostasis and increases susceptibilities to bacterial infection.Thus,our findings reveal an important role of circular RNA in the regulation of innate lymphoid cell homeostasis.Benyu Liu Nian Liu Xiaoxiao Zhu Liuliu Yang Buqing Ye Huimu Li Pingping Zhu Tiankun Lu Yong Tian Zusen Fan 2021Cellular & Molecular Immunology2021,18,6:2

5Synthesis,structure and nonlinear optical properties of two novel two-photon absorption chromophores显示文摘Two triphenylamine-based derivatives that can be used as two-photon absorption chromophore,tris{4-[4-(3-trifluoromethyl-3-oxopanoyl)]phenyl}amine (1) and tris{4-[4-(3-phenyl-3-oxopanoyl)] phenyl} amine (2) were successfully synthesized and fully characterized by elemental analysis,IR,1H NMR and MS. The single crystal X-ray diffraction analysis showed that the molecules possess D-(π-A)3 structures. One-and two-photon absorption and fluorescence in various solvents were experimentally investigated. 6Insights into the mechanism of Sub3 inhibiting Fusarium moniliforme infection in maize显示文摘Fusarium moniliforme(F.moniliforme) and its secondary metabolite fumonisin pose a severe threat to food safety,and searching for effective antimicrobial agents is a focus of current research.In this study,the secondary structure of Sub3 was analyzed by circular dichroism,meanwhile,the inhibition rate of Sub3 against spores,mycelia of F.moniliforme and infected maize was studied.To explore the possible inhibition mechanisms,morphological and structural changes of spores treated with Sub3 at0,1/2 MIC(minimum inhibitory concentration) and MIC were observed by scanning electron microscopy and transmission electron microscopy;the cell wall integrity,membrane integrity,reactive oxygen species,mitochondrial membrane potential,ATP synthase activity,redox reactions,and the nuclear damage of F.moniliforme were also investigated.The results showed that Sub3 was mostly in the state of random in deionized water,while mainly showed the β-sheet structure in the hydrophobic environment of 50% Trifluoroethanol(TFE) solution,indicating that Sub3 might generate partial structure deformation when acting on the cell membrane;and its MIC on F.moniliforme spores was 0.2 g/L.Under the 1/2 MIC and MIC,the inhibition rates of Sub3 against F.moniliforme infected maize were 34.3% and75.6%,respectively.The results of inhibition mechanisms revealed that the defective pathogenicity of F.moniliforme caused by Sub3 was attributed to damages on both the cell wall and the cell membrane,which might upset balance of intracellular redox system and mitochondrial energy metabolism and trigger nucleus damage,ultimately leading to cell death.Meanwhile,Sub3 could diminished ATP synthase enzyme activity in a dose-dependent manner.The results provided direct evidence for inhibition of F.moniliforme infection of maize by Sub3,and useful knowledge applicable for food preservation.Haojie Yang Wei Zhang Pingping Tian Bangbang Li Shan Wei Shuaibing Zhang Na Li Yangyong Lyu Yuansen Hu 2022Grain & Oil Science and Technology2022,5,1:1

8Anti-fungal properties of Puroindoline A against Aspergillus glaucus显示文摘Aspergillus glaucus can grow in low moisture grain,and is one of the main fungi responsible for agricultural product losses.Puroindoline A(PINA)is a tryptophan-rich alkaline adiponectin that can effectively inhibit numerous plant bacteria and fungi.However,the mechanism of PINA against A.glaucus remains unclear.Herein,we found that recombinant PINA(rPINA)could inhibit A.glaucus mycelia growth on salt Czapek dox agar(SCDA)medium and spore germination on Czapek dox(CD)medium.Scanning electron microscopy revealed that incomplete morphological characteristics of both A.glaucus spores and mycelia occurred following rPINA treatment.Laser scanning confocal microscopy(LSCM)showed that rPINA could enter the interior of spores.Flow cytometry and propidium iodide(PI)staining illustrated membranes of spores were severely damaged,especially after treatment with 0.9 mg/mL rPINA for 12 h,and spores with intact membranes were reduced by 30.7%.Additionally,rPINA reduced the mitochondrial membrane potential(MMP)by 5,5′,6,6′-tetrachlorr-1,1′,3,3′-tetraethyl-imidacarbocyanine iodide(JC-1)staining,and caused DNA damage by 4′,6-diamidino-2-phenylindole(DAPI)staining.These results indicated that rPINA may damage cell membranes and DNA structure and reduceMMP,thereby inhibiting the growth of A.glaucus.The antifungal mechanism has been demonstrated in this study,and results show that rPINA has application potential in preventing postharvest loss in the agricultural industry.Wenjing Yuan Pingping Tian Ang Lyu Yangyong Lyu Wei Zhang Shan Wei Yuansen Hu 2020Grain & Oil Science and Technology2020,3,1:0
9Identification of prognostic indicators,diagnostic markers, and possible therapeutic targets among LIM homeobox transcription factors in breast cancer显示文摘Background:Breast cancer(BRCA)is the most common malignant tumor among women worldwide.Despite advances in treatment,many patients still die from a lack of effective diagnostic and prognostic markers and powerful therapeutic targets.LIM homeobox genes(LHXs)play vital roles in regulating the development of various organisms.However,there are limited reports regarding their roles in the diagnosis,prognosis,and treatment of BRCA.Methods:UALCAN,Kaplan–Meier plotter,cBioPortal,GeneMANIA,STRING,DAVID 6.8,TRRUST v2,LinkedOmics,and TIMER were utilized to analyze differential expression,prognostic value,genetic alteration,neighbor gene network,transcription factor targets,kinase targets,and immune cell infiltration of LHXs in BRCA patients.Results:LHX gene expression patterns are clear in BRCA and its different subtypes.Further analyses indicated that this altered expression is possibly affected by genetic and/or epigenetic changes.The prognostic and diagnostic values of certain LHXs are unique to different BRCA subtypes.LHXs are mainly involved in the regulation of differentiation and development,and their neighbor genes are primarily involved in cancer‐related pathways.Moreover,most LHXs are closely correlated with immune cell infiltration.Furthermore,LHXs may exert their functions by regulating a series of transcription factor and kinase targets.Conclusions:LHXs are unique diagnostic and prognostic markers and participate in cancer through different signaling pathways and/or regulatory mechanisms in BRCA.This study provides potential applications of LHXs for the diagnosis,prognosis,and treatment of BRCA and its different subtypes.Pingping Gao Na Sun Tingting Zhao Yuanyuan Sun Jing Gu Dandan Ma Hao Tian Zaihui Peng Yi Zhang Fei Han Xiaowei Qi 2022Cancer Innovation2022,1,3:0
10Developmental competence and ultrastructural changes of heat-stressed mouse early blastocysts produced in vitro显示文摘Mouse early blastocysts were exposed to temperatures of 39℃ and 41℃ for 2 h,respectively,to determine their developmental competence and ultrastructural changes. The results showed that heat stress at 41℃ for 2 h,significantly reduced the percentages of expanded and hatched blastocysts,but not at 39℃ for 2 h. The average cell numbers in expanded blastocysts,which developed from early blastocysts heat-stressed at temperatures of 39℃ and 41℃,were significantly reduced. The average cell numbers in hatched blastocysts subjected to heat stress were no different from those in the control group cultured at 37℃. The mitochondria of the early blastocysts heat-stressed at 39℃ for 2 h,were slightly swollen,but they had recovered after culturing at 37℃ for 2 h. However,the mitochondria in the blastocysts heat stressed at 41℃ for 2 h were severely swollen,and their number increased. The ribosomes shed from the rough endoplasmic reticulum,and the number of secondary lysosomes in the plasma increased. The integrity of desmosomes was disrupted. The space between the nuclear envelope and the perivitelline membrane enlarged. The fibre fraction and the particulate fraction of nucleoli were separated. The heterochromatin in nucleoli was also increased in its quantity. There were some lamellar-shape structures and heterogeneous dense materials exhibiting in the cytoplasm. The ultrastructural changes induced by heat shock at 41℃ for 2 h were not reversible. 12Role of Natural Product Eriocalyxin B in Promoting Apoptosis of Colon Cancer Cells through ERK1/2 Pathway显示文摘[Objectives] To investigate the role of Eriocalyxin B (EriB) in promoting colon cancer cell apoptosis through ERK1/2 pathway in vitro, and to provide a natural candidate drug for colon cancer treatment. [Methods] Colon cancer cells treated with different concentrations of EriB were detected by CCK-8 assay;cell scratch assay and crystal violet staining were used to detect the invasion and migration of colon cancer cell;cell apoptosis was detected by Annexin V/PI double staining, and cell cycle was detected by PI staining;Western Blotting was used to detect epithelial-mesenchymal transition and apoptosis-related proteins in colon cancer cells treated with EriB. [Results] After EriB treatment, the proliferation, migration and apoptosis of colon cancer cells were significantly inhibited, and the ratio of P-ERK1/2 to ERK was significantly decreased.
