| 2 | Multiple transgenes Populus xeuramericana 'Guariento' plants obtained by biolistic bombardment显示文摘A JERF36 regulation gene, a selection marker gene (NPT-II), and the foreign genes levansucrase (SacB), Vitreoscilla hemoglobin (vgb), and Binary coleopterus insect resistance (BtCry3A+OC-I) were co-transferred into Populus xeuramericana 'Guariento' using biolistic bombardment; 25 kanamycin resistant plants were obtained. The results of PCR and Southern hybridization showed that the foreign genes had been integrated into the genome of P. xeuramericana 'Guariento' and 5 genes were all transferred into 7 poplar plants. | 4 | Research on analysis method for temperature control information of high arch dam construction显示文摘Temperature control,which is directly responsible for the project quality and progress,plays an important role in high arch dam construction.How to discover the rules from a large amount of temperature control information collected in order to guide the adjustment of temperature control measures to prevent cracks on site is the key scientific problem.In this paper,a mathematic logical model was built firstly by means of a coupling analysis of temperature control system decomposition and coordination for high arch dam.Then,an analysis method for temperature control information was presented based on data mining technology.Furthermore,the data warehouse of temperature control was designed,and the artificial neural network forecasting model for the highest temperature of concrete was also developed.Finally,these methods were applied to a practical project. | 5 | Epigenetic modification in histone deacetylase deletion strain of Calcarisporium arbuscula leads to diverse diterpenoids显示文摘Epigenetic modifications have been proved to be a powerful way to activate silent gene clusters and lead to diverse secondary metabolites in fungi. Previously, inactivation of a histone H3 deacetylase in Calcarisporium arbuscula had led to pleiotropic activation and overexpression of more than 75% of the biosynthetic genes and isolation of ten compounds. Further investigation of the crude extract of C. arbuscula Δhda A strain resulted in the isolation of twelve new diterpenoids including three cassanes(1-3), one cleistanthane(4), six pimaranes(5-10), and two isopimaranes(11 and 12) along with two know cleistanthane analogues. Their structures were elucidated by extensive NMR spectroscopic data analysis. | 7 | Diurnal variation of nitrated polycyclic aromatic hydrocarbons in PM_(10) at a roadside site in Xiamen,China显示文摘Intensive daytime and nighttime sampling was carried out from 23 Oct to 31 Dec 2008 to investigate the occurrence of nitrated polycyclic aromatic hydrocarbons(NPAHs) in PM 10 at a roadside site in Xiamen,China.At the same time,six PM 10 samples were collected from a nearby roadway tunnel for comparison.Six NPAHs,namely 9-nitroanthracene,2-and 3-nitrofluoranthene,1nitropyrene,7-nitrobenz[a]anthracene,and 6-nitrobenzo[a]pyrene,were identified and quantified using GC/MS in negative ion chemical ionization mode.The average total concentration of six NPAHs(∑ NPAHs) in the cold season(26 Nov-31 Dec) was 2.3(daytime) and 9.9(nighttime) times higher than those in the warm season.Significant statistical difference(p < 0.01,2-tailed) of ∑ NPAHs between daytime and nighttime was found during both the warm and cold seasons.NPAHs were significantly positively correlated with their parent PAHs and nitrogen dioxide but negatively correlated with ambient temperature.The ratio of 2 + 3-nitrofluoranthene to 1nitropyrene exhibited a similar diurnal pattern as ∑ NPAHs and was generally greater than 5,indicating the importance of secondary atmospheric formation.The diurnal variations of NPAHs were all influenced by the diurnal variations of PAHs,nitrogen dioxide,sunlight,and temperature.The daily inhalable exposure to the six NPAHs in the tunnel was much higher than the roadside values in the warm season but only slightly higher than those in the cold season. | Shuiping Wu Bingyu Yang Xinhong Wang Huasheng Hong Chungshin Yuan | 2012 | Journal of Environmental Sciences2012,24,10: | 5 |

| 9 | Artemisinin ameliorated proteinuria in rats with adriamycin-induced nephropathy through regulating nephrin and podocin expressions显示文摘OBJECTIVE:To investigate the effects of artemisinin against proteinuria and glomerular filtration barrier damage in rats with adriamycin-induced nephropathy,and the potential mechanism underpinned the action.METHODS:Forty adriamycin rats were randomly divided into two groups with the ratio of 1︰3;the small-number group served as control group(n=10),and the rats in the large-number group were treated with adriamycin to induce nephropathy;then they were further randomly assigned into 3subgroups:benazepril group(n=10),artemisiningroup(n=10),and adriamycin group(n=10).The benazepril group and artemisinin group were treated with benazepril suspl(5.0 mg/kg daily)and artemisinin suspl(150 mg/kg daily)respectively after being modeled;those in the control group and adriamycin group were intragastrically administered an equivalent volume of distilled water every day.The treatment after model establishment lasted for a total of 4 weeks.The 24 h uric protein,blood biochemicals,renal pathological changes,renal ultrastrutural changes,Nephrin and Podocin proteins and gene expressions were measured by Coomassie brilliant blue assay,completely automatic biochemical analyzer,light microscope,electron microscopy,Western blot and reverse transcription polymerase chain reaction,respectively.RESULTS:The rats in adriamycin group showed a significant increase in 24 h uric protein excretion,serum total cholesterol(TC),triglyceride(TG),blood urea nitrogen(BUN),serum creatinine(Scr)and decrease in albumin(Alb)(P<0.05 or P<0.01).Compared with adriamycin group,artemisinin could reduce uric protein excretion,decrease the serum TC,TG elevation,increase the serum Alb level,up-regulate the expressions of Nephrin and Podocin(P<0.05 or P<0.01),but no statistical significance effects on the levels of BUN,Scr in artemisinin group(P>0.05).The renal pathological and ultrastrutural observation indicate that artemisinin could attenuate the severity of foot process effacement and fusion in the nephropathic rats.CONCLUSION:Artemisinin might have an effect on the nephropathy in rats caused by adriamycin,which may be at least partly correlated with attenu-ation of the severity of foot process effacement and fusion,up-regulation of the expressions of Nephrin and Podocin in the glomeruli in the rats. | Xili Wu Peng An Bingyu Ye Xingmin Shi Huimin Dang Rongguo Fu Chenglin Qiao | 2014 | Journal of Traditional Chinese Medicine2014,34,1: | 5 |

| 11 | Extraction Process Optimization and Moisturizing Performance of Fucoidan from Sargassum fusiforme显示文摘[Objectives] The research aimed to optimize extraction process of fucoidan from Sargassum fusiforme( FSF) and study its moisturizing performance. [Methods]Extracting condition of FSF by cellulase hydrolysis-ultrasonic assisted extraction method was optimized. The influences of solvent p H,enzyme dosage,extraction temperature,cellulose hydrolysis time,ultrasonication time,and the ratio of material to liquid on FSF were investigated by single factor and orthogonal experiments. [Results] The optimum extraction conditions were as followings:p H,4. 5; enzyme dosage,1%; extraction temperature,40℃; cellulose hydrolysis time,2 h; ultrasonic time,15 min; and the ratio of material to liquid,1∶ 10( g∶ m L). Under the optimal condition,the extraction yield of FSF was 8. 50%,RSD = 2. 74%. The short-time hygroscopicity( within 8 h) of crude extract of fucoidan from S. fusiforme( CEFSF) was better than glycerin,butanediol,and sodium alginate,and the moisture retention capacity of 1% CEFSF aqueous solution was better than 1% butanediol or 1% sodium alginate,and was equal to 5% glycerin under relative humidity of 43% and 81%. The determination results of skin moisture content and transepidermal water loss rate( TEWL)showed that: 5% CEFSF solution had good moisturizing effect. | 13 | Effect of rare earth on the performance of Ru/MgAl-LDO catalysts for ammonia synthesis显示文摘A series of MgAl-layered double oxides(LDO) doped with different rare-earth elements(Y, La, and Ce)were synthesized by the calcination of Mg-Al layered double hydroxides, and Ru, which were used to prepare ammonia synthesis catalysts. The as-obtained oxides and catalysts were characterized by XRD,TEM, TPD, TPR and XPS to understand their catalytic performances in ammonia synthesis. The H_2-TPR and HRTEM studies reveal that Ru/Y-LDO catalyst possesses more active Ru metal and small particle size.The XPS demonstrates that the electronic interaction between Y and Ru metals is stronger, which can be tentatively explained by most of Y inserted into the hydrotalcites structure. CO_2-TPD demonstrates that Ru/Y-LDO catalyst shows stronger basic site densities than catalysts doped with Ce and La. | 14 | Experimental Study on the Protection of Agrimony Extracts from Different Extracting Methods against Cerebral Ischemia-Reperfusion Injury显示文摘Objective To study the protective effect of agrimony extracts from different extracting methods on cerebral ischemia-reperfusion injury in rats, in order to optimize the extraction scheme of agrimony.Methods Male rats were randomly assigned into seven groups: 1. Sham-operated group, 2. Untreated MCAO group (MCAO), 3. Petroleum ether extract of Agrimonia pilosa treated MCAO group (PEA), 4. Ethyl acetate extract of Agrimonia pilosa treated MCAO group (EAEA), 5. Ethanol extract of Agrimonia pilosa treated MCAO group (EEA), 6. Water extract of Agrimonia pilosa treated MCAO group (WEA), 7. Nimodipine treated MCAO group (NP). Intragastrical drug administration (i.g) was performed at 0 and 6 hours after MCAO.Neurological function tests were performed after reperfusion for 24 hours, then the brain was removed for the evaluations of the cerebral infarction volume (percentage of total brain volume) by immunohistochemistry,histological changes (hematoxylin-eosin staining), Na+/K+-ATPase, Ca2+-ATPase (modified method of Svoboda and Mosinger), mRNA expression of Tumor suppressor gene (P53) and hot shock protein (HSP70)(quantitative real-time PCR).Results The neurological function of MCAO group had significantly higher scores than the sham group (P<0.01). The WEA group showed a significantly lower neurological score than the MCAO group (P<0.05),indicating the protective effect of WEA on neurological deficits. The mean infarction volumes of WEA (13.5±6.6%, F=4.75, P<0.01), EEA (19.90±6.90%, F=5.23, P<0.01), PEA (20.40±5.30%, F=4.68,P<0.01) and EAEA (22.50±10.50%, F=6.25, P<0.05) group were all significantly smaller than that of MCAO group (29.40±6.50%). HE staining demonstrated that, compared to the treated groups, the infarcted cerebral tissue of MCAO group had more swelling neural cells, lighter stained nucleus, fewer and irregularly distributed neurons. The activity of Na+/K+-ATPase and Ca2+-ATPase reduced in the MCAO group (3.67±0.48 U/mg,1.28±0.26 U/mg, respectively), and were significantly higher in WEA group (7.56±0.85 U/mg, F=12.65,P=0.010; 3.59±0.22 U/mg, F=8.32, P=0.041, respectively). The MCAO group showed significantly elevated P53 and HSP70 mRNA expressions compared to the sham group (P<0.01, P<0.05). P53 mRNA expressions in Agrimony extracts treated groups were significantly lower than that of the MCAO group (all P<0.01), with the WEA group showing the greatest difference from MCAO group. | 16 | Quantifying the effects of stripe rust disease on wheat canopy spectrum based on eliminating non-physiological stresses显示文摘The wheat canopy reflectance spectrum is affected by many internal and external factors such as diseases and growth stage. Separating the effects of disease stress on the crop from the observed mixed signals is crucial for increasing the precision of remote sensing monitoring of wheat stripe rust. The canopy spectrum of winter wheat infected by stripe rust was processed with the difference-in-differences(DID) algorithm used in econometrics. The monitoring accuracies of wheat stripe rust before and after processing with the DID algorithm were compared in the presence of various external factors, disease severity, and several simulated satellite sensors. The correlation between the normalized difference vegetation index processed by the DID algorithm(NDVI-DID) and the disease severity level(SL) increased in comparison with the NDVI before processing. The increase in precision in the natural disease area in the field in the presence of large differences in growth stage, growth, planting, and management of the crop was greater than that in the controlled experiment. For low disease levels(SL < 20%), the R2 of the regression of NDVI-DIDon SL was 38.8% higher than that of the NDVI and the root mean square error(RMSE) was reduced by 11.1%. The increase in precision was greater than that for the severe level(SL > 40%).According to the measured hyperspectral data, the spectral reflectance of three satellite sensor levels was simulated. The wide-band NDVI was calculated. Compared with the wide-band NDVI and vegetation indexes(VI) before DID processing, there were increases in the correlation between SL and the various types of VIS-DID, as well as in the correlation between SL and NDVI-DID. It is feasible to apply the DID algorithm to multispectral satellite data and diverse types of VISfor monitoring wheat stripe rust. | 17 | Human scFv antibody fragments specific for hepatocellular carcinoma selected from a phage display library显示文摘AIM: To identify the scFv antibody fragments specific for hepatocellular carcinoma by biopanning from a large human naive scFv phage display library.METHODS: A large human naive scFv phage library was used to search for the specific targets by biopanning with the hepatocellular carcinoma cell line HepG2 for the positiveselecting and the normal liver cell line L02 for the counterselecting. After three rounds of biopanning, individual scFv phages binding selectively to HepG2 cells were picked out. PCR was carried out for identification of the clones containing scFv gene sequence. The specific scFv phages were selected by ELISA and flow cytometry. DNA sequences of positive clones were analyzed by using Applied Biosystem Automated DNA sequencers 3 730. The expression proteins of the specific scFv antibody fragments in E. coli HB2151were purified by the affinity chromatography and detected by SDS-PAGE, Western blot and ELISA. The biological effect of the soluble antibody fragments on the HepG2 cells was investigated by observing the cell proliferation.RESULTS: Two different positive clones were obtained and the functional variable sequences were identified.Their DNA sequences of the scFv antibody fragments were submitted to GenBank (accession nos: AY686498 and AY686499). The soluble scFv antibody fragments were successfully expressed in E. coli HB2151. The relative molecular mass of the expression products was about 36 ku,according to its predicted Mr value.
