| 1 | Astragalus polysaccharide regulates brown adipogenic differentiation through miR-1258-5p-modulated cut-like homeobox 1 expression显示文摘Astragalus polysaccharide(APS)is the major natural active component of Astragalus membranaceus,which has been recognized as one of the most popular herbal medicines worldwide.Enhancing the formation and function of brown adipose tissue increases energy expenditure and hence may potentially be used against obesity and type 2 diabetes.The aim of the present study was to explore the effect and mechanism of APS on brown adipocyte formation.Mouse C3H10T 1/2 cells were subject to APS,and both proliferation and brown adipogenic differentiation were determined.The results showed that APS exhibits a decreased proliferation ability,which is accompanied by downregulated proliferating cell nuclear antigen,cyclin D1,and cyclin-dependent kinase 4.APS promotes the differentiation of C3H10T 1/2 cells into brown adipocytes and induces the expressions of key brown adipogenic transcriptional factors,including CCAAT/enhancer-binding proteinβ,uncoupling protein 1,and PR domain-containing 16.Importantly,APS enables insulin sensitization in brown adipocytes,which may proceed through activation of the canonical phosphatidylinositol-3-kinase(PI3K)/protein kinase B(AKT)signaling pathway and AMP-activated protein kinase(AMPK).Furthermore,the level of cut-like homeobox 1(CUX1)is positively related to brown adipogenic differentiation,while APS regulates Cux1 expression through interaction with miR-1258-5p.Notably,the promotional effect of APS on brown adipogenic differentiation was abolished by Cux1 knockout.Collectively,our results suggest that APS enhances the differentiation of C3H10T 1/2 cells into brown adipocytes through regulating Cux1 via miR-1258-5p. | Yuxin Cao Buhao Deng Shihe Zhang Hongmei Gao Pengkang Song Jianxin Zhang Junxing Zhao | 2021 | Acta Biochimica et Biophysica Sinica2021,53,12: | 1 |
| 2 | Tracking the reactivity of ozonation towards effluent organic matters from WWTP using two-dimensional correlation spectra显示文摘The characteristics of effluent organic matter(EfOM) from a wastewater treatment plant(WWTP) during ozonation were investigated using excitation and emission matrix(EEM)spectra, Fourier transform infrared spectroscopy(FT-IR) and high-performance size exclusion chromatography(HPSEC) at different ozone dosages. The selectivity of ozonation towards different constituents and functional groups was analysed using two-dimensional correlation spectra(2D-COS) probed by FT-IR, synchronous fluorescence spectra and HPSEC.The results indicated that ozonation can destroy aromatic structures of EfOM and change its molecular weight distribution(MWD). According to 2D-COS analysis, microbial humiclike substances were preferentially removed, and then the protein-like fractions. Terrestrial humic-like components exhibited inactivity towards ozonation compared with the above two fractions. Protein-like substances with small molecular weight were preferentially reacted during ozonation based on 2D-COS probed by HPSEC. In addition, the selectivity of ozone towards different functional groups of EfOM exhibited the following sequence:phenolic and alcoholic C\O groups > aromatic structures containing C_C double bonds >aliphatic C\H. | 5 | Two-dimensional correlation spectroscopic analysis on the interaction between humic acids and aluminum coagulant显示文摘In this study, two-dimensional correlation spectroscopy integrated with synchronous fluorescence and infrared absorption spectroscopy was employed to investigate the interaction between humic acids and aluminum coagulant at slightly acidic and neutral p H. Higher fluorescence quenching was produced for fulvic-like and humic-like fractions at p H 5. At p H 5, the humic-like fractions originating from the carboxylic acid, carboxyl and polysaccharide compounds were bound to aluminum first, followed by the fulvic-like fractions originating from the carboxyl and polysaccharide compounds. This finding also demonstrated that the activated functional groups of HA were involved in forming the Al-HA complex, which was accompanied by the removal of other groups by co-precipitation.Meanwhile, at p H 7, almost no fluorescence quenching occurred, and surface complexation was observed to occur, in which the activated functional groups were absorbed on the amorphous Al(OH)3. Two-dimensional FT-IR correlation spectroscopy indicated the sequence of HA structural change during coagulation with aluminum, with IR bands affected in the order of COOH> COO->NH deformation of amide Ⅱ> aliphatic hydroxyl C\OH at p H 5, and COO->aliphatic hydroxyl C\OH at p H 7. | 6 | Application of a hybrid gravity-driven membrane filtration and dissolved ozone flotation(MDOF)process for wastewater reclamation and membrane fouling mitigation显示文摘This study proposed a novel membrane filtration and dissolved ozone flotation integrated(MDOF) process and tested it at pilot scale. Membrane filtration in the MDOF process was operated in gravity-driven mode, and required no backwashing, flushing, or chemical cleaning. Because ozone was added in the MDOF process, ozonation, coagulation, and membrane filtration could occur in a single reactor. Moreover, in situ ozonation occurred in the MDOF process, which differs from the conventional pre-ozonation membrane filtration process. Significant enhancement of turbidity removal was further achieved through the addition of membrane filtration. Membrane fouling was mitigated in the MDOF process compared to the MDAF process. In situ ozonation in the MDOF process decreased the fluorescence intensity and transformed the high MW dissolved organics into small MW compounds. For the fouling layer, the extracellular polymeric substance(EPS) contents and cake layer morphology were analyzed. The results indicated that the contents of EPS decreased. Furthermore, a thinner and more loosely structured cake layer formed in the MDOF process. | 7 | Vitamin A injection at birth improves muscle growth in lambs显示文摘Vitamin A and its metabolite,retinoic acid(RA)play important roles in regulating skeletal muscle development.This study was conducted to investigate the effects of early intramuscular vitamin A injection on the muscle growth of lambs.A total of 16 newborn lambs were given weekly intramuscular injections of corn oil(control group,n=8)or 7,500 IU vitamin A palmitate(vitamin A group,n=8)from birth to 3 wk of age(4 shots in total).At 3 wk of age and weaning,biceps femoris muscle samples were taken to analyze the effects of vitamin A on the myogenic capacity of skeletal muscle cells.All lambs were slaughtered at 8 months of age.The results suggest that vitamin A treatment accelerated the growth rate of lambs and increased the loin eye area(P<0.05).Consistently,vitamin A increased the diameter of myofibers in longissimus thoracis muscle(P<0.01)and increased the final body weight of lambs(P<0.05).Vitamin A injection did not change the protein kinase B/mammalian target of rapamycin and myostatin signaling(P>0.05).Moreover,vitamin A upregulated the expression of PAX7(P<0.05)and the myogenic marker genes including MYOD and MYOG(P<0.01).The skeletal muscle-derived mononuclear cells from vitamin A-treated lambs showed higher expression of myogenic genes(P<0.05)and formed more myotubes(P<0.01)when myogenic differentiation was induced in vitro.In addition,in vitro analysis showed that RA promoted myogenic differentiation of the skeletal muscle-derived mononuclear cells in the first 3 d(P<0.05)but not at the later stage(P>0.05)as evidenced by myogenic gene expression and fusion index.Taken together,neonatal intramuscular vitamin A injection promotes lamb muscle growth by promoting the myogenic potential of satellite cells. | Pengkang Song Xiaoyou Chen Jiamin Zhao Qiang Li Xinrui Li Yu Wang Bo Wang Junxing Zhao | 2023 | Animal Nutrition2023,,3: | 0 |

| 9 | Vitamin A regulates mitochondrial biogenesis and function through p38 MAPK‑PGC‑1α signaling pathway and alters the muscle fiber composition of sheep显示文摘Background Vitamin A(VA)and its metabolite,retinoic acid(RA),are of great interest for their wide range of physiological functions.However,the regulatory contribution of VA to mitochondrial and muscle fiber composition in sheep has not been reported.Method Lambs were injected with 0(control)or 7,500 IU VA palmitate into the biceps femoris muscle on d 2 after birth.At the age of 3 and 32 weeks,longissimus dorsi(LD)muscle samples were obtained to explore the effect of VA on myofiber type composition.In vitro,we investigated the effects of RA on myofiber type composition and intrinsic mechanisms.Results The proportion of type I myofiber was greatly increased in VA-treated sheep in LD muscle at harvest.VA greatly promoted mitochondrial biogenesis and function in LD muscle of sheep.Further exploration revealed that VA elevated PGC-1αmRNA and protein contents,and enhanced the level of p38 MAPK phosphorylation in LD muscle of sheep.In addition,the number of type I myofibers with RA treatment was significantly increased,and type IIx myofibers was significantly decreased in primary myoblasts.Consistent with in vivo experiment,RA significantly improved mitochondrial biogenesis and function in primary myoblasts of sheep.We then used si-PGC-1αto inhibit PGC-1αexpression and found that si-PGC-1αsignificantly abrogated RA-induced the formation of type I myofibers,mitochondrial biogenesis,MitoTracker staining intensity,UQCRC1 and ATP5A1 expression,SDH activity,and enhanced the level of type IIx muscle fibers.These data suggested that RA improved mitochondrial biogenesis and function by promoting PGC-1αexpression,and increased type I myofibers.In order to prove that the effect of RA on the level of PGC-1αis caused by p38 MAPK signaling,we inhibited the p38 MAPK signaling using a p38 MAPK inhibitor,which significantly reduced RA-induced PGC-1αand MyHC I levels.Conclusion VA promoted PGC-1αexpression through the p38 MAPK signaling pathway,improved mitochondrial biogenesis,and altered the composition of muscle fiber type. | Pengkang Song Jiamin Zhao Fanqinyu Li Xiaoyi Zhao Jinxin Feng Yuan Su Bo Wang Junxing Zhao | 2024 | Journal of Animal Science and Biotechnology2024,15,2: | 0 |
| 10 | A comprehensive simulation approach for pollutant bio-transformation in the gravity sewer显示文摘Presently, several activated sludge models (ASMs) have been developed to describe a few biochemical processes. However, the commonly used ASM neither clearly describe the migratory transformation characteristics of fermentation nor depict the relationship between the carbon source and biochemical reactions. In addition, these models also do not describe both ammonification and the integrated metabolic processes in sewage transportation. In view of these limitations, we developed a new and comprehensive model that introduces anaerobic fermentation into the ASM and simulates the process of sulfate reduction, ammonification, hydrolysis, acidogenesis and methanogenesis in a gravity sewer. The model correctly predicts the transformation of organics including proteins, lipids, polysaccharides, etc. The simulation results show that the degradation of organics easily generates acetic acid in the sewer system and the high yield of acetic acid is closely linked to methanogenic metabolism. Moreover, propionic acid is the crucial substrate for sulfate reduction and ammonification tends to be affected by the concentration of amino acids. | 11 | Serum Procalcitonin Levels on Admission Predict Death in Severe and Critical COVID-19 Patients in Wuhan, China显示文摘Background:We evaluated whether the serum procalcitonin(PCT)level could predict death in severe and critical coronavirus disease 2019(COVID-19)patients.Methods:This study included 129 COVID-19 patients.PCT levels on admission,treatment,and death were collected.The outcomes were compared.Results:The optimum cutoff value of the PCT level determined by receiver operator characteristic curve analysis to predict all-cause death was 0.085 ng/mL,with sensitivity of 95.7%and specifi city of 72.6%.Overall,78 patients had a PCT level below 0.085 ng/mL and 51 patients had a PCT level of 0.085 ng/mL or greater.High-PCT-level patients had lower levels of lymphocytes(P=0.001)and albumin(P=0.002)and higher levels of creatinine(P=0.024),D-dimer(P=0.002),and white blood cells,neutrocytes(P<0.001),high-sensitivity C-reactive protein(P<0.001),interleukin-6(P<0.001),interleukin-8(P=0.001),interleukin-10(P=0.001),tumor necrosis factor(P<0.001),erythrocyte sedimentation rate(P=0.001),and ferritin(P=0.001).During the 30-day observation period,23 patients died.Mortality was significantly higher in high-PCT-level patients than in patients with low PCT levels(43.1%vs.1.3%;P<0.001).The risks of death(P<0.0001)and ventilator use(P<0.0001)were increased in patients with PCT levels of 0.085 ng/mL or greater.Conclusions:A PCT level of 0.085 ng/mL or greater on admission could effectively predict death and ventilator use in severe and critical COVID-19 patients. | Han Jin Shengwen Yang Pengkang He Haoyu Weng Shengcong Liu Fan Yang Long Zhang Fangfang Fan Yan Zhang Jing Zhou Jianping Li | 2020 | Cardiovascular Innovations and Applications2020,,3: | 0 |
