| 1 | Matrix metalloproteinase-2 and tissue inhibitor of metallo-proteinase-2 in colorectal carcinoma invasion and metastasis显示文摘AIM: To explore the relationship between matrix metalloProteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-2 (TTMP-2) in the development of colorectal carcinoma and to provide a valuable marker for clinical diagnosis.METHODS: Twenty-five patients with colorectal carcinoma underwent surgical resection. Samples were taken from tumor sites and normal tissues. MMP-2 activity was determined by gelatin zymography. Western blot and ABC immunohist ochemical staining were used to detect the expression levels of MMP-2 and TIMP-2 in normal and colorectal carcinoma tissues. Statistical analyses were performed using the Student's ttest and one-way ANOVA. P<0.05 was considered statistically .significant. All the statistical analyses were performed using SPSS 10.0software. RESULTS: MMP-2 activity could be detected in both normal and colorectal carcinoma tissues. MMP-2 actMty in colorectal carcinoma tissues was much higher than that in normal tissues (P<0.05, t =3.916, 4.227). MMP-2 adivity was positively related to the colorectal carcinoma invasion depth, lymph node metastasis and Duke's stage. Western blot and ABC immunohistochemical staining demonstrated that the expression level of MMP-2 in colorectal carcinoma tissues was much higher than that in normal tissues (P<0.05, t = 9.429), but the expression level of TIMP-2 in colorectal carcinoma tissues was much lower than that in normal tissues (P<0.05, t = 7.329). The MMP-2/TIMP-2 ratio of colorectal carcinoma was much higher than that of normal tissues. With the progression of invasion depth, lymph node metastasis and tumor Duke's stage, the activity and expression level of MMP-2 and TIMP-2 gradually increased, but the MMP-2/TIMP-2 ratio gradually decreased. | 2 | Expression of Epstein-Barr virus genes in EBV-associated gastric carcinomas显示文摘AIM: To understand the expression of latent and lytic genes of Epstein-Barr virus (EBV) in EBV-associated gastric carcinoma (EBVaGC) and to explore the relationship between EBV-encoded genes and development of EBVaGC at molecular level.METHODS: One hundred and seventy-two gastric carcinoma tissues and 172 corresponding para-carcinoma tissues were tested for EBV genome by polymerase chain reaction (PCR)-Southern blotting. EBV-encoded small RNA (EBER)1 of the PCR positive specimens was detected by in situ hybridization (ISH). Gastric carcinomas with positive EBER1signals were classified as EBVaGCs. RT-PCR and Southern hybridization were applied to the detection of expression of nuclear antigen (EBNA) promoters (Qp, Wp and Cp),EBNA 1 and EBNA 2, latent membrane proteins (LMP) 1,2A and 2B and lytic genes (immediate early genes BZLF1and BRLF1, early genes BARF1 and BHRF1, late genes BcLF1and BLLF1) in EBVaGCs.RESULTS: Eleven EBV positive samples existed in gastric carcinoma tissues (6.39%). No EBV positive sample was found in corresponding para-carcinoma tissues. The difference between EBV positivity in carcinoma tissues and corresponding para-carcinoma tissues was significant(x2 = 9.0909, P = 0.0026). Transcripts of Qp and EBNA1were detected in all the 11 EBVaGCs, while both Wp and Cp were silent. EBNA2, LMP1 and LMP2B mRNA were absent in all the cases, while LMP2A mRNA was detected in 4 of the 11 cases. Of the 11 EBVaGCs, 7 exhibited BcLF1 transcripts and 2 exhibited BHRF1 transcripts. The transcripts of BZLF1and BARF1 were detected in 5 cases, respectively. No BLLF1and BRLF mRNA were detected.CONCLUSION: The latent pattern of EBV in gastric carcinoma corresponds to the latency Ⅰ/Ⅱ. Some lyric infection genes are expressed in EBVaGCs tissues. | 3 | Relationship between Epstein-Barr virus-encoded proteins with cell proliferation,apoptosis,and apoptosis-related proteins in gastric carcinoma显示文摘AIM: To investigate the interrelationship between EpsteinBarr virus (EBV)-encoded proteins and cell proliferation, apoptosis and apoptosis-related proteins in gastric carcinoma, and to explore their role in gastric carcinogenesis.METHODS: Tissues from 13 cases of EBV-associated gastric carcinoma (EBVaGC) and 45 cases of matched EBV-negative gastric carcinoma (EBVnGC) were collected, and then subjected to analysis for apoptotic index (AI) using the terminal deoxynucleotidyl transferase (TdT)mediated dUTP-biotin nick end-labeling (TUNEL) assay. Nuclear cell proliferation-associated antigen ki-67 index (KI), bcl-2, and p53 expression were examined by immunohistochemistry. p53 mutation in exons 5-8 of 13 EBVaGC cases was determined by single-strand conformation polymorphism (SSCP) and DNA sequencing. RT-PCR and Southern hybridization were used to detect the expression of nuclear antigens (EBNAs) 1 and 2, latent membrane protein (LMP) 1, immediately early gene BZLF1 and early genes BARF1 and BHRF1 in 13 EBVaGC cases. RESULTS: The percentage of AI, KI and p53 overexpression was significantly lower in the EBVaGC group than in the EBVnGC group. However, bcl-2 expression did not show significant difference between the two groups. p53 gene mutations were not found in 13 EBVaGCs. Transcripts of EBNA1 were detected in all 13 EBVaGCs, while both EBNA2 and LMP1 mRNA were not detected. Six of the thirteen cases exhibited BZLF1 transcripts and two exhibited BHRF1 transcripts. BARF1 mRNA was detected in six cases. CONCLUSION: Lower AI and KI may reflect a low biological activity in EBVaGC. EBV infection is associated with p53 abnormal expression but not bcl-2 protein in EBVaGC. | 4 | Reversal of 5-flouroucial resistance by adenovirus-mediated transfer of wild-type p53 gene in multidrug-resiatant human colon carcinoma LoVo/5-FU cells显示文摘AIM: To observe the reversal effects of wide-type p53 gene on multi-drug resistance to 5-FU (LOVO/5-FU).METHODS: After treatment with Ad-p53, LOVO/5-FU sensitivity to 5-Fu was investigated using tetrazolium dye assay. Multidrug resistance gene-1 (MDR1) gene expression was assayed by semi-quantitative reverse transcriptionpolymerase chain reaction and the expression of p53 protein was examined by Western blotting.RESULTS: The reversal activity after treatment with wide type p53 gene was increased up to 4.982 fold at 48 h. The expression of MDR1 gene decreased significantly after treatment with wide-type p53 gene, and the expression of p53 protein lasted for about 5 d, with a peak at 48 h, and began to decrease at 72 h.CONCLUSION: Wide-type p53 gene has a remarkable reversal activity for the high expression of MDR1 gene in colorectal cancers. The reversal effects seem to be in a time dependent manner. | 13 | Effects of Grape Balanced Nutrition Fertilizer on Yield and Quality of Wine Grapes显示文摘In this study,the wine grapes balanced nutrient fertilizer(BNF)was designed according to the wine grape nutritional requirements in different growth stages,and objected to reduce fertilizer application rate of wine grape cultivation,improve fertilizer utilization rate,achieve stable yield and improve the quality of wine grapes.The experiment was carried out at the Great Wall Sanggan Winery,and the Aristocratic Winery.The experiment was designed with two treatments:Namely BNF treatment,and the traditional fertilization(CK).We investigated the effects of BNF on the quality of wine grapes by examining the plant traits of wine grapes in different experimental areas during the growing season,the fruit traits at maturity,and determined the contents of VC,organic acids,amino acids,flavonoids,soluble total sugars,soluble solids,tannins,total phenols and catechins.The results show that:①The application of grape BNF greatly reduced the application amount of nitrogen,phosphorus and potassium,while the vine had no fertilizer deficiency phenomenon;②It increased the single fruit weight,the ear weight and yield to varying degrees,and the yield increase range was between 12% and 41%;③The wine grapes with BNF had the following characteristics:tight infructescence,even color,small fruit less,no fruit dropped,strong fruit colloid feeling,better aroma and taste;④It greatly improved the quality of wine grapes,and increased the content of soluble solids,soluble total sugar,organic acids,VC,total phenols,flavonoids,amino acids and tannins in fruits;⑤It greatly increased the total catechin content of'Riesling'and'Syrah',and had no significant effects on the catechin content of'Merlot'and'Cabernet Sauvignon'.In summary,the application of grape BNF has certain practical significance for guiding the production of local wine grapes,ensuring the yield while improving the quality,increasing the income and reducing the environmental pollution caused by excessive fertilization. | Jianhua LUO Haisen YU Lin SHI Xin KE Yu WANG Jixian DONG Zhigang GUO Donglian LI Pengzhao GAO | 2020 | Agricultural Biotechnology2020,9,4: | 1 |
| 14 | Characteristics and source apportionment of ambient volatile organic compounds and ozone generation sensitivity in urban Jiaozuo,China显示文摘In recent years,many cities have taken measures to reduce volatile organic compounds(VOCs),an important precursor of ozone(O_(3)),to alleviate O_(3) pollution in China.116 VOC species were measured by online and offline methods in the urban area of Jiaozuo from May to October in 2021 to analyze the compositional characteristics.VOC sources were analyzed by a positive matrix factorization(PMF)model,and the sensitivity of ozone generation was determined by ozone isopleth plotting research(OZIPR)simulation.The results showed that the average volume concentration of total VOCs was 30.54 ppbv and showed a bimodal feature due to the rush-hour traffic in the morning and at nightfall.The most dominant VOC groups were oxygenated VOCs(OVOCs,29.3%)and alkanes(26.7%),and the most abundant VOC species were acetone and acetylene.However,based on the maximum incremental reactivity(MIR)method,the major VOC groups in terms of ozone formation potential(OFP)contribution were OVOCs(68.09μg/m^(3),31.5%),aromatics(62.90μg/m^(3),29.1%)and alkene/alkynes(54.90μg/m^(3),25.4%).This indicates that the control of OVOCs,aromatics and alkene/alkynes should take priority.Five sources of VOCs were quantified by PMF,including fixed sources of fossil fuel combustion(27.8%),industrial processes(25.9%),vehicle exhaust(19.7%),natural and secondary formation(13.9%)and solvent usage(12.7%).The empirical kinetic modeling approach(EKMA)curve obtained by OZIPR on O_(3) exceedance days indicated that the O_(3) sensitivity varied in different months.The results provide theoretical support for O_(3) pollution prevention and control in Jiaozuo. | Pengzhao Li Chun Chen Dan Liu Jie Lian Wei Li Chuanyi Fan Liangyu Yan Yue Gao Miao Wang Hang Liu Xiaole Pan Jing Mao | 2024 | Journal of Environmental Sciences2024,,4: | 0 |

| 17 | Protective performance of shear stiffening gel-modified foam against ballistic impact:Experimental and numerical study显示文摘As one of the most widely used personal protective equipment(PPE),body armors play an important role in protecting the human body from the high-velocity impact of bullets or projectiles.The body torso and critical organs of the wear may suffer severe behind-armor blunt trauma(BABT)even though the impactor is stopped by the body armor.A type of novel composite material through incorporating shear stiffening gel(STG)into ethylene-vinyl acetate(EVA)foam is developed and used as buffer layers to reduce BABT.In this paper,the protective performance of body armors composed of fabric bulletproof layers and a buffer layer made of foam material is investigated both experimentally and numerically.The effectiveness of STG-modified EVA in damage relief is verified by ballistic tests.In parallel with the experimental study,numerical simulations are conducted by LS-DYNA®to investigate the dynamic response of each component and capture the key mechanical parameters,which are hardly obtained from field tests.To fully describe the material behavior under the transient impact,the selected constitutive models take the failure and strain rate effect into consideration.A good agreement between the experimental observations and numerical results is achieved to prove the validity of the modelling method.The tests and simulations show that the impact-induced deformation on the human body is significantly reduced by using STG-modified EVA as the buffering material.The improvement of protective performance is attributed to better dynamic properties and more outstanding energy absorption capability of the composite foam. | Huan Tu Haowei Yang Pengzhao Xu Zhe Yang Fan Tang Cheng Dong Yuchao Chen Lei Ren Wenjian Cao Chenguang Huang Yacong Guo Yanpeng Wei | 2024 | Defence Technology(防务技术)2024,32,2: | 0 |

However,its low density and porous structure caused by the sintering mechanism in the absence of shrinkage,restrict its wide applications in engineering.This paper reviews the research progress and related technologies on the preparation of high-density RSi C and its composites. RSi C with relative high density up to 2. 75g·cm- 3can be obtained by a combination of pretreatment to Si C raw materials such as reshaping,modification and particle size distribution,and appropriate forming method. Post treatments such as cyclic pyrolysis and impregnation- recrystallization,and slurry impregnation- recrystallization are needed for the further density increase of RSi C( 2. 99 g·cm- 3). | 19 | Effect of CeO_(2) on carbon deposition resistance of Ni/CeO_(2) catalyst supported on SiC porous ceramic for ethanol steam reforming显示文摘Ni/CeO_(2) catalysts(nCeO_(2):n_(Ni)=0,1,4,7,10)supported on SiC porous ce ramics for ethanol steam reforming(ESR)were investigated with respect to hydrogen production performance and growth of carbon deposition.The oxygen released from CeO_(2) enables the oxidation of CH_(x) species to serve as carbon precursors,thus providing Ni/CeO_(2) catalysts with stronger resistance to carbon deposition compared with Ni catalysts.The Ni/CeO_(2) catalysts prepared by inverse microemulsion and impregnation methods exhibit regular semicircular spherical shape on SiC porous ceramics.Under 500℃for 25 h of ESR reaction,the ethanol conversion rate over Ni/CeO_(2) catalysts(n_(CeO_(2)):n_(Ni)=7)is sustained up to 100%and H_(2) selectivity is essentially kept at 74%.The by-product selectivity declines stepwise with increasing content of CeO_(2),which is attributed to the adsorption and oxidation of CO and of CH_(x) species as CH_4 precursor from CeO_(2).The scanning electron microscopy(SEM)and transform electron microscopy(TEM)results reveal that further loading of CeO_(2) on the surface of Ni catalysts can alleviate both migration and sintering of Ni particles.Furthermore,carbon deposition on Ni/CeO_(2) catalysts preferentially outgrow filamentous rather than amorphous carbon,with a tendency for the latter to be more deactivated. | Tingting Bao Hui zhou Yi Zhang Chenxu Guo Wenming Guo Hang Qin Pengzhao Gao Hanning Xiao Weixiong Liu | 2023 | Journal of Rare Earths2023,41,11: | 0 |
| 20 | Efficient regeneration system applicable to five Musa cultivars显示文摘Banana(Musa spp.) is an important staple food, economic crop, and nutritional fruit worldwide.Hybridization is seriously hampered by the long generation time, polyploidy, and sterility of most cultivars.Establishment of an efficient regeneration and transformation system for banana is critical for their genetic improvement. An efficient and reproducible transformation system for banana using direct organogenesis was developed. Media containing benzylaminopurine(BA)combined with one of four other growth regulators was evaluated for the regeneration efficiency of five Musa cultivars and the ability to induce/support development of new banana shoots. The result indicated that the greatest number of shoots per explant for all five Musa cultivars was obtained using MS medium supplemented with8.9 mmol$L–1BA and 9.1 mmol$L–1thidiazuron(TDZ).In 240–270 d, one immature male flower could regenerate between 380 and 456, 310–372, 200–240, 130–156, and100–130 well-developed shoots for Gongjiao, Red banana, Rose banana, Baxi, and Xinglongnaijiao, respectively.
