| 3 | Spontaneous T-symmetry breaking and exceptional points in cavity quantum electrodynamics systems显示文摘Spontaneous symmetry breaking has revolutionized the understanding in numerous fields of modern physics. Here, we theoretically demonstrate the spontaneous time-reversal symmetry breaking in a cavity quantum electrodynamics system in which an atomic ensemble interacts coherently with a single resonant cavity mode. The interacting system can be effectively described by two coupled oscillators with positive and negative mass, when the two-level atoms are prepared in their excited states. The occurrence of symmetry breaking is controlled by the atomic detuning and the coupling to the cavity mode,which naturally divides the parameter space into the symmetry broken and symmetry unbroken phases.The two phases are separated by a spectral singularity, a so-called exceptional point, where the eigenstates of the Hamiltonian coalesce. Because of its rapid growth, the efficiency and robustness of AM-based product development processes should be improved. Artificial intelligence(AI) is a powerful tool that has outperformed humans in numerous complex tasks. AI-enabled intelligent agents can reduce the workforce required to scale up AM production and achieve higher resource utilization efficiency. This study provides an introduction of AI techniques. Then,the current development of AI-enabled AM product development is investigated. Existing intelligent agents are used for problems in product design, process design and production stages. Based on the review, current research gaps and future research directions are identified. To guide future development of more efficient and comprehensive intelligent agents, a smart AM framework based on cloud-edge computing is proposed. | 7 | Achieving Safe Deep Reinforcement Learning via Environment Comprehension Mechanism显示文摘Deep reinforcement learning(DRL), which combines deep learning with reinforcement learning, has achieved great success recently. In some cases, however,during the learning process agents may reach states that are worthless and dangerous where the task fails. To address the problem, we propose an algorithm, referred as Environment comprehension mechanism(ECM) for deep reinforcement learning to attain safer decisions. ECM perceives hidden dangerous situations by analyzing object and comprehending the environment, such that the agent bypasses inappropriate actions systematically by setting up constraints dynamically according to states. ECM, which calculates the gradient of the states in Markov tuple, sets up boundary conditions and generates a rule to control the direction of the agent to skip unsafe states. ECM is able to be applied to basic deep reinforcement learning algorithms to guide the selection of actions. | 8 | AIF-regulated oxidative phosphorylation supports lung cancer development显示文摘Cancer is a major and still increasing cause of death in humans. Most cancer cells have a fundamentally different metabolic profile from that of normal tissue. This shift away from mitochondrial ATP synthesis via oxidative phosphorylation towards a high rate of glycolysis, termed Warburg effect, has long been recognized as a paradigmatic hallmark of cancer, supporting the increased biosynthetic demands of tumor cells. Here we show that deletion of apoptosis-inducing factor (AIF) in a Kras^G12D-driven mouse lung cancer model resulted in a marked survival advantage, with delayed tumor onset and decreased malignant progression. Mechanistically, Aif deletion leads to oxidative phosphorylation (OXPHOS) deficiency and a switch in cellular metabolism towards glycolysis in non-transformed pneumocytes and at early stages of tumor development. Paradoxically, although Aif-deficient cells exhibited a metabolic Warburg profile, this bioenergetic change resulted in a growth disadvantage of Kras^G12D-driven as well as Kras wild-type lung cancer cells. Cell-autonomous re-expression of both wild-type and mutant AIF (displaying an intact mitochondrial, but abrogated apoptotic function) in Aif-knockout Kras^G12D mice restored OXPHOS and reduced animal survival to the same level as AIF wild-type mice. In patients with non-small cell lung cancer, high AIF expression was associated with poor prognosis. | 15 | STX5 Inhibits Hepatocellular Carcinoma Adhesion and Promotes Metastasis by Regulating the PI3K/mTOR Pathway显示文摘Background and Aims:Syntaxin 5(STX5)is a member of the syntaxin or target-soluble SNAP receptor(t-SNARE)fam-ily and plays a critical role in autophagy.However,its function and molecular mechanism in tumor cell migration are still un-known.The role of STX5 in influencing hepatocellular carci-noma(HCC)is an important topic in our research.Methods:By using quantitative reverse transcription polymerase chain reaction(qPCR),western blotting,and immunohistochemical analysis of RNA and protein in tissues,we comprehensively evaluated data sets from public databases and clinical patient cohorts for STX5.The correlation of STX5 expression with the clinicopathological characteristics of HCC patients were assessed.In addition,we predicted signal pathways from dif-ferentially expressed genes(DEGs)and the Cancer Genome Atlas(TCGA)databases,and confirmed the prediction using integrated transcriptome and RNA-seq.We further investi-gated the underlying mechanisms of STX5 in the migration and adhesion of HCC cells both in vitro and in vivo.Results:In the TCGA dataset and our patient cohort,STX5 levels were significantly higher in HCC tissues than in adjacent normal liver tissues.At the same time,high expression of STX5 pre-dicted worse prognosis in patients with liver cancer.High ex-pression of STX5 indicates the decrease of adhesion and the increase of migration of HCC cells,and the conversion of epi-thelial-mesenchymal transition(EMT)in vitro via PI3K/mTOR pathway activation.Conversely,when Sirolimus,a phospho-inositide 3-kinase(PI3K)/AKT/mechanistic target of rapa-mycin(mTOR)inhibitor acts on cells simultaneously,STX5 overexpression-mediated enhancement of HCC metastasis is reversed.Double-negative regulation of STX5 and mTOR further enhanced the inhibitory effect of STX5 on HCC me-tastasis.In vivo,STX5 knockdown inhibited the metastasis of HCC cells.Conclusions:Our study demonstrates a novel research result that STX5 promotes HCC metastasis through PI3K/mTOR pathway.We believe that combined inhibition of STX5 and mTOR is a potential treatment for effectively pro-longing patient survival and inhibiting HCC metastasis. | Bin Zhang Ziyin Zhao Youpeng Wang Tingting Guo Mingyang He Ge Guan Pai Peng Jinzhen Cai Bingyuan Zhang Xutao Liu Qiaoling Song | 2023 | Journal of Clinical and Translational Hepatology2023,11,3: | 1 |
