1Tuning Superhydrophobic Materials with Negative Surface Energy Domains显示文摘Hydrophobic/superhydrophobic materials with intrinsic water repellence are highly desirable in engineering fields including antiicing in aerocrafts,antidrag and anticorrosion in ships,and antifog and self-cleaning in optical lenses,screen,mirrors,and windows.However,superhydrophobic material should have small surface energy(SE)and a micro/nanosurface structure which can reduce solid-liquid contact significantly.The low SE is generally found in organic materials with inferior mechanical properties that is undesirable in engineering.Intriguingly,previous theoretical calculations have predicted a negative SE forθ-alumina(θ-Al_(2)O_(3)),which inspires us to use it as a superhydrophobic material.Here,we report the experimental evidence of the small/negative SE ofθ-Al_(2)O_(3) and aθ-Al_(2)O_(3)-based superhydrophobic coating prepared by one-step scalable plasma arcing oxidation.The superhydrophobic coating has complete ceramic and desired micro/nanostructure and therefore exhibits excellent aging resistance,wear resistance,corrosion resistance,high-temperature tolerance,and burning resistance.Owing to the rarity of the small/negative SE in inorganic materials,the concept to reduce SE byθ-Al_(2)O_(3) may foster a blowout to develop robust superhydrophobicity by complete inorganic materials.Zhongzhen Wu Liangliang Liu Shunning Li Shunping Ji Pinghu Chen Suihan Cui Zhengyong Ma Yuchang Weng Qian Huang Zhongcan Wu Hao Wu Yuan Lin Ricky KYFu Hai Lin Xiubo Tian Paul KChu Feng Pan 2019Research2019,,1:6

3In Vitro Anti-hepatitis B Virus Activity of 2',3'-Dideoxyguanosine显示文摘2',3'-dideoxyguanosine(DoG) has been demonstrated to inhibit duck hepatitis B virus(DHBV) replication in vivo in a duck model of HBV infection. In the current study, the in vitro antiviral effects of DoG on human and animal hepadnaviruses were investigated. Our results showed that DoG effectively inhibited HBV, DHBV, and woodchuck hepatitis virus(WHV)replication in hepatocyte-derived cells in a dose-dependent manner, with 50% effective concentrations(EC50) of 0.3 ± 0.05, 6.82 ± 0.25, and 23.0 ± 1.5 lmol/L, respectively. Similar to other hepadnaviral DNA polymerase inhibitors,DoG did not alter the levels of intracellular viral RNA but induced the accumulation of a less-than-full-length viral RNA species, which was recently demonstrated to be generated by RNase H cleavage of pgRNA. Furthermore, using a transient transfection assay, DoG showed similar antiviral activity against HBV wild-type, 3TC-resistant rtA181 V, and adefovirresistant rtN236T mutants. 4Emergence of a novel reassortant avian influenza virus(H10N3) in Eastern China with high pathogenicity and respiratory droplet transmissibility to mammals显示文摘Decades have passed since the first discovery of H10-subtype avian influenza virus(AIV) in chickens in 1949,and it has been detected in many species including mammals such as minks,pigs,seals and humans.Cases of human infections with H10N8viruses identified in China in 2013 have raised widespread attention.Two novel reassortant H10N3 viruses were isolated from chickens in December 2019 in eastern China during routine surveillance for AIVs.The internal genes of these viruses were derived from genotype S(G57) H9N2 and were consistent with H5N6,H7N9 and H10N8,which cause fatal infections in humans.Their viral pathogenicity and transmissibility were further studied in different animal models.The two H10N3 isolates had low pathogenicity in chickens and were transmitted between chickens via direct contact.These viruses were highly pathogenic in mice and could be transmitted between guinea pigs via direct contact and respiratory droplets.More importantly,these viruses can bind to both human-type SAα-2,6-Gal receptors and avian-type SAα-2,3-Gal receptors.Asymptomatic shedding in chickens and good adaptability to mammals of these H10N3 isolates would make it easier to transmit to humans and pose a threat to public health.Kaituo Liu Pingyun Ding Yuru Pei Ruyi Gao Wenwen Han Huafen Zheng Zhuxing Ji Miao Cai Jinyuan Gu Xiuli Li Min Gu Jiao Hu Xiaowen Liu Shunlin Hu Pinghu Zhang Xiaobo Wang Xiaoquan Wang Xiufan Liu 2022Science China(Life Sciences)2022,65,5:1
5The Inhibitory Effects of Mouse ICOS-Ig Gene-Modified Mouse Dendritic Cells on T Cells显示文摘The main approach to reduce graft rejection has been focused on the development of immunosuppressive agents at present. Although these strategies have reportedly reduced graft rejection, there has been a reciprocal increase in more severe immunosuppression and lethal infections, as well as severe side effects. Blockade of costimulatory T cell response has been proved as one of useful strategies to reduce graft rejection. Furthermore,it has been shown that infusion of dendritic cells (DCs) with a potent negative regulatory ability for T cells could prolong allograft survival. In this study mouse DCs (mDCs) were transfected with the recombinant plasmid pcDNA3.0 containing mouse inducible costimulator-Ig (mICOS-Ig) cDNA by electroporation. The transient expression of mICOS-Ig in mDC could be detected by ELISA and SDS-PAGE. Mouse ICOS-Ig fusion protein expressed in mDC and mICOS-Ig gene-modified mDC could inhibit lymphocyte proliferation in mixed lymphocyte culture (MLC) in vitro. Furthermore, mICOS-Ig gene-modified mDC could inhibit lymphocyte proliferation in recipient mice. 6Enhanced pathogenicity and transmissibility of H9N2 avian influenza virus in mammals by hemagglutinin mutations combined with PB2-627K显示文摘H9N2 avian influenza viruses(AIVs)circulate globally in poultry and have become the dominant AIV subtype in China in recent years.Previously,we demonstrated that the H9N2 virus(A/chicken/Eastern China/SDKD1/2015)naturally harbors a mammalian-adaptive molecular factor(627K)in the PB2 protein and is weakly pathogenic in mice.Here,we focused on new markers for virulence in mammals.A mouse-adapted H9N2 virus was serially passaged in mice by infecting their lungs.As expected,infected mice showed clinical symptoms and died at passage six.A comparison between the wild-type and mouse-adapted virus sequences identified amino acid substitutions in the hemagglutinin(HA)protein.H9N2 viruses with the T187P t M227L double mutation exhibited an increased affinity to human-type(SAα2,6Gal)receptors and significantly enhanced viral attachment to mouse lung tissues,which contributed to enhancing viral replication and virulence in mice.Additionally,HA with the T187P t M227L mutation enabled H9N2 viral transmission in guinea pigs via direct contact.AIV pathogenicity in mice is a polygenic trait.Our results demonstrated that these HA mutations might be combined with PB2-627K to significantly increase H9N2 virulence in mice,and this enhanced virulence was achieved in other H9N2 AIVs by generating the same combination of mutations.In summary,our study identified novel key elements in the HA protein that are required for H9N2 pathogenicity in mice and provided valuable insights into pandemic preparedness against emerging H_(9)N_(2)strains.Kaituo Liu Yaqian Guo Huafen Zheng Zhuxing Ji Miao Cai Ruyi Gao Pinghu Zhang Xiaowen Liu Xiulong Xu Xiaoquan Wang Xiufan Liu 2023Virologica Sinica2023,38,1:0
7Phylogenetic,Demographic and Dating Analyses of Bufo gargarizans Populations from the Zhoushan Archipelago and China's Mainland显示文摘Relatively little is known about the relationship between Bufo gargarizans populations from Zhoushan Archipelago and nearby continental regions on the Pacific coast of eastern China.In this paper,155 new specimens of B.gargarizans from Zhoushan Archipelago and adjacent continents and 71 published specimens of B.gargarizans from China's Mainland were studied.Phylogeographical and dating analyses of B.gargarizans were performed using mitochondrial DNA sequencing with a length of 1436 bp.A mt DNA tree that indicated seven major clades was obtained.The earliest split in the mt DNA tree corresponding to the divergence of populations from the western highland region occurred approximately 4.0 million years ago(mya).A subsequent clade occurred about 3.4 mya,with cladogenesis continuing toward the end of the Pleistocene.The continental clades were distributed in the western,central and northeastern regions of China.Zhoushan Archipelago clades consisted of two largely geographically overlapping subclades with the mt DNA divergence time of 0.73 mya.These results indicated there was extensive dispersal after vicariance.The B.gargarizans populations on Zhoushan Archipelago most probably originated from populations in nearby eastern continental regions of China.It was concluded that geological uplifting during the Pliocene and several sea-level changes in Pleistocene might have influenced the divergence and population demographical history of this species.Haojie TONG Yubin WO Pinghu LIAO Yuanting JIN 2017Asian Herpetological Research2017,18,3:0
8Phylogenetic Relationships among Chinese Rice Frogs within the Fejervarya limnocharis Species Complex (Amphibia: Dicroglossidae)显示文摘We present a molecular assessment of the widely-distributed ricefrog(Fejervarya limnocharis)which provides many newsamples that add to knowledge of their phylogeography within China and considersgenetic support forfive Chinese species withinthis complex.TwomtDNAfragments from 270 individuals and eight nuclearDNAloci(105 individuals)were sequenced from specimens sampled from across China.Data from nine specimens from China,Indonesia and Japan were also retrieved from previous studies.MtDNAwas informativeabout population divergence within China and indicated one major clade(with four subclades)from South China and the Zhoushan Archipelago,Zhejiang,China,and a second major clade(with eight subclades)from other parts of China.Recent demographic expansions(less than 50ka ago)were detected within three of these 12 subclades,potentially associa ted with lowered sea-levels after marine transgressions.Nota bly,most frogs from the previously unstudied Zhoushan Archipelago(eastern China)were found to be closely related to Japanese populations.BPPand STACEY species delimitation analyses of the multilocus data favoured five candidate species within the complex.Previous work had described Fejervaryamultistriata and F.kawamurai from the Chinese mainland although here we detected considerable genetic divergence within the latter and found that this may be indicative of two species.One of these corresponds to the Zhoushan Archipelago,Zhejiang,China and Japan,and the other from most parts of Chinese mainland.This study provides a large multilocus dataset that contributes to the systematics of this species complex.Kun YANG Yubin WO Gang SHAO Pinghu LIAO Haojie TONG Richard P.BROWN Yuanting JIN 2022Asian Herpetological Research2022,13,4:0

11FADDdel-GFP-Modified Mouse Insulinoma Cells Counteract the Cytotoxicity of Reactive T Cells显示文摘IDDM results from pancreatic beta cell destruction by islet-reactive T cells, a process that involves beta cell apoptosis. FasL-Fas pathway plays a major role in pancreatic beta cell death. Fas-associated death domain protein (FADD), the component of the tumor necrosis factor receptor type 1 (TNF-R1) and Fas signaling complexes, is involved in TNF-R1- and Fas-induced apoptosis. Inhibiting the function of FADD will lead to blocking downstream apoptosis signal, which protects pancreatic beta cells from destruction by FasL-Fas pathway. In this study we constructed eukaryotic expressing vector of fusional protein FADDdel-GFP named pFADDdeI-GFP. After pFADDdel-GFP was transfected into NIT, the expression of FADDdel-GFP in NIT was detected by fluorescence microscopy and the resistance of NIT transfected with pFADDdel-GFP to cytotoxicity mediated by special T cells was detected by FACS and MTT. The results showed that NIT modified by pFADDdel-GFP obviously resisted cytotoxicity mediated by special T cells. Therefore, it may be useful in the prevention or treatment of IDDM by intervening FasL-Fas pathway.
