| 1 | CYP2E1 mediated isoniazid-induced hepatotoxicity in rats显示文摘AIM: To investigate the role of CYP2E1 in isoniazid (INH)-induced hepatotoxicity and the influence of rifampicin(RFP) on INH-induced liver injury. METHODS: Rats were treated with INH alone (100 mg/kg, ip) or co-administeredwith RFP (100 mg/kg, ig) for 10 d and 21 d. Hepatotoxicity was assayed by plasma enzymes (sALT, sAST) andhistopathological examinations. Hepatic CYP2E1 activity was measured by aniline hydroxylase (ANH), and CYP2E1mRNA expression was determined by RT-PCR. Plasma hydrazine concentration was determined by RP-HPLC.RESULTS: For a 10 d INH-treatment, hepatic CYP2E1 level was increased to 3.7-fold over the control; liverimpairment appeared after 21 d treatment, while CYP2E1 and plasma hydrazine were, respectively, increased to 4.6-fold and 1.7-fold. However, in INH-RFP group for 10 d, CYP2E1 and plasma hydrazine were, respectively,decreased by 13 % and 18 % over INH group; similarly, hepatic injury is equal to INH group appeared after 21 d,and CYP2E1 was further decreased by 26 %. Correlation analysis showed that sALT had a positive correlation withplasma hydrazine and with CYP2E1 activity; CYP2E1 activity was also markedly correlated with plasma hydrazine.And compared with control, there is no difference in changes of CYP2E1 mRNA expression in INH and INH-RFPtreatment for 21 d. CONCLUSION: The metabolite of INH, hydrazine, plays an important role in INH-inducedhepatotoxicity in rats. The induction of CYP2E1 by hydrazine is involved in the hepatotoxicity of INH. | 2 | Establishment of intestinal organoid cultures modeling injury-associated epithelial regeneration显示文摘The capacity of 3D organoids to mimic physiological tissue organization and functionality has provided an invaluable tool to model development and disease in vitro.However,conventional organoid cultures primarily represent the homeostasis of self-organizing stem cells and their derivatives.Here,we established a novel intestinal organoid culture system composed of 8 components,mainly including VPA,EPZ6438,LDN193189,and R-Spondin 1 conditioned medium,which mimics the gut epithelium regeneration that produces hyperplastic crypts following injury;therefore,these organoids were designated hyperplastic intestinal organoids(Hyper-organoids).Single-cell RNA sequencing identified different regenerative stem cell populations in our Hyper-organoids that shared molecular features with in vivo injury-responsive Lgr5+stem cells or Clu+revival stem cells.Further analysis revealed that VPA and EPZ6438 were indispensable for epigenome reprogramming and regeneration in Hyper-organoids,which functioned through epigenetically regulating YAP signaling.Furthermore,VPA and EPZ6438 synergistically promoted regenerative response in gut upon damage in vivo.In summary,our results demonstrated a new in vitro organoid model to study epithelial regeneration,highlighting the importance of epigenetic reprogramming that pioneers tissue repair. | Molong Qu Liang Xiong Yulin Lyu Xiannian Zhang Jie Shen Jingyang Guan Peiyuan Chai Zhongqing Lin Boyao Nie Cheng Li Jun Xu Hongkui Deng | 2021 | Cell Research2021,31,3: | 9 |
| 3 | Genesis and Uranium Sources of Leucogranite-hosted Uranium Deposits in the Gaudeanmus Area, Central Damara Belt, Namibia: Study of Element and Nd Isotope Geochemistry显示文摘This paper presents a systematic study of major and trace elements and Sm-Nd isotopes in leucogranites closely related to uranium mineralization in the Gaudeanmus area, Namibia. The results illustrate that the uraniferous leucogranites possess high Si_O_2(68.8 wt%–76.0 wt%, average 73.1 wt%) and K(4.05 wt%–7.78 wt%, average 5.94 wt%) contents, and are sub-alkaline and metaluminous to weakly peraluminous, as reflected by A/CNK values of 0.96–1.07 with an average of 1.01. The leucogranites are rich in light rare earth elements(LREE/HREE = 2.53–7.71;(La/Yb)N = 2.14–10.40), have moderate Eu depletion and high Rb/Sr ratios(2.03–5.50 with an average of 4.36); meanwhile, they are enriched in Rb, K, Th, U and Pb, and depleted in Ba, Nb, Ta, and Sr. The ε_(Nd)(t) values of uraninites range from -14.8 to -16.5, and the two-stage Nd model ages are 2.43–2.56 Ga. Detailed elemental and Sm-Nd isotopic geochemical characteristics suggest that the leucogranites were formed in a postorogenic extensional environment. | 4 | Experiment research on grain drying process in the heat pump assisted fluidized beds显示文摘A heat pump assisted fluidized bed grain drying experimental system was developed. Based on this system, a serial of ex- periments was performed under four kinds of air cycle conditions. According to the experimental analysis, an appropriate drying medium-air cycle for the heat pump assisted fluidized bed drying equipment was decided, which is different from the commonly used heat pump assisted drying system. The experimental results concerning the drying operation performance of the new system show that the averaged coefficient of performance (COP) can reach more than 2.5. | 5 | New technology to examine and improve the purity of hybrid rice with herbicide resistant gene显示文摘The herbicide resistant bar gene has been widely used as selectable marker genes in the study on plant genetic transformation. Owing to the integration of the gene into rice genome, transgenic rice was resistant to the herbicide Basta. Therefore, selection of transgenic plant and genetic analysis became easier. In the studies, bar gene was introduced as a genetic marker gene into rice restore line variety of two-line or three-line. Combined with conventional breeding method, the good herbicide resistant individual plant was derived as the new restore line for hybrid combination. After sprayed with Basta at seedling phase, the real hybrid rice that had herbicide resistance could grow normally, while the fake hybrid rice and weeds, having no bar gene, were killed by herbicide. | 6 | Correlations Based on CFD and Their Applications in Optimization for Staggered and Parallel Plate Fin Heatsinks显示文摘Both parallel and staggered plate fin arrays have shown promise tbr use in high performance heatsinks regard of its individual manufacturing costs. The geometrical and operational parameters are very important to their cooling performance as heatsinks in prac- tical applications. Fluent 5.0 commercial CFD (computational tluid dynamic) code is used to simulate the flow and heat transfer of those heatsinks of different realistic parameters. Based on those simulations, two correlations, concerning Nusselt number and friction factor as the functions of geometrical and operational parameters, FB (tin-base area ratio), PR?(ratio of spanwise pitch to lengthwise pitch) and Re, were developed. From the both, the perfonnance comparisons for optimizing geometrical and operational parameters of a fixed di- mension heatsink are shown at constant pumping power and constant thermal resistance. Several optimized parameters were obtained with the discussion to various goals in real application. | 8 | Effects of Suction Side Squealer Tip on the Performance of a Low-Speed Axial Compressor显示文摘This paper presents the investigation of the effects of suction side squealer tip on the performance of an axial compressor.The experiment is carried out in a single-stage large-scale low-speed compressor.The investigated tip geometries include flat tip as the baseline and suction side squealer tip.The tip clearance of the baseline is 0.5% of the blade span.The static pressure rise characteristic curves of both the rotor and the stage are measured.The flow field at the exit of the rotor is measured by a 5-hole probe under design and off-design conditions.The static pressure on the endwall of the rotor passage is also obtained.The results show that the pressure rise characteristic curves obtained by measuring the pressure on the end wall are almost unchanged by using the suction side squealer tip.The measuring results of the 5-hole probe show the static pressure and the total pressure in tip region is slightly greater than that of the flat tip at the design condition at the exit of the rotor.It also leads to greater av-eraged static pressure rise and total pressure.At the near stall condition,the averaged static pressure and total pressure is lower than the baseline which is related to the redistribution of the blade load caused by the suction side squealer tip. | Jun Zhang Hongwei Ma Jingyang Li | 2012 | Journal of Thermal Science2012,21,3: | 5 |

| 11 | Mechanical and thermal properties of RE4Hf3O12(RE=Ho,Er, Tm) ceramics with defect fluorite structure显示文摘The thermal and environmental barrier coatings (T/EBC) are technologically important for advanced propulsion engine system. In this study, RE4Hf3Oi2 (RE=Ho, Er, Tm) with defect fluorite structure was investigated for potential use as top TBC layer. Dense pellets were fabricated via a hot pressing method and the mechanical and thermal properties were characterized. RE4Hf3Oi2 (RE=Ho, Er, Tm) possessed a high Vickers hardness of 11 GFa. The material retained high elastic modulus at elevated temperatures up to 1773 K, which made it attractive for high temperature application. The coefficient of thermal expansion (CTE) of RE4Hf3Oi2 (RE = Ho, Er, Tm) laid in the range between 7× 10^-6K^-1 to 10×10^16K^-1 from 473 K to 1673 K.
