1Expression of calreticulin is associated with infiltration of T-cells in stage ⅢB colon cancer显示文摘AIM: To investigate the correlation between expression of calreticulin and infiltration of lymphocytes in stage ⅢB colon cancer. METHODS: Sixty-eight pathologically-confirmed speci-mens were obtained from stage ⅢB (T3N1M0) colon cancer patients who underwent radical resection between January 1999 and May 2002 at the Cancer Center of Sun Yat-Sen University, Guangzhou, China. Immuno-histochemical analysis was performed to show infiltration of lymphocytes and expression of calreticulin in colon cancer. Association between calreticulin expression, infiltration of lymphocytes, and 5-year survival rate of patients was assessed. RESULTS: The expression level of calreticulin was lower in cancer nest than in its adjacent normal epithelium since 61.8% (42/68) of the samples were stained with calreticulin in colon cancer. The expression of calreticulin in colon cancer was associated with the infi ltration of CD45RO+ cells rather than with that of CD3+ cells. In addition, the stronger expression of calreticulin and the higher infiltration of CD3+ and CD45RO+ cells in colon cancer were associated with the higher 5-year survival rate of patients. 2Role of myeloid-derived suppressor cells in autoimmune disease显示文摘Myeloid-derived suppressor cells(MDSCs) represent an important class of immunoregulatory cells that can be activated to suppress T cell functions. These MDSCs can inhibit T cell functions through cell surface interactions and the release of soluble mediators. MDSCs accumulate in the inflamed tissues and lymphoid organs of patients with autoimmune diseases. Much of our knowledge of MDSC function has come from studies involving cancer models, however many recent studies have helped to characterize MDSC involvement in autoimmune diseases. MDSCs are a heterogeneous group of immature myeloid cells with a number of different functions for the suppression of T cell responses. However, we have yet to fully understand their contributions to the development and regulation of autoimmune diseases. A number of studies have described beneficial functions of MDSCs during autoimmune diseases, and thus there appears to be a potential role for MDSCs in the treatment of these diseases. Nevertheless, many questions remain as to the activation, differentiation, and inhibitory functions of MDSCs. This review aims to summarize our current knowledge of MDSC subsets and suppressive functions in tissue-specific autoimmune disorders. 3Role of CD56-expressing immature biliary epithelial cells in biliary atresia显示文摘AIM: To analyze the clinical and pathological parameters and expression of the neural cell adhesion molecule(CD56) in patients with biliary atresia(BA).METHODS: Established clinical laboratory markers of hepatic function, including enzyme activity, protein synthesis, and bilirubin metabolism, were evaluated in patients with BA and compared with those in patients with choledochal cysts and neonatal hepatitis. Pathological changes in tissue morphology and fibrosis were examined by histological and tissue collagen staining. Immunohistochemical staining for the biliary epithelial cell markers CD56 and CK19 together with the Notch signaling related molecules Notch1 and Notch2 was performed in the context of alterations in the structure of intrahepatic biliary ducts.RESULTS: Differences in some clinical laboratoryparameters among the three diseases examined were observed, but they did not correlate with the pathological classification of fibrosis in BA. Immunohistochemical staining showed the presence of CD56-positive immature bile ducts in most patients(74.5%) with BA but not in patients with choledochal cysts or neonatal hepatitis. The number of CD56-expressing cells correlated with disease severity, with more positive cells present in the later stages of liver damage(81.8% vs 18.2%). Furthermore, bile plugs were mainly found in CD56-positive immature biliary ducts. Notch signaling was a key regulatory pathway in biliary duct formation and played a role in tissue fibrosis. Notch1 was co-expressed in CD56-positive cells, whereas Notch2 was found exclusively in blood vessels in the portal area of patients with BA. 4A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 7Physiological and Biochemical Characteristics of Lactation in Half-lactating Yak显示文摘Yaks of two lactating types, the half-lactating yak and the total lactating yak, were investigated in their milk compositions, milk coagulation properties, and contents of three kinds of hormones, glucose,and protein in plasma. The half-lactating Maiwa yak and Jiulong yak contained significantly higher contents of protein, fat, and activities of alkaline phosphatase and γ-glutamyl transpeptidase in milk than that of the corresponding total-lactating yak breeds, with reduced milk yield and similar lactose level and relative percentages of main milk protein components. The half-lactatin yak resembled yaks in a late stage of lactation in their biochemical composition of milk; however, significant differences were also observed, which indicated that the former was in a special stage of lactation quite different from dairy cows. Milk of the total-lactating or halflactating yaks could be coagulated normally by adding chymosin, with a similar coagulation time. 9Synthesis, Crystal Structure and Calculated β Value of a Tetrahedral Zinc (II) Complex——Zn(2-NH_2py)_2Br_2显示文摘A new zinc(II) complex, Zn(2-NH2py)2Br2 (py=pyridine), has been synthesized, and its molecular structure has been confirmed by IR, elemental analysis and -ray crystal structure analysis. Its space group is P21/n with Mr=413.43 (C10H12Br2N4Zn), a=7.435(2), b=12.865(3), c=14.186(4)? b=94.08(2)? V=1353.5(5) 3, Z=4, Dc=2.029 g/cm3, F(000)=328, ?2.378mm-1, R=0.0283, wR=0.0501. The total observed reflections with I≥2?I) were 5314, of which the independent reflections were 3106. The complex structure contains a distorted tetrahedron formed by four atoms coordinated to zinc atom, namely two bromide atoms and two nitrogen atoms. The two ZnBr bond lengths within one molecule, 2.3763(6) and 2.4002(5)? respectively, are not equal; and so are the two ZnN bond lengths, which are 2.031(2) and 2.044(2)?respectively. The calculation results using PM3 method through MOPAC software package in Chem 3D show that its first molecular hyperpolarizability b value is 5.210-30esu, which is comparable with that of p-nitroaniline. 10Functional analysis of tomato calmodulin gene family during fruit development and ripening显示文摘Calmodulin is a ubiquitous calcium sensor to recognize the different developmental and/or stimulus-triggered calcium changes and regulate plant growth and development.However,the function of calmodulin remains elusive for fleshy fruit development.We performed expression studies of a family of six calmodulin genes(SlCaMs)in tomato fruit.All calmodulins showed a double peak expression pattern.The first flat peak appeared at 10–30 days after anthesis,but their expression rapidly declined at mature green and breaker.Then a sharp and even higher peak came at turning/pink stages.Among six calmodulins,SlCaM1 had the highest expression during fruit enlargement,whereas SlCaM2 was the major calmodulin during fruit ripening.However,SlCaMs showed different patterns in three ripening mutants rin,Nor and Nr.In particular,at the stages corresponding to mature green and breaker,the expression levels of SlCaMs in those mutants were significantly higher than wild-type.Furthermore,SlCaMs,especially SlCaM2 were upregulated by ethylene.Transiently overexpressing SlCaM2 in mature green fruit delayed ripening,while reducing SlCaM2 expression accelerated ripening.Our results suggest that SlCaMs play double roles to regulate fruit ripening.Prior to the ethylene burst,the ethylene-independent repression of SlCaMs might be critical for fruit to initiate the ripening process.After the ethylene burst,SlCaMs could participate in the ethylene coordinated rapid ripening.Tianbao Yang Hui Peng Gary R Bauchan 2014Horticulture Research2014,1,1:3

12Geochemistry of Permian Coal and Its Combustion Residues in Huainan Coalfield, China显示文摘INAA (instrumental neutron activation analysis) was used to determine the contents of trace elements in coal samples from 11 workable Permian coal seams in Huainan coalfield. With XRFS (X-ray fluorescence spectrometry), the geochemical composition of its combustion residues from Tianjiaan Power Plant and Luohe Power Plant was analyzed. In addition, the electron microprobe was employed to study the chemical composition of some fly ashes and bottom ashes. Compared with those of the coals around the world, most trace elements of the Huainan coal are characterized by a normal abundance. Like most coals worldwide, the elements Se, S, As, Sb, Br, U and Cl are enriched in Huainan coal according to dark’s value, while the other elements tend to be diluted. The large fluctuation in the contents of trace elements in this kind of coal demonstrates great differences in coal seams. On the other hand, the contents of these elements are very similar in different bottom and fly ash samples. The elements with low organic affinity tend to concentrate in fly and bottom ash than in coal, especially those elements associated with Fe. However, the elements with strong organic affinity tend to be volatile during the combustion. Moreover, the abundance of the elements in fly ash and bottom ash changes with different densities and with different particle sizes. The bottom and fly ashes consist of glass, lime, mullite, coal residues and ferrite minerals.
