| 1 | 美国临床肿瘤学会IV期非小细胞肺癌化疗的临床实践指南更新显示文摘本文旨在为IV期非小细胞肺癌患者的治疗提供更新版推荐。本文资料检索源自2002年以来公布的相关随机试验文献。此指南范围限于化疗与生物治疗。更新委员会对这些文献进行了总结并提供了推荐更新。162篇文献符合标准被纳入参考。本推荐基于可改善总生存期的治疗方法。仅改善无进展生存期的治疗方法推动了对毒性及生存质量的监测。对于体力状态评分为0分或1分患者的一线治疗,可推荐以铂类为基础的细胞毒性药物的两药联用。对铂类治疗有禁忌的患者,可采用非铂类细胞毒性两药联合。对于体力状态评分为2分的患者,单一细胞毒性药物即可。对于疾病进展或经过4个周期的治疗仍对治疗无反应的患者,应停止一线细胞毒性化疗。即使在6个周期后患者对治疗仍有反应,亦应停止两药细胞毒性化疗。对于伴有明确的表皮生长因子受体(epidermal growth factor receptor,EGFR)突变的患者,可推荐一线采用吉非替尼治疗;对于EGFR突变为阴性或不明确的患者,细胞毒性化疗更佳。除具有特定临床特征的患者外,可推荐贝伐单抗与卡铂-紫杉醇联用。对于通过免疫组化证实EGFR阳性的肿瘤患者,可推荐西妥昔单抗与顺铂-长春瑞滨联用。多西紫杉醇、厄洛替尼、吉非替尼或培美曲塞被推荐作为二线治疗。对于未曾接受过厄洛替尼或吉非替尼治疗的患者,可推荐厄洛替尼作为三线治疗。现有数据不足以推荐常规三线采用细胞毒性药物。已有的证据也不足以推荐常规应用分子标记物选择化疗。 | Christopher G. | 2 | Optimization of phenol degradation by Candida tropicalis Z-04 using Plackett-Burman design and response surface methodology显示文摘Statistical experimental designs were used to optimize the process of phenol degradation by Candida tropicalis Z-04,isolated from phenol-degrading aerobic granules.The most important factors influencing phenol degradation (p<0.05),as identified by a two-level Plackett-Burman design with 11 variables,were yeast extract,phenol,inoculum size,and temperature.Steepest ascent method was undertaken to determine the optimal regions of these four significant factors.Central composite design (CCD) and response surface analysis were adopted to further investigate the mutual interactions between these variables and to identify their optimal values that would generate maximum phenol degradation.The analysis results indicated that interactions between yeast extract and temperature,phenol and temperature,inoculum size and temperature affected the response variable (phenol degradation) significantly.The predicted results showed that the maximum removal efficiency of phenol (99.10%) could be obtained under the optimum conditions of yeast extract 0.41 g/L,phenol 1.03 g/L,inoculum size 1.43% (V/V) and temperature 30.04°C.These predicted values were further verified by validation experiments.The excellent correlation between predicted and experimental values confirmed the validity and practicability of this statistical optimum strategy.This study indicated the excellent ability of C.tropicalis Z-04 in degrading high-strength phenol.Optimal conditions obtained in this experiment laid a solid foundation for further use of this microorganism in the treatment of highstrength phenol effluents. | Jiangya Zhou Xiaojuan Yu Cong Ding Zhiping Wang Qianqian Zhou Hao Pao Weimin Ca | 2011 | Journal of Environmental Sciences2011,23,1: | 48 |

| 6 | Effects of GS3 and GL3.1 for Grain Size Editing by CRISPR/Cas9 in Rice显示文摘Grain size is one of key agronomic traits associated with grain yield and grain quality. Both major quantitative trait loci GS3 and GL3.1 play a predominant role in negative regulation of grain size. In this study, a CRISPR/Cas9-mediated multiplex genome editing system was used to simultaneously edit GS3 and GL3.1 in a typical japonica rice Nipponbare. In T1 generation, we found that gs3 formed slender grain with lower chalkiness percentage, while gs3gl3.1 produced larger grain with higher chalkiness percentage. In terms of other agronomic traits, flag leaf size, grain number and grain yield of both gs3 and gs3gl3.1 mutants were affected. It is noteworthy that gs3 and gs3gl3.1 mutants both led to dramatical reduction of grain number, thereby decreased grain yield. | 7 | Characterization of high-yield performance as affected by genotype and environment in rice显示文摘We characterized yield-relevant characters and their variations over genotypes and environments(locations and years) by examining two rice varieties(9746 and Jinfeng) with high yield potential. 9746 and Jinfeng were planted in two locations of Shanghai,China,during 2005 and 2006. The results show that there was a large variation in grain yield between locations and years. The realization of high yield potential for the two types of rice was closely related to the improved sink size,such as more panicles per square meter or grains per panicle. Stem and leaf biomasses were mainly accumulated from tillering stage to heading stage,and showed slow decline during grain filling. Meanwhile,some photosynthetic characters including net photosynthesis rate(Pn),leaf area index(LAI),specific leaf area(SLA),fluorescence parameter(maximum quantum yield of PSII,Fv/Fm),chlorophyll content(expressed as SPAD value),as well as nutrient(N,P,K) uptake were also measured to determine their variations over genotypes and environments and their relationships with grain yield. Although there were significant differences between years or locations for most measurements,SLA at tillering and heading stages,Fv/Fm and LAI at heading stage,stem biomass at heading and maturity stages,and leaf nitrogen concentration at tillering and heading stages remained little changed,indicating their pos-sible applications as selectable characters in breeding programs. | 9 | Assessing the genetic diversity of the critically endangered Chinese sturgeon Acipenser sinensis using mitochondrial markers and genome-wide single-nucleotide polymorphisms from RAD-seq显示文摘As a living fossil, the endangered Chinese sturgeon(Acipenser sinensis) has been considered a national treasure in China. Here,the famous Gezhouba Dam and Three Gorges Dam on the Yangtze River were built in 1988 and 2006, for economic purposes.The natural population of Chinese sturgeon has declined since then, as these dams block its migratory route to the original spawning grounds in the middle reaches of the Yangtze River. In 2013 and 2014, there was an absence of spawning where it typically happened near the Gezhouba Dam. Nevertheless, from April to June in 2015, over 1,000 larvae with different body lengths(10–35 cm) were detected along the Shanghai Yangtze Estuary; but only little is currently known about the population genetic structure of the Chinese sturgeon. Herein, we inferred population genetic parameters from 462 available Chinese sturgeon specimens based on a 421-bp fragment of the mitochondrial DNA(mtDNA) D-loop region and 1,481,620 SNPs(singlenucleotide polymorphisms) generated by restriction site-associated DNA sequencing(RAD-seq). For the D-loop dataset, 15 haplotypes were determined. Randomly picked 23 individuals, representing the 15 D-loop haplotype groups, were subsequently used for further RAD-seq validation. The average nucleotide diversity calculated from the mtDNA and RAD datasets was 0.0086 and 0.000478, respectively. The overall effective female population size was calculated to be 1,255 to 2,607, and the long-term effective population size was estimated to range from 11,950 to 119,500. We observed that the genetic variability and the effective female population size of the current population in the Yangtze River are severely low, which are similar to the data reported over 10 years ago.
